1.An experimental vaccine against tetrodotoxin with longer term of validity

Objective:To explore the possibility of immune prevention and cure for tetrodotoxin (TTX) intoxication and develop an antitoxin vaccine against TTX.Methods:TTX was conjugated to Tachypleus tridentatus hemocyanin(TTH) in presence of formaldehyde and this conjugate(TTX-TTH) was used as an immunogen to immunize Balb/C mice.The quality of antisera in animal was detected on schedule by enzyme linked immunosorbent assay(ELISA) and competition-inhibited enzyme immunoassay(CIEIA).Mice immunized with TTX-TTH were challenged intraperitoneally with lethal doses of TTX(1LD=13.5 ?g/kg).Results:The high titer and affinity of antisera could last for a time as long as more than one year.The immunized mice were ip challenged with 1?LD of TTX once and again at a fixed period,there was a affirmative antitoxic effect in about 12 months(total 15?LD),and a partial effect in following time;about one fourth of animal survived till 24 months post initial immunization(total 26?LD),and which was at a stage of senescence in mice.The anti-TTX poisoning effect of animal was approximately consistent with the antisera quality tested.Conclusion:The experimental vaccine of TTX could effectively protect animal from TTX intoxication and its effect was of longer duration of validity.Immunoprophylaxis would be the hopeful means for detoxification of TTX. [
2.Two new sesquiterpene lactones from the pericarp of Illicium macranthum.

Silica gel column chromatography was used for the isolation and purification of the chemical constituents of the pericarp of Illicium macranthum. From dichloromethane-EtOAc (1:1) fraction and EtOAc fraction of the methanol extracts, eleven compounds were identified on the basis of chemical and spectral data. Two new compounds were elucidated to be 6-deoxyneomajucin (1) and 2-oxo-6-deoxyneomajucin (2), along with nine known compounds 6-deoxypseudoanisatin (3), pseudoanisatin (4), anisatin (5), pseudomajucin (6), protocatecheuic acid (7), shikimic acid (8), shikimic acid methylester (9), beta-sitosterol (10) and daucosterol (11). Compounds 1 and 2 are new majucin-type sesquiterpene lactones.

Information research methods were used to collect and analyze relevant literature. The development of African traditional medicine has been effectively promoted through various measures taken by African organizations and governments. In recent years, Africa has achieved remarkable results in promoting the development of traditional medicine. Many other countries will benefit from studying their examples and adopting similar measures.
4.HPLC fingerprint analysis of Crocus sativus L. and comparative study on its different sources

AIM: To establish the method of fingerprint analysis of Crocus sativus L. and compare their HPLC-FPS of eight kinds of Crocus sativus L. from different sources. METHODS: HPLC with ZORBAX SB-C 18 column was used, the mobile phase was a linear gradient of methanol containing 1% acetic acid and water containing 1% acetic acid in 40min, the detection wavelength at 312nm. RESULTS: 12 marker peaks were separated. The methodological evaluation showed that the method had a good repeatability, and the marker peak area ratio of different samples were different. CONCLUSION: This method is simple and accurate with good reproducibility and can be used for a quality control for Crocus sativus L.

7.Clinicopathological analysis for synchronous colorectal carcinoma

Objective To analyze clinicopathological features of synchronous colorectal carcinoma. Methods Data of colorectal cancer patients admitted to our hospital from June 1994 to December 2003 were analyzed retrospectively. Patients were divided into multiple synchronous colorectal carcinoma group ( MCG) and single colorectal cancer group ( SCG). Clinicopathological features and prognosis were compared between the two groups. SPSS 10. 0 was used for data analysis. Results Of all colorectal cancer(CRC) patients,3. 2 % (39/1225) had multiple synchronous CRCs and 939 patients had sporadic single CRC. In MCG, two patients had hereditary nonpolyposis colorectal cancer ( HNPCC) syndrome. No significant differences were found between MCG and SCG with regard to demographic features, Dukes stage and differentiation of index CRC. More patients in MCG had metachronous CRC (x2 = 4. 545, P= 0.033) and colorectal polyps ( x2 = 12. 013, P = 0.001) compared with SCG. Forty-six percent of multiple synchronous CRCs located in right colon in MCG, which was higher than that in SCG ( x2 = 25. 757 ,P = 0. 0001). Malignancy in adenoma was the frequent event accompanying cancer in MCG. Five-year survival rate in MCG was 57% compared with 64% in SCG ( x2 =0.084, P = 0.772 ). Conclusion Patients with right colon cancer seem easily to have multiple synchronous CRCs and malignancy in adenoma is most frequently accompanying the cancer. For patients with multiple synchronous CRCs, the prognosis is equivalent to that of patients with SCG.
8.J-pouch coloanal anastomosis after low anterior resection for the middle and low rectal carcinoma

Objective To evaluate J-pouch coloanal anastomosis after low anterior resection for the middle and low rectal carcinoma. Methods From January 1998 to July 2002, 120 patients undergoing low anterior radical resection for the middle or low rectal carcinomas were divided into groups of coloanal anastomosis and that of 5 cm colonic J-pouch-anal anastomosis. WT5”HZResults These two groups were well matched for gender, age and histologic stage. There were no significant differences in operative time, hospital stay, complications, postoperative recurrence rate and postoperative survival time between the two groups as founded by an average follow-up of 18 months. The mean distance from the inferior edge of the tumor to the dentate line was (3 6?1 5) cm in the J-pouch group, significantly less than that in coloanal anastomosis group of (5 2?1 9) cm, ( P =0 000). Defecation frequency, urgency and incontinence were significantly improved at 3 months and 12 months after operation in the J-pouch group ( P 0 05). Conclusion J-pouch coloanal anastomosis after low anterior resection for the middle and low rectal carcinoma significantly improves the short-term bowel function after operation.

10.Simultaneous Determination of Eight Effective Compounds of Fufang Xueshuantong ;Capsule in Rat Plasma by LC-MS/MS

Objective To establish a sensitive and specific LC-MS/MS method for measurement of notoginsenoside R1, ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1, ginsenoside Rd, tanshinone Ⅰ, astragaloside Ⅳ and harpagosidein of Fufang Xueshuantong Capsule in rat plasma. Methods The HPLC separation was performed on Thermo Hypersil GOLD column (2.1 mm× 100mm, 5 μm) at 30 ℃, injecting 10 μL and using acetonitrile-water (0.1% formic acid) as the mobile phrase (B was acetonitrile, A was 0.1%formic acid;0-10 min, 25%-55%B;10-20 min, 55%-70%B) with the flow rate of 0.2 mL/min. Detection was performed on a tandem quadrapole mass spectrometer using positive electrospray ionization, SRM scan mode. Results The eight compounds showed good linearity in wide ranges (notoginsenoside R1 1.00-800 ng/mL, ginsenoside Rg1 0.950-760 ng/mL, ginsenoside Re 1.44-1440 ng/mL, ginsenoside Rb1 1.33-1330 ng/mL, ginsenoside Rd 9.90-990 ng/mL, harpagosidein 1.01-1010 ng/mL, astragaloside Ⅳ 1.16-928 ng/mL, tanshinone Ⅰ 10.0-800 ng/mL). In addition, the accuracy and recovery were around 85%-115%and 50%-70%. The RSD of intra and inter day precision were lower than 15%. Conclusion The method is specific, rapid and sensitive. Therefore, it can be applied to pharmacokinetic study of eight effective compounds in Fufang Xueshuantong Capsule.
