1. Research progress of microRNA in prostate cancer

Prostate cancer (PCa) is one of the most common malignant tumors in the male genitourinary system and it increasingly poses a threat to men's health. As a post-transcriptional regulator, microRNAs (miRNAs) play important roles in the evolution and development of life and the occurrence and progress of tumor. Researches have shown that the abnormal expression of miRNAs exerts an indispensable part in the initiation and progression of PCa. In this review we summarized the changes of the miRNAs expression profiling in PCa, the progression of castration resistant PCa, and chemotherapeutic drug resistance and its clinical application.
2. Efficacy of different anti-platelet aggregation regimens before percutaneous transluminal angioplasty and stenting

Objective To use thromboelastograph (TEG) technique for examining the platelet inhibition effect by aspirin and clopidogrel before percutaneous transluminal angioplasty and stenting (PTAS), so as to search for the optimal timing, dosage for clinical practice. Methods The clinical data of 93 patients were divided into 4 groups according to the aspirin history and different doses of aspirin before PTAS; the 4 groups were; non-medication history with aspirin+lower dose aspirin (aspirin 100 mg+clopidogrel 75 mg, Group 1), non-medication history with aspirin + higher dose aspirin(aspirin 300 mg+clopidogrel 75 mg, Group 2), medication history with aspirin+lower dose aspirin(Group 3), and medication history with aspirin+higher dose aspirin(Group 4). The blood samples were collected on day 1, 3 after anti-platelet aggregation drugs; TEG technique was used to detect arachidonic acid (AA)-induced inhibition rate of platelet aggregation and adenosine diphosphate (ADP) receptor-induced inhibition rate of platelet aggregation. Results For different time points; in group 1, the inhibition rates of platelet aggregation of aspirin and clopidogrel were significantly higher on day 3 (AA: \[89. 09 + 17. 42]%, ADP: \[57. 02 + 23. 97]%) as compared with those on day 1 (AA: \[82.24 + 22.25]%, ADP: \[49.62 + 25.44]%; P<0. 05); in group 3, the inhibition rates of platelet aggregation were also significantly higher on day 3 (AA: [95. 06 + 8. 05]%, ADP: \[47. 76 + 24. 95]%) than those on day 1 (AA: \[88.88 + 14.66]%, ADP: \[36.17 + 22.71)%]%; P<0. 05). For different doses: the AA-induced inhibition rates were not significantly different between group 1 and group 2 or between group 3 and group 4 on day 1 and 3. Conclusion Without aspirin history, the inhibitory effect of platelet aggregation of aspirin before PTAS for 3 d is better than that for 1 d, and there is no difference between those of lower and higher doses.

5.The infection status and genotype distribution of high risk human papillomavirus in Chongqing women

Objective To evaluate the infection status and genotype distribution of high risk human papillomavirus (HPV) in Chongqing women with different healthy status of cervix .Methods A retrospective analysis including 3 118 cases from June 2013 to March 2014 were performed ,945 of which were carried out multiplex real time PCR ,and data of cervix healthy status and HPV positive cases were subjected to chi‐square test .Results The general positive percentage of high risk HPV was 27 .49% ,16 .35%in healthy check‐up group ,and 24 .42% in cervicitis group and 39 .87% in cervical neoplasia lesion ,there was a significant difference of HPV positive ratio between these three groups(P< 0 .05) ;and HPV positive ratio was higher in older women without significant difference .In Chongqing region ,the three most common HPV types were HPV52 (21 .97% ) ,HPV16 (20 .00% ) and HPV58 (14 .75% ) .However ,the three most common HPV types in various groups are different :HPV52 (26 .20% ) ,HPV58 (14 .3% ) and HPV51 (10 .70% ) in health check‐up group ; HPV52 (21 .10% ) ,HPV58 (20 .00% ) and HPV16 (18 .90% ) in cervicitis group while HPV16 (29 .00% ) ,HPV52 (19 .80% ) and HPV58 (11 .50% ) in cervical neoplasia lesion group .The multiple infec‐tion rate of HPV was 5 .86% ,as the cervix status gets worse ,HPV concurrent infection increases ,and concurrent HPV types vary :mostly with HPV 52 in healthy check‐up group while with HPV 16 in cervical neoplasia lesion group .Conclusion The prevalence and genotype distribution of HPV in Chongqing women with different healthy status of cervix are significant difference.
6.Regulation mechamion of mesenchymal stem cells to immunoeffector cells--review.

Mesenchymal stem cells have two main properties: self renewal and the ability to differentiate multiple lineage. Because MSCs exhibit low immunogenicity and demonstrate significant suppressive activity in cell cultures containing alloreactive T cells, they play an important role in transplantation immunology, but the exact mechanism remains unknown. This article focuses on the immunoregulatory feature of MSCs to immunoeffector cells, such as T cells, B cells, and NK cells. The role of MSC in transplantation immunoregulation, regulatory mechanism of MSC in cellular immunity (direct contact of cells with cells, apoptosis and immunoregulation of MSC on lymphocytes), immunoregulation of MSC on DC and NK cells were reviewed.

7.ACTH-independent macronodular adrenal hyperplasia: report of 12 cases

Objective To explore the diagnosis and treatment of adrenocorticotropin-independent macronodular adrenal hyperplasia (AIMAH). Methods The clinical symptoms, endocrinological and radiological characteristics, treatment modalities, and postoperative clinical course of 12 AIMAH patients were retrospectively reviewed. There were 5 men and 7 women with an average age of 48 years old. Results All cases had some clinical features and biochemical evidence of Cushing's syndrome. The circadian rhythm of serum cortisol was abnormal. Low- and high-dose dexamethasone suppression tests failed to suppress cortisol secretion. CT or MRI scan showed bilateral enlargement of the adrenal glands with multiple macronodules. Bilateral adrenalectomy was performed in three patients and unilateral adrenalectomy combined with contralateral subtotal adrenalectomy was performed in one patient. Seven patients underwent unilateral adrenalectomy of the larger gland. The removed adrenal glands weighted within 8-96 g range and pathologic diagnoses were nodular or adenomatoid hyperplasia of adrenal cortex. The duration of postoperative follow-up was 14 to 124 months. Clinical symptoms of Cushing's syndrome disappeared after surgery in all cases and there was no Nelson's syndrome after bilateral adrenalectomy. Urinary free cortisol and serum cortisol were within normal ranges. No further enlargement of the contralateral gland has been noticed in patients who received unilateral adrenalectomy during the follow-up.Conclusions AIMAH had unique endocrinological and pathological features. Unilateral adrenalectomy appears to be an effective and safe alternative treatment for AIMAH. Bilateral adrenalectomy or unilateral adrenalectomy combined with contralateral subtotal adrenalectomy may be performed if the symptoms had not been improved or recurred after unilateral adrenalectomy.
8. Hypoxia inducible factor in postmenopausal osteoporosis: Recent progress

Hypoxia inducible factor (HIF) plays a key role in the cellular response to hypoxia, and is involved in a variety of pathological and physiological processes such as glucose metabolism, vascular remodeling and erythropoiesis. Growing research suggests that HIF has an important effect on osteogenesis, bone resorption and angiogenesis; while the deterioration of bone vascular structures and functions is closely related to the development and progression of postmenopausal osteoporosis. In this review, we summarized the relationship between HIF and postmenopausal osteoporosis based on the roles of HIF in osteogenesis, bone resorption and angiogenesis, hoping to evaluate its therapeutic prospect on postmenopausal osteoporosis.
9. Robotic-assisted laparoscopic nephrectomy (right) combined with inferior vena caval thrombectomy for level II tumor thrombus: The first clinical case in China

Objective: To report the first clinical case of robotic-assisted laparoscopic nephrectomy(right) combined with inferior vena caval thrombectomy for level II tumor thrombus in China. Methods: A 73-year-old female patient presented with "gross hematuria and flank discomfort for 4 months" was admitted to our hospital. Preoperative CT scans revealed a 4.9 cm×4.7 cm right renal cancer, grossly extending into the inferior vena cava (IVC, length of the IVC tumor thrombus: 4.6 cm). After three months of neoadjuvant targeted molecular therapies (TKIs), the tumor size reduced to 4.3 cm×4.4 cm on CT and the IVC tumor thrombus length reduced to 3.3 cm. IVC filter was placed cephalad 2 days before surgery under DSA intervention. The patient was placed in recumbent position during the operation; a 12-mm optical port was placed to the right of the rectus abdominis at 2 cm above the umbilicus. Two 8-mm robotic ports and three 12-mm assistant ports were placed percutaneously under direct visualization through a stab incision. The robotic system was then docked, with the colon reflected medially and the duodenum kocherized. The IVC sheath was opened, and the IVC was dissected circumferentially above and below the insertion of the right renal vein to the extent dictated by the length of the thrombus. With the right renal artery dissected and transected, the vessel loops were used to create modified-Rummel tourniquets. The vena cava below and above tumor thrombus, and the left renal vein were cross-clamped. With the vessel loops cinched down completely and secured by hem-o-lok clips, the wall of the IVC was then incised longitudinally for approximately 5 cm. The tumor thrombus was delivered intact along with the invaded IVC wall. The IVC was closed and tourniquet was loosened. After adequate hemostasis was achieved, the specimen was placed into an entrapment sac and incisions were closed by layers. Results: The procedure was smoothly completed. The total operating time was 363 min and the total IVC cross-clamp time was 47 min. The estimated blood loss was 1 200 mL. The amount of intraoperative transfusion was 1 200 mL and 1 000 mL postoperative blood transfusion was required for a low level of hemoglobin. The bowel function recovered at day 3 after operation and drainage was maintained for 3 days. The patient was discharged 16 days after operation. Conclusion: We successfully completed the first clinical case of robotic-assisted laparoscopic nephrectomy combined with inferior vena caval thrombectomy for level II tumor thrombus in China; it is a safe and feasible procedure, but has great technical difficulty, so the patients should be chosen with great care.
10. Establishment of two renal carcinoma A498 cell lines stably expressing CXCR4

Objective To transfect CXCR4 and enhanced green fluorescent protein (EGFP)-CXCR4 plasmids into renal carcinoma cell line A498 cells to preparecell lines stably expressing CXCR4. Methods Two specific plasmids containing CXCR4 or EGFP-CXCR4 were transfected into renal cell carcinoma cell line A498. Then the cells stably expressing CXCR4 were screened by using G418. Confocal microscopy was used to observe the changes of EGFP-CXCR4 fusion protein in A498 cells before and after stimulation with SDF-1. Western blotting analysis was used to determine CXCR4 expression after transfection. Proliferation of A498 cells was detected by MTT and the invasion ability of cells was detected by transwell assay. Results The sequencing result of two plasmids was consistent with CXCR4 DNA sequence, and two cell lines were screened out by G418 screening after the plasmids were transfected into A498 cells. EGFP-CXCR4 fusion protein was found in the cell membrane and cytoplasm of EGFP-CXCR4 transfection group under confocal microscopy. EGFP-CXCR4 migrated into cells after SDF-1 stimulation. Western blotting analysis revealed higher CXCR4 expression in A498 cells stably transfected with CXCR4 plasmids compared with normal A498 cells. The proliferation of cells in pCNDA-CXCR4 and pEGFP-CXCR4 groups were significantly higher than that in normal A498 cell group (P<0. 01). Transwell assay showed that the cell invasion ability of cells with stable CXCR4 expression was significantly increased compared with that in the normal A498 cell group (P<0. 01). Conclusion We have successfully established A498 cell lines stably expressing CXCR4, which have enhanced proliferation levels and higher invasive ability.
