1.RELATIONSHIP BETWEEN NEURONAL INJURIES AND REACTION OF ASTROCYTES FOLLOWING CEREBRAL ISCHEMIA REPERFUSION IN RAT

The involvement of astrocytes and correlation between neuronal injury and astrocyte response were studied. Blockingmiddle cerebral artery and reperfusing o. 5~48 h, H-E staining, immunoccytochemistry single-and double-labeling, dotble label-ing combined with TUNEL and GFAP immunocytochemistry were used to investigate neuronal injury and astrocyte response.The is chemic area peaked at 24 h of reperfusion. The neurons presented irreversible degeneration at 6 h of reperfusion. At24 h,ischemic area in the preoptic area developed into infarcted area; astrocytes exhibited differential morphological features: reactive,malnourished and degenerative changes. At 48 h of reperfusion, the number of astrocytes began to go up. The astrocytes in is-chemic area didn't proliferate within 48 h. By contrast, a few astrocytes underwent apoptosis. In conclusion, these data indicatethat the reaction of astrocytes is closely connected with the extent of neuronal injuries. The reactive astrocytes imply that astro-cytes positively respond to the neuronal injuries, which might play a role in promoting neuronal survival.
2.Clinical value of combined detection of serum HE4 and CA125 in patients with ovarian cancer

Objective To investigate the application value of the combined detection of the serum levels of tumor-associated car-bohydrate antigen 125(CA125) and human epididymis epithelial secretory protein 4 (HE4) in the diagnosis and the differentiation diagnosis of ovarian cancer .Methods Serum levels of CA125 and HE4 in 51 patients with ovarian cancer ,37 patients with benign ovarian tumor and 40 healthy females as the healthy control group were measured by chemiluminescent immunoassay (CLIA) and enzyme linked immunosorbent assay(ELISA) respectively .The detection results combined with the clinical data were performed the statistical analysis .Results The serum levels of CA125 and HE4 in the ovarian cancer group were significantly higher than those in the benign ovarian tumor group and the healthy control group (P<0 .01) .Serum concentration in the stage Ⅲ - Ⅳwere higher than that in the stage Ⅰ - Ⅱ(P<0 .01) .In the combined detection of HE4 and CA125 ,the sensitivity ,specificity and accuracy for diag-nosing ovarian cancer were 84 .6% ,83 .9% and 89 .4% respectively .Their sensitivity and accuracy were higher than those of any single marker detection ,the specificity was consistent with that in the single CA 125 detection .Conclusion The combined detection of serum CA125 and HE4 is conducive to increase the diagnosis efficiency and has certain application value in the differential diag-nosis of benign and malignant ovarian tumor .
3.Rapid Identification of Staphylococcus aureus in Positive Blood Culture Bottles by Loop-mediated Isothermal Amplification

OBJECTIVE To establish a method of loop-mediated isothermal amplification(LAMP) for detecting Staphylococcus aureus in positive blood culture bottles.METHODS Genomic DNA in 293 positive blood culture bottles was extracted by guanidine hydrochloride and benzenemethanol,then genes ssa and mecA were amplified by LAMP to identify S.aureus.Finally,the results of LAMP were compared with the results of traditional method.RESULTS Twenty-two strains of S.aureus were detected in 293 positive blood culture bottles by LAMP method.Compared with traditional method,the sensitivity and specificity of LAMP method were both 100%,respectively,the detection could be finished in an hour.CONCLUSIONS The LAMP-based assay is simple,rapid,sensitive and specific which can be used to detect S.aureus in positive blood culture bottles rapidly.
