1.Discussion and challenge of development of Yunnan medicine in the international environment of Traditional Chinese Medicine

This paper introduced and analyzed the unique geographical location and diverse climatic conditions in the yunnan region, its rich reservation of regional medical plant resources, and the local history, doctors, literatures and medical schools of Traditional Chinese Medicine (TCM). Besides, we combined the characteristics with the current national development strategy "The Belt and Road Initiative" , "cultural soft power" and the Yunnan’s development strategy "big health industry" and "healthy life destination" . This paper discussed the current situation and development prospect of the international development of TCM in Yunnan and the results helped to provide suggestion for the internationalision of TCM in Yunnan.
2.Effect of early interventions with He-Ne laser and infrared radiation for preventing neuralgia Following herpes custer

Objective To investigate the effect of early interventions with helium-neon (He-Ne) laser and infrared physical therapy in the treatment of herpes coster. Methods Sixty-six patients with herpes zoster and concurrent severe neuralgia were randomly divided into treatment and control groups (n=33). The patients in the control group received routine treatment, and early He-Ne laser and infrared treatment was administered in addition to the routine treatment in the treatment group. The visual analogue scale (VAS) was used to evaluate the therapeutic effects in the two groups. Results Early intervention with laser and infrared treatment produced significantly better therapeutic effects than the routine treatment (U=193.520,P=0.000). In the treatment group, the rate of excellent or good therapentic effects and the response rate was 90.9% and 100.0%, respectively, without occurrence of postherpetic neuralgia;in the control group, the rate of good therapeutic effects and the response rote was 63.6% and 81.8%, respectively, and postherpetic neuralgia occurred in 3 eases (9.1%) after the treatment. Conclusion Early intervention with He-Ne laser and infrared treatment in patients with herpes coster helps achieve rapid control of the condition, shortens the treatment course, and reduces the occurrence of postherpetic neuralgia.
3.Effect of integrin-?1 antisense oligodeoxynucleotides on human pancreatic cancinoma transplanted subcutaneously in nude mice

Objective To investigate the effect of integrin-?1 antisense oligodeoxynucleotide(ASODN) on(human) pancreatic cancinoma transplanted subcutaneously in nude mice.Methods The models of human(pancreatic) cancinoma transplanted subcutaneously were established in nude mice,then divided randomly into 3 groups and different treatment was given respectively(control group,random oligodeoxynucleotide group and ASODN group).After treatment,the weight of nude mice and tumor volume were observed,and the tumor growth inhibitory rate and the tumor response rate were calculated.The expressions of integrin-?1 mRNA and protein in tumor tissue were determined by RT-PCR and Western-blot.Results The tumor growth inhibitory rate in the random oligodexynucleotide group and the ASODN group was 4.75% and 72.70%,respectively.The tumor decrease rate of the ASODN group was 10.91%.The expression level of integrin-?1 mRNA and protein was decreased in the ASODN group compared with other 2 control groups. Conclusions Our findings suggest that integrin-?1 antisense oligodeoxynucleotides result in marked inhibition of human pancreatic(cancinoma) growth in nude mice.It may be a novel treatment approach for human pancreatic carcinoma.
4.Expression and Antigenic Characterization of the Epitope-G1 of the Bovine Ephemeral Fever Virus Glycoprotein in Pichia pastoris

The epitope-G1 gene of Bovine ephemeral fever virus (BEFV) glycoprotein was synthesised by PCR and cloned into expression vector pPIC9K to construct recombinant plasmid pPIC9K-G1. Then the pPIC9K-G1 was linearized and transformed into Pichia pastoris GS 115. The recombinant P. pastoris strains were selected by a G418 transformation screen and confirmed by PCR. After being induced with methanol, an expressed protein with 26 kDa molecular weight was obtained, which was much bigger than the predicted size (15.54 kDa). Deglycosylation analysis indicated the recombinant G1 was glycosylated. Western blot and ELISA tests, as well as rabbit immunization and specificity experiments indicated that the target protein had both higher reaction activity and higher immunocompetence and specificity. The recombinant G1 protein could be used as a coating antigen to develop an ELISA kit for bovine ephemeral fever diagnosis.
5.Recent progresses of magnetic resonance imaging of subthalamic nucleus

Subthalamic nucleus (STN) is the main target nucleus for deep brain stimulation (DBS) treatment in patients with Parkinson disease.To implant the electrode on the sensorimotor part of STN individually and accurately,the boundary of STN is required to be clarified clearly on the magnetic resonance imaging (MRI) without geometric distortion.At present,there are three categories of MRI sequences:spin echo sequence including T2-weighted imaging (T2WI),inversion recovery (IR),diffusion tensor imaging (DTI),and fractional anisotropy (FA);magnetization transfer technique including magnetic susceptibility weighted imaging (SWI) and T2-weighted magnitude imaging (T2 * WI);image reconstruction technique such as quantitative susceptibility mapping (QSM).It is found that QSM can provide optimal signal-noise ratio to identify the boundary of STN,T2 * technique comes second.T2WI has high geometric accuracy when the patients wear frame,which is appropriate for direct DBS implantation on STN with frame.
6.Effects of laminarin sulphate on the expressions of PTEN and P271kip1 in prostate cancer PC-3 cells.

OBJECTIVETo study the effect of laminarin sulphate (LAMS) on the expressions of PTEN and P27kip1 in androgen-independent prostate cancer PC-3 cells in vitro, and investigate the mechanism of its anti-tumor action.
METHODSThe inhibitory effects of different concentrations of LAMS (0, 50, 100, 200 microg/ml) on androgen-independent prostate cancer PC-3 cells were detected by WST-8 assay. The morphology of PC-3 cells was observed under the fluorescence microscope, and the cell cycle and apoptosis were analyzed by flow cytometry. The mRNA and protein levels of PTEN and P27kip1 were measured by RT-PCR and Western blot.
RESULTSLAMS inhibited the proliferation of androgen-independent prostate cancer PC-3 cells in a dose- and time-dependent manner, and the cell cycle analysis showed that PC-3 cells were arrested in the S phase after treated with different concentrations of LAMS. The rate of apoptosis was increased and many typical apoptotic morphological features were observed under the fluorescence microscope. The PTEN and P27kip1 expressions at mRNA and protein levels were increased in a dose-dependent manner.
CONCLUSIONLAMS can inhibit the proliferation, arrest the cell cycle in the S phase and induce apoptosis of prostate cancer PC-3 cells. The significantly increased expressions of PTEN and P27kip1 may be one of the mechanisms for LAMS inhibiting prostate cancer PC-3 cells.

8.Expression of vascular endothelial growth factor (VEGF) C and VEGF receptor 3 in non-small cell lung cancer.

OBJECTIVETo study the relationship between angiogenesis and lymphangiogenesis with the expression of vascular endothelial growth factor C (VEGF-C) and VEGFR-3 in human non-small cell lung cancer (NSCLC).
METHODSSamples of 76 NSCLC cases with the neighboring noncancerous tissue were studied using anti- VEGF-C, VEGFR-3 and CD34 antibodies. Assessment of lymphatic vessel density and microvessel density (MVD) were performed.
RESULTSVEGF-C expression in NSCLC was associating with the differentiation of tumor cells (P = 0.009). Expression of VEGF-C and VEGFR-3 was significantly associated with lymph node metastasis (P = 0.008 and P = 0.013 respectively) and lymphatic invasion (P = 0.027 and P = 0.020 respectively). A significant positive correlation was found between VEGF-C in cancer cells and VEGFR-3 in lymphatic endothelial cells (P = 0.009). The number of lymphatic vessels (P = 0.006) and microvascular (P = 0.046) in VEGF-C positive tumors was significantly larger than in VEGF-C-negative tumors. Lymphatic vessel density was closely related to lymph node metastasis (P = 0.010), lymphatic invasion (P = 0.019) and clinical stages (P = 0.015). MVD was closely related to blood metastasis (P < 0.001) and clinical stages (P < 0.001). Patients with positive VEGF-C expression had a worse prognosis than those with a negative VEGF-C expression (P < 0.001).
CONCLUSIONSVEGF-C/VEGF-D in NSCLCs, are related to lymphangiogenesis and angiogenesis, as well as to the occurrence and the development of lung cancers. VEGF-C promotes intratumoral lymphangiogenesis via VEGFR-3, resulting facilitated invasion of cancer cells into the lymphatic vessels. VEGF-C expression can be a useful predictor of poor prognosis in NSCLC.

9.Three PHEX gene mutations in Chinese subjects with hypophosphatemic rickets and literature review.

The clinical data of three Chinese children who had been definitely diagnosed with X-link dominate hypophosphatemic rickets (XLH) by gene mutation analysis of phosphate-regulating gene with homologies to endopeptidases on the X chromosome (PHEX) were retrospectively studied and the relevant literature was reviewed. PHEX gene mutations were detected in all 3 XLH children; a nonsense mutation (c.58C>T) in one case and splicing mutations (c.1645+1G>A, c.436+1G>A) in the other two cases. Among these mutations, c.436+1G>A was novel. As of January 2014, a total of 329 PHEX gene mutations were reported, primarily within three mutation hot spots, throughout the world. Missense mutations accounted for the highest proportion (24%) among all mutations. There is literature showing geographic differences in the total number of XLH subjects and PHEX mutation types across the world. In the current literature, 89 cases of XLH with 28 types of PHEX mutations have been reported in the population of mainland China. Exon 22 is the most frequent mutation site (18%) and missense mutations are the most common type of mutations (61%). It is concluded that exon 22 is the mutation hot spot and missense mutation is the most common type of mutation in the PHEX gene in Chinese XLH patients and that c.436+1G>A detected in this study is a novel PHEX gene mutation in Chinese with XLH.

10.Study on Inclusion Behavior and Properties of Cordycepin with Hydroxypropyl-β-Cyclodextrin

The inclusion complex of cordycepin ( COR ) with hydroxypropyl-β-cyclodextrin ( HPβCD ) was prepared by the method of saturated solution. The inclusion of HPβCD with COR in aqueous solution was studied by UV-vis spectroscopy, and the inclusion ratio of COR/HPβCD complex was determined with the Job plots. The COR/HPβCD complex was characterized and determined by means of 1 H NMR and 2D NMR, differential scanning calorimetry ( DSC ) , thermogravimetric analysis ( TG ) , X-ray diffraction ( XRD ) , Fourier transform infrared spectroscopy ( FTIR) and scanning electron microscope ( SEM) . The results showed that the COR/HPβCD complex ratio was 1:1 and the water solubility and stability of COR were obviously increased in the inclusion complex with HPβCD. The COR/HPβCD complex will be potentially useful for its medical application.
