1.Research and application of biomaterial in related territory of surgery

With the development of the cross-correlation technique in surgery, the transplantation and vi-cariousness have become more and more important in repair surgery and reconstructive surgery. A nice bio-material could replace the related tissue or organ of human body, support the homologous function of organ-ism,and accelerate the process of self recovery and autotherapy. This review is about the status of research and application of biomaterial in surgery.
2.Study on the biocompatibility and the application in cardiothoracic surgery of biomaterial

A nice biomaterial,which could replace the correlated tissue or organic of human body,supports the homologous function of organism,and accelerates the process of self recovery and autotherapy in the same time.The application of biomaterial in clinic,not only have the chemical characters and function of vitodynamics to match,but also be with nice biocompatibility and practical safety.Some development of biomaterial and the biocompatibility of it in this paper was reviewed,and the application status in the cardiothoracic surgery in recent years.
4.Research on the parameters of oxidative stress in T2DM patients combined with acute coronary syndrome

Objective To investigate the variation of the oxidative stress status in type 2 diabetes mellitus(T2DM)patients with acute coronary syndrome (ACS)and significance.Methods Eighty -nine patients hospitalized in our department from September 2012 to April 2014 were divided into three groups: T2DM control (group DM), T2DM with coronary artery disease(group DM-CAD) and T2DM with acute coronary syndrome (group DM-ACS).Blood glucose,HbA1C,Lipids,serum Malondialdehyde (MDA) level, su-peroxide dismutase(SOD) activity, glutathione peroxidase (GSH-Px) activity and total anti-oxidation capacity(T-AOC) were deter-mined.Results Blood glucose、HbA1C and Lipids were similar in three groups ( P >0.05).Compared to group DM and group DM-CAD, serum MDA levels in group DM-ACS were significantly higher( P <0.05), while SOD、GSH-Px activities and T-AOC were sig-nificantly lower than those in group DM and group DM -CAD( P <0.05).Conclusions The oxidative stress level being significantly increased in T2DM patients with ACS.
6.Comparison Study on Effects of Electroacupuncture on Glucose Metabolism of Cerebral Motor Function Areas

Objective To compare the effect of electroacupuncture at head points and body points on glucose metabolism of the cerebral motor function regions in normal subjects.Methods To observe the change of glucose metabolism of cerebral motor area in normal subjects between before and after acupuncture during the movement by PET.Talairach coordinates(Atlas of brain) and statistical parametric mapping(SPM) software were used to deal with the acquired imaging data.Results ① Acupuncture at Baihui(GV20) and left Qubin(GB7) could increase metabolism of glucose in bilateral Lps and precuneus,the activation of left area in the brain being more significant.②Acupuncture at right Quchi(LI11) and Zusanli(ST36) change metabolism of glucose in left gyrus precentralis,right loblus paracentralis,right gyrus frontalis medialis,both cerebellums and both putarnens.Conclusion All acupoints can change glucose metabolism in cerebral structures related to motor function in the bilateral cerebral hemispheres,different acupoints active different motor areas.The function of acupuncture is a kind of comprehensive regulative process.
7.Analysis on the occurrence of malignant tumors after kidney transplantation

Objective To study the incidence of malignant tumors in the patients undergone renal transplantation, and to explore the mechanism of higher incidence. Methods A retrospective study was performed on 829 patients undergone renal transplantation followed by immunosuppression therapy from 1998-2006. 15 cases developed malignant tumors among the 829 patients with incidence of 1.9%. The incidence of urinary system tumor was 66.7%, of hepatoma was 13.3%, and 6.7% for lymphoma, cutaneous cancer and pulmonary carcinoma respectively. Results 15 patients suffered from renal tumors 11-73 months after kidney transplantation, in whom renal function was normal in 14 patients, and in 1 patient the grafted kidney showed dysfunction. No tumor metastasis was found in those cases, except in 3 patients ureteral lumen infiltration was found. Most patients accepted operations, with the dose of the immuosurpression drugs adjusted or reduced, and supplemented with chemotherapy or rediotherapy and immunotherapy. Conclusions The pathogenesis might be related with immunosuppression drugs postoperatively, and also the effects of transplantation itself and the primary disease before transplantation, for instance, uroepithelium malignancy was prolonged medication was frequently the reason for renal transplantation.
8.Marginal adaptation of metal crowns:comparison of two production methods

BACKGROUND:At present, the confrontation of CAD/CAM systems and the lost wax casting method for marginal adaptation has been widely concerned, but the results are controversial. OBJECTIVE:To compare the marginal adaptation of single cobalt-chromium PFM crowns produced by 3 Shape CAD/CAM systems and the lost wax casting method. METHODS: A maxilary first premolar was prepared according to the standards. 50 resin-generation models were created by bi-color silicone rubber method and randomly divided into two groups: CAD/CAM group (A1-A25) and cast group (B1-B25). Al the models were cast in plaster. In CAD/CAM group, the digital impressions were obtained by optical scanning, then designed through computer and cut directly by NC miling machine. In cast group, metal ceramic crowns were made by the lost wax casting method. When crowns were al seated on the working dies, the marginal gaps were copied with light silicone rubber and then embedded with occlusal silicone rubber. The thickness of crown marginal gap was measured with a stereo microscope. RESULTS AND CONCLUSION:The average vertical clearance was (34.67±7.48)μm of CAD/CAM group, while (53.70±9.64)μm of the cast group; the difference was statisticaly significant between two groups (P < 0.05). The average level clearance was (37.61±7.63)μm of CAD/CAM group, while (53.02±10.65)μm of the cast group, and the difference was statisticaly significant between two groups (P < 0.05). Experimental findings suggest that, the marginal adaptation of single cobalt-chromium PFM crowns cast produced by 3 Shape CAD/CAM systems is clinical acceptable (< 120μm), and CAD/CAM systems of metal casting is obviously better than the lost wax casting in building metal based crowns with adaptable margin.
9.Expression of mutated insulin gene in HepG-2 cell line

Objective To express a mutated insulin gene in HepG-2 cell line to further research of insulin gene therapy. Methods Native human insulin cDNA was obtained from fetus pancreas with RT-PCR. Furin consensus cleavage sequence was introduced into proinsulin cDNA with site-directed mutagenesis (overlap extension PCR), and the new sequence was named as INS/furin. Subsequently, INS/furin was subcloned into the multiple clone sites of plasmid p(G1RE)3BP-1Luc. The new plasmid p(G1RE)3BP-11?furin was identified with the method of enzyme digestion by Hind Ⅲ and EcoR V. HepG-2 cells were transfected with the plasmid p(G1RE)3BP-11?furin by liposome-mediated method. The transfected HepG-2 cells were incubated for 48h in a glucose-containing medium (25mmol/L), and then the conditioned media were collected and HepG-2 cells were harvested respectively. The expression of INS/furin mRNA in transfected HepG-2 cells was examined by RT-PCR, the regained DNA was sequenced and insulin in conditioned media was investigated by radioimmunoassay. Results Two enzymes, Hind Ⅲ and EcoR V, digested p(G1RE)3BP-11?furin, and 2 fragments with length of 260 bp and 4 700bp, were obtained. The 260bp fragment was identified as insulin/furin, indicating that the target gene had been successfully inserted in specific sites. RT-PCR showed that insulin/furin mRNA was expressed in transfected HepG-2 cell, and the regained DNA was confirmed as insulin/furin by sequencing; while insulin was detected by radioimmunoassay in conditioned media. Conclusion The recombinant mammalian expression plasmid p(G1RE)3BP-11?furin has been successfully constructed, and transfected into HepG-2 cells, which therefore may efficiently secrete bioactive insulin.
