2.Correlation between inflammatory factors and subclinical atherosclerosis in type 2 diabetic patients with metabolic syndrome

ELISA technique was employed to determne the concentration of mannose binding lectin (MBL),plasminogen activator inhibitor-1 (PAI 1),and high sensitive ()eactive protein (hs-CRP).hs CRP level m patients with subclinical atherosclerosis was significantly higher (P < 0.01),MBL level was of no significant difference (P>0.05) ; PAI-1 was significantly higher (P<0.01) only m those with metabolic syndrome(MS).Taking no account of subclinical atherosclerosis,MBL of diabetic patients with MS was significantly lower while hs-CRP and PNI 1 were signific amlv higher (P < 0.01).Inflammatory faetors MBL,hs-CRP,and PAI-1 involved in the -ube linical atherosclerosis development in the type 2 diabetic patients with MS.
3.Comparison of the immunoregulatory effects of Radix Millettia Speciosa and Radix Millettia Championi on immunosuppressed mice

Objective To compare the immunoregulatory effects of Radix Millettia Speciosa (RM Speciosa) and Radix Millettia Championi (RM Championi)on immunosuppressed mice.Methods Kunming mice were randomly divided into normal control group,CTX model group,LMS positive group,RM Speciosa groups and RM Championi groups (20,10 and 5 g/kg).The mice were treated respectively with drug or NS once a day for consecutive 20 days.Mice in the other groups were injected intraperitoneally with CTX at days 8,10 and 12 to establish immunosuppressed mice model except the normal group.The changes of body weight,immune organ weight,white blood cell (WBC)number,carbon particle clearance capability of macrophages and delayed type hypersensitivity (DTH)of mice in all groups were determined and compared.Results Compared with that in CTX group,the WBC number was significantly increased (P<0.01 or P<0.05)and the degree of DTH (P<0.01) was enhanced at different doses in RM Speciosa groups and RM Championi groups.The activity of macrophages and the index of thymus and spleen were also improved in the treatment groups (P<0 .0 5 or P<0 .0 1 ).The index of thymus and the degree of DTH in RM Speciosa groups (20 and 10 g/kg)were slightly higher than those in the corresponding RM Championi groups (P<0.01 or P<0.05).For other indicators,RM Speciosa groups did not significantly differ from RM Championi groups (P>0.05).Conclusion Both RM Speciosa and RM Championi can improve the immune function of CTX-induced immunosuppressed mice,and RM Speciosa is slightly superior to RM Championi in improving specific cellular immunity.
4.Correlationship between TCM Syndromes and Carotid Atherosclerosis Plaque, Adiponectin Level of Hypertension Complicated with Carotid Atherosclerosis

Objectives To study the relationship between TCM syndromes and adiponectin (APN) level, carotid atherosclerosis plaque of hypertension complicated with carotid atherosclerosis, and provide evidence for clinical diagnosis and treatment. Methods One hundred patients of hypertension complicated with carotid atherosclerosis were divided into phlegm syndrome group, stasis syndrome group, and intermingled phlegm and blood stasis syndrome group, and compared with 30 healthy people as control. Carotid atherosclerosis plaque was detected with GE LOGIQ500 color ultrasound system. ELISA was used to determine the serum APN. Results APN of the intermingled phlegm and blood stasis syndrome group was obviously lower than other groups, and there were obvious differences among them. There were more plaques in the intermingled phlegm and blood stasis syndrome group. A negative correlationship was showed between carotid artery intima media thickness (IMT) and APN of hypertension complicated with carotid atherosclerosis. Conclusion There is correlation between TCM syndrome and APN level, carotid atherosclerosis plaque of hypertension complicated with carotid atherosclerosis. The anomalous change of APN and plaque formation of the patients can be preliminarily estimated with the syndrome of TCM.
5.Content determination of Forsythoside A in Yinqiaosan granules by HPLC

Objective To establish a method for determination of the content of Forsythoside A in Yinqiaosan granules . Methods HPLC method was adopted to carry on the determination .Welch Ultimate XB‐C18 column (250 mm × 4 .6 mm ,5μm) was used with the mobile phase of methanol‐0 .2% Glacial acetic acid solution by gradient elution . The flow rate was 1 .0 ml/min ,the detection wavelength was 330 nm ,and the column temperature was 20 ℃ .Results Forsythoside A showed a good linear relationship at the range from 0 .080 3~3 .211 μg (r=1 .000) .The average recovery rates were 103 .78% ,and RSD=0 .82% .Conclusion The method was convenient and accurate .Meanwhile ,it also has good specificity and reproducibili‐ty .It can be used effectively for the quality control of Yinqiaosan granules .
6.Optimizing seizure detection by quantitative EEG in paediatric refractory status epilepticus

Background & Objective: Continuous electroencephalography (cEEG) is valuable in the diagnosis
and management of refractory status epilepticus (RSE) but requires intensive skilled interpretation.
Density spectral array (DSA) is a quantitative analytic tool used to screen cEEG recordings for seizures.
This study aims to determine the optimal amplitude setting and to compare the use of single-averagetrendgraph display and eight-trendgraph display in seizure detection with DSA. Methods: Five excerpts
from pediatric cEEG recordings with RSE were identified. In Phase 1 of the study, each of 4 readers
determined the DSA amplitude setting one most preferred for each excerpt, and marked all seizures
in one excerpt using one’s preferred setting. Inter-rater agreement in seizure detection was measured.
In Phase 2, readers marked all seizures in all excerpts, first using single-average-trendgraph display,
and then using eight-trendgraph display after a wash-out period. Intra-rater agreement in seizure
detection between the two display methods was calculated. Results: In Phase 1, DSA readers’ choice
of preferred amplitude settings varied widely but inter-rater agreement in seizure detection was high.
In Phase 2, seizure detection using single- and eight-trendgraph displays showed high agreement with
each other and, where they disagreed, single-average-trendgraph was more sensitive. Additionally,
low seizure-to-background amplitude ratio in EEG recordings was associated with worse detection
sensitivity/specificity.
Conclusions: DSA amplitude settings do not affect seizure detection. Single-trendgraph display is
comparable to eight-trendgraph display in screening cEEG for seizures. Seizure detection with DSA
performs better in cEEG recordings with high seizure-to-background amplitude ratio.
8.Clinical research about heating infusion for premature infants to promote their restoration of physiological function

Objective In order to know the effect of heating infusion for premature infants to pro-mote their restoration of physiological function. Methods Divided 160 premature infants who need infu-sion into the experimental group and the control group randomly, there were 80 cases in each group. Normal temperature infusion was used in the control group, while heating infusion was used in the experimental group, compared the body temperature, pluse, respiration, blood pressure, oxygen saturation and the blood ghcose between the two groups. Results There were significant difference about the indexes which had mentioned above between the two groups. Conclusions Heating infusion can effective promote the reha-bilitation of physiological function of premature infants.

10.Polymorphism analysis on chemokine SDF-1α transcript being the ligand for HIV-1 coreceptor CXCR4

Objective To determine the polymorphism in CXC chemokine SDF-1α transcript and its effects on HIV infection.Methods Three groups of study subjects lived in Hang Kong were recruited:278 HIV-heahhy donors of Chinese origin.49 HIV+Caucasians and 13 Chinese with high risk behavior to HIV but kept uninfected.Genomic DNA and RNA were extracted from eripheral blood mononucleal cell. The PCR and RT-PCR reaction were set up accordingly.Sequence of the SDF-1α promoter,the open reading frame(ORF)and the 3'untranslate region(3'UTR)were analyzed.Two steps PCR reaction using two reverseprimers with mismatched nucleic acid were employed to screen the frequency of a novel mutation. Results equencing analysis from 100 subjects indicated that non mutation happened in tlle promoter and ORF of SDF-1α.A novel mutation Was detected from 3'UTR of SDF-1α.It is a "GA" insertion in "G" rich region near the stop code of SDF-1α.The mutation Was named as SDF-1-3’GA+and submitted to GenBank (AY874118).The mutation happened in three roups.with allele frequency of 15.1% in the healthy Chinese.of 30.7% in the high risk Chinese group.Conclusions our results confirm that SDF-1 genes arerelatively conserved.None noteworthy mutation is identified in the promoter and ORF regions of SDF-1α However.a novel mutation is identified from the 3'UTR of SDF-1α. It would be worthwhile to etermine effect of the novel mutation on HIV infection.
