1.Impact of citreoviridin on mRNA expression of mitochondrial respiratory chain synthesis related transcriptional regulation gene, membrane potential and reactive oxygen species in rat cardiomyocytes

Objective To investigate the impact of citreoviridin(CIT) on mRNA expression of mitochondrial respiratory chain synthesis related transcriptional regulation gene,mitochondrial membrane(MMP) potential and reactive oxygen species (ROS) in cardiomyocytes of rat.Methods Viability of rat primary cardiomyocytes treated with different concentrations of CIT (0,1,2,3,4,5,6,7,8,9,10 μmol/L) for 24 h was determined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method.Based on the MTT curve,median inhibitory concentration(IC50) was calculated using SPSS 13.0.High-,medium-and low-dose groups of CIT(1.650,1.234,0.715 μmol/L) were defined corresponding to 99%,95% and 90% of cardiomyocyte viability,respectively.CIT was not added in as the control group.After 24 hours,the mRNA expression levels of peroxisome-proliferator-activated receptor γcoactivator(PGC-1α),nuclear respiratory factor 1 (Nrf1) and nuclear respiratory factor 2(Nrf2) in cardiomyocytes were detected by reverse transcriptase polymerase chain reaction(RT-PCR).Changes of MMP and intracellular ROS were determined by a fluorescence microplate reader using 5,5',6,6'-tetrachloro-1,1',3,3'-tetraethylbenzimidazolcarbocyanine iodide (JC-1) and 2,7-dichlorofluorescein diacetate (DCFH2-DA) as fluorescent probes.Results Compared with 0 μmol/L CIT group [(89.4 ± 17.6)%],viabilities of rat primary cardiomyocytes treated with 2-10 μmol/L CIT groups[(80.2 ± 20.2)%,(74.4 ± 18.7)%,(63.2 ± 8.9)%,(51.5 ± 18.8)%,(39.0 ± 15.7)%,(22.6 ± 10.5)%,(19.9 ± 4.9)%,(20.7 ± 4.8)%,(18.5 ± 3.3)%] decreased significantly(all P < 0.05).The IC50 value of cardiomyocytes after24 h treatment with CIT was 4.6 μmol/L The PGC-1α mRNA expressions ofhigh-,medium-and low-dose groups(0.431 ± 0.041,0.619 ± 0.031,0.653 ± 0.037) were significantly lower compared to that of the control group(0.776 ± 0.081,all P < 0.05).The Nrf1 mRNA expression of high-dose group(0.358 ± 0.05) was significantly lower compared to that of the control group(0.580 ± 0.098,P < 0.05).Nrf2 mRNA expressions of the high-and medium-dose groups(0.352 ± 0.041,0.472 ± 0.011) were significantly lower than that of the control group (0.667 ± 0.091,all P< 0.05).Compared with the control groups[(100.00 ± 0.00)%,(100.00 ± 0.00)%],the MMP levels of high-,medium-and low-dose groups[(55.3 ± 3.3)%,(69.8 ± 4.7)%,(81.8 ± 2.7)%] were significantly lower and the ROS levels[(606.0 ± 46.3)%,(275.0 ± 53.5)%,(158.9 ±29.5)%] were significantly higher(all P < 0.05).Conclusions CIT inhibits the biosynthesis of mitochondria in primary cardiomyocytes and induces oxidative stress.Myocardial injury is caused by cardiomyocyte apoptosis through mitochondrial pathway,which leads to myocardial injury.
2.Analysis of Cosmetics Safety in China

In recent years, various studies and analyses related to cosmetics safety were conducted according to routine tests in Hygienic Standard of Cosmetics (2007), and the eligible rates of tested cosmetics were high. But the other prohibited and limited use components, such as antibiotics were analyzed rarely, and meanwhile some kinds of cosmetic related dermatitis cases appeared dramatically. Several dermatitis, especially contact dermatitis and hormone dependent dermatitis symptoms were not contained in Diagnostic Criteria and Principles of Management of Skin Diseases Induced by Cosmetics-General Guideline, GB 17149.1—1997. So it indicated the standard, GB 17149.1—1997 should be revised and some prohibited and limited use components such as hormone and antibiotic testing should be appended to the safety analysis of cosmetics.

To study the multidrug resistance activity of 1-alkyl-2-acetyl-1, 2, 3, 4-tetrahydroisoquinoline derivatives. Methods: A series of novel tetrahydroisoquinoline derivatives bearing at C-1 position a carbon chain derived from fatty acids were prepared through the Bischler-Napieralski cyclization reaction. Their multidrug resistance (MDR) reversal cancerous multidrug resistance activities were evaluated against K562 and K562/DOX cell lines in vitro by MTT assay with verapamil as a control. Results and Conclusion: The structures of these tetrahydroisoquinolines were confirmed by extensive spectroscopic methods(1H NMR, MS, IR and elemental ana-lyses). MDR results showed that compounds 7 and 10 exhibited moderate reversal activities, and were slightly less potent than those of verapamil against K562 cell line. It is believed that compounds 7 and 10 have MDR activity.
5.Exploration on the essence of TCM syndrome with systems approach.

6.The preliminary study on mortality prediction for patients in surgical intensive care unit with protein C

Objective To determine whether anticoagulation markers can improve mortality prediction in patients of surgical intensive care unit (ICU).Methods A case-control study was adopted,252 patients from Tongji hospital's surgical ICU and 30 healthy control individuals were investigated.The protein C,antithrombin,thrombomodulin,and other coagulation/ inflammatory markers were detected.The Acute Physiology and Chronic Health Evaluation Ⅱ (APACHE Ⅱ) score were obtained.Markers level comparison among survivors,non-survivors and controls were conducted with single factor variance analysis,Kruskal-Wallis test or Mann-Whitney U test.Results Between survivors and non-survivors after 28-day hospitalization,there were significant difference on protein C levels[(70.2 ±22.7)% vs (48.6 ±29.8)%,t=2.84,P<0.01],APACHE Ⅱ scores[(21.0±8.2) vs (29.5 ±10.9),t =-2.51,P<0.05] and prothrombin times[(12.9-± 3.5) s vs (18.8 ± 10.2) s,t =-2.13,P < 0.05].Combining protein C levels with APACHE Ⅱ score could obtain a higher mortality prediction efficiency in patients of surgical ICU than any single marker (AUC =0.806).That protein C concentration less than 47.5% [OR =6.40,95%confidence interval(CI) 2.526-16.216,P <0.001] and APACHE Ⅱ score (OR =1.123,95% CI 1.012 -1.250,P < 0.05) were the independent risk factors for surgical ICU death.Conclusion Decrease of protein C levels predict increase of mortality risk in patients of surgical ICU,combining protein C with APACHE Ⅱ score can improve the prognostic accuracy for patients of surgical ICU.(Chin J Lab Med,2013,36:339-342)
7.Bond strengths of absorbable polylactic acid root canal post with three different adhe-sives

Objective:To find absorbable adhesives with suitable bonding properties for the absorbable polylactic acid root canal post. To test and compare the bond strengths of absorbable polylactic acid root canal post with three different adhesives. Methods:The absorbable polylactic acid root canal posts were used to restore the extracted teeth, using 3 different adhesives: cyanoacrylates, fibrin sealant and glass ionomer cement. The teeth were prepared into slices for micro-push-out test. The bond strength was statistically analyzed using ANOVA. The specimens were examined using microscope and the failure mode was divided into four categories:cohesive failure between absorbable polylactic acid root canal posts and adhesives, cohesive failure between dentin and adhesives, failure within the adhesives and failure within the absorbable polylactic acid root canal posts. Results:The bond strength of cyanoacrylates [(16. 83 ± 6. 97) MPa] and glass ionomer cement [(12. 10 ± 5. 09) MPa] were significantly higher than fibrin sealant [ ( 1 . 17 ± 0 . 50 ) MPa ] , P <0 . 001 . There was no significant difference between cyanoacrylates and glass ionomer cement (P =0. 156). In the group of cyanoacrylates, the cohesive failure between the absorbable polylactic acid root canal posts and the adhesives was 25 . 0%, the cohe-sive failure between the dentin and the adhesives was 16. 7%, the failure within the adhesives was 33. 3%, and the failure within the absorbable polylactic acid root canal posts was 25 . 0%. In the group of fibrin sealant , the cohesive failure between the absorbable polylactic acid root canal posts and the adhesives was 66 . 7%, the cohesive failure between the dentin and the adhesives was 22 . 2%, the failure within the ad-hesives was 11. 1%. In the group of glass ionomer cement, the cohesive failure between the absorbablepolylactic acid root canal posts and the adhesives was 87. 5%, the failure within the adhesives was 12. 5%. The major failure mode in fibrin sealant and glass ionomer cement was the cohesive failure between the absorbable polylactic acid root canal posts and the adhesives. No major failure modes were found in the group of cyanoacrylates. Conclusion:The bond strength of fibrin sealant is low, which cannot meet the requirement of clinical use. The bond strengths of cyanoacrylates and glass ionomer cement are suitable for clinical use. The cyanoacrylates are a kind of absorbable adhesive which has suitable bonding proper-ties for the absorbable polylactic acid root canal post.

Objective To investigate the therapeutic effects of Smad7 and urokinase-type plas- minogen activator(uPA)co expression on CCl_4-induced rat liver fibrosis.Methods Forty SD rats were subcutaneously injectied of 40% CCl_4 every three days for 8 weeks.The rats were then divided into model group,AdSmad7/uPA group(injected with AdSmad7/uPA via tail vein),AdSmad7 group(injec- ted with AdSmad7 via tail vein)or AdGFP group(injected with AdGFP via tail vein).Ten healthy rats were served as control.The serum levels of procollagenⅢ(PCⅢ)and laminin(LN)were determined by radioimmunoassay,and the hydroxyproline level in liver tissues were examined by alkaline hydrolysis. The expressions of Smad7 and uPA in tissue were examined by immunohistochemistry and fibrosis area was stained with Sirius red.Results The expressions of Smad7 and uPA protein were significantly higher in AdSmad7/uPA group than that in AdGFP group after 3 days.Serum levels of ALT,AST,PCⅢand LN were significantly decreased in AdSmad7/uPA group compared to Smad7 and AdGFP groups (all P value
9.Mapping and analysis of HPV16 integration sites in a cervical cancer cell line

Objective To explore the chromosome aberration of cervical cancers induced by human papillomavirus(HPV16)virus.Methods The change of change of chromosomes of Hela cells induced by HPV-16 infection HPV-16 in fection was detected by FISH.Results The integration sites of HPV-16 were not observed in Hela cells.But HPV16 virus was integrated on chromosomes 3.Conclusion HPV16 could cause chromosome 3 aberration in Hela cells and result in the occurance of cervical cancers.
10.Effect of Cholecalciterol Cholesterol Emulsion on Liver of Zymosan-induced Multiple Organ Failure in Mice

Objective To study the effect of cholecalciterol cholesterol emulsion(CCE)in the zymosan(Z)-induced acute hepatic injury. Meth-ods A total of 36 C57 BL/6 mice were randomly divided into 4 groups,namely negative control(CON)group,CCE group,Z group and CCE+Z group,respectively. There were 9 mice in each group. Mice from CON group and Z group were fed with pure water. Mice from CCE group and CCE+Z group were fed with cholecalciterol cholesterol emulsion 20μL dissolved in 200 mL pure water which was kept in darkness. After 14 days, Z group and CCE+Z group were injected with zymosan at a dose of 500 mg/kg. After 18 hours,all the mice in each group were sacrificed. The liver tissues were harvested for histopathological examination. The serum ALT levels were determined. The molecular expression of IL-6 and IL-18 in liv-er tissue of mice were evaluated by Western blotting and real-time quantitative PCR method. Results The results of histopathological examination showed that liver tissue damage in CCE+Z group was lighter than that of Z group ,and heavier than that of the CON group. Compared to the CON group,Z group had the highest serum ALT level,followed by CCE+Z group,while in Z group was significantly lower than that in CON group(all P<0.05). The expression of IL-6 and IL-18 protein and mRNA showed level of Z group was apparently higher than those of CON group and CCE+Z group(all P<0.05). Conclusion Cholecalciterol cholesterol emulsion can play certain protective effect on zymosan-induced liver injury in mice.
