1.Changes of serum thyroid hormone levels in patients with polycystic ovary syndrome induced by ovulation disorder

Objective To analyze the relationship and significance between plasma level of thyroid hormone and ovulation disorder of (polycystic ovary syndrome)PCOS.Methods Eighty-nine cases with PCOS were induced ovulation by CC+HCG.During menstrual cycle of induced ovulation,PCOS patients were divided into two groups according to Ovulation (No ovulation 40,ovulation 49).The level of thyroid hormone at different follicular development period and ovulatory time were detected.Result Levels of thyroid hormone at two groups were no difference at 5 mm,10 mm,15 mm size follicular phase(P>0.05).But there was significant at follicular 20mm size(F inner groups=4.38,50.34,48.65;P<0.05).The level of T3 an T4 at No ovulation group was lower than those in ovulation group(T3:(2.69±0.15) nmol/L vs.(1.97±0.17) nmol/L;T4:(124.38±0.29) nmol/L vs.(97.43±0.16) nmol/L).The level of TSH at No ovulation group was (3.43±0.10) mU/L,higher than that in ovulation group ((1.38 ± 0.18) mU/L;P < 0.05).Conclusion The level of thyroid hormone in patients of ovulation group is at a normal level when the follicle is mature,and the level of thyroid hormone can affect the normal ovulation of patients with PCOS.
2.Effect of C2 gene on tumorigenesis of human gastric cancer cells.

Objective To investigate the effect of a new cloned full length gene, C2 gene, which encodes human eukaryotic translation initiation factor, on tumorigenesis of human gastric cancer cells in vivo and in vitro. Methods A constructed eukaryotic vector carrying the full length of C2 gene was amplified, purified, and transfected into a gastric cell line SGC7901 cell. The expression of C2 protein was examined with fluorescence activated cell sorting (FACS) and Western Blot. The effect of C2 gene on tumorigenesis of human gastric cancer cells was investigated in vitro and in vivo with methods of clone formation in plate and oncogenesis in nude mouse. Results FACS and Western Blot showed that C2 protein was expressed highly and stably in transfected cells. The average clone formation rate of C2 gene transducted cells is 43.8%, the rate which is lower than that of vector transducted cells (76.9%). In vivo, the time of tumorigenesis of C2 gene transducted cells in nude mouse is prolonged from 1 week to 2 weeks, and the volume of tumor node is smaller than that of vector transducted cells, that is, 1.20 and 5.58 cm 3 respectively ( P
3.Clinical observation on the treatment of anemia of hypothyroidism with complex melanteritum pill

Objective To observe the curative effect of complex melanteritum pill in treating anemia of hypothyroidism.Methods A total of 50 patients were randomly recrutied into a control group(20 patients)and a treatment group(30 patients).The treatment group was treated by Levothyroxine Sodium Tablets,50 μg qd,and complex melanteritum pill,nine pills/time,three times daily;while the control group was treated by levothyroxine sodium tablets,50 μg qd.Both groups were treated for 8 weeks,without blood transfusion,adding iron and folic acid and vitamin B12,etc;Results The basic cure rate and the total effective rate in the treatment group was 50.0%and 80.0%respectively;the basic cure rate and the total effective rate in the control group was 30.0%.and 50.0%respectively.There was a significant difference between the two groups by comparison(P<0.05).Conclusion Complex melanteritum pill was effective in treating hypothyroidism with few side effects.
4.Cohort study for the prevalence of the hyperlipidemia and the plasma lipids level in group of staff and workers in China Ocean Petroleum

Objective To study dynamic change for the prevalence of the hyperlipidemia and the plasma lipids level of staff and workers in group in China Ocean Petroleum so as to provide the evidence for the prevention of the patients suffered from hyperlipidemia. Method Four thousand eight hundred and nin-ty-nine workers fmished blood fat test and blood fat data separately for two times in 1998 and 2004. Results The TC average value of males rose to (5.68 ± 0.99) mmol/L from (5.46 ± 1.08) mmol/L and the TC av-erage value of females rose to (5.79 ± 0.99) mmol/L from (5.42 ± 1.04) mmol/L,both were statistical sig-nificant difference(P < 0.01 ).The TG average value of males rose to ( 1.65 ± 1.31 ) mmol/L from ( 1.48 ± 1.29) mmol/L and the TG average value of females rose to (1.46 ± 1.09) mmol/L from (1.28 ± 1.17) mmol/L,both were statistical significant difference (P < 0.01 ). The gross hyperlipidemia,HTC,HTG rate rose to 58.9%,45.1%,32.4% from 46.2%,32.1%,25.7% for the males and those rose to 58.0%,49.6%,26.2% from 42.0%,31.2%,19.3% for the females ,all were statistical significant difference (P < 0.01 ).After 6 years,the males control rate of the HTC and HTG were 27.5% and 29.9%,the females control rate of the HTC and HTG were 25.1% and 34.0%. Conclusions The staff and workers suffered from high hyperlipidemia and lipid level is higher in man group in China Ocean Petroleum and also faster. Treatment control rate of hyperlipidemia was very lower .Therefore,the prevention measure must be taken as fast as possible to reduce lipid level and hyperlipidemia patient's rate actively and improve control rate.
5.Application of potassium titanyl phosphate (KTP) laser delivered via bronchofiberscope in the treatment of endobronchial tuberculosis.

To explore the application of potassium titanyl phosphate (KTP) laser delivered via bronchofiberscope in the treatment of endobronchial tuberculosis. 36 patients with a diagnosis of endobronchial tuberculosis, with age ranging from 15 to 40 y were treated with KTP laser between Dec. 2002 and July 2004 (designated as treatment group). The other 36 patients diagnosed as having endobronchial tuberculosis (aged 18 to 42 y, with a mean age of 33. 5 y) without having received KTP laser treatment were included in a control group. Our results showed that the effective rates, in terms of recovery of bronchial lumen and cleanup of caseous necrotic mass were significantly higher in the treatment group 8 weeks after the treatment (P<0.01), and the healing rates of atelectasis and obstructive infection were also significantly higher in the treatment group (P<0.05 and P<0.01), but the incidence of complication after 8 weeks was no significant difference (P >0.05). No significant changes were found in SaO2 and HR before, during and after the operation in the treatment group (P>0.05). It is concluded that KTP laser is an effective therapy for endobronchial tuberculosis.
6.Effects of stimulating cerebellar fastigial nucleus on internal diameter of brachial artery in the elderly with transient ischemic attack

BACKGROUND: Stimulating cerebellar fastigial nucleus can regulate, dilate blood vessels and greatly increase local blood flow. However, if its mechanism is to improve the function of vascular endothelium is still uncertain.OBJECTIVE: To study the effects of stimulating cerebellar fastigial nucleus by electricity to vascular relaxation of transient ischemic attack(TIA) patients which is induced by blood flow and explore its protective mechanism to blood vessels of TIA patients.DESIGN: Randomized case control study to patients based on diagnosis SETTING: Department of general diseases, rehabilitation room and ultrasound department of a university hospital.PARTICIPANTS: Forty-four TIA patients(> 60 years old) admitted into Department of General Diseases of Tongji Hospital of Tongji Medical College of Huazhong University of Science and Technology during February 2001 to October 2002 were selected after excluding cerebral hemorrhage or cerebral infarction, atrium fibrillation and other arrhythmia or heart failure and blood system diseases. They were randomly divided into treatmentgroup(22 cases)and control group(22 cases) which were conducted stimulating fastigial nucleus + routine treatment and only routine treatment respectively.METHODS: High resolution ultrasound technique and brachial artery congestion method were used to test the internal diameter change rate of brachial artery in TIA patients before and after treatment.MAIN OUTCOME MEASURES: The internal diameter change rate of brachial artery of TIA pat ients.RESULTS: The internal diameter change rate of brachial artery was (4.59 ± 3.32) % and ( 10. 34 ± 3.13 ) % respectively before and after conducting electric stimulus to fastigial nucleus with significant difference ( t = 5.91, P < 0.01 ). There was significant difference on internal diameter change rate between treatment group and control group( t =5.41, P < 0.01 )while there was no difference in control group before and after treatment [(4.68±3.20) %,(5.10±3.29) %](t=1.72, P> 0.05).CONCLUSION: Conducting electric stimulating to fatigial nucleus can greatly improve the reactively vascular congestive function of brachial artery internal diameter and it can improve the vascular endothelial functions of TIA patients.
7.Effect of C2 gene on humen gastric cancer cell cycle

Objective To investigate the effect of a new cloned full length gene, C2 gene which encodes a translation initiation factor, on cell cycle and the relationship between C2gene and apoptosis. Methods Construct a eukaryotic vector carrying the full length of C2 gene, then transfected it into a gastric cell line-SGC7901 cells and gained cells expressing C2 protein transiently or stably. Test the expression of C2 protein and the change of cell cycle of the transfected cells using the flurescence activatied cell sorting(FACS) and the changes of their ultra structure electromicroscopically. Results The C2 gene eukaryotic expression vector was successfully constructed. FACS showed the C2 protein expression ratio of transfected cells was 32.3%, while the empty control group was 0.9%. The test of cell cycle showed that there was appeared apoptosis peak in transiently or stably transfected cells. The apoptosis cells can be seen electromicroscopically among the C2 transfected cells. Conclusions C2 gene transfection can lead SGC7901 cells to apoptosis, but the mechanism has to be further study.
8.Randomized controlled trial of icotinib concurrent with thoracic radiotherapy for treating advanced non-small cell lung cancer (NSCLC)

Objective To compare the efficacy and safety of icotinib therapy alone versus icotinib combined with thoracic radiotherapy for the treatment of advanced non-small cell lung cancer (NSCLC) patients with an activating epidermal growth factor receptor (EGFR) gene mutation.Methods A total of 83 patients with advanced NSCLC harboring an activating EGFR gene mutation was enrolled in this study.All the patients were randomly divided into 2 groups.Patients in group A (n =41) received thoracic radiotherapy (prescribed at 60-66 Gy) combined with icotinib (three times per day,125 mg once).Patients in group B (n =42) were given icotinib therapy alone (three times per day,125 mg once).Treatment was continued until disease progression or unacceptable toxicity or death.The primary end points were median progression-free survival (mPFS) and 12 month-PFS rate.The secondary end points included objective response rate (ORR),disease control rate (DCR) and adverse events.Results With a median follow-up of 18.2 months,mPFS was 15.2 months (95% CI:12.2-17.4) in group A and 13.2 months (95% CI:10.8-14.9) in group B (x2 =4.29,P=0.036).PFS rates of 12 months for group A and group B were 70.3% and 61.2%,respectively.The ORR were 78.0% vs.57.1% (x2 =5.16,P =0.028),and the DCR were 95.1% vs.92.9% (P>0.05) in groups A and group B,respectively.No grade 3-4 adverse events was observed in both groups except the rashes (4 cases in each group).Besides,10 patients had grade 1-2 radiation-related pneumonitis and 15 patients suffered grade 1-2 radiation-related oesophagitis in group A.Conclusions In advanced NSCLC patients with an activating EGFR gene mutation,the combination of thoracic radiotherapy and icotinib had achieved an improvement on ORR and PFS with good tolerance.Clinical trial registration Chinese clinical trial registry,ChiCTRINR-16010262.
9.Peptidoglycan modulation of chemokine production in normal human epidermal kerafinocytes and the role of Toll-like receptor 2 in this process

Objective To investigate the effect ofpeptidoglycan from Staphylococcus aureus on the release of several chemokines including intedeukin 8 (IL-8), regulated upon activation, normal T cell expressed and secreted (RANTES), macrophage-derived chemokine (MDC) by normal human epidermal keratinocytes (KCs) and the role of Toll-like receptor 2 (TLR2) in this process. Methods KCs were derived from the foreskin of a healthy boy and propagated. After 2 - 4 passages, KCs were collected and treated with various concentrations (3, 10, 30 and 100 mg/L) of peptidoglycan for 24 hours or with peptidoglycan of 100 mg/L for varying durations (3, 6, 12, 36 hours). A fi'action of KCs were pretreated with functional grade purified anti-TLR2 monoclonal antibody before the treatment with peptidoglycan of 100 mg/L. After additional 12-hour culture following the treatment, enzyme linked immunosorbent assay was used to detect the level of IL-8, RANTES and MDC in culture supernatants of KCs. Results KCs spontaneously released IL-8 and RANTES. Peptidoglycan increased the production of IL-8 but decreased that of RANTES by KCs. The levels of IL-8 were 209.96 ± 10.31 ng/L, 250.28 ± 9.52 ng/L, 285.11 ± 10.28 ng/L, 359.40 ± 6.93 ng/L in KCs treated with peptidoglycan of 3, 10, 30, 100 mg/L, respectively, compared to 135.41 ± 14.37 ng/L in untreated KCs (all P < 0.05). On the contrary, a significant decrement was seen in the secretion of RANTES by KCs treated with peptidoglycan of 10, 30, 100 mg/L compared with untreated KCs (110.72 ± 8.51 ng/L, 90.50 ±2.45 ng/L, 49.89 ± 13.74 ng/L vs 149.94 ± 18.71 ng/L, all P < 0.05). The monoclonal antibody to TLR-2 could markedly suppress the promotion of IL-8 production by peptidoglycan, but had no obvious influence on the inhibition of RANTES production by peptidoglycan. MDC could not be detected in the culture super-natants of KCs with or without peptidoglycan stimulation. Conclusion Peptidoglycan could inhibit RANTES secretion but induce IL-8 production by KCs likely via TLR2.
10.Relationships between constitutional types of traditional Chinese medicine and motion sickness in 145 ocean sailors.

Objective: To investigate the relationships between constitutional types of traditional Chinese medicine (TCM) and motion sickness. Methods: A survey of TCM constitutions in ocean sailors participating in a voyage was performed by using the TCM Constitution Questionnaire developed by Beijing University of Traditional Chinese Medicine, while the survey of motion sickness was operated by Graybiel's diagnostic criteria. The incidences of motion sickness among sailors with different types of constitutions were compared. Results: Prior to the voyage, 50.3% of sailors exhibited a gentleness constitution, 14.5% were of dampness-heat constitution, 10.3% were of qi-stagnation constitution, whereas the percentages of qi-deficiency, yang-deficiency, yin-deficiency, blood-stasis and special diathesis constitutions were 6.2%, 7.6%, 6.2%, 4.1% and 0.7%, respectively. None exhibited a phlegm-dampness constitution. By the end of the 176-day voyage, the percentages of gentleness, dampness-heat, qi-depression, qi-deficiency, yang-deficiency, yin-deficiency, blood-stasis, special diathesis and phlegm-dampness constitutions were 33.8%, 13.8%, 13.1%, 11.0%, 6.9%, 9.7%, 4.1%, 0.7% and 6.9%, respectively. The incidence of motion sickness was 69.7% (101 sailors) during this voyage. The incidences of motion sickness among sailors with different types of constitutions before the voyage showed significant difference (P<0.001). The incidence of motion sickness was higher in the sailors with dampness-heat constitution than in those with gentleness constitution. Conclusion: Types of Chinese medical constitution can be related to susceptibility to motion sickness. Furthermore, ocean voyage may have an effect or influence on the type of Chinese medical constitution of sailors involved.
