2.The efficacy of open occluded coronary artery branch through outside stent balloon expansion technique

Objective To evaluate the efficacy and safety of open occluded coronary artery branch through outside stent balloon expansion technique.Methods A retrospective analysis of 26 patients,26 interventional treat-ment of coronary bifurcation lesions were taken.All patients initially used conventional interventional techniques, branch reserve protection guidewire and main branch stent.When branch occlusion occured during operation and failed again through guidewire,small outer diameter of the balloon branch guidewire which was squeezed could be used to branch opening.The blocked branch opening was expanded outside of the stent,and the balloon was expanded after the guide wire enter into branch again.The main branch/branches kissing balloon inflation or main branch /branch double stenting would be used when necessary.Results Among 26 cases of bifurcation branch occlusion,25 cases successfully completed the outer stent balloon,achieved a branch balloon expansion after reentry guidewire,saved blocked branches and the rate of success was 96.2%,and 6 cases of the main branch and the branch kissing balloon dilatation,2 cases of remedial double stenting.Following -up for 1 -12(5.3 ±6.8)months after operation in 25 cases of patients,there was no death and myocardial infarction and other adverse cardiovascular events.Conclusion Out-side stent balloon expansion technique can improve the success rate of coronary occlusion branch opening,and it have fewer complications and worth of clinical application becasuse of its satisfactory results.
3.Tuina along the meridians combined with Chinese medicine for motor functions and activities of daily living in patients with post-stroke upper limb spasticity

Objective:To observe the effect of combining Chinese medicine and tuina along the meridians on motor function and activities of daily living (ADL) in patient with post-stroke upper limb spasticity. Methods:A total of 220 patients with post-stroke upper limb spasticity were randomly allocated into a treatment group (n=110) and a control group (n=110). Patients in the treatment group received tuina along the meridians combined with spasticity-alleviating and collateral-unblocking Chinese medicine, whereas patients in the control group received routine rehabilitation therapy. Patients in both groups were treated for 3 weeks. Then the patients’ motor function, ADL and muscle tone were evaluated before and after treatment using the Fugl-Meyer assessment scale (FMA), modified Barthel index (MBI) and modified Ashworth scale (MAS). Results:After treatment, the FMA scores, MBI scores, and muscle (shoulder intortor, elbow flexors and wrist flexors) tones were significantly improved (P<0.05), but the improvement was more significant in the treatment group than that in the control group (P<0.05). Conclusion:Tuina along the meridians combined with spasticity-alleviating and collateral-unblocking Chinese medicine can substantially alleviate muscle tone on the affected side and remarkably improve the patients’ motor function and ADL.

Aim To investigate the protecting role and mechanisms of crude flavones of pueraria loblata(FP) on human umbilical vein endothelial cells(HUVEC) in high glucose medium.Methods ①HUVECs were incubated in 20% FCS RPMI-1640 culture medium containing 0,5,10,30,60,90,120 mmol?L~(-1) glucose for 48 hours.The proliferation of HUVEC was analyzed with MTT method(490 nm).② HUVECs were incubated in 20% FCS RPMI-1640 culture medium containing 0 or 30 mmol?L~(-1) glucose,either alone or in the presence of FP(final concentration 10~(-3),10~(-4),10~(-5),10~(-6) mol?L~(-1) respectively) for 48 hours.At the same time,30 mmol?L~(-1) mannitol was added.The proliferation of HUVEC was analyzed using MTT method.The cell cycle of HUVEC was analyzed using flowcytometry.The contents of SOD,NO and ICAM-1 in HUVEC supernatant were measured using test kits.Results HUVEC in 5 mmol?L~(-1) glucose medium proliferated well,but the proliferation of HUVEC in 10,30,60,90 and 120 mmol?L~(-1) glucose media was disturbed,showing a negative correlation.Proliferation of HUVEC was inhibited in 30 mmol?L~(-1) glucose,compared with that in 0 mmol?L~(-1) glucose and 30 mmol?L~(-1) mannitol(P
5.Qualitative and quantitative study on the apoptosis of cervical cancer cells treated by tetrandrine

Objective To study the effects of tetrandrine on apoptosis of HeLa cervical cancer cells qualitatively and quantitatively. Methods We measured tetrandrine-induced inhibition of HeLa cell proliferation at different concentrations and time points by MTT assay. The rate of Hela cell apoptosis induced by tetrandrine was detected by flow cytometer and confocal laser scanning microscope (CLSM). Results Tetrandrine inhibited the proliferation of HeLa cells in dosage- and time-dependent manners. Flow cytometry showed that the apoptosis rate was (51.8±0.97)% at the concentration of 15μmol/L, which was significantly higher than that in the control group (24.3±1.23)% (P<0.05). The cells treated with tetrandrine showed typical apoptotic morphology under CLSM. Conclusion Tetrandrine can inhibit proliferation and induce apoptosis of HeLa cervical cancer cells.
6.Clinicopathologic characteristics of triple-negative breast cancer

Objective To study the clinicopathologic characteristics of triple-negative breast cancer (TNBC) and its value in the prediction of prognosis. Method In this study,500 cases of female breast cancers were examined immunohistochcmically for the TNBC. The clinicopathologic characteristics of the 243 TNBC cases were inspected. Results TNBC accounted for 17.6% (88/500) of the 500 breast cancers. The histological types of the TNBC included mainly infihrative ductal carcinoma, metaplastic carcinoma and medullar carcinoma. Among those, histological grade Ⅲ accounted for 72.7% (64/88) of all the TNBC and was more common than that in hormone receptor positive breast cancers (HR+ group ) and Her-2 overexpression breast cancers (Her-2 group)(P=0.000). The positive rates of CK5/6 and EGFR in the TNBC were 30.7% (27/88) and 34.1% (30/88), respectively. The positive rates of ERCC1 and KIT in the TNBC were 28.4% (25/88) and 34.1% (30/88), respectively, Both of which were higher than those in the HR + group and Her-2 group, respectively (P=0.032 and P=0.026). 3-year survival rate of the TNBC was 71.5% and it was lower than that of HR group (P=0.021) and not significantly different from that of Her-2 group (P=0.474). Conclusions TNBC is the breast cancer with high aggressive pathologic futures and poor prognosis. EGFR and ERCC1 expression were positive in a portion of TNBC cases.
7.Study on the Correlation between CYP2C19 Gene Polymorphisms and Antiplatelet Reactivity of Clopidogrel in Xinjiang Uygur Patients with Acute Coronary Syndrome

OBJECTIVE:To investigate the correlation between CYP2C19 gene polymorphisms and antiplatelet reactivity of clopidogrel in Xinjiang Uygur patients with acute coronary syndrome (ACS). METHODS :Totally 90 Uygur patients with ACS who were admitted to the cardiovascular department of Xinjiang Kashi Second People ’s Hospital from Jan. 2018 to Jan. 2019 were selected as the study subjects. They were given anti-platelet therapy of asprin+clopidogrel ,and received the treatment continuously for one year after discharge. The platelet aggregation inhibition (DPAI)rate of the patients were determined ,and the response to clopidogrel was evaluated. PCR-fluorescence probe method was used to detect genotype of CYP2C19*2 and * 3,and PCR-direct sequencing method was used to detect genotype of * 17. Multivariate Logistic regression analysis was adopted to investigate the correlation of gene and non-gene factors with DPAI of patients. RESULTS :Among 90 patients,there were 10 patients with clopidogrel resistance (CR)and 80 patients with non-CR. There were 58,28 and 4 patients with CYP2C19*2 G/G,G/A,A/A genotype,respectively;there were 88 and 2 patients with CYP2C19*3 G/G,G/A genotype ,respectively;there were 64 and 26 patients with CYP2C19*17 C/C,C/T genotype ,respectively;the genotype frequency of each genotype was consistent with Hardy-Weinberg equilibrium (P>0.05). After treatment ,DAPIs of patients with CYP2C19*2 G/A,A/A genotype were decreased significantly,while those of the patients with A/A genotype were significantly lower than patients with G/A genotype patients (P< 0.05). DAPIs of patients with CYP2C19*17 C/T genotype were increased significantly ,compared with C/C genotype 2016D01C087) patients (P<0.05). There was no statistical significance in @fudan.edu.cn DAPIs between CYP2C19*3 G/A and A/A genotype patients (P>0.05). Multivariate Logistic regression analysis showed 85775264@qq.com that CYP2C19 gene pol ymorphism was independently related to DPAI in Xinjiang Uygur patients with ACS [OR =2.314,95%CI(1.569,3.144),P=0.009],while age ,gender,smoking and other non-gene factors were not related to DPAI (P>0.05). CONCLUSIONS :CYP2C19 gene polymorphism is associated with antiplatelet reactivity of clopidogrel in Xinjiang Uygur patients with ACS. * 2 wild type patients may have higher DPAI ,while * 17 wild type patients may have lower DPAI.

We report a complete genomic sequence of rare isolates (minor genotype) of the SARS-CoV from SARS patients in Guangdong, China, where the first few cases emerged. The most striking discovery from the isolate is an extra 29-nucleotide sequence located at the nucleotide positions between 27,863 and 27,864 (referred to the complete sequence of BJ01) within an overlapped region composed of BGI-PUP5 (BGI-postulated uncharacterized protein 5) and BGI-PUP6 upstream of the N (nucleocapsid) protein. The discovery of this minor genotype, GD-Ins29, suggests a significant genetic event and differentiates it from the previously reported genotype, the dominant form among all sequenced SARS-CoV isolates. A 17-nt segment of this extra sequence is identical to a segment of the same size in two human mRNA sequences that may interfere with viral replication and transcription in the cytosol of the infected cells. It provides a new avenue for the exploration of the virus-host interaction in viral evolution, host pathogenesis, and vaccine development.
