Objective To explore the causes of death of patients with cerebral hemorrhage of hypertension in recent 6 years and the countermeasure of decrease death rate.Methods The clinical data,different kinds of treatment and curative effects of the patients of two groups,admitted in recent six years,with cerebral hemorrhage of hypertension was refrospectively analysied.Results Of 157 cases of cerebral hemorrhage of hypertension in group A,there were 69 cases died(44%).Of 158 cases of cerebral hemorrhage of hypertension in group B,there were 48 cases died(30.4%).Death rate between the two of group had significant difference(P

AIM: To investigate the effects of octreotide on metabolism in the A549 cells treated with lipopo-lysaccharides ( LPS).METHODS: The technologies of gas chromatography/mass spectrometry (GC/MS) and liquid chromatography/mass spectrometry (LC/MS) were used to test the metabolism of lung A549 cells subject to different treat-ment with LPS and/or octreotide.The results were visualized and checked by chromatogram, and the corresponding intensi-ty data were analyzed by the principal component analysis(PCA)method.The metabolites with different expression and the underlying interaction network were resolved.RESULTS: The metabolism analysis by LC/MS method indicated that there were different expression levels between different treated groups.Further analysis was carried out by orthogonal projections to latent structures-discriminant analysis (OPLS-DA) and the different expressed metabolites were obtained, which were mainly amino acids and phospholipids.By analyzing with GC/MS method and t-test, the different expressed metabolites were mainly organic acid, saccharides and amino acid metabolite.The interaction network diagram was constructed about the response of A549 cells induced by LPS and/or octreotide, including glycolysis/gluconenogenesis, tryptophan metabo-lism, galactose metabolism, urine cycle and citrate cycle.Fourteen key components were found such as serotonin, indole, threonine, serine, glucose, phenylalanine, lactose, fumarate, 4-hydroxyphenyllactate, aspartate, asparagine, putrescine, proline and succinate.CONCLUSION: In octreotide treated LPS-induced A549 cells, the main metabolites are organic acid, saccharides, amino acids and phospholipids.The interaction network is constructed, including 5 metabolic pathways
and 14 key components.
3.High glucose induces H9 c2 cardiomyocyte hypertrophy through Ca2+-CaN-NFAT3 signaling pathway

AIM:To study the morphological changes of cardiac H9c2 cells during the developmental process of fetal rat.METHODS:Embryonic rat heart-derived H9c2 cells were maintained in DMEM supplemented with 10%fetal bovine serum.The H9c2 cells were plated at a density of 6 000 cells/cm and divided into 5 groups:H9c2 cells were trea-ted with 5 mmol/L glucose, 25 mmol/L glucose, 50 mmol/L glucose, Norvasc (25 nmol/L) +25 mmol/L glucose, or Norvasc (25 nmol/L)+50 mmol/L glucose for 48 h.The morphology of H9c2 cells was observed.The cell surface area was measured by Image-Pro Plus 6.1 software.Fluorescence spectrophotometry was used to detect the concentration of in-tracellular calcium ion ( [ Ca2+] i ) in the cardiomyocytes.The concentration of CaN in the cell was measured by ELISA. The mRNA expression of CaNAβ, NFAT3 and β-MHC in the cells was detected by real-time PCR.The protein levels of CaNAβ, NFAT3 and β-MHC in cultural H9c2 cells were detected by Western blot.RESULTS: The mean area of the cells, the mean fluorescence value of [ Ca2+] i and the concentration of CaN in 25 mmol/L glucose group were higher than those in 5 mmol/L glucose group, and those were lower than those in 50 mmol/L glucose group.After treated with Nor-vasc, those results decreased significantly.The expression of CaNAβ, NFAT3 andβ-MHC at mRNA and protein levels in 25 mmol/L glucose group was higher than those in 5 mmol/L glucose group, but was lower than those in 50 mmol/L glu-cose group .The expression of CaNAβ, NFAT3 andβ-MHC at mRNA and protein levels decreased significantly in Norvasc treatment group.CONCLUSION:Ca2+-CaN-NFAT3 signaling pathway is perhaps involved in high glucose-induced H9c2 cardiomyocyte hypertrophy.
4.Morphology and distribution of CD44+/Oct4+colorectal cancer stem cells

BACKGROUND:More and more studies employ CD44 as a specific marker of colorectal cancer stem cells. Oct4 is a transcription factor of embryonic stem cells, and it has been discovered recently that there is a higher expression in primary colorectal carcinoma.
OBJECTIVE:To investigate the quantity, location and distribution of CD44+/Oct4+cells in primary colorectal carcinoma.
METHODS:A total y of 108 cases of human colorectal carcinoma and 18 cases of normal mucosa, 18 cases of adenoma were col ected and made into three tissue microarrays, each containing of 48 dots. The locations of CD44+/Oct4+cells were detected by double-label immunohistochemical staining and hematoxylin-eosin staining. The morphologic features of them were investigated on hematoxylin-eosin staining at the same position.
RESULTS AND CONCLUSION:The results of double-label immunohistochemical staining demonstrated that there were no CD44+/Oct4+cells in normal intestine mucosa and a very smal amount of CD44+/Oct4+cells in adenoma, and double-positive cells could also be seen in colorectal carcinoma. The number of CD44+/Oct4+cells was rare and the cells were scattered or distributed focal y along the basement of gland basal side. The cells with scarce cytoplasm were square, and its nucleus was oval or high cylindrical, deeply stained and homogeneous. The quantity of CD44+/Oct4+cells was negatively correlated with the differentiation of colorectal cancer (r=-0.579, P<0.01), and was associated with the depth of tumor invasion (r=0.236, P<0.05). These findings indicate that CD44+/Oct4+cells may be colorectal cancer stem cells.
5.Voxel based morphometric study of brain structure in patients with posttraumatic stress disorder.

Voxel based morphometry (VBM) methods are used to detect the difference in brain structures between the posttraumatic stress disorder (PTSD) sufferers and the normal controls. Standard VBM method can detect the difference of the gray matter or white matter densities while the optimized VBM method can detect the difference of gray matter or white matter volumes in the whole brain. The experiments showed that for the patient group, gray matter density or volumes significantly increased in the right frontal lobe, middle frontal gyrus, vermis, left caudate and parietal lobe, compared with the normal controls. However, in the left frontal lobe and middle frontal gyrus, gray matter density significantly decreased. There is no significant difference in white matter between the two groups. These results are consistent with those of the fMRI, which not only provide the evidence for further study of the pathogeny in PTSD but also validate the efficiency of the VBM methods for detecting the difference in the whole brain structure.
