Fifty-one patients with bee-sting poisoning were divided into slight poisoning group (n =22),moderate poisoning group (n =20) and severe poisoning group (n =9).And 30 healthy persons were selected as control group. The levels of interleukin-8 (IL-8),Von Willebrand factor (vWF),prothrombin time (PT),activated partial thromboplastin time (APTT),fibrinogen (FIB),thrombin time (TT) and D-dimer were measured. The levels of all parameters in slight poisoning group were no significantly different from with control group (P > 0.05).The levels of PT,APTT,IL-8 and vWF were significantly higher in moderate and severe poisoning groups than those in the control group (P <0.01).But the levels of APTT,IL-8 and vWF in severe poisoning group were much higher than those in moderate poisoning group (P <0.01).The injury of vascular endothelial cell by IL-8 and vWF caused the change of coagulative function and promoted the progress of inflammation.

Objective To use the matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) technique for detecting the mutation gene of neonatal non-syndromic hereditary hearing impairment gene in Guangxi and to investigate its effectiveness and feasibility in clinical application. Methods A total of 7 100 newborns were performed the hearing preliminary screening and secondary screening by adopting AABR.The genomic DNA was extracted by the heel blood spot.Twenty mutation characteristics of 4 deaf predisposing genes were detected by MALDI-TOF-MS.Results The pass rate of hearing screening in 7 100 newborns was 97.11% (6 895/7 100),the positive rate of neonatal gene mutation was 3.54% (251/7 100),in which the GJB2 gene mutation was in 131 cases, the carrying rate was 1.84%,235delC heterozygous mutation was in 108 cases.SLC26A4 gene mutation was in 93 cases, which dominated by 1229C>T heterozygous mutation and IVS7-2A>G heterozygous mutation,mtDNA12SRNA gene mutation was in 16 cases and GJB3 gene mutation was in 11 cases. Conclusion Adopting the MALDI-TOF-MS screening technique can increase the detection rate of hot point mutation in common deaf related genes and discover neonatal genetic NSHI from molecular level and provides the corresponding geneticconsulting guidance for early finding and predicting deaf occurrence, and formulating the interventional measures.

Objective To analyze the content of active components in fresh Danshen Radix Salviae Miltiorrhizae,Red Sage Root) from different producing areas, and review reasonably the medicinal quality of fresh Danshen. Methods The content of 5 salvianolic acids and 12 tanshinones in fresh Danshen, from 7 different producing areas, were determined by applying LTQ-Orbitrap XL high-resolution mass spectrometer. On this basis, all content were analyzed by using principal component analysis (PCA) and partial least square (PLS) analysis. Results The content of active components in fresh Danshen from the same producing area were basically the same, and difference was significant in content of active components among fresh Danshen from different producing areas. The content of salvianolic acids was the highest in fresh Danshen from Hubei, and content of tanshinone Ⅰ and tanshinone ⅡA were the highest in fresh Danshen from Sichuan and Henan. The analysis results of PCA and PLS showed that the distribution of most active components had a distinct regional specificity. Conclusion The difference is significant in quality of fresh Danshen from different producing areas, and they have a distinct regional specificity.Using fresh Danshen as quality evaluation materials, that is a useful supplement to review quality of raw medicinal materials based on dry herbs. It explored a new and more comprehensive approach for reviewing other Chinese medicinal materials.

BACKGROUND:The anterior cruciate ligament has unique nonlinear mechanical properties under a complex physiological loading environment.Elastin is an important contributor to the mechanical properties of the anterior cruciate ligament, but its mechanical response to the anterior cruciate ligament under axial tension is not clear. OBJECTIVE:To quantitatively analyze the effect of elastin on the tensile mechanical response of the anterior cruciate ligament. METHODS:Elastase solution and control buffer were prepared. The porcine anterior cruciate ligament samples were prepared into small-size samples and randomly soaked in 0,0.1,1.0,2.0,5.0,and 10.0 U/mL elastase solution for 6 hours, and other small samples of the same size were soaked in 2 U/mL elastase solution for 0,1,3,6,9,and 12 hours. To determine suitable soaking conditions for elastin-targeted enzymes and verify the digestive effect, histological staining was used to compare the effects of enzyme treatment on tissue structure and composition. The ligament samples were randomly divided into elastase-treated group and PBS group, and immersed in 2 U/mL elastase solution and PBS buffer for 6 hours, respectively.A mechanical tensile test was performed before and after immersion. RESULTS AND CONCLUSION:(1)The biochemical results showed that being treated in 2 U/mL elastase solution for 6 hours could reduce the elastin content by 31.1%,and had no significant effect on other mechanical-related components in the tissue.(2)The histological results showed that elastase was able to penetrate the tissue, and the loose degree of tissue increased after treatment.(3)In the mechanical results before and after treatment, the mechanical properties of the PBS group decreased significantly, only the low-tension elastic modulus increased significantly and the initial length increased significantly in the elastase-treated group.(4)The intergroup comparison results showed that there was no significant difference between the two groups in pre-treatment,but the low-tension elastic modulus, initial slopes, saturated slopes, and initial length of the elastase-treated group after treatment were significantly higher than those in the PBS group.(5)These results suggest that elastin degradation significantly affects the biomechanical properties of the anterior cruciate ligament and further complements our understanding of the structure-function relationship of the anterior cruciate ligament.
