1.Correlation Study Between Serum Platelet Derived Growth Factor Level and Ultrasound Imaging Features of Coronary Atherosclerotic Plaques in Relevant Patients

Objective: To detect the features of atherosclerotic plaques and their correlation to serum platelet derived growth factor (PDGF) level in patients with different types of coronary artery disease (CAD) by intravascular ultrasound (IVUS). Methods: A total of 106 patients admitted in our hospital with coronary angiography from 2011-01 to 2013-10 were enrolled and divided into 2 groups: Acute coronary syndrome (ACS) group,n=60 and Stable angina pectoris (SAP) group, n=46. Basic information as gender, age, history of smoking, hypertension and diabetes were recorded in all patients; serum levels of PDGF were measured by ELISA. IVUS was conducted to examine coronary atherosclerotic plaque morphology, property, rupture, thrombosis and refactoring in criminal vessels; meanwhile, external elastic membrane area, lumen area, plaque area and eccentric index were determined at the lesion site. The above parameters were compared between 2 groups and the correlation of different types of plaques to serum levels of PDGF were studied. Results:①Serum PDGF level in SAP group was higher than ACS group (3.64±0.60) ng/L vs (2.12±0.51) ng/L,P<0.05.②The patients in SPA group were mainly having hard plaque and in ACS group were mainly having soft plaque. Compared with SAP group, ACS group had the higher incidences of plaque rupture, thrombosis and positive refactoring; ACS group was mainly with eccentric plaque with low eccentric index allP<0.05-0.01; ACS group had the larger external elastic membrane area at the lesion site,P<0.05.③The patients with fibrous plaque, calcified plaque and mixed plaque (hard plaque) had the lower serum levels of PDGF than those with soft plaque,P<0.05. Conclusion: PDGF had been involved in atherosclerotic plaque formation and it played an important role in ACS pathogenesis. The patients with unstable plaques had the higher serum PDGF level, therefore PDGF detection may help ACS diagnosis and prediction.

OBJECTIVE To investigate the antimicrobial-resistant profile of sulfamethoxazole-trimethoprim(SXT) resistant Staphylococcus aureus.METHODS The strains of S.aureus(SAU) were isolated from clinical specimens by routine methods.The identification and susceptibility test of the isolates were determined by Automated Microbiology Analyzer.Disk-diffusion was used for detecting meticillin-resistant S.aureus(MRSA).?2 Test was made to identify the significance of difference.RESULTS 68.0% Of isolates were referred as MRSA.The resistant rates of the isolates of SAU to SXT was 30.8%.The resistant rate of the isolates of MRSA and MSSA to SXT were 27.4% and 38%.60.5% And 71.3% of isolates were referred as MRSA from the SXT-resistant group and MRSA from the SXT-sensitive group.The resistant rate of MRSA from the group of SXT-resistant to tetracycline,levofloxacin and rifampicin were all significantly lower than those of isolates from the SXT-sensitive group(all P

Recurrent spontaneous abortion (RSA) was one of the most difficult infertile diseases which was usually defined as consecutive still birth for more than three times during the first 20 weeks of gestation. Various factors and pathogeneses were thought to play a role in RSA. Recent studies had indicated that the disorders of blood vessel growth and the secondary abnormal blood perfusion may contribute to the occurrence of RSA. However, blood vessel growth was linked with the regulation of angiogenesis related factors. To provide references for the clinical prevention and treatment, the purpose of this paper was to evaluate the association between solubility vascular endothelial growth factor receptor-1(sflt-1), thrombospondin 1(TSP-1) and ovary, endometrial abnormal blood perfusion in cases of RSA.
6.Rapid detection of isoniazid resistance in clinical Mycobacterium tuberculosis isolates by high-resolution melting curve analysis

We detected the isoniazid resistance in clinical Mycobacterium tuberculosis isolates by high-resolution melting (HRM) curve analysis and assessed the application value of the assay.The isoniazid resistance of 49 M.tuberculosis isolates preserved in laboratory was analyzed by the drug sensitivity test (traditional proportion method).Further analysis was made on the sequencing of the isoniazid resistance determining region in these test strains,and their mutation sites were screened.Specific primers used in the HRM curve analysis were designed based on the screened mutation sites,DNA mutations were assayed in the isoniazid-resistant gene determining region by the HRM curve analysis,and an assessment was made of the detection efficiency of the assay in isoniazid resistance in M.tuberculosis.Results of the drug sensitivity test (proportion method) showed that,of the 49 test strains,there were 20 isoniazid-resistant strains,29 isoniazid-sensitive strains.Results of the sequencing analysis showed that:1) KatG gene had four mutation patterns,i.e.,point mutations at site 234,at sites 234 and 315,at sites 234 and 463,and at sites 234,315 and 463;2) there were three mutations were detected in inhA gene,i.e.,mutations in inhA-8,-15 and-152.Analysis of gene mutation in drug-resistant strains found that of the 20 isoniazid-resistant strains,11 (55 %) were mutated at codon 315 of KatG gene;6 (30%) were mutated in inhA-15 (4/20),-8 (1/20) and-153 (1/20) of inhA gene;two (10%) were mutated at codon 315 of KatG gene and in inhA-15;in one strain (5%),no mutation was detected in KatG and inhA genes.Through the gene mutation detection,the sensitivity and specificity of isoniazid resistance in M.tuberculosis were 95 % and 100 %,respectively.Results of HRM curve analysis of drug-resistance gene mutations in test strains showed gene mutations were present in 18 strains and absent in 24 ones;referring to DNA sequencing results,the sensitivity and specificity of the assay were 94.7% and 80%,respectively.Judged by mutations as drug-resistance via the HRM curve analysis,19 resistant and 24 sensitive strains were tested.With the drug sensitivity test results by the proportion method as controls,the sensitivity and specificity of the assay were 95 % and 82.76 %,respectively.Use of the HRM curve in the detection of resistance of M.tuberculosis to isoniazid is characterized by good sensitivity and short time consuming,and has certain value in the rapid diagnosis of isoniazid-resistant tuberculosis.
7.Effect of Psychology Intervention and Family Rehabilitation Guides on Patients with Spinal Cord Injury

Objective To observe the influence of psychology intervention and family rehabilitation guides on psychological status and quality of life of patients with spinal cord injury(SCI). Methods 24 SCI patients were divided randomly into treatment(n=12) and control(n=12) groups. The treatment group was given to psychology intervention and family rehabilitation, the psychological status and ADL were evaluated individually by SCL-90 and Barthel index on pre-guide and post-guide of six months.Results The discrepancy got statistic meaning on degree of SCL-90 and Barthel index on pre-guide and post-guide(P<0.05). Conclusion Effective psychology intervention and family rehabilitation guides could impove psychological status and enhance quality of life of patients.

Objective To investigate the expression of transforming growth factor beta (TGF-β) and Smad signaling in ankylosing spondylitis (AS) and to explore their roles in the pathogenesis. Methods Fiftythree patients with AS were included in the study. In these 53 cases, 30 patients were performed computed tomography-guided needle biopsy in sacroiliac joint. Serum TGF-β_1 was determined by enzyme-linked immunosorbent assay (ELISA). Immunohistological studies were performed with the streptavidin-peroxidase conjugated methods to assess the expression of TGF-β_1, p-smad3 and Smad7 in sacroiliac joint tissue sample.One-way ANOVA, two independent samples t test and kolmogoorov-Simimov test were used to do statistical analysis. Results In 53 cases patients with AS, 20 cases were with high level Erythro-cyte sedimentation rate(ESR) and C-reactive protein (CRP), while those of the other 33 cases were normal. Serum average TGF-β_1level [ (15.9±5.6) ng/ml ], in patients with high level ESR/CRP [(5.4±5.8) ng/ml ] was significantly increased as compared to the controls and patients with normal ESR/CRP [(4.1±3.6) ng/ml] (P<0.05). There was no expression of TGF-β_1 could be detected in the pannus and bone marrow in SI joints tissue of 30 cases with AS, while decreased level of smad7 expression was detected. In addition, p-smad3 expression was found in the nuclear. Conclusion TGF-β_1 signaling may play an important role in the inflammatory erosion and cartilage fibrosis of sacrojlitis in AS.

Objective To investigate the expression of connective tissue growth factor(CTGF),coll agen I and collagen Ⅲ in sacroiliac joint(SIJ)of patients with spondyloarthropathy(SpA).Methods Thirty patients with SpA,including 17 patients with grade Ⅱ saeroiliitis and 13 patients with grade Ⅰ sacroiliitis,were performed on CT guided needie biopsy of SIJ.After sacroiliitis were confirmed by staining with hematoxylin and eosin in sacroiliac joint tissue sample,immunohistochemical assay was performed to determine the expression of CTGF,collagen Ⅰ and collagen Ⅲ in sacroiliac ioint tissue.Univariate Chi-square test was used for data comparison between multiple groups and t-test was used for two group data comparison.Results Contrast to healthy controls,CTGF were found upexpressed on the cytoplasm of inflammatory cells in pannus and bone marrow of sacroiliac tissue samples of patients with SpA,while collagen I and collagen Ⅲ were found up-expressed in bone,cartilage and ligament tissue[(57.9±42.4)/HP vs(2.7±2.5)/HP P<0.05,0.298±0.080 vs 0.044±0.024 and 28.254±41.165 vs 0.105±0.054.P<0.05 respectively].Conclusion CTGF,collagen Ⅰ and collagen Ⅲ are up-expressed in SIJ of SpA patients.CTGF may play an important role in articular cartilage fibrosis and ossification of SpA.
10.Targeting Prnp in bovine fibroblasts by promoter-trap strategy.

Promoter-trap strategy for enriching targeted colonies has been usually used to elevate the gene targeting efficiency in somatic cells. Knocking out Prnp in animals by gene targeting can render them resistant to Prion diseases. We constructed a bovine Prnp promoter-less targeting vector BoPrPneo, then transfected the linearized vector into the bovine fetal fibroblasts BFF through electroporation. After selecting in cell culture medium with 250 microg/mL G418, we obtained 99 drug-resistant cell colonies, 4 of them were positive for targeted events after PCR screening, and the targeted colonies were further confirmed by sequencing and Southern blotting. This suggests that one allele of Prnp has been successfully knocked out in bovine fetal fibroblasts. This research supplies a simple, safe and effective method to targeting bovine Prnp.
