1.Resveratrol attenuates LPS-induced cell injury and TLR4/NF-κB activation in human periodontal ligament cells

Objective:To investigate the protective effect of resveratrol on lipopolysaccharide (LPS)-induced cell injury in human periodontal ligament cells(hPDLCs).Methods:hPDLCs were cultured and identified.The cultured hPDLCs were divided into 5 groups:control group and LPS(10 μg/ml) + RES(0/30,60 and 90 μmol/L respectively) groups.The cell proliferation was detected by MTI assay.The secretion of TNF-α and IL-6 of hPDLCs was detected by ELISA kit.The expression of TLR4/NF-κB mRNA and protein was determined by PCR and Western blot analyses,respectively.Results:The cultured cells were negative for cytokeratin and positive for vimentin staining.Compared with the control group,cell proliferation was decreased,the secretion of TNF-α/IL-6 levels and the expression of TLR4/NF-κB mRNA and protein were increased after treatment with LPS.Whereas,with 30-90 μmol/L resveratrol pretreatment,the proliferation ability of hPDLCs was enhanced(P < 0.05),the secretion levels of TNF-α and IL-6 and the expression of TLR4/NF-κB mRNA and protein were reduced (P < 0.01) in a dose-dependent manner.Conclusion:Resveratrol may attenuate LPSinduced cell injury by inhibiting TLR4/NF-κB pathway in hPDLCs.
2.EFFECTS OF FRUCTOOLIGOSACCHARIDE ON CHOLESTEROL METABOLISM IN FINISHING PIGS

Objective: To study the effects of dietary fructooligosaccharide (FOS) on cholesterol metabolism in finishing pigs. Methods: A total of 96 finishing pigs (Duroc?Landrance?Jia) were randomly alotted to four groups each of which was replicated three times with eight pigs per replicate. The groups received the same basal corn-soybean meal diet and FOS was added to the basal diet at 0,0.5%,1 0%,1.5% at the expense of corn, respectively. At the end of the feeding trial, eight pigs with similar body weights from each treatment were slaughtered. Results: (1) Supplementation with FOS significantly improved average daily gain and feed efficiency. (2) Pigs fed 1.0% and 1.5% FOS had significantly lower serum TC and LDL-C concentrations and greater HDL-C/LDL-C ratio compared with the control. Pigs fed 1.5% FOS had significantly lower serum VLDL-C and ApoB concentrations and Apo B/Apo A-1 ratio. (3) Supplementation with 1.0% and 1.5% FOS significantly decreased the activities of HMG-CoA reductase. (4) Supplementation with 1.5% FOS significantly increased the colonic acetate and butyrate and plasma acetate. Supplementation with 0.5%,1.0% and 1.5% FOS significantly increased colonic propionate. (5) Fecal cholesterol excretion was significantly greater in the pigs fed 1.0% and 1.5% FOS and fecal bile acid excretion was significantly greater in the pigs fed 1.5% FOS than the control. Conclusion: FOS may improve cholesterol metabolism by alteration in the absorption and synthesis of cholesterol resulted from the changes in colonic fermentation, and by increase in the fecal excretion of cholesterol and bile acid.

4.Ticagrelor versus clopidogrel in patients with acute coronary syndromes and diabetes mellitus

Patients diagnosed to have acute coronary syndromes( ACS) were included in the study. All ACS patients were divided into non-diabetes group(40 cases) and diabetes group(80 cases) . Blood coagulation function was de-termined for all patients. In the diabetes group eighty patients were randomly divided into ticagrelor group (48 ca-ses) and clopidogrel group (32 cases) . After 5 days′treatment, platelet function was detected. Our study aimed to examine the effects of ticagrelor and clopidogrel on the platelet function.
5. Muscarinic cholinergic receptor antagonist inhibits the growth and angiogenesis of small cell lung cancer in vivo

Objective: To investigate the effects of muscarinic cholinergic receptor 3 (M3R) antagonist 4-diphenylacetoxy-N -methylpiperidine methiodide (4-DAMP) on the growth of small cell lung cancer and angiogenesis in nude mice. Methods: The mRNA and protein expressions of M3R in human small cell lung cancer SBC3 cells were detected by RT-PCR and Western blotting, respectively. The subcutaneous transplantation tumor model of SBC3 cells in nude mice was established. The model mice were randomly divided intofour groups, then intraperitoneally injected with 0.9% sodium chloride solution (as the control group) and different doses of 4-DAMP (0.5, 1 and 2 mg/kg), respectively (once a day, for 15 days). The size of xenograft tumor was measured every 2 days. After the nude mice were sacrificed, the tumor mass was measured, and the tumor inhibitory rate was calculated. The expressions of M3R and vascular endothelial growth factor (VEGF) in the tumor tissues were detected by real-time fluorescent quantitative PCR and Western blotting, respectively. The microvessel density (MVD) and VEGF expression were detected by immunohistochemistry. Results: Human small cell lung cancer SBC3 cells expressed M3R mRNA and protein. After the subcutaneous transplantation tumor model of SBC3 cells in nude mice was successfully established and treated with different doses (0.5-2 mg/kg) of 4-DAMP, the sizes of xenograft tumors were significantly decreased as compared with the control group (all P < 0.05), and the weights of tumor tissues were significantly reduced (all P < 0.01). The tumor growth inhibitory rates in 0.5, 1 and 2 mg/kg 4-DAMP-treated groups were 15.82%, 33.54% and 55.06%, respectively. Furthermore, the relative expression levels of M3R and VEGF as well as MVD in the tumor tissues of 0.5, 1 and 2 mg/kg 4-DAMP-treated groups were significantly down-regulated as compared with the control group (all P < 0.05). Conclusion: M3R antagonist 4-DAMP can inhibit the growth and angiogenesis of human small cell lung cancer in nude mice.
6.EFFECTS OF PROPIONATE ON CHOLESTEROL METABOLISM IN FINISHING PIGS

Objective: To study the effects of dietary sodium propionate on cholesterol metabolism in finishing pigs. The groups received the same basal corn-soybean meal diet and sodium propionate was added to the basal diet in the concentrations of 0.5、1.0、1.5 % at the expense of corn, respectively. At the end of the feeding trial, eight pigs from each treatment were slaughtered. Results: (1) Pigs supplemented with 1.0% and 1.5% sodium propionate had significantly lower serum cholesterol and low density lipoprotein cholesterol concentrations and greater ratios of high density lipoprotein/low density lipoprotein cholesterol compared to the control. Pigs supplemented with 1.5% sodium propionate had significantly lower serum very low density lipoprotein cholesterol. (2) Supplementation with 1.0% and 1.5% sodium propionate significantly decreased the cholesterol content in liver and the activity of hepatic 3-hydroxy-3-methylglutary coenzyme A (HMG-CoA) reductase. (3) Supplementation with sodium propionate had no significant effects on growth performance, colonic or serum short-chain acid concentrations, fecal cholesterol or bile acid excretion. Conclusion: Sodium propionate may decrease the activity of hepatic HMG-CoA reductase and suppress the synthesis of cholesterol.
8.Analysis of problems existing in using process of micro-perfusion pump by low birth weight infants and the improvement measures

Objective To make a concrete analysis of micro-perfusion pump and compatible syringes used in neonatal intensive care unit (NICU) for low birth weight infants,and to find solutions to the defects in application.Methods 20 ml sterile syringes were set different infusion rates (1ml/h,5ml/h,6ml/h,10ml/h),different infusion time (5min,30min,lh).Then,the card slot on the micro-perfusion pump where the end of 20ml sterile syringe was placed was regulated.The regulated syringes were also set different infusion rates (5ml/h,6ml/h,l0ml/h) and time as before.The error accuracy was calculated.Results The results showed that at the end stage of 20ml sterile syringes pumping,there was always 0.5~1.5 ml residual liquid which could not be pumped out,but perfusion pump displayed injection was completed.Before regulation,the error accuracy between different infusion rates of 20ml sterile syringes was 6.00%,3.80%,2.33%,1.30% respectively.However,after improvement,the regulated syringes could completely pump all liquid out,and the error accuracy between different infusion rates and different infusion time was also superior to pre-improvement.Conclusions Improved micro-infusion pump and sterile solution prefilled syringe have no drug residual,and display high accuracy and small error accuracy.
9.Hearing Thresholds at Extended High Frequencies in Civilian Student Pilotswith Normal Hearing at Conventional Frequencies

Objective To study extended high frequency (EHF) audiometry for early detection of hearing loss in student pilots in civil aviation.Methods A total of 175 student pilots (all male,18~25 years old, mean 20.2±0.92 years) from a university flight academy were surveyed and underwent EHFA.All subjects had hearing thresholds ≤25 dB HL at conventional frequencies (0.25~8 kHz).The results were compared with the corresponding recommended standards in other countries.According to the use of personal listening devices, all the subjects were divided into the low risk group (non-use, 121 cases) and the high risk group (using>1 year, day>1 hour, 52 cases).The differences of hearing thresholds and detection rates at EHF between the two groups were compared.Results The hearing thresholds of 173 subjects (1 case of middle ear disease and 1 case with family history of hearing loss were excluded) at 9~20 kHz were slightly higher than the reference equivalent threshold sound pressure level (RETSPL) prescribed by American National Standardization Association and the age-matched thresholds recommended by a foreign literature (P<0.05 or P<0.01).The hearing thresholds in the high risk group elevated dramatically than that in the low risk group at 9,12.5,16 and 18 kHz(P<0.05 or P<0.01).As frequencies increased, the detection rate of hearing thresholds in the high risk group decreased gradually, and at 18 and 20 kHz it was significantly lower than that in the low risk group (P<0.01).Conclusion EHF audiometry is a helpful tool for early detection of noise-induced hearing loss in student pilots.Hearing health care should be emphasized in civilian student pilots.It is recommended to avoid the use of personal listening devices or reduce the use time of them.
10.Management of femoral artery pseudoaneurysm caused by heroin injection

Objective To study the management of femoral artery pseudoaneurysm caused by heroin (injection).Methods 11 cases of femoral artery pseudoaneurysm were treated. 4 cases presented with ruptured aneurysm and were treated emergently by direct ligation of the aneurysmal artery through a femoral incision. 2 cases (underwent) elective operation in which the iliac artery was initially exposed through an extraperitoneal (lower) (abdominal) incision to control bleeding, and then the aneurysmal artery was ligated through a femoral (incision).Results All of the operations were successful, and no gangrene of the affected limb occurred.(Conclusions) Arterial ligation for femoral artery pseudoaneurysm caused by heroin injection is simple and (effective). (Intraoperative) observation of distal arterial backflow,measurement of (arterial) pressure and (intraoperative) (arteriography) can improve the safety of the operation.
