1.Mechanism of HAG priming protocols inducing apoptosis of leukemia cell lines in vitro

Objective To investigate the effect and the mechanism of different G-CSF-priming protocols on leukemia cell lines (HL-60 and U937) in vitro and provide the clinical guidance to clinical treatment of acute leukemia.Methods The leukemia cell lines HL-60 and U937 were used as model to detect the effects of three drugs alone and combined two drugs (HA) or three drugs (HAG) respectively.Cell viability and cell growth inhibition were performed by cell count kit-8 (CCK-8) assay.Apoptotic marker AnnexinV/PI,cell membrane surface antigen CD11b,cell cycle,mitochondrial membrane potential (JC-1) and Caspase-3 were determined by flow cytometry.Results After using of HAG for 48 h,HL-60 and U937 cells counts were decreased significantly and the apoptotic marker Annexin V was significantly increased. To compare the single drug group with two drug combination group,the result was significantly different (P <0.05),and the apoptosis of U937 cells was higher than HL-60 cell line.CD11b expression among the three groups did not change (P > 0.05).Using of CAG and MAG,the mitochondrial nembrane potential of HL-60 and U937cells was increased,the three-drug combination group was significantly higher than single-drug group and control group (P <0.05); Caspase-3 was activated,the fluorescence intensities of Caspase-3 of the three-drug combination group and single drug group were significantly higher (P <0.05) comparing with the control group.Conclusion HAG regimen could induce leukemia cells to apoptosis through the reduction of mitochondrial membrane potential and the activation of Caspase-3 to induce apoptosis of leukemia cells.
2.Effect of finasteride on reducing hemorrhage in holmium lasterenucleation of prostate

Objective To evaluate the effect of preoperative oral finasteride on patients with benign prostate hyperplasia (BPH) undergoing holmium lasterenucleation of prostate (HoLEP).Methods A total of 156 BPH patients from Department of Urology in Shanghai Ninth People's Hospital from Sep.2010 to Mar.2012 was analyzed retrospectively.79 patients receiving oral 5 mg/d finasteride before operation were selected as medication group,and the 77 patients without taking finasteride were selected as control group.The perioperative data,including operation time,amount of washing fluid during operation,preoperative and postoperative changes of hemoglobin level and postoperative bladder washing time were compared between two groups.Results Compared with control group,the changes of hemoglobin level after HoLEP,amount of washing fluid during operation,and postoperative bladder washing time with normal saline were significantly decreased in experimental group[(1.08±0.27) g/L vs.(1.55±0.32) g/L,(27.51±3.67) L vs.(36.89±6.47) L,(24.85±4.17) h vs.(35.87±5.10) h,all P<0.05].Conclusions Oral finasteride before HoLEP can reduce perioperative bleeding and the volume of bladder irrigation with normal saline.

4.Dynamic spore outputs and antibiotic activities of asexual fungi (strain CY-8202) isolated from Cordyceps sinensis

Object To study the growth speed and dynamic spore production of CY-8202, which is an asexual strain of entomopathogenic fungi isloated from Cordyceps sinensis (Berk.) Sacc., to explore the method of artificial culture of CY-8208 strain, to assay its antibiotic activity and spectrem and to provide the experimental basis for studies of its active components. Methods Variations in colony diameter of the cordyceps hypha cultured on the fungi media were measured. The spore-count was used to determine the dynamic colony spore outputs of the hypha on several fungi media and water agar. The agar-piece method was used to test its antibiotic activity.Results There were linear relationships between the colony extensions and the culture times on common fungi media such as PDA medium, etc.. The amounts of spore produced by the single colony of the fungi were more than 10 7 and gradually increased, but the rates of increase tended to be gentle after 200 h. Antimicrobial tests against 22 microbial strains showed strong inhibition of gram positve bacteria including Bacillus subtilis, Micrococcus tetragenus and Staphylococcus albus, and to gram negative bacteria, including Proteus vulgaris, Salmonella typhi, Aerobacter aerogenes and Salmonella sp., as well as weak inhibition of three mold strains and one actinomycetes strain, but no inhibition was observed in four yeast tested.Conclusion The growth activity and the spore-production ability of Cordyceps hypha are two important factors to infect validly its insect-host. For the growth activity, its strong penetration seems more important than its fast growth. The ability to product numerous spores of Cordyceps hypha may be an important mark of its strong infectivity to insect-host. The antimicrobial tests show that CY-8202 may secrete some metabolites which have a more broad-spectrum antibacterial activity than cordycepin isolated initially from Cordyceps militaris.

BACKGROUND:Previous research has proved good therapeutic effect on the acute hepatic failure by the treatment of polylactic acid-O-carboxymethylated chitosan(PLA-O-CMC) nanoparticles-cultured porcine hepatocytes embedded in type I collagen.OBJECTIVE:To investigate the effect of PLA-O-CMC nanoparticles on immunological rejection in intraperitoneal porcine hepatocyte xenotransplantation for treatment of acute liver failure rats.DESIGN,TIME AND SETTING:Randomized control animal experiments were carried out at the Institute of Hepatobiliary Surgery and Laboratory of Nerve Regeneration in Nantong University from May 2005 to May 2006.MATERIALS:In situ recirculating collagenase perfusion method was used to isolate porcine hepatocytes.64 SD rats were intraperitoneally injected with D-galactosamine to induce acute hepatic failure models.METHODS:Totally 64 rats were divided into four groups:Model group received no intervention;Nano-collagen hepatocytes group was transplanted with PLA-O-CMC nanoparticles-cultured porcine hepatocytes embedded in collagen gel,which was then wrapped up using greater omentum;Porcine hepatocytes embedded in collagen gel(collagen hepatocytes group) and free porcine hepatocytes suspension(pure hepatocytes group) were transplanted into peritoneal cavity of SD rats,respectively.At 48 hours following the modeling,the porcine hepatocytes transplantations were performed and the number of transplanted cells was 5.0?107 hepatocytes in all groups.MAIN OUTCOME MEASURES:The serum interleukin-2,interferon-?,IgG and IgM levels of rats were determined at days 1,2,3,5 and 7.The pathological changes of transplants were observed under microscope at days 1,3 and 7.RESULTS:There were no significant differences in serum interleukin-2 and interferon-? levels of all groups during 7 days post-transplantation.Serum IgG levels increased with a peak at day 3 after hepatocytes transplantation,and then began to decline.Serum IgG levels in nano-collagen hepatocytes group were lower than those in collagen hepatocytes group and pure hepatocytes group at days 2 and 3(P

Iatrogenic gastrointestinal perforation is one of the severe adverse events of endoscopic therapeutic procedure. For acute iatrogenic perforation,management by endoscopic techniques is a simple and rapid modality to close the perforation with minimal invasiveness and avoiding the traditional surgical trauma. Endoclips,suture with special instruments,covered stents,degradable sheets combined with tissue adhesive,and combined endoscopic techniques such as snares combined with endoclips,are the major endoscopic therapeutic modalities for closure of iatrogenic gastrointestinal perforation. In this article,the current status and progress of endoscopic management for acute iatrogenic gastrointestinal perforation were reviewed.
10.Analysis of prognosis of collapsing focal segmental glomerulosclerosis in children

Objective To analyze the long-term prognosis and prognostic factors of idiopathic collapsing focal segmental glomerulosclerosis (FSGS) and not otherwise specified FSGS in children. Methods The clinical, pathology and follow-up data of patients with idiopathic collapsing FSGS and not otherwise specified FSGS were analyzed retrospectively by Kaplan-Meier method, univariate and multivariate Cox regression analysis. Results A total of 64 patients (29 idiopathic collapsing FSGS and 35 not otherwise specified FSGS) were diagnosed by renal biopsy. The 4-year renal survival rate of idiopathic collapsing FSGS and not otherwise specified FSGS were 48.3%, 74.3% respectively. Univariate analysis revealed that the renal survival time were 25.41±3.28 months in idiopathic collapsing patients, and 35.53±2.73 months in not otherwise specified patients. The different is significant (χ2=4.07,P=0.044). Multivariate Cox regression analysis showed that poor treatment response (HR=5.92, P<0.05) and renal insufficiency at early stage (HR=2.45, P<0.05) were independent risk factors of prognosis. Conclusions Compared with patients with not otherwise specified FSGS, the renal survival time is shorter in idiopathic collapsing FSGS patients. Patients with renal insufficiency and poor response to treatment have poorer prognosis.
