1.Protective Effect of Mouse Nerve Growth Factor on Amikacin -Induced Ototoxicity

Objective To investigate the effects of mouse nerve growth factor on amikacin (AK)-induced ototoxicity in guinea pigs .Methods 45 guinea pigs were randomly divided into three groups with 15 guinea pigs in each group .The control group received no drug treatment ,the poisoning group received an intraperitoneal injection of AK (400 mg · kg -1 · d-1 ) for 10 days ,and the treatment group received an intraperitoneal injection of AK (400 mg · kg -1 · d-1 ) for 10 days and an intramuscular injection of NGF (1 500 AU · kg -1 · d-1 ) for 14 consecutive days .Auditory brainstem responses were tested on the day before administration ,the 10th day and 20th day after administration .All the guinea pigs were sacrificed when the last ABR test was finished on the 20th day whose Corti organs were observed as well .Results The ABR thresholds of each group showed no significant difference before administration .The ABR threshold of the treatment group was lower than that of the poisoning group .With light microscopy ,the hair cells with normal morphology were observed in the control group .However ,there was an ex-tensive loss of outer hair cell in the poisoning group .For the treatment group ,the miss rate of hair cell was lower compared with the poisoning group .Conclusion Our data shows that NGF has played a protective role on amikacin-induced ototoxicity .
2.Expression and significance of Notch1 and Bmi-1 protein in lung cancer tissue

BACKGROUND:Studies have found that Notch pathway and Bmi-1 gene both have the ability to regulate stem cel self-renew. Functional dysfunction of the both may have a great relationship with tumorigenesis. OBJECTIVE:To investigate the expression and clinical significance of Notch1 and Bmi-1 protein in lung tissue. METHODS:Eighty-seven lung cancer tissue samples (lung cancer group) and forty pathologicaly confirmed normal lung tissue samples (normal group) were obtained from related surgeries and included as research objects. The protein expression of Notch1 and Bmi-1 in specimens of these two groups was measured by immunohistochemistry SP method. The relationship between Notch1 and Bmi-1 protein expression and clinicopathological features of lung cancer patients was analyzed. RESULTS AND CONCLUSION: The positive rate of Notch1, Bmi-1 protein expression was respectively 61% and 47%, which was significantly higher in the lung cancer group than that in the normal group (P < 0.05). In the lung cancer group, Notch1 protein expression was significantly positively correlated with Bmi-1 protein expression (r=0.567,P < 0.001). There were no significant differences in Notch1 and Bmi-1 protein expression rates between different genders and different pathological types of patients (P < 0.05). The Notch1 and Bmi-1 protein positive expression rates in poorly-differentiated, TNM stage III-IV lung cancer patients with lymph node metastasis were significantly higher than those in wel- and moderately-differentiated, TNM stage I-II lung cancer patients without lymph node metastasis (P < 0.05). These results demonstrate that Notch1 and Bmi-1 protein may have certain relationship with the occurrence and development of lung cancer.
3.Effect of periodontal basic treatment combined with coenzyme Q10 on cytokines in serum and gingival crevicular fluid of periodontitis with coronary heart disease

Objective To investigate the effect of periodontal basic treatment combined with coenzyme Q10 on cytokines in serum and gingival crevicular fluid of periodontitis with coronary heart disease.Methods A total of 68 patients with periodontitis with coronary heart disease were collected and randomly divided into 2 groups with 34 cases in each group.Patients in the control group were treated by periodontal basic treatment, patients in the experimental group were treated on the basis of the control group with coenzyme Q10 injection, continuous treatment for 2 weeks, the inflammatory factors levels in serum and gingival crevicular fluid were detected and the periodontal indexes and clinical efficacy were observed.Results Compared with before treatment, the TNF-α, IL-1β, IL-6 and IL-8 levels of serum and gingival crevicular fluid were decreased (P<0.05), the plaque index (PLI), sulcus bleeding index (SBI), periodontal depth (PD) and clinical attachment loss (CAL) were decreased (P<0.05);compared with the control group, the TNF-α, IL-1β, IL-6 and IL-8 levels in serum and gingival crevicular fluid of the experimental group were lower ( P<0.05 ) , PLI, SBI and PD periodontal indexes were lower (P<0.05), the clinical curative effect was higher (P<0.05).Conclusion Coenzyme Q10 injection in the treatment of patients of periodontitis with coronary heart disease could significantly improve the periodontal inflammation, and reduce the IL-6 and TNF-αlevels in serum and gingival crevicular fluid.

5.Analysis of the Results of Drug Sampling and Testing in Hubei Province from 2013 to 2015

OBJECTIVE:To evaluate the quality of sampling drugs of Hubei province,and to provide reference for drug enter-prises and supervision department. METHODS:The data obtained by Hubei Institute for Food and Drug Control about provincial drug sampling and testing from 2013 to 2015 were summarized,the overall quality and unqualified items were statistically analyzed according to current standard. RESULTS & CONCLUSIONS:The number of total sampling batches was 7721 and that of the un-qualified batches was 333 (4.3%) during 2013-2015. Most of the unqualified samples came from distribution units (2.8%-9.0%) and medical institutions (5.2% -11.7%). Among all sample types, the herbal medicine has the highest unqualified rate (12.1%-37.4%),with the largest number of unqualified items(content determination,supplement inspection item,property,resid-ual amount of sulfur dioxide,ash content). The quality of pharmaceutical preparations is relatively stable,and the unqualified items correlated with drug types and dosage forms. The current issues of provincial drug sampling and testing are the variety of standard levels,unreasonable sampling resource allocation and the low level of the industry. Supervision department is proposed to readjust drug standard,reasonably plan sampling and testing process and strengthen production source controlling.
6.The mechanism of glutamine against liver injury

Glutamine(Gln) is the most abundant free amino acid in the circulation. However, Gln depletion is obviously increased in critically ill patients. The liver plays a key role in glutamine metabolism since it can either generate or consume Gln. A number of studies have demonstrated that administration of Gln can protect from liver damage during sepsis or chemotherapy. The mechanism may be that glutamine can be more efficiently transported across cell membranes for glutathione synthesis. Supplement of Gln to PN has been proven to maintain intestinal integrity and reduce bacterial translocation. As a gluconeogenic amino acid, Gln can decrease the portal insulin/glucagon ratio and prevent hepatic steatosis through stimulating glucagon secretion.
7.Chondrogenesis of adipose tissue-derived stem cells induced by auricular chondrocytes from microtia in vivo

BACKGROUND:Due to quantity and quality deficiencies, chondrocytes from microtia are difficult to act as seed cells to construct an ear cartilage scaffold with the normal human auricle size. OBJECTIVE: To test the hypothesis that auricular chondrocytes from microtia can promote chondrogenic differentiation and chondrogenesis of human adipose tissue-derived stem cells (ADSCs) at non-chondrogensis sitein vivo, which is the preparatory work for preparation of human tissue-engineered auricle cartilage scaffold. METHODS: Human ADSCs at passage 3 and auricular chondrocytes at passage 2 were mixed at a ratio of 7:3 and 5.0×1010/L mixed cells were suspended in 0.2 mL of 30% Pluronic F-127, and then the mixture was injected subcutaneously into Balb/c nude mice as experimental group. Auricular chondrocytes or ADSCs at the concentration of 5.0×1010/L were mixed with 0.2 mL of Pluronic F-127 and injected respectively as positive and negative control groups. 1.5×1010/L auricular chondrocytes were mixed and injected as low-concentration chondrocyte control group. All specimens were collected at the 8th week post-injection. Newborn tissues in nude mice were taken out for morphological examination, wet weight measurement, determination of glycosaminoglycans, histological and immunohistochemical staining. RESULTS AND CONCLUSION:The wet weight of specimens in the experimental group was over 80% of that in the positive control group, and the wet weight of specimens in the low-concentration chondrocyte control group was less than 30% of that in the positive control group. The average wet weight and glycosaminoglycan content were significantly higher in the experimental and positive control group than in the negative control and low-concentration chondrocyte control groups (P < 0.05). In all the groups except for the negative control group, mature cartilage lacunas could be observed by histological staining and collagen type Ⅱ could be detected for expression by immunohistochemistry to different extents. In the low concentration chondrocyte control group, cartilage lacunas were incompact and inhomogeneous, and the extracellular matrix was slightly stained. In the negative control group, mature cartilage lacunae and collagen type Ⅱ could not be detective. To conclude, auricular chondrocytes from microtia can promote chondrogenic differentiation and chondrogenesis of ADSCs at the non-chondrogenesis sitein vivo.
8.Study on the ability of specific-binding peptide modified liposome loaded paclitaxel targeting A549 lung cancer stem cell

Objective To prepare CD133 specific-binding peptide conjugated liposome loaded paclitaxel and evaluate the efficiency of cellular uptake and the ability of inhibiting A549 lung cancer stem cell.Methods Liposomes were prepared by film-ultrasonic method.The partical size,zeta-potential and entrapment efficiency of liposomes were evaluated.Cellular uptake effciency of A549 lung cancer stem cell for liposomes were explored.The anti-proliferation efficiency of TLP-PTX to A549 lung cancer stem cell was evaluated by MTT assay.Tumor spheroids were used to evaluate anti-tumor ability of TLP-PTX to A549 lung cancer stem cell. Results The particle diameter of TLP-PTX was (115.8 ±8.3)nm and the entrapment efficiency of PTX was 88.5%.CD133 specific-binding peptide could enhance the efficiency of cellar uptake.The uptaken efficiency of TLP by A549 lung cancer stem cell were 2.6 times higher than that of LP(P<0.05 ).The MTT Results showed that the toxicity of TLP-PTX on A549 lung cancer stem cell was significantly stronger than LP-PTX and paclitaxel solution(P<0.05 ).The tumor inhibition test results showed that TLP-PTX has good anti-tumor effect. Conclusion TLP-PTX can specifically recognize the surface marker CD133 of A549 lung cancer stem cell,facilitate liposomes into cells and inhibit A549 lung cancer stem cell proliferation.TLP-PTX is an effective drug delivery system targeting to A549 lung cancer stem cell.
9.A study on the treatment of depression and anxiety in elderly patients by Shu gan jie yu capsule combined with ascitalopram

Objective To study the treatment of elderly depression and anxiety in the treatment of depression and anxiety. Methods 76 cases of senile depression and anxiety co-morbidity were selected from our hospital from January 2016 to January 2017, and were randomly divided into control group and experimental group, with 38 cases in each group. The patients in the control group were treated with escitalopram, and the patients in the experimental group were treated with ascitalopram. The treatment time for both groups was two months. Comparative analysis was made on the clinical indicators of the experimental group and the control group. Results After treatment, the score of HAMD score and HAMA score in the experimental group was significantly lower than that in the control group, and the score of the experimental group was significantly lower than that in the control group, the differences were statistically significant (P< 0.05). The effective treatment efficiency of the patients in the experimental group was 86.84 % compared with the control group (73.68%), the differences were statistically significant (P< 0.05). There were 9 cases and 10 cases of adverse reactions between the experimental group and the control group, but there was no significant difference in the incidence of adverse reactions. Conclusion The liver capsule joint resolve depression escitalopram citalopram treatment of senile depression and anxiety comorbidities better clinical effect, can significantly improve the condition, improve the treatment efficiency, further promotion and application of clinical significance.
10.Magnesium deficiency and the occurrence and progression of type 2 diabetes mellitus
