1.From the features of JCEM to consider the future of CJEM

Journal of Clinical Endocrinology and Metabolism (JCEM), sponsored by U.S. Endocrine Society, is particularly appreciated by clinical endocrine doctors worldwide. It is intimately related to clinical practice while still catching up state of art research methods and recent discoveries and its creativity, novelty, scientific reliability, feasibility and readability are highly appraised. Compared to JCEM, our Chinese Journal of Endocrinology and Metabolism seems to be inadequate in certain aspects and needs to be improved in the future.
2.Content Determination of Ginsenoside Rg1, Ginsenoside Rb1 and Notoginsenoside R1 in Tianqi Tongjing Capsule by HPLC

Objective To establish an HPLC method for content determination of Ginsengnoside Rg1, Ginsengnoside Rb1 and Notoginsenoside R1 inTianqi Tongjing Capsule.Methods An Agilent ZORBAX SB-C18 column (4.6 mm × 250 mm, 5μm) was used. The mobile phase was composed of acetonitrile (A) and water (B) with gradient elution (0-20 min, 19%→21%A;20-50min, 21%→36%A) at a flow rate of 1.0 mg/mL;the wavelength of detector was 203 nm;the temperature of the column was 25℃.Results The calibration curves of Ginsengnoside Rg1, Ginsengnoside Rb1 and Notoginsenoside R1 showed good linearity within the range of 0.442 4-3.981 6μg (r2=1.000 0), 0.524 8-3.673 6μg (r2=0.999 4) and 0.203 2-1.016μg (r2=0.999 2), respectively. The average recoveries (n=9) were 99.49%, 99.02% and 99.98%, and RSD were 2.44%, 2.45% and 2.14%, respectively.Conclusion The method is simple, reliable, rapid and with good repeatability, and can effectively control the quality ofTianqi Tongjing Capsule.
3.COMPUTER ANALYSIS OF EEG ON OVERTRAINED ATHLETES AND ITS CLASSIFICATION BASED ON SPECTRAL DIFFERENCES

This paper focuses on investigating the spectral changes of EEG in overtrained athletes through self-normal controls and thus establishing a digital quantitative approach to recognize the EEGs of overtrained ones. In addition, the VEPs of those with overtraining were analysed.During a period of 11 months, EEGs of each of 20 athletes were recorded separately at their overtrained and normal states. A control group was formed of 19 normal athletes, each of whom two EEG exams were made too. The EEG was taken at rest, during periodic flash stimulation and after hyperventilation (HV). Paired t test was performed on the power spectra of EEGs and VEPs, and Fisher classifier were established by the differences of spectral features (DSF) between the two EEGs in each of almost all of the 30 cases.The relative powers of the EEGs in the overtrained cases showed a significant increase (P
4.Construction and expression of Runx1-shRNA expressiong plasmid.

This study aims to construct the plasmid of human Runx1 and observe its possible effects on Runx1 gene expression in human pulmonary adenocarcinoma cells (A549). The shRNA sequence targeting human Runx1 was designed and synthesized, then inserted into pSuper plasmid by DNA recombination technology. The recombinant plasmid was confirmed by bacterial colonies PCR, enzyme digestion analysis and DNA sequencing. A549 cells were transfected by Runx1 shRNA plasmid. The inhibition efficiency of pSuper-Runx1-shRNA plasmid on Runx1 at mRNA level and protein level were measured with real-time PCR and Western blot. The results of real-time PCR and Western blot indicated that the mRNA and protein levels of Runx1 in A549 cells were inhibited by the pSuper-Runx1- shRNA expression plasmid, and the inhibition rate were 33% and 50%, respectively.

5.Practice of X-ray Examination Table Controlled by PLC

Objective By the modern manufacture regulation of original equipment manufacture(OEM),the quality of large scale X-ray equipment is ensured by means of assembling the purchased key components,such as high frequency and high voltage generators and X-TV systems,etc.Methods The technology of Programmable Logic Controller(PLC)was utilized to improve the reliability of X-ray examination table.Results The design of PLC-controlled X-ray examination table was used in the digital X-ray equipment yeas ago.The malfunction probability of mechanic parts was very low,whereas the electric circuit part was almost zero.Conclusion The practice of PLC improves the reliability and control level for X-ray equipment.
6.Digital Practice of Large Scale X-Ray Equipment

Objective To improve X-ray equipment for better medical treatment by digitally transform normal X-ray equipment.Methods Digital image workstation was equipped for digital photography so that ordinary X-ray equipment has the function of a digital X-ray equipment.Results The digital transformation enhance the clinical performance of X-ray equipment.Conclusion It has been proved that some ordinary X-ray equipment can be equipped with digital workstation.
7.Relationship between exon deletion frequency of CDKN2/P16 and pathological type,metastasis,sex in osteosarcoma

Objective To study on the CDKN2/P16 gene in primary osteosarcoma.Method By using molecular biological methods that inclued genome DNA extraction from paraffined tissue and PCR SSCP analysis technique, we studied alternations of CDKN2/P16 gene in 25 primary osteosarcomas.Results (1)The deletions frequency in differentiation degree of osteosarcomas was① bone brood cell, 16.7% ;② cartilage brood cell,12.5% ;③ Fiber brood cell:20% ,(P >0.05).(2)The deletion frequency in male patients was 17.6% , female patients 12.5% ,(P >0.05).(3)In early metastatic osteosarcomas the deletion rate was 33.3% ,which was significantly higher than that of the control group with the rate of 10.5% (P< 0.05).(4)The deletion rate was 16% and the mutations were not found.Conclusion (1)The deletion rate was 16% and the mutations were not found.This suggests that the deletions of CDKN2/P16 gene were closely related to the genesis of primary osteosarcoma and that the main type of the alternation of CDKN2/P16 gene was deletion.(2)In early metastatic osteosaarcomas the deletion rate was 33.3% , which was significantly higher than that of the control group with the rate of 10.5% .This indicates to great extend that the deletions of CDKN2/P16 gene were closely related to the metastatic ability.(3)The deletions frequency had no significant relationship with differentiation degree of osteosarcomas, so was with the sex of the patient.
8.The enantioselective pharmacokinetic study of desvenlafaxine sustained release tablet in Chinese healthy male volunteers after oral administration.

A chiral LC-MS/MS method for the simultaneous analysis of desvenlafaxine (DVS) enantiomers in human plasma was developed and applied to a pharmacokinetic study on 12 Chinese healthy volunteers. d6-Desvenlafaxine was used as internal standard (IS). Chromatographic separation was performed on the Astec Chirobiotic V chiral column (150 mm x 4.6 mm, 5 μm). The assay was linear over the concentration range of 0.500-150 ng x mL(-1) for both enantiomers (r2 > 0.99). The method was successfully applied to a stereoselective pharmacokinetic study of 100 mg desvenlafaxine sustained release tablets on 12 Chinese healthy volunteers under fasting conditions. The results showed that the pharmacokinetic parameters were similar to both enantiomers in Chinese healthy volunteers. The AUC(0-t), and C(max) of the two enantiomers were about 1.5 times higher than those of blacks and whites reported in the literature.
9.Simultaneous determination of erdosteine and its active metabolite in human plasma by liquid chromatography-tandem mass spectrometry with pre-column derivatization.

A sensitive, rapid and accurate liquid chromatography-tandem mass spectrometric (LC-MS/MS) method with pre-column derivatization was developed for the simultaneous determination of erdosteine and its thiol-containing active metabolite in human plasma. Paracetamol and captopril were chosen as the internal standard of erdosteine and its active metabolite, respectively. Aliquots of 100 microL plasma sample were derivatized by 2-bromine-3'-methoxy acetophenone, then separated on an Agilent XDB-C18 (50 mm x 4.6 mm ID, 1.8 microm) column using 0.1% formic acid methanol--0.1% formic acid 5 mmol x L(-1) ammonium acetate as mobile phase, in a gradient mode. Detection of erdosteine and its active metabolite were achieved by ESI MS/MS in the positive ion mode. The linear calibration curves for erdosteine and its active metabolite were obtained in the concentration ranges of 5-3 000 ng x mL(-1) and 5-10 000 ng x mL(-1), respectively. The lower limit of quantification of erdosteine and its active metabolite were both 5.00 ng x mL(-1). The pharmacokinetic results of erdosteine and its thiol-containing active metabolite showed that the area under curve (AUC) of the thiol-containing active metabolite was 6.2 times of that of erdosteine after a single oral dose of 600 mg erdosteine tables in 32 healthy volunteers, The mean residence time (MRT) of the thiol-containing active metabolite was (7.51 +/- 0.788) h, which provided a pharmacokinetic basis for the rational dosage regimen.
10.Serum Levels and Clinical Significance of MMP-9 and NGAL in Patients with Gastric Cancer

Background:Matrix metalloproteinase-9(MMP-9)is known to play a key role in tumor invasion and metastasis by mediating the degradation of extracellular matrix. It has been revealed that neutrophil gelatinase-associated lipocalin ( NGAL)is capable of protecting MMP-9 from degradation and thereby preserving its enzymatic activity. Aims:To explore the MMP-9 and NGAL levels in peripheral blood of gastric cancer patients and their clinical significance. Methods:Serum levels of MMP-9 and NGAL were determined by ELISA in 76 gastric cancer patients and 50 healthy controls. Correlations of serum MMP-9 and NGAL levels with clinicopathological features of gastric cancer were analyzed,and their diagnostic performance for screening gastric cancer was evaluated by ROC curve. Results:Serum levels of MMP-9 and NGAL in patients with gastric cancer were significantly higher than those in healthy controls(P<0. 01),and either of them was positively correlated with the TNM stage of gastric cancer. Furthermore,MMP-9 was correlated with lymph node metastasis and serosal invasion,while NGAL was correlated with distant metastasis. However,tumor differentiation was not correlated with either MMP-9 or NGAL. By ROC curve,608. 19 ng/mL and 14. 71 ng/mL were identified as the cutoff value of serum MMP-9 and NGAL,respectively,for screening gastric cancer. Sensitivity and specificity of MMP-9 were 64. 5% and 90. 0%,and those of NGAL were 85. 5% and 90. 0%,respectively. When parallel test was taken,the sensitivity would be increased to 94. 7% and specificity being 82. 0%. Conclusions:Serum levels of MMP-9 and NGAL are elevated and associated with tumor progression in patients with gastric cancer. Both of them might be used as potential markers for screening,disease assessment and prognosis prediction of gastric cancer.
