1.Role of EGF family in development of endocardial cushion

During early embryonic development,these epithelial cells of cardiac atrioventricular and outflow tract undergo an epithelial-mesenchymal transformation to become mesenchymal cells that invade cardiac jelly,forming endocardial cushion.Epidermal growth factor family plays an important role in regulating epithelial-mesenchymal transformation.The family is composed of multiple ligands and four tyrosine kinase receptors ( ErbB),among which ErbB2 and ErbB3 are necessary for normal development of endocardial cushion.In addition,other receptors and ligands also play important roles in heart development.This review discusses the role of receptors and ligands of the EGF family in heart endocardial cushion development.
2.Transcatheter hepatic arterial thermo-chemotherapy and thermo-lipiodol embolization for the treatment of hepatic metastases from colorectal carcinoma

Objective To evaluate the clinical efficacy of transcatheter hepatic arterial thermo-chemotherapy and thermo-lipiodol embolization in the treatment of hepatic metastases from colorectal carcinoma.Methods Sixty-eight cases with hepatic metastases from colorectal carcinoma were equally and randomly divided into two groups.The patients in study group were treated with transcatheter hepatic arterial thermo-chemotherapy and thermo-lipiodol embolization,while the patients in control group were treated with conventional(normal temperature) transcatheter hepatic arterial chemotherapy lipiodol embolization.Results The effective rate of study group and control group was 65%(22 / 34) and 32%(11 / 34) respectively,the difference between two groups was statistically significant(P

4.Influence of hypoxia-inducible factor-1 alpha on the susceptibility of oral squamous cell carcinomas cell lines against chemotherapeutic drugs and radiation

BACKGROUND: The transcription factor, hypoxia-inducible factor-1al pha (HIF-1α), is the key regulator that controls the hypoxic response of mammalian cells. However, the role of HIF-1α in the therapeutic efficacy of chemo-radiotherapy in oral squamous cell carcinomas (OSC) is poorly understood. OBJECTIVE: To investigate the effect of HIF-α on the susceptibility of OSC cell lines against chemotherapeutic drugs and radiation. DESIGN: AN observational comparative experiment. SETTING: Beijing Chaoyang Hospital Affiliated to Capital Medical University. MATERIALS: OSC-2, OSC-4, OSC-5, and OSC-6 cell lines were estab lished from oral squamous cell carcinoma (The cell lines were from the De partment of Oral Oncology, Kochi Medical School, Japan). METHODS: The experiments were completed in Beijing Chaoyang Hospi tal affiliated to Capital Medical University from September 2004 to August 2006. The cells were cultured in Dulbecco's modified Eagle's medium (DMEM) containing fetal bovine serum (FBS), L-glutamine, penicillin and streptomycin. ① Total cell lysates were extracted from untreated OSC cells and those treated with 30 Gy γ-rays, 100 μmol/L cis-dichlorodiamine plat inum (CDDP), or 100 μmol/L 5-fluorouracil (5-FU). The expressions of HIF-1α protein and mRNA were determined with western blotting analysis and real-time polymerase chain reaction (PCR). ② Inhibition of cell prolif eration after different interventions: OSC cells were seeded in 96-well mi croplates (1.0×104) and cultured for 48 hours after the irradiation with 30 Gy γ-rays or in the presence of 100 μmol/L CDDP or 100 mmol/L 5-FU, and then the inhibition of cell proliferation was detected with 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. ③ Apoptosis after different interventions: OSC cell lines were cultured for 24 hours after the irradiation with 30 Gy γ-rays or in the presence of 100 μmol/L CDDP or 100 μmol/L 5-FU. The cells were then stained with annexin V-FITC and propidium iodide, and the numbers of apoptotic cells were analyzed by a FACScan cytometer (only for the OSC cells treated with γ-rays and CDDP). ④ Plasmid construction and transfection: Human HIF 1α cDNA was cloned into the pcDNA3.1/V5-His TOPO expression vector. OSC-2 cells were temporarily transfected with HIF-1α cDNA. OSC-5 cells were temporarily transfected with HIF-1α siRNA (Sense sequence is 5' CUGAUGACCAGCAACUUGAtt 3'). The blank control group was also set, the inhibition of cell proliferation was observed, and the apoptosis was ana lyzed (only in the CDDP-treated group) by using the methods mentioned above. MAIN OUTCOME MEASURES: ① Expressions of HIF-1α protein and mRNA in different OSC cell lines; ② Inhibition of proliferation and the apoptosis of OSC cell lines after different interventions; ③ Overexpression and knockdown of HIF-1α by expression vector and siRNA; ④ Influence of overexpression and knockdown of HIF-1α on the susceptibility of OSC cell lines against γ-rays and chemotherapeutic drugs. RESULTS: ① Expressions of HIF-1α protein and mRNA in different OSC cell lines: The relative expression levels of HIF-1α mRNA were 1.0, 2.2, 4.3 and 4.0 in OSC-2, OSC-4, OSC-5 and OSC-6 cells, respectively. Simi larly, the expression of HIF-1α total cell lysates proteins and nuclear proteins were much lower in OSC-2 and OSC-4 cells than in OSC-5 and OSC6 cells. ② Inhibition of proliferation and the apoptosis in OSC cell lines after different interventions: After the treatment with g-rays, CDDP, and 5-FU, the cell numbers decreased obviously to (39.5±3.2)%, (39.2±1.2)%,and(47.9±3.6)% in OSC-2 cells, (53.9±6.6)%, (54.3±1.4)%, (54.8±3.8)%in OSC-4 cells. Those decreased to (74.1±3.8)%, (76.5±9.1)%, (69.6±7.7)%in OSC-5 cells and (71.4±7.4)%, (84.4±8.8)%, (82.0±4.5)% in OSC-6 cells.After the OSC cells were treated of with CDDP, the numbers of apoptotic cells were (50.9±1.3)%, (67.3±2.2)%, (12.2±0.8)% and (38.6±0.9)% in OSC-2, OSC-4, OSC-5 and OSC-6 cells. G-rays induced the apoptosis were (21.2±1.1)%, (14.6±0.9)%, (9.7±1.0)% and (10.4±0.8)% in OSC-2, OSC-4,OSC-5 and OSC-6 cells. ③ Inhibition of proliferation and the apoptosis in the transfected cell after different interventions: The cell numbers of HIF-1 α-transfected OSC-2 cells treated with γ-rays, CDDP and 5-FU were much more than those in the control group (t=-4.693 ,-8.617,-6.721, P < 0.01),whereas the cell numbers of OSC-5 cells transfected with siRNA were obviously fewer than those in the control group (t=5.800, 5.595, 4.253, P < 0.05-0.01). After the incubation of HIF-1α-transfected OSC-2 cells with CDDP for 24 hours, apoptosis was detected in (34.0±1.9)% of the cells, which was lower than that in the control group [(49.6±3.4)%, t=6.937, P < 0.01]. After the incubation of HIF-1α siRNA transfected OSC-5 cells with CDDP for 24hours, apoptosis was detected in (27.7±2.3)% of the HIF-1α siRNA transfected OSC-5 cells, which was obviously higher than that in the blank control group [(11.4±2.1)%, t=-8.941, P < 0.01].CONCLUSION: The expression level of HIF-1α correlates negatively with the susceptibility of OSC cells against chemotherapeutic drugs and radiation. The down-regulation of HIF-1α expression enhances the susceptibility of OSC cells against chemotherapeutic drugs and radiation. Therefore, the down-regulation of HIF-1α may be a potentially effective strategy for cancer treatment.
5.Effects of health interventions for patients with hyperuricemia on knowledge-attitude-performance in community

A total of 446 hyperuricemic patients were divided randomly by community-based cluster sampling into study (n =228) and comparison (n =218) groups.After 12-month health interventions,the Knowledge-Attitude-Performance (KAP) questionnaire [(19.0±4.7) points],lifestyle (balanced diet,modest physical exercises & regular monitoring) self-report questionnaire [attitudes prevention awareness:(8.3 ± 6.1) points,disease self-management behaviors:(13.2 ± 9.8) points],uricacid index [(395 ± 46)mmol/L] and body mass index (BMI) [(23.1 ± 2.3)kg/m2] were better than those at pre-intervention in study group [(9.3 ± 2.7),(4.6 ± 3.9),(7.4 ± 6.4)],[(420 ± 44)mmol/L,(24.9 ± 1.6) kg/m2] (P < 0.05).Also the inter-group difference had statistical significance (P < 0.05).It suggests that health intervention management is both feasible and effective in improving the KAP of patients,modulating lifestyles and lowering the levels of uric acid index and BMI.
6.Application of bionics urine bags in stroke patients with dysuria

Objective To explore the effect of bionic urine bags on dysuria in stroke patients and improve the effect of bladder function training. Methods Forty patients with urinary incontinence were selected and randomly divided into control group (n=46) and observation group (n=44) by random number table. The patients in the control group were treated with Kangwei anti-reflux urine bags and the urine was discharged every 3 hours at daytime, once every 4 hours at night. The patients in the experimental group were treated with OT-U bionic urine bags for drainage urine, the threshold of urine bag pressure set:when the bladder pressure reached more than 35cm H2O, the bladder urine flowed into the urine bag. The two groups were compared in view of catheter catheterization time, urination after urination time, self-urination, re-intubation rate, urine overflow, urinary tract infection. Results The duration of indwelling catheters in the observation group was significantly higher than that of the control group (P <0.05). The rates of re-intubation and urinary tract infection were significantly lower as well (P<0.05). Conclusion Biomimetic urine bag can effectively promote the training of bladder function, shorten the time of indwelling catheter and reduce the occurrence of urinary tract infection after extubation.
7.Observing the influences of liver function and virus outcome with telbivudine combined with adefovir dipivoxil

Objective To observe the clinical effects of anti-hepatitis B virus with telbivudine combined with adefovir dipivoxil.Methods 82 cases of patients diagnosed with chronic hepatitis B from January 2014 to February 2016, were randomly divided into two groups, the control group were treated with adefovir ester treatment, the observation group were treated with telbivudine combined with adefovir dipivoxil treatment, patients were followed up and recorded Child-Pugh score, aspartate aminotransferase ALT, total bilirubin TBIL, aspartate aminotransferase AST, albumin ALB, HBV-DNA negative rate, HbeAg seroconversion rate case, the use of statistical methods for data analysis.Results After treatment, the observation group Child-Pugh score, ALT values were(4.01±0.79)points,(37.19±4.82)U/L, were better than the control group(5.46±1.16)points,(61.49±9.78)U/L, and the differences were statistically significant (P<0.05).Observation group after treatment TBIL, AST, ALB, respectively(34.91±5.49)μmol/L,(56.49±5.28)U/L,(45.51±4.24)g/L, were better than the control group(61.62±10.06)μmol/L,(78.64±8.16)U/L,(38.76±3.21)g/L, and the differences were statistically significant (P<0.05).The observation group after treatment for HBV-DNA negative rate and HbeAg seroconversion rate(82.93%, 58.54%)respectively were higher(58.54%, 29.27%), and the difference was significant (P<0.05).Conclusion Telbivudine combined with adefovir dipivoxil anti-HBV effect is good, better than medication alone is worthy of further research and application.
8.Change of survivin gene expression in gastric cancer cell line BGC-823 induced by hydroxycamptothecine

Background and purpose:The antitumor drugs produce a marked effect through the induction of apoptosis. The resistance to chemotherapy has been found to be associated with abnormal expression of genes related to apoptosis. We designed this experiment to investigate the role of survivin gene in the resistance to chemotherapy. Methods:After being treated with Hydroxycamptothecine (OPT,21.96mmol/L), the apoptosis in gastric cancer cell line BGC-823 cells were evaluated by either flow cytometry(FCM) or the terminal deoxynucleotidy transfer mediated dUTP-biotin nick end labeling(TUNEL). The expression of Survivin was detected by reverse transcription-Polymerase chain reaction(RT-PCR) and immunohistochemistry.Results:Compared to control group, the OPT can induced apoptosis in BGC-823[(12.68?0.17)% vs (3.35?0.13)% by FCM,(16.00?1.23)% vs (2.78? 0.84)% by TUNEL[, OPT could upregulate survivin expression in BGC-823 cells (1.489?0.041)% vs (0.756?0.037)% in terms of level of mRNA,(0.928?0.046) vs (0.303?0.032) in terms of protein expression).Conclusions:The survivin gene may be associated to the resistance to chemotherapy through the induction of gene expression.
9.Study on the peri-puerperium outcome and associated factors of patients with primary Sj?grenˊs syndrome

Objective To analyze the peripartum complications,frequency of neonatal abnormalities and the associated factors in patients with primary Sj?grenˊs syndrome (SS). Methods The chart of 39 patients with primary SS who were admitted to the hospital for delivery were retrospectively reviewed. The clinical data, obstetrical outcome and the frequency of neonatal abnormalities as well as the possible associated factors were analyzed. Results There were 76 pregnancies and 41 deliveries among these 39 patients. Two patients (5%) had pregnancy-related hypertension, 1 (3%) had gestational diabetes and 1 (3%) had eclampsia. Twenty-eight(72%) patients had at least one episode of complication. In which, 27(68%) were induced abortion, 2 (5%) were premature birth, 6 (21%) were amniotic fluid volume abnormalities, 2 (5%) were post-partum bleeding, 5 (13%) were premature rupture of membrane, 6 (21%) were intra-uterine distress and 3 (8%) had intra-uterine growth retardation. Fetal abnormality was detected in 7(18%) patients, in which 3 fetus (8%) died before delivery, 2 fetus (5%) were small than gestational age and 2 (5%) had fetal deformity. Compared to the reports in the literature, the rate of abortion and fetal death was higher, but the rate of growth retardation , induced abortion was lower. Conclusion The frequency of peri-partum complication and fetal abnormality is increased in patients with primary SS. Pregnancies in patients with primary SS should beclosely monitored by rheumatologists.
