1.Cloning and Bioinformatics Analysis of Human Lung Fibrosis-inducing Factors

Chitinases are ubiquitous chitin-fragmenting hydrolases, however until a few years ago that chitinase like proteins have been found in mammalian. A silica-induced bronchoalveolar lavage protein (iSBLP58) with fibroblast growth promoting activity in silicotic rat, which had high sequence homology with members of the mammalian chitinase protein family has been previously purified and characterized. Bioinformatics analysis showed that several human EST clones from pooled colon, kidney or stomach matched the rat protein sequence. Thereafter clone from human kidney RNA samples with several pairs of primers was managed and a set of sequences was obtained, whose cDNA and amino acid sequences have high similarity with each other and several human chitinases in GenBank. Comparison with human genome sequence suggests that these molecules may be from variant transcripts of same a Pre-mRNA. Here the characterization of these sequences is reported.
2.Expressions of annexins and epidermal growth factor receptor in pediatric middle ear cholesteatoma

Objective To explore the expressions of annexin and epidermal growth factor receptor(EGFR) in pediatric middle ear cholesteatoma.Methods Twenty-three children with middle ear cholesteatoma and 26 children with normal skin of external auditory canal(control group) were selected from the children enrolled in the First Affiliated Hospital of Zhengzhou University from August 2014 to March 2016.The expressions of annexin A1 (AnxA1),AnxA2 and EGFR mRNA in cholesteatoma and normal tissues were analyzed by quantitative real-time PCR (qPCR).Protein expressions of AnxA1,AnxA2 and EGFR were evaluated by using Western blot and immunohistochemistry methods.Results The expressions of AnxA1,AnxA2 and EGFR mRNA were significantly increased in cholesteatoma compared with the control group (AnxA1:4.68 ± 1.77 vs.2.65 ± 0.96,U =111.5,P < 0.001;AnxA2:3.89 ± 1.00 vs.2.4 7 ± 0.81,U =84.5,P < 0.001;EG FR:4.97 ± 1.85 vs.3.50 ± 0.95,U =15 3.5,P =0.004).AnxA1 and AnxA2 mRNA expressions were positively correlated with EGFR mRNA (AnxA1 and EGFR:r2 =0.283 2,P =0.009;AnxA2 and EGFR:r2 =0.213 5,P =0.027).Compared with the control group,protein expressions of AnxA1,AnxA2 and EGFR were markedly enhanced (AnxA1:0.450 ±0.031 vs.0.320 ±0.026,U =102.4,P <0.001;AnxA2:0.568 ±0.024 vs.0.365 ±0.028,U =94.6,P <0.001;EGFR:0.397 ±0.021 vs.0.228 ±0.017,U =128.4,P <0.001).The results of immunohistochemistry analysis showed that the ratio of AnxA1,AnxA2 and EGFR positive cells were higher than those in the control group(AnxA1:65.22% vs.38.46%,x2 =9.296,P =0.026;AnxA2:69.57% vs.46.15%,x2 =8.378,P =0.039;EGFR:69.57% vs.50.00%,x2 =10.574,P =0.014).Conclusions The expressions of AnxAl,AnxA2 and EGFR are upregulated in pediatric cholesteatoma,with AnxA1 and AnxA2 expressions positively correlated with EGFR.
3.Clinical efficacy of massage combined with Astragalus and eucommia decoction in the treatment of periarthritis of shoulder

Objective To study and analyze the clinical efficacy of massage combined with Astragalus and eucommia Decoction in the treatment of periarthritis of shoulder.Methods100 patients with periarthritis of shoulder treated in our hospital from July 2015 to July 2016 were selected.They were randomly divided into the control group and the experimental group, with 50 patients in each group.Patients in the control group were treated with massage, and the experimental group was treated with massage combined with astragalus root and Eucommia ulmoides decoction.The 2 groups were treated with 2 or two courses, the 1 course of treatment for 15 days.The treatment effect and relative shoulder index score of the experimental group and the control group were compared and analyzed.ResultsAfter the corresponding treatment, the effective number of patients in the experimental group was 48 cases.The effective rate of treatment was 96%.The number of effective cases in the control group was 44, and the effective rate was 88%.The effective rate of the experimental group was significantly higher than that of the controlgroup, with statistical difference (P<0.05).After treatment, the shoulder function, activity and pain in the experimental group and the control group were significantly better than those before the treatment,The scores of shoulder functional performance index and shoulder pain index in the experimental group were significantly lower than those in the control group, with statistical difference (P<0.05).ConclusionMassage combined with Huangqi Eucommia Decoction in the treatment of periarthritis of shoulder can improve the therapeutic effect to a great extent, reduce the pain of patients with shoulder joint, to help with the recovery of shoulder joint function, with further clinical promotion and application significance.
4.Effect of MicroRNA-21 in Proliferation and Apoptosis in Pediatric Middle Ear Cholesteatoma Keratinocyts

Objective This study aims to investigate the post-transcriptional regulatory effects that control proliferation, apoptosis, and invasion in pediatriccholesteatoma keratinocytes.In particular, the potential role of miR-21was focused on in this study.Methods A total of 23 pediatric cholesteatoma tissues were processed for cell culture.Pediatriccholesteatoma keratinocytes were transfected with miR-21 inhibitors, or negative control miRNAs.RT-PCR was used to assess the expression levels of miR-21.EdU incorporation assay and TUNEL staining were used to assess the proliferation and apoptosis of pediatric cholesteatomakeratinocytes, respectively.Results MiRNA-21 was downregulated when pediatric cholesteatoma keratinocytes were transfected with miR-21 inhibitors.Furthermore, the number of proliferative EdU+cells decreased in cholesteatoma keratinocytes transfected with miR-21 inhibitors;and the number of TUNEL-positive cells also increased in cells transfected with miR-21 inhibitors, compared with cells transfected control miRNA.Conclusion MiRNA-21 promotes the proliferation and inhibits apoptosis of pediatric cholesteatoma keratinocytes.
5.Transabdominal ultrasonography in the diagnosis of achalasia Transabdominal ultrasonography in the diagnosis of achalasia

Objective: To assess the clinical application of transabdominal ultrasonography for the diagnosis of achalasia. Methods: The subjects were divided into 3 groups, with 8 patients suffered from achalasia as patient group, 10 normal individuals and 8 patients with cardiac carcinoma as controls. All subjects received water provocative test to study the lower esophagus and cardia. Sonographic findings were compared between the 3 groups. After treatment by administration of drugs, pneumatic balloon dilation, or intrasphincteric injection of botulinum toxin, the 8 patients were re-examined by TAUS for longitudinal comparison. Results: (1)The normal subjects had no dilation of cardia and lower esophagus before drinking water, but their cardias opened rapidly with the drinking action. (2)All the patients of achalasia were found to have dilation of lower esophagus before taking water. During 10 min continuous TAUS scanning, 5 of 8 demonstrated absence of relaxation of cardias and absence of water from the esophagus into the stomach. The other 3 showed slight and intermittent relaxation of cardias with water flow into the stomach. Thirty minutes after water provocative test, all the 8 patients had water retardation within the lower esophagus. (3)The patients of cardiac carcinoma had an ultrasound feature of cardiac ring enlargement and thickening of cardiac wall without dilation of lower esophagus before water provocative test. Water passed the cardia more slowly. Conclusion: TAUS is a readily accepted examination. It can quickly give reliable diagnostic clues to achalasia. TAUS can also be used for making assessment of therapeutic effect on achalasia.
6.Establishment of the UPLC Fingerprint and Analysis of Principal Component of Qinghou Liyan Granule

OBJECTIVE:To establish the UPLC fingerprint for Qinghou liyan granule,and provide reference for its quality control by combining with principal component analysis (PCA). METHODS:UPLC was performed on the column of ACQUITY UPLC BEH C18 with mobile phase of acetonitrile(A)-0.1% aqueous phosphoric acid solution(B)(gradient elution)at a flow rate of 0.4 mL/min,the detection wavelength was 280 nm,column temperature was 30℃,and injection volume was 2μL. Using ellag-ic acid as a reference,12 batches of samples were analyzed,Similarity Evaluation Software for Chromatographic Fingerprint of Traditional Chinese Medicinewas used for the similarity analysis and identification of the common peaks,and the PCA was used for common peaks. RESULTS:There were 18 common peaks in the fingerprints of 12 batches of samples,and 6 principal peaks (gallic acid,ellagic acid,naringin,hesperidin,neohesperidin and baicalin) were identified;the similarity degree of 12 batches and reference fingerprints were no less than 0.984. According to PCA,the 18 peaks can be integrated into 3 principal components, with cumulative contribution rate of principal component of 78.277%;baicalin and 16 peaks were the discriminating factors of the fingerprint of Qinghou liyan granule. CONCLUSIONS:The method can provide reference for the quality control of Qinghou liyangranule.
7.Comparison of heparin binding protein and procalcitonin in the evaluation of prognosis of patients with sepsis

Objective To compare heparin binding protein (HBP)and procalcitonin(PCT)in the evalua-tion of in prognosis of patients with sepsis in ICU.Methods 80 sepsis patients were selected.According to the sever-ity of the disease,they were divided into general sepsis group (n =31),severe sepsis group (n =29)and death group (n =20).The differences of HBP and PCT in three groups were compared.Results Compared with HBP and PCT in the general sepsis group[(3.91 ±0.36)ng/mL,(1.26 ±0.35)ng/mL)],PCT of the severe sepsis group (11.81 ± 0.54)ng/mL was significantly increased(F =2 777.87,P =0.000);HBP and PCT of the death group[(15.28 ± 1.21 )ng/mL,(36.68 ±0.36)ng/mL)]were significantly increased compared with the general sepsis and severe sep-sis groups (F =49 789.02,P =0.000).Conclusion The sensitivity of PCT was significantly higher than HBP in the groups,PCT was better in judging the prognosis of sepsis.
8.Cytomorphological analyses of malignant lymphomas in serous effusions

Objective To study the effect of cytomorphological analyses of malignant lymphomas in serous effusions .Methods Aspirating serous effusion were analyzed immediately ,then centrifugated ,stained with Wright-Giemsa ,and observed under micro-scope .The cytomorphological characteristics of lymphomas in serous effusions was analysed .Results 62 cases of malignant lym-phomas were found in 1 440 cases of the serous effusion .The diagnosis efficiencies of lymphomas were 89 .55% for sensitivity , 99 .85% for specificity ,96 .77% for positive expectant value ,99 .49% for negative expectant value and 99 .38% for total effective rate .Lymphoma cells were watched more in pleural effusion(46 cases) than in ascites(18 cases) and pericardial effusion(3 cases) . Cytomorphology of lymphomas included vacuoles ,large cells ,small cells and irregular cells .Conclusion The cytomorphological analysis of malignant lymphomas in serous effusion has an instructive significance to the clinical auxiliary diagnosis .

10.Hospital Infection among Psychiatric Inpatients in State of Protective Constraint

OBJECTIVE To explore the affection of protective constraint on infection among psychiatric inpatients and it′s preventive methods.METHODS A retrospective study of infection among psychiatric patients was carried by medical records investigation.Inpatients in states of protective constraint and non-constraint in confining units of psychiatric department were divided into study group and control group.Case-control study was carried up on rate of hospital infection and rate of infection of parts of body between groups.RESULTS The infective rate of study group was significantly higher than control one,in which,infective rates of lower respiratory tract,urinary tract,dermatological and mucous parts of study group were significantly higher than control group.CONCLUSIONS Protective constraint for psychiatric patients is one of risk factors of hospital infection,to which should be pay attention,and preventive methods might be taken.
