1.Impact of tetramethylpyrazine on experimental colitis of mice by cDNA microarray

Objective To study the impact of tetramethylpyrazine on genes expression in colitis mice by means of cDNA microarray.Methods Thirty mice were divided into normal control group,0.9% NaCI solusion group and tetramethylpyrazine group with 10 in each group.Except the mice in normal group,all mice were treated with oxazolone enema,mRNA was extracted from colons of 0.9% NaCI solusion group and tetramethylpyrazine group.The eDNA probes were prepared and labeled with dyes Cy3 or Cy5.Then the fluorescent probe was hybridized with eDNA microarray and scanned for fluorescent intensity.The differently expressed genes were identified by analysis of gene expression profile.Two differently expressed genes (interleukin-4 and -10) were further validated by fluorogenic quantitative RT-PCR.Results There were 432 (2.86%) differently expressed genes in tetramethylpyrazine group.Of which,307 genes were up-regulated and 125 genes were down-regulated.The function of some genes were well known.Conclusions Tetramethylpyrazine,which can depress inflammatory reaction,anti-adhesive molecule and improve immune systems,is a potential therapy for ulcerative colitis although more detailed curing targes of such medicine need to be further study.
2.Effect of continuous passive motion on the healing of experimental full thickness defects of articular cartilage in knee of rabbit

Objective To study the effect of continuous passive motion (CPM) on the healing of man made full thickness defects of articular cartilage in the knee of rabbit. Methods A diameter of 3.2mm full thickness defect of articular cartilage in the facies patellaris of femur were made in 60 knees of rabbits. Then the rabbits were randomly divided into groups A,B and C. The animals in groups A and B were treated with CPM for 8 hours and 2 hours daily, respectively, for 4 weeks, while those in group C kept in cage for free activity only. Half of the animals of every group were sacrificed at the end of the 4th and 12th weeks. The H.E. staining, safranin O staining and immunohistochemical analysis of collagens I and II were conducted, the gross appearance and histological change were observed and scored. Results There was significant difference with regard to the score of gross appearance and histological change among groups A, B and C. Conclusion The healing of full thickness defects of articular cartilage in rabbits treated by CPM is better than that in rabbits without CPM treatment. CPM can promote tissue repair and stimulate synthesis of collagenⅡand the glycosaminoglycan.
3.A preliminary study on the relationship between duodenogastric reflux and Helicobacter pylori infection

Objective To study the relationship between contents of duodenogastric reflux and Helicobacter pylori (H.pylori) infection in patients with peptic ulcer. Methods Seventy patients with peptic ulcer diagnosed by endoscopy entered the study. Contents of duodenogastric reflux was detected by 99M TC- EHIDA and H.pylori status was determined by both histology with Giemsa staining and serum anti-H.pylori-IgG level using ELISA method. 4.Effect of achalasia with temporary metal internal stent dilation on esophageal motility

Objective To explore the effect of achalasia with temporary metal internal stent dilation on esophageal motility.Methods Twenty nine patients with achalasia were undertaken temporary metal internal stent placement under X ray control (stents were of 20 25 mm in diameters). After stent dilation 3 7 days, the stents were removed by endoscopy. We measured the lower esophageal sphincter (LES) residual pressure and carried out 24h pH value monitor on the patients, and only LES pressure with relaxation rate for 12 healthy subjects (HS). Results LES residual pressure before stent dilation were markedly higher than that after stent dilation ( P 0.05).Conclusions Temporary metal internal stent dilation shows LES pressure and higher rate of GER for achalasia
5.Synergy of taxol combined with irradiation on apoptosis induction in human nasopharyngeal carcinoma cell line

Objective To study the synergistic effect of taxol and irradiation on programmed cell death induction in the human nasopharyngeal carcinoma cell line (CNE) in vitro.Methods Cell viability was evaluated by clonogenic assay. Cell cycle perturbation was assessed by flow cytometric analysis and apoptosis was determined with TUNEL.Results The intensity of cytotoxicity depended on the taxol dose used to CNE cells during the first 24 hrs. A G 2+M block was present with exposure to the drug of 100 nmol/L. When CNE cells were irradiated after 24 hrs of taxol exposure, synergistic effect was demonstrated with a taxol SER of 1.23. The same level apoptosis was induced by 10 ?mol/L taxol for 24 hrs or 2 Gy X-ray radiation in CNE cells.By both,the increase of induced apoptotic cells was over their additive results. Conclusion Taxol combined with radiation is able to enhance the radiosensitivity in the CNE cell line.
6.Minimally invasive plate fixation in metaphyseal comminuted fractures of long tubular bones in lower extremities

Objective To assess the clinical value of the mi nimally invasive plate fixation technique in treatment of metaphyseal comminute d fractures of long tubular bones.Methods Sixty-eight patients with metaphyseal comminuted fractures o f femur or tibia were treated with min imally invasive percutaneous plati ng and early recovery movement from April 1999to October 2001.Results All the patients were followed up for a mean time of 10months.The postopera tive radiograph showed that reducti on and internal fixation were satisfactory in sixty-five patients.Lateral angular deformity of 8?occurred in one c ase,and 2cm shortage of the lower lim b in two cases.Sixty-eight patients obt ained bone union and satisfactory clinical function.There were no deep i nfec-tion,nonunion or other complicatio ns.Conclusion For metaphyseal comminuted fractur es of long bones of lower extremities,minimally invasive percutaneous plate fixation is o f significant value due to the advant ages of less iatrogenic trauma,rigid fixation and early movement.[
7.Tentative application of the anatomical plate in treatment of distal tibia fractures

Objective To discuss our tentative application of the anatomic plate in the treatment of distal tibia fractures in order to develop a new effective fixation technique for this kind of fracture. Methods From November 2000 to September 2001, the authors applied Linkdistal tibia anatomical plates in the treatment of distal tibia fractures, which involved metaphysis in 12 cases, including 8 males and 4 females, aged from 18 years ~66 years, with 38 years on average. All the cases were of high energy injuries and complicated with ipsilateral fibular fracture. According to AO/ASIF classification: three were type A1, five were A2, three were A3, and one was B1; three of them were close fractures, five were type IO1 open fractures, and four were IO2. Results All the cases healed at one stage without infection or exposure of the implant. Bone callus could be found 2 3 weeks after operation. Bone union was achieved within 3~6 months, with 4 months on average. No delayed union or non union occurred. Functions of ankle joints recovered completely. Conclusion The anatomical plate for distal tibia is supperior to the traditional plates in both biomechanical and biological aspects. It is the best choice for the treatment of distal tibia fractures.
8.Comparison of hemiarthroplasty and DHS fixation for the treatment of unstable intertrochanteric hip fractures in senile patients

[Objective]To discuss a reasonable treatment for unstable intertrochanteric hip fractures in senile patients by comparing the effects of hemiarthroplasty and DHS fixation.[Method]Totally 126 intertrochanteric hip fractures in senile patients with complete clinical data were retrospectively analyzed,who were treated with hemiarthroplasty or DHS fixation from June 2002 to October 2005.Of the 126 patients,53 were treated with hemiarthroplasty,73 were treated with DHS fixation.All of them had multiple medical co-morbidities and were combined with severe osteoporosis.According to modified Even-Jensen classification,all of them belonged to unstable fractures.Comparison was made between the two treated groups in terms of operative time,blood transfusion during or after surgery,time for bed rest postoperatively,postoperative complications and St.Michael hip score one year after surgery.[Result]The average duration of follow-up for hemiarthroplasty and DHS fixation was 16 and 18 months respectively.Compared with the group of DHS fixation,the group of hemiarthroplasty experienced shorter time for bed rest postoperatively,fewer postoperative complications and higher St.Michael score 1 year after surgery,and the differences between two groups had statistical significance(P0.05).[Conclusion]Cemented hemiarthroplasty has the advantages of allowing early full weight bearing after surgery,low complication rate and satisfactory functional recovery.It is a reasonable treatment for unstable intertrochanteric hip fractures in senile patients.
9.Two treatment methods for obstructive sleep apnea hypopnea syndrome

OBJECTIVE To investigate the two treatment methods for obstructive sleep apnea hypopnea syndrome(OSAHS)patients.METHODS 1.Four patients with habitual snoring and two patients with mild OSAHS accepted pillar palatal implant.2. Eleven patients with habitual snoring and fifteen patients with mild to moderate OSAHS accepted temperature- controlled bipolar radiofrequency(RF)ablation in both tonsils.RESULTS The postoperative examination was performed after 6 months.Pillar palatal implant: snoring VAS was reduced from 6.9?0.68 preoperatively to 3.8?0.54 postoperatively in 4 patients with habitual snoring,snoring VAS was down from 9.2?1.21 to 3.1?0.52 in 2 patients with mild to moderate OSAHS.Seven of 11 cases with habitual snoring were cured by RF,and 2 cases were effective,the other 2 cases were failed. Ten of fifteen cases with mild to moderate OSAHS were cured,3 cases were effective,and 2 cases were failed. The effective rate was 84.6%(22/26).CONCLUSION 1.The pillar palatal implant system is a safe method for patients with simple snoring and mild OSAHS in a short time observation.2.Temperature-controlled bipolar radiofrequency ablation is effective for mild to moderate OSAHS duo to hypertrophy of the tonsils.3.According to the clinical characteristics of the patients to select suitable method is the key to get a satisfactory curative effect.
10.Role and mechanism of sinomenine and sulfasalazine in experimental colitis of mice model

Objective To investigate the role and mechanism of sinomenine and sulfasalazine (SASP) in experimental colitis of mice.Methods Seventy SPF grade Kunming mice were evenly divided into seven groups.Except control group,all mice were treated with oxazolone to create experimental colitis model.After modeling,the model group,low dose sinomenine group,high dose sinomenine group,low dose sinomenine combined group,high dose sinomenine combined group and SASP group was gavaged once daily with 0.9% NaC1 solution 1 mL,40 mg/kg sinomenine,120 mg/kg sinomenine,40 mg/kg sinomenine and 400 mg/kg SASP,120 mg/kg sinomenine and 400 mg/kg SASP and 400 mg/kg SASP alone for seven days.The mouse stool characters,changes in body weight and fecal occult blood were recorded and disease activity index (DAI) was scored.The next day after the end of the intervention,all mice were sacrificed and specimens of the colon were obtained aud injury was scored.The specimens of inflammation part and ulcer site of colon were taken for histological examination and injury scoring.The expression of mitogen-activated protein kinase kinase 5 (MKK5),extracellular signal-regulated kinase 5 (ERK5),mitogen-activated protein kinase kinase 7 (MKK7),Jun N-terminal kinase (JNK),nuclear factor κB (NF-κB) at mRNA level were detected by reverse transcription-polymerase chain reaction (RT-PCR).The t-test was performed for comparison between groups.Results The DAI of low dose sinomenine group,high dose sinomenine group,low dose sinomenine combined group,high dose sinomenine combined group and SASP group was 2.33±0.77,1.03±0.73,2.70±0.67,1.60±0.66 and 2.03±0.79,respectively,the score of injury was 5.50±1.43,4.00±1.49,6.80±1.75,4.80±1.32 and 5.40±1.58,respectively,all were lower than those of model group (3.40±0.66 and 11.40±1.71) and the differences were statistically significant (tDA1 =3.33,7.61,2.34,6.08 and 4.18,t score of injury =8.35,10.31,5.94,9.66 and 8.15 ; all P<0.05).The score of injury of high dose sinomenine group,high dose sinomenine combined group and SASP group was 1.40 ±± 1.26,1.70 ± 1.06 and 1.80 ± 1.32,respectively,which were lower than that of model group (3.00 ± 1.05) and the differences were statistically significant (t = 3.07,2.75 and 2.25,all P<0.05).The expression of MKK5,ERKS,MKK7,JNK and NF-κB at mRNA level of SASP group was 24.29±3.40,34.74±3.05,21.34±3.74,18.71±4.12 and 21.68±2.96,respectively,all were lower than those of low dose sinomenine group (51.94±9.16,50.71±11.09,57.98±17.22,55.99±9.65 and 67.41±10.21) and low dose sinomenine combined group (72.03±17.44,119.91±47.26,74.09±21.24,71.83±16.91 and 86.51±18.61).However,those of SASP group were higher than those of high dose sinomenine group (6.53±0.85,17.87±2.74,13.52±2.56,10.41±2.62 and 13.79± 1.43) and high dose sinomenine combined group (16.80±7.15,21.09±3.92,15.81±1.35,14.11±3.10 and 16.62±3.15).All of low dose sinomenine group were higher than those of high dose sinomenine group,all of low dose sinomenine combined group were higher than those of high dose sinomenine combined group,all of low dose sinomenine combined group were higher than those of low dose sinomenine group and all of high dose sinomenine combined group were higher than those of high dose sinomenine group and the differences were statistically significant (tMKK5=8.95,8.49,16.01,2.99,15.61,9.26,3.22 and 4.51,tERK5=4.41,5.69,13.02,12.81,7.82,6.78,4.50 and 2.13,tMKK7 =6.58,7.73,5.80,4.40,8.11,6.32,1.86 and 2.94,tJNK=10.59,7.57,5.37,2.82,13.21,7.57,2.57 and 2.88,tNF-κB =13.60,10.88,7.60,3.70,16.44,11.71,2.85 and 2.59 ; all P<0.05).Conclusions Sinomenine can efficiently improve the inflammatory reaction in the mouse model of colitis and the mechanism may be related with ERK5,JNK and NF-κB signaling pathways.The higher the dose,the more significant the efficacy.The antagonism may exist between sinomenine and SASP.
