1.The effects of sodium phenylbutyrate(SPB)on the expression of p21 and survivine genes on human tongue squamous cancer cell line Tca8113

Aim To investigate the effects of sodium phenylbutyrate on cell proliferation,apoptosis and its expression of p21 and survivin genes in human tongue squamous cancer Tca8113 cell line.Methods The cellular proliferation inhibitory ratio was evaluated by MTT assay and the apoptosis and cell cycle of the Tca8113 cell line was detected by FCM.The expression of p21 and survivin genes was analyzed with Western blot and RT-PCR.Results Sodium phenylbutyrate could inhibit the Tca8113 cells proliferation,promote cell apoptosis and arrest the cells at G_1/G_0 phase.The expression of p21 gene in Tca8113 cell line treated by sodium phenylbutyrate was increased,and one of survivin gene was decreased.Conclusions Sodium phenylbutyrate induces up-regulation of p21 gene and down-regulation of survivin gene,which inhibits Tca8113 cell proliferation and induces its apoptosis and arrests the cells at G_1/G_0 phase.And the increase of p21 mRNA expression is negatively correlated with the decrease of survivin mRNA expression(r_s=-0.548,P<0.01),and so is its protein expression(r_s=-0.514,P<0.01).

There was a correlation among the level of TGF-β1, immune cell cytokines and HBV replication during antiviral therapy. It has an important clinical significance for the researchers to investigate dynamic change of TGF-β1 in chronicviral hepatitis type B (CHB) immunological response and antiviral treatment.
3.Evaluation of the Transcatheter Intra-arterial Hyperthermo-chemotherapy for Advanced Ovarian Cancer

Objective To explore the efficacy and safety of transcatheter intra-arterial hyperthermo-chemotherapy for advanced ovarian carcinoma. Methods 36 patients with unresectable or recurred ovarian carcinoma were divided into two groups randomly: treatment group(n=20) and control group(n=16). Transcatheter intra-arterial infusion warmed to 62 degree chemo-agents via the ovarian tumor supplied artery was performed in the treatment group while routine intraarterial chemotherapy was done in the control group. The CA125 level dynamic change, short-term efficacy and side effect after the interventional procedure were compared between two groups.Results The CA125 level decreased significantly in the treatment group more than that in the control group(?
4.MRI diagnosis of complications related to pregnancy

Objective To discuss the value of MRI in diagnosis of complications related to pregnancy.Methods MRI were per-formed in 22 cases of late pregnancy,7 cases of postpartum with vagina bleeding,and 1 case with eight weeks of gestation following irregular vagina bleeding,respectively.The position of placenta and umbilical cord were observed in the cases of late pregnancy.The shape and signal features of embryo were evaluated in the case of eight weeks gestation following irregular vagina bleeding.Residual placenta and its relationship with the wall of uterus were analyzed in the cases of postpartum with vagina bleeding.Results Among the 22 cases of late pregnancy,normal position of placenta were in 4 cases,placenta previa were in 18 cases,umbilical cord winding around fetal neck in 8 cases.9 cases of placenta previa complicated with placenta accreta were comfirmed by cesarean delivery,how-ever,only 6 cases of placenta accreta were detected on MRI.The residual placenta with placenta accreta was found in all 7 cases of postpartum patients.Hydatidiform mole was proved by pathology in the case of eight weeks gestation following irregular vagina bleeding,and it was showed as a mass with hypointensity on T1 WI and hyperintensity on T2 WI in uterine cavity.Conclusion The complications related to pregnancy such as placenta previa,umbilical cord winding around fetal neck,residual placenta,placenta ac-creta could be correctly displayed on MRI.Hydatidiform mole also could be diagnosed by combining the clinical symptoms with MR images.
5.Construction and expression of human disulfide-stabilized Fv fragment gene to Rabies Virus

Objective To realize the construction and expression of human dsFv genes to rabies virus. Methods The genes of dsFv VH and VL were amplified by PCR-based point mutagenesis strategy. Then the genes were cloned into plasmid pET-22b (+). E. coli BL21(DE3) was transformed with the recombinant expression plasmid and the protein was induced in Luris-Bertani medium by addition of IPTG. The inclusion body proteins of VH and VL were denaturalized by GuHCl, and then re-naturalized in refolding solution to form dsFv fragments. Afterwards, we evaluated the antigen-binding activity of the dsFv and its stability as compared with its originative scFv. Results The fragment genes of dsFv to rabies virus were constructed successfully by PCR-based point mutagenesis strategy. Sequence analysis proved that cysteines were introduced into the position 44 aa of VH and position 100 aa of VL. The dsFv VH and VL genes were expressed in PET22b(+)/BL21(DE3). The VH and VL protein folded into the active dsFv antibody fragments which showed specific binding capability to rabies virus. Conclusion We succeeded in achieving the stabilization of the human scFv to rabies virus and obtaining the active human dsFv antibody fragments. This established a solid basis for further study of the biological activaty and clinical application of dsFv to rabies virus.
6.Tissue expansion in treatment of facial congenital giant nevus

Objective To investigate the effect of tissue expansion in facial congenital giant nevus.Methods From October 2014 to October 2016,7 cases of facial congenital giant nevus patients were treated with expanded skin flaps,including single expansion and secondary expansion.Partial excision and skin tissue expansion were used to drastically remove giant pigmented nevi on body or extremities of infants.There were some points for attention during the surgery:the size and location of the partial excision and expander should be designed reasonably;no tumor-manipulation and non-tension principle should be taken great notice,and expanders should be linked up with one another when several expanders were used at the same time,so as to reduce the possibility of forming the envelop,which was favorable for the second surgery of transfer of a skin flap.Results All cases of giant pigmented nevi excision were on the face,which were completely removed from the patients.The areas of the nevi were from 7 cm × 5 cm to 15 cm × 13 cm.All patients were followed up for 3-12 months (mean 6 months).All flaps survived well.Expander exposure was happened in 2 cases with no bad results.One case of scar received late repair due to surgical scars and all got satisfactory results.Conclusions Tissue expansion is an ideal method in the treatment of facial congenital giant nevus.
7.Clinical Study of Coronary Artery Lesion in Patients with Angina Pectoris Using Virtual Histology Intravascular Ultrasound

Purpose To observe the characteristics of coronary artery lesion in patients with unstable angina pectoris (UAP) and stable angina pectoris (SAP) using virtual histology intravascular ultrasound (IVUS-VH).Materials and Methods A total of 199 patients with angina pectoris were enrolled and divided into SAP group (101 cases) and UAP group (98 cases) according to clinical symptoms, ECG and myocardial enzyme level. All the patients underwent coronary angiography to determine the criminals vessels, grayscale intravascular ultrasound was used to measure and compare the external elastic membrane area (EEMA), lumen cross-sectional area (lumen CSA), plaque area (PA), plaque burden (PB), remodeling index (RI), plaque eccentricity index (EI) of the criminals vessels. IVUS-VH method was used to measure and compare the area and percentage of calcified tissue,fibrous tissue, lipid tissue and necrotic tissue in the plague components of the criminals vessels between the two groups.Results There was no significant difference (t=1.392,-0.345, 1.921, 0.378 and 0.857,P>0.05) of EEMA, lumen CSA, PA, EI and RI measured at the location with smallest lumen area between the two groups. Necrotic core area and percentage of lesion composition in UAP group was significantly higher than that in SAP group (t=2.361,P<0.05). There was no statistically significant difference (t=1.045, 1.884 and 0.787,P>0.05) between the two groups on the area of fiber, lipid and calcification. On the distribution of plaque components, fibrous plaque area percentage of UAP group was lower than that in SAP group (t=-2.418, P<0.05), while the necrotic core area was significantly higher than SAP group (t=2.602, P<0.05), there was no significant difference (t=-0.551 and 0.085,P>0.05) between the two groups on lipid and calcification area.Conclusion Necrotic core area and percentage of plaque composition in the UAP group is larger than the other group. Criminals lesions are more unstable and more easily to be complicated with acute cardiovascular events.

Objective To investigate the effects of isoflurane and sevoflurane at the same dose on expression of Caspase-3 of primary somatosensory cortex (SI) and expression of micmmbule associated protein 1B (MAP1B)in cortical neuronsin neonatal SD rats.Methods Fifty-five neonatal SD rats at postnatal day 7 (eleven rats each litter,altogether 5 litters)were assigned randomly into control group(C group),isoflurane group (I group)and sevoflursne group(S group)in average.The rats in I group,S group or C group were exposed to 1.1% isoflurane or 1.8% sevoflurane (equivalent to 0.5MAC)or air 4h.The brain of neonatal rats were perfused and embedded by paraffin,Caspase-3 positive expression in the SI cortex of brain was detected by immunohistochemistry staining.Besides,the fresh cortex was dissected at O h in C group and at 2h,4h in I group and S group,microtubule associated protein 1 B expression was detected by West blot staining.Results Caspase-3 positive cells in the SI cortex were increased by 561.23%in I group(t=4.45,P<0.01)and 194.46% in S group(t=5.17,P<0.01)when compared with C group,and increased by 124.45% in I group(P<0.05)when compared with S group.The MAP1B protein was increased by 557.15%at 2h(t=16.54 P<0.01)and 475.21% at 4h(t=32.97,P<0.01)in I group while increased by 693.11%at 2h(t=9.45,P<0.001)and 268.15% at 4h(t=2.79,P=0.049) in S group when compared with C group.In S group,MAP1B protein at 4h reduced by 53.65%(P<0.01) when compared with that at 2h.Conclusion 0.5 MAC isoflurane can induce more apoptosis in the cortex in the neonatal rats'brain at postnatal day 7 than sevoflurane.They can both significantly promote the expression of MAP1B in the cortex to start the self-reparation.
9.Clinical study of hypospadias accompanied by congenital ptosis

Objective To study the concurrence of congenital ptosis and hypospadias and the relationship between the two. Methods The incidence rate and degree of ptosis were recorded for 254 cases of hypospadias. Results The morbidity of ptosis in hypospadias cases was 4.72%.The posterior hypospadias group had a higher ptosis rate than the anterior and middle hypospadias groups,P < 0.05. While there was no statistical difference of severe ptosis morbidity rates between these two groups. Conclusions Congenital ptosis is a common concurrent anomaly of hypospadias outside the urogenital system.The severity of hypospadias is relevant with the morbidity of ptosis but not with the degree of ptosis.Hypospadias and ptosis may share common pathogenic or susceptive factors.
10.Endobronchial ultrasound-transbronchial needle aspiration (EBUS-TBNA) for staging of lung cancer

Objective To evaluate the clinical effects of endobronchial ultrasound - transbronchial needle aspiration (EBUS-TBNA) in the evaluation of staging of lung cancer.Methods Between July 2008 to March 2010,the first 128 patients selected by CT or PET/CT scanning with lung cancer in whom metastatic carcinoma in the hilar and/or mediastinal lymph nodes underwent EBUS-TBNA and were clinically followed up.There were 102 males and 26 femals with the age of 37 - 85 years,average 60.1 years.Review the performance in check and the result of biopsy.Results From 128 patients of mean age 60.1 years ( range 37 - 85 ),189 lymph nodes were punctured.The mean diameter of the nodes was 12.3 mm and the range was 6-16 mm.There were no procedural complications.Accuracy,sensitivity,and specificity for EBUS-TBNA were 98.53%,98.50%,and 100%,respectively.Conclusion EBUS-TBNA allows real-time visualization of mediastinal and hilar lymph nodes,allowing sampling safely and efficiently.It has great potential for diagnosis of staging of lung cancer.
