1.Molecular biological markers on diagnosis of early gastric cancer
Chinese Journal of Disease Control & Prevention 2001;5(2):139-141
Gastric cancer(GC) is deeply threatening the health of our people, the mortality rate of GC is the second death cause of carcinoma i n our country. Since the lack of special clinical symptom at the early state of GC, detecting GC at the molecular biological level is expected to diagnose early gastric cancer. This article discusses some markers such as PCNA, p53, telomera se, ras protein and so on to assess the possibility of diagnosing the early gas tric cancer with these molecular biological markers. According to analyzing rela ted papers published, PCNA and p53 were found to be the molecular biological mar kers which should be firstly taken into account in diagnosing gastric cancer. Bo th of them can be easily detected by using immunohistochemical method. Telomeras e is also a good marker, but because of its difficult technology and high cost, it is not so practicable as the former two as the diagnostic marker of early gas tric cancer. Besides, ras potein can also be taken into account as the early det ecting auxiliary marker for GC.

BACKGROUND:Pedicle is the strongest bone structure for the connection between vertebrae and lamina, and the screw fixation through pedicle can provide reliable mechanical basis for the reconstruction of spinal stabilization.
OBJECTIVE:To analyzed the three-dimensional analogue measurement parameters of lumbar pedicle, in order to improve the accuracy and stability of the clinical application of pedicle screw fixation.
METHODS:The original CT data of the spine (L 1-L 5 ) related diseases patients who treated with pedicle screw were analyzed, and the three-dimensional multiplanar reconstruction imaging model was established with Mimics software. By the using of the Mimics software, the lumbar three-dimensional construction was conducted, and the screw length, diameter, transverse section angle and sagittal section angle were predicted, and then statistical y compared with the actual data of the patient postoperative.
RESULTS AND CONCLUSION:Case analysis showed that the position and the length of the screw were good without screw offset and fracture after internal fixation. There were no significant differences in the screw length, diameter, transverse section angle and sagittal section angle between the preoperative predicted parameters and the actual parameter after fixation (P>0.05), while the sagittal section angle of partial segments were smal er than the actual measure value (P<0.05). The lumbar three-dimensional reconstruction and simulation of pedicle screws placement through Mimics software can precisely guide the actual screw placement and improve the safety of pedicle screws placement.

Vasculogenic mimicry (VM) is a process by which aggressive tumor cells generate non-endothelial cell-lined channels in malignant tumors including hepatocellular carcinoma (HCC).It has provided new insights into tumor behavior and has surfaced as a potential target for drug therapy. The molecular events underlying the process of VM formation are still poorly understood. In this study,we attempted to elucidate the relationship between Notch4 and VM formation in HCC.An effective siRNA lentiviral vector targeting Notch4 was constructed and transfected into Be17402,a HCC cell line.VM networks were observed with a microscope in a 3 dimensional cell culture system.Cell migration and invasion were evaluated using wound healing and transwell assays.Matrix metalloproteinases (MMPs) activity was detected by gelatin zymography.Furthermore, the role of Notch4 inhibition in Be17402 cells in vivo was examined in subcutaneous xenograft tumor model of mice. The results showed that downregulation of Notch4 destroyed VM network formation and inhibited migration and invasion of tumor cells in vitro (P<0.05).In vivo, tumor growth was also inhibited in subcutaneous xenograft model (P<0.05).The potential mechanisms might be related with down-regulation of MT1-MMP,MMP-2,MMP-9 expression and inhibition of the activation of MMP2 and MMP9.These results indicated that Notch4 may play an important role in VM formation and tumor invasion in HCC.Related molecular pathways may be used as novel therapeutic targets for HCC antiangiogenesis therapy.

Objective To explore the method and effect of transinfection of rabbit early knee osteoarthritis models via chitosan microsphere with gene of recombined human IL-1Ra gene and TGF-β1 gene. Methods Chitosan microspheres with plasmids of IL-1Ra gene and TGF-β1 gene, and rabbit early knee osteoarthritis models were prepared. Rabbits in different groups had intra-articular injections of chitosan microsphere containing IL-1Ra gene and / or TGF-β1 gene, and chitosan solution as control group before being executed regularly and randomly. The joint specimens were evaluated by HE staining, lycopene red O staining and immunohistochemical analysis and Mankin's score. ELISA was used for detection of IL-IRa and TGF-β1 concentration of articular cavity fluid in each group. Results The control group was consistent with the pathological changes of early OA. In co-transinfection group, judging from the appearance and staining of cartilage, the OA damage of the specimens was less serious than other groups'. Its Mankin's score was significantly lower than single-gene transinfection group (P < 0.05), and the latters Mankin's score were significantly lower than control group (P < 0.05). Conclusion Intra-articular injection of chitosan microspheres containing both IL-1Ra gene and TGF-β1 gene could inhibit the degeneration of cartilage and promote cartilage repair.
