1.Design, synthesis and biological evaluation of the novel trehalose derivatives.

Using brartemicin as the leading compound, fifteen novel trehalose derivatives were designed and synthesized, and the structures were determined by 11H NMR, MS and element analysis. Inhibitory effects of the target compounds on the proliferation of A549, HepG2 and HUVEC cells were detectec by MTT assay. The abilities of adhesion, invasion and migration of A549 and HepG2 cells inhibited by the synthesized compounds were evaluated through Matrigel experiment and Transwell assay. The results showed that, the target compounds had no significant cytotoxicity (compared with the control, P>0.05) to A549, HepG2 and HUVEC cells at the dose range of 1-32 µmol.L-1. At the above dose range, the inhibitory effects of A549 cells adhesion, invasion and migration and HepG2 cells adhesion and invasion by compounds 79 and 82 are better than brartemicin.

Objective To compare the reliability of acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) and APACHE Ⅲ to estimate mortality of critical patients in abdominal surgery. 4.Regulatory effects of hippocampus extracts on PI4-K activity

Objective The hippocampus has been used in Chinese medicine for a long time.Although it has the anti-decrepitude,anti-fatigue and anti-tumor effect,the molecular mechanism is not yet known.PI4-K,the first protein kinase in the PI pathway,should be the target molecular for drug screening.Methods We prepared antigen protein IOV-PI4-K through inside-out ghost,then the antibody-antigen reaction between IOV-PI4-K and its antibody A6D was tested by ELISA.Results Two different function groups,which could increase or decrease the binding affinity of PI4-K to its antibody,were screened out from the extracts of hippocampus.This fact indicated that there might be two different components to up-regulate or down-regulate the activity of PI4-K respectively.Conclusion Derivatives of the components could be developed into anti-tumor or anti-decrepitude drugs.Meanwhile,a new way for performing drug screen through ELISA was established.
5.Perioperative Use of Antibacterials in Patients with Caesarean Section:Analysis of 360 Cases

OBJECTIVE:To investigate the preventive use of antibacterials in patients undergoing cesarean section in our hospital.METHODS:A total of 360 medical records of patients undergoing cesarean section were randomly sampled in 2006(30 cases per month)for an analysis of the correlation between choice of antibacterials,combination rate and course of treatment etc and curative effects.RESULTS:Of the 360 cases,86.8% used second and third generation cephalosporins,6.3% fourth generation cephalosporin(cefepime),3.8% cephradine.CONCLUSION:No positive correlation was noted between choice of antibacterials and course of treatment and the curative effects in the prevention of postoperative infection.
6.Prevention and treatment of urethral stricture after hypospadias repair in children

Urethral stricture is one of the common complications after hypospadias repair,and it is related with the blood supply of materials used in urethra formation.The more severe and proximal hypospadias is,the higher possibility of stricture is.The patients with urethral stricture have the symptoms of stranguria and urinary retention,and they usually need re-surgery.The repair with well vascularized flap can decrease the occurrence of urethral stricture after hypospadias repair.
9.Clinical analysis of 15 patients suffered from advanced ascetic schistosomia-sis with tuberculous pleurisy

Objective To discuss the clinical characteristics of advanced schistosomiasis combined with tuberculous pleuri?sy,so as to reduce misdiagnosis and mistreatment. Methods The clinical data of 15 patients suffered from advanced schistoso?miasis combined with tuberculous pleurisy were collected and analyzed retrospectively. Results The 15 patients all showed var?ious degrees of gastrointestinal symptoms with the performance of abdominal distension,digestive function and fatigue,14 pa?tients showed various degrees of edematous,9 patients showed stuffiness,4 patients had cough and expectoration,and 2 pa?tients had low fever and night sweats. Three cases were diagnosed within 2 days after admission,9 cases were diagnosed within one week after admission,and 3 cases were diagnosed after one week. Fifteen patients all received anti?tuberculosis treatment based on routine liver protection,diuresis,and symptomatic and supportive treatment. One patient with severe liver and kidney dysfunction died and one with gastrointestinal bleeding died. The remaining 13 patients were clinically cured. Conclusions The patients suffered from advanced schistosomiasis combined with tuberculous pleurisy do not show obvious tuberculosis poison?ing symptoms,and are easily misdiagnosed and missed?diagnosed. Therefore,physicians should pay much attention to the pa?tients whose pleural effusion cannot subside effectively or whose symptom cannot improve.
10.Establishment of a real-time PCR method for quantitative detection of a novel metabolic syndrome related gene (MSRG)

Objective To establish a Taqman real-time PCR assay for quantitative detection of the expression of metabolic syndrome related gene (MSRG) mRNA, and to study the function of a new gene. Methods Specific primers and probes were designed for real-time PCR according to the MSRG cDNA sequence. The plasmid standard preparations were constructed by T-A clone, and serial 10-fold dilutions of the extracted plasmid standard preparations were prepared for plotting the standard curve which was used for relative quantification of real-time PCR. The sensitivity, specificity and reproducibility of real-time PCR assay were detected. To compare with the semi-quantitative RT-PCR, the expression levels of MSRG were measured by real-time PCR and semi-quantitative RT-PCR, respectively, in HepG2 cells which were incubated with glucose in different concentrations [5.6mmol/L (G5.6), 22mmol/L (G22) and 33.3mmol/L(G33.3)]. Results An effective real-time PCR assay was established for detection of MSRG mRNA expression levels. Significant differences existed in MSRG expression in HepG2 cells between the G33.3, G22 and G5.6 groups detected by real-time PCR assay. The expression levels of MSRG in HepG2 cells increased significantly in G33.3 group, whereas no significant difference on the expression level of MRSG mRNA was found between G22 and G5.6 groups when semi-quantitative RT-PCR was used for detection. It suggested that the real-time PCR assay was more sensitive and even more precise than that of semi-quantitative RT-PCR. Conclusion The real-time PCR assay was a sensitive, specific, quantitative, and reproducible tool for studying the function of MSRG at the mRNA expression levels of gene.
