2.Validation of zebrafish embryo toxicity evaluation method

OBJECTIVE In this study,nine different levels of known teratogenic embryos toxic com-pounds (aceta minophen,methi mazole,indo metacin,methotrexate,ascorbic acid,isoniazid,penicillin G,saccharin ,5-fluorouracil )were used to validate the established evaluation method of zebrafish e mbryo develop mental toxicity in our laboratory via zebrafish e mbryonic develop ment toxicity tests. METHODS By water bath method,zebrafish embryos of post-fertilization 2 h (2 hpf)were exposed to different concentrations of compounds.Observe and record the nu mber of malformations and dead e mbryo in 24,48,72,144 hpf.After three parallel experiments,get EC50 ,LC50 through software and calculate the teratogenic indices (TI =LC50 /EC50 )of the positive drug and recorded it as teratogens.If there is no teratogenic index recorded it as non-teratogens.Co mpare the results with the existing animal experiments and clinical trials to obtain the sensitivity and specificity.RESULTS Based on the results, exposure to aceta minophen (TI =2.07),methi mazole (TI =2.91 ),indo methacin (TI =1 .67),5-flu-orouracil (TI =8.31 ),methotrexate (TI =1 .31 ),showed various levels of teratogenic effects,and the corresponding TI′s were calculated.Methi mazole appeared more obvious skeletal deformities;indo m-ethacin toxicity is mainly reflected in abnormal heart,blood circulation and other endpoints;5-FU at lower doses that result in higher mortality rates,but also took place over the lacking of pig mentation;aceta min-ophen for the heart,liver and other target organ toxicity,pericardial cyst;methotrexate appears signifi-cant so mites,skeletal deformity.Since the IT′s for ascorbic acid,isoniazid,penicillin G,saccharin can-not be calculated,these compounds are considered to show negative results,indicating that the data fro m the nine compounds in zebrafish test are consistent with traditional animal experiments and clinical information,the sensitivity of 100% and specificity of 100%.CONCLUSION Though validation test,it has been de monstrated that the established zebrafish embryos develop mental toxicity evaluation method can be applied to early screening of drug toxicity.
3.A study of disability adjusted life year about patients with epilepsy

Objective To evaluate of the disability adjusted life years (DALY) and its influencing factors in patients with epilepsy.Methods Eight hundred and seventy-four cases with epilepsy were investigated in 9 pilot sites in Henan province.Make up the life expectancy of residents in Henan province for the DALY estimates.Using the R 2.10.0 software,the quality-adjusted life years and its influencing factors were analyzed by univariate and multivariate analyzes.Results There was 16.64 per capita loss of all cases of epilepsy DALY,and DALY loss value was 3.41 per 1000 populations.Comprehensive intervention and no sense of control group patients were per capita DALY loss of 8.56 person-years and 24.19 person-years respectively.There was significant difference between the two groups DALY loss (Z =23.146,P < 0.01).Moreover,non-conditional Logistic regression equation was also statistically significant (x2 =365.83,P < 0.01).Age,duration,seizure frequency,compliance,and anxiety situation were the main factors of the disease burden.Conclusions In the future,enhanced interventions should be focused on the priority groups and the modifiable factors.Additionally,the ‘standard treatment + follow-up management and health education + psychological intervention' should be strengthened as a primary means of comprehensive intervention.
5.Marginal adaptation of metal crowns:comparison of two production methods

BACKGROUND:At present, the confrontation of CAD/CAM systems and the lost wax casting method for marginal adaptation has been widely concerned, but the results are controversial. OBJECTIVE:To compare the marginal adaptation of single cobalt-chromium PFM crowns produced by 3 Shape CAD/CAM systems and the lost wax casting method. METHODS: A maxilary first premolar was prepared according to the standards. 50 resin-generation models were created by bi-color silicone rubber method and randomly divided into two groups: CAD/CAM group (A1-A25) and cast group (B1-B25). Al the models were cast in plaster. In CAD/CAM group, the digital impressions were obtained by optical scanning, then designed through computer and cut directly by NC miling machine. In cast group, metal ceramic crowns were made by the lost wax casting method. When crowns were al seated on the working dies, the marginal gaps were copied with light silicone rubber and then embedded with occlusal silicone rubber. The thickness of crown marginal gap was measured with a stereo microscope. RESULTS AND CONCLUSION:The average vertical clearance was (34.67±7.48)μm of CAD/CAM group, while (53.70±9.64)μm of the cast group; the difference was statisticaly significant between two groups (P < 0.05). The average level clearance was (37.61±7.63)μm of CAD/CAM group, while (53.02±10.65)μm of the cast group, and the difference was statisticaly significant between two groups (P < 0.05). Experimental findings suggest that, the marginal adaptation of single cobalt-chromium PFM crowns cast produced by 3 Shape CAD/CAM systems is clinical acceptable (< 120μm), and CAD/CAM systems of metal casting is obviously better than the lost wax casting in building metal based crowns with adaptable margin.
6.Progress in the study of Her2-targeted cancer therapeutic antibodies.

Tumor surface antigen human epidermal growth factor receptor 2 (Her2) is a type I receptor tyrosine kinase, which belongs to human epidermal growth factor receptor family. Her2-overexpression is associated with tumorigenesis and metastasis. Due to significant clinical effects, Her2-targeted cancer therapy especially therapeutic antibody has become the hot spot in the field of cancer treatment. Anti-Her2 antibody drugs include monoclonal antibodies, antibody-drug conjugates, bispecific antibodies and emerging "two in one" antibody. Based on structure and function of Her2, this review focuses on recent advances in active mechanisms and clinical researches of these antibodies.

7.Interventional effect of Yufeng capsule on function of platelet aggregation and contents of thromboxane B2 and 6-ketoprostaglandin F1 alpha in blood plasma of gerontic rats with blood stasis

BACKGROUND: Clinical experiments indicates that Yufeng capsule (composed of milkvetch root, angelica, szechwan lovage rhizome, senega and salvia miltiorrhiza) is effective for benefiting vital energy, circulating blood flow,dispelling gores and killing inflammation. The mechanism may be associated with the function of platelet aggregation, changes of contents of thromboxane B (TXB) and 6-Ketoprostaglandin F1 alpha (6-keto-PGF1 α) in plasma.OBJECTIVE: To observe the influence of Yufeng capsule on the platelet aggregation and its mechanism.DESIGN: Randomized controlled animal trial.SETTING: Department of Pathology, Daqing Branch of Harbin Medical University.MATERIALS: The experiment was performed at the Laboratory of Pharmacology, Daqing Branch of Harbin Medical University from May 2000 to July 2004. A total of 40 Wistar male gerontic rats with mean age of (8-10) weeks and mean body mass of (550±150) g and 8 Wistar male young rats with mean age of 4-6 weeks and mean body mass of (230±120) g were selected.METHODS: The 8 Wistar male youth rats were selected as youth control group. The 40 Wistar male gerontic rats were selected as the blood stasis model, and according to the numeration table method they were classified into 5 groups: aspirin group, low-dose Yufeng capsule group, the middle-dose Yufeng capsule group, high-dose Yufeng capsule group and gerontic model group, with 8 rats in each group. The Yufeng capsule was constituted by milkvetch root, angelica, szechwan lovage rhizome, senega and salvia miltiorrhiza, and provided by the Preparation Office, Second Affiliated Hospital,Heilongjiang University of Chinese Medicine. Respectively the rats were injected with aspirin solution (5.5 g/kg), Yufeng capsule (0.33 g/kg, 0.66 g/kg and 1.32 g/kg), successively for 22 days by gastric perfusion, twice a day.Those in the gerontic group and youth control group were not treated with any drug One hour after last time administration, function of platelet aggregation(PAG), contents of thromboxane B2 (TXB2) and 6-Ketoprostaglandin F1 alpha (6-keto-PGF1 α) in plasma were examined with platelet aggregation method and radioimmunoassay.MAIN OUTCOME MEASURES: Function of platelet aggregation; contents of TXB2 and 6-keto-PGF1 α as well as ratio of TXB2 to 6-keto-PGF1α.RESULTS: Totally 48 animals were involved in the result analysis. ①The PAG was higher in the gerontic model group than other groups (P < 0.01).②One minute platelet aggregation rate (PAG1), five minutes platelet aggregation rate (PAG5)and the maximal platelet aggregation rate (PAGM) in the high-dose Yufeng capsule group were lower than those in the aspirin group[(7.96±2.17), (6.38±1.66), (14.33±2.87) ng/L and (31.95±3.94),(35.29±6.54), (38.68±7.46) ng/L, respectively(P < 0.01 )]. PAG5 in the middle-dose group was lower than that in the aspirin group [(13.96±1.66),(35.29±6.54) ng/L, (P < 0.01 )]. ③TXB2 content in the low-dose Yufengcapsule group, middle-dose Yufeng capsule group, high-dose Yufeng capsule group, aspirin group and youth control group was lower than that in the gerontic model group (P < 0.01). ④The TXB2 content in the high-dose Yufeng capsule group was lower than that in the low-dose Yufeng capsule group [( 100.13±8.90), (127.00±20.99) ng/L, (P < 0.01 )]. ⑤TXB2/6-ke-to-PGF1 α in the high-dose Yufeng capsule group, middle-dose Yufeng capsule group and youth control group was lower than that in the gerontic model group [0.62±0.17,0.69±0.14,1.75±0.17,1.11±1.12, (P < 0.05 )].CONCLUSION: High-dose and middle-dose Yufeng capsule is able to decrease the content of TXB2 and increase the content of 6-keto-PGF1 α, and improve their ratio. It is important to enhance the balance of TXB2 and 6-keto-PGF1 α. The effect in the low-dose Yufeng capsule group is lower as compared with that in the high-dose and middle-dose Yufeng capsule groups. It is thought that the low-dose Yufeng capsule has preventive effect.

10.An evaluation scale of medical services quality based on "patients' experience".

An evaluation scale of medical services quality was developed on the basis of "patients' experience". The questionnaires were developed by, among others, searching relevant literature, collecting well-established assessment scales, measuring patients' experience and satisfaction, brain-storming, literature analysis. Delphi method was adopted for expert consultation. Scale items were screened and revised. The key indexes were converted. Field surveys were conducted for testing the reliability and validity of the scale. Our modified evaluation scale for measuring medical services quality based on "patients' experience" included 6 dimensions (tangibility, reliability, responsiveness, assurance, empathy and continuity), and 50 items. The novel scale based on "patients' experience" may better serve the purpose of assessing medical services quality.
