1. A review on intestinal lymphatic drug transport

OBJECTIVE: Available information on the background of the current state and advances of the intestinal lymph trans port of medicines were reviewed. Drug transported by the intestinal lymphatic system can avoid hepatic first-pass metabolism, increase the bioavalibitlty of the highly lipophilic drug, and it is of importance for immunomodulatory, anticancer and anti-infective drugs. METHODS: We conducted a systematic literature review of articles published recent years, analysis and summaries are made upon the advances and methods applied for the increase of the intestinal lymph transport. RESULTS AND CONCLUSION: Intestinal lymphatic transport can increase the bioavailability via a reduction in the first-pass metabolism and the possibility of specifically targeting drugs to regions of the lymphatics, thus it is very advantageous for drugs that are under significant first-pass metabolism. The present review embodies a brief background of the mechanism of access of drugs to the intestinal lymph, a discussion on the links between lipid absorption and transport of highly lipophilic drugs, and approaches for enhancing lymphatic drug transport. Finally, experimental models of the lymphatic transport are also discussed.
2.Vowel selection effects on the parameters of acoustic analysis

Objective:To investigate if there is difference about the parameters of acoustic analysis with the different vowe |a|、|i|、| | and decide which vowels is best fitted to the acoustic analysis. Method:40 normal and 130 pathologic voice patients accepted acoustic analysis. Each acoustic parameters such as jitter,shimmer、NNE、 SDF0、SNR were compared with different vowels. Result:Jitter、shimmer、NNE with |i| were much lower than that with |a| and | | in normal cases and light hoarseness group. While the acoustic parameters with |i| were much higher than that with |a| and | | in moderate and heavy hoarseness group. Conclusion: |a| and | | were the preferred vowels for normal and light hoarseness group,while the |i| could supply some useful suggestion for the moderate and heavy hoarse patients.
3.HPLC fingerprint of Fructus aurantii micropowder by methanol extraction

AIM: To provide a HPLC fingerprint of Fructus aurantii micropowder in order to create a basis for(identification.) METHODS: With the help of computer similarity evaluation,Seventeen kinds of Fructus aruantii sample were pulverized and micronized separately,then were extracted by methanol and compared correlation between both extracts. RESULTS: In methanol extraction,contents of aurantiamarin,hesperidin and neohesperidin in micropowder were more than that in pulverized powders,correlation of both HPLC fingerprint was 0.9~1.0. CONCLUSION: The method established can be used for identifying and evaluating crud drug of Fructus aurantii,and further prove that micronization is beneficial to increasing flavonoid dissolution.
5.Effects of ovariectomy on the expression of estrogen receptor beta immunoreactivity in the basal forebrain of adult female mice

Objective To study the effect of ovarian estrogen on the expression of estrogen receptor beta (ER beta) of the basal forebrain of mice. Methods Ovariectomized (OVX) mice were used as animal model, and nickel ammonium sulfate intensified immunohistochemical SP technique was used to detect the changes of ER beta expression in the basal forebrain. Results The expression of ER beta in the basal forebrain of mice decreased dramatically, after OVX decreased to the lowest levels at 3 d after OVX, increased gradually, and recovered to normal level 20 d later. Conclusion Ovarain estrogen can regulate the expression of ER ? of the murine basal forebrain, indicating that estrogen, via ER ?, can regulate the nerous structure and function of the basal forebrain, which may be the mechanism of estrogen replacement therapy on Alzheimer′s brain.
6.Endovascular brachytherapy with Liquid ~(188) Re affect cell proliferation, phenotype of smooth muscle cell and secretion of transforming growth factor

Objective To investigate the effect of endovascular brachytherapy with Rhenium-188-filled catheter balloon on cell proliferation,phenotype of smooth muscle cell and secretion of transforming growth factor; to explore the possible mechanism of this method employed in restenosis prevention. Methods After the right common carotid arteries were overstretched by 3.5?20 mm balloon,twenty New Zealand White rabbits were randomized into irradiation (n=10) and control group (n=10). Endovascular bachytherapy was performed wth a 188[KG*2/2]Re filled 3.0?20 mm balloon catheter delivering a dosage of 16 Gy (0.5 mm underneath the luminal surface) in the irradiation group, whereas isovolume normal-sodium-filled balloon catheter was used in the control group for an equal period. After thirty days, analysis of the carotial arteries was made by measns of histological and immunohistochemical staining. Results (1) The positive staining index of proliferation cellular nuclear antigen (PCNA) and transforming growth factor (TGF) of the irradiation group is significantly less than the control group (0.028?0.005 vs 0.054?0.011, 0.033?0.008 vs 0.063?0.014, respectively); (2) The positive staining of ?-actin index in intima has no significant difference between the irradiatin group and the control group. Conclusion (1) The preventive effect on restenosis with endovascular brachytherapy is possibly achieved by suppression of cellular proliferation and transforming growth factor secretion. (2)The phenotype of smooth muscle cell is possibly unaffected by endovascular brachytherapy.
7.The effect and mechanism of allicin on the migration and invasion properties of human osteosarcoma U87 cells

ABSTRACT:Objective To investigate the anti-metastatic effect of allicin on glioma cell line U87 and related mechanisms.Methods In this study,we employed MTT assay to test the anti-proliferative effect of allicin. Transwell assay was used to test the anti-metastatic ability of allicin.Real-time PCR and Western blotting were employed to test the effect of allicin on the expressions of matrix metalloproteinase-2 (MMP-2 ) and matrix metalloproteinase-9 (MMP-9).Western blotting was employed to test the phosphorylated level of p38.Results Allicin could significantly inhibit the proliferation and invasion of U87 cells (concentration>8 μg/mL,P <0.05). Meanwhile allicin (concentration<8μg/mL)could inhibit the invasion of U87 cells.After treatment with allicin for 24 hours,the expressions of MMP-2 and MMP-9 were decreased significantly (P < 0.05 ).Moreover,allicin treatment decreased the phosphorylated level of p38 obviously (P < 0.05 ).Conclusion Allicin inhibits the invasion and migration of glioma cell line U87 by reducing the expressions of MMP-2 and MMP-9 via suppressing the activity of p38 signal pathway,suggesting that allicin is a potential therapeutic agent for glioma.
8.Detection of p16 gene methylation in population exposed to arsenic in Inner Mongolia

Objective To investigate the occurrence of p16 gene methylation in the population chronically exposed to arsenic in Inner Mongolia,and to study the molecular mechanisms of carcinogenesis associated with arsenic exposure.Methods The study group was composed of 40 cases of typical arseniasis selected from the epidemic area,and the two control groups consisting respectively of 40 non-arseniasis cases selected from the same epidemic area,and 40 healthy persons enrolled from non-epidemic area.Methylation of p16 gene in the blood specimens were analyzed for all the subjects with MS-PCR techniques,and statistical analysis was performed using chi square test.Results The positive rates of p16 hypermethylation in blood specimens were 65.0%,47.5% and 20.0% respectively in study group and two control groups,and the rate of hypermethylation increased with the increase in arsenic exposure with drinking water in epidemic areas.The positive rate of p16 hypermethylation showed significant differences(P

10.EFFECT OF ADRIAMYCIN ON LEAKAGE OF LACTATE DEHYDROSENASE(LDH) & CONTENT OF MALONDIALDEHYDE (MDA) IN CULTURED MYOCARDIAL CELLS

Cardiotoxic effects of adriamycin were investigated in cultured heart cells. Three-day cultured myocardial cells were treated with the different doses of adriamycin (0.3mg/L, 1 mg/L, 3 mg/L) , and incubated in verieus times times ( 4 h, 8 h, 24h ) , and, then the content of lactate dehydrogenase ( LDH ) in media and malondialdehyde ( MDA ) in cells were measured. The results showed that the content of LDH in media increased with the time in culture. The content of LDH in media and MDA in cells increased markedly with the increase in adriamycin concentration. It suggested that the adriamycin has direct damage to myocardial cells, which is time-dependent and dose-dependent.
