1.Establishment and evaluation of prognostic model of severe traumatic brain injury

Objective To build the prognostic model of STBI and provide guidance for making clinical decision .Methods A total of 486 patients with STBI was collected .Six months after injury ,according to GOS ,the patients were divided into five levels :good , mild-to-moderate disability ,serious disability ,persistent vegetative state and death .Ordinal regression analysis were used to find out the main factors of different populations .Results There were significant differences between different levels in nine factors .The re-sult of ordinal regression analysis showed that GCS ,age ,rescue time and reaction of pupil to light stood as independent variables in prognosis of patients with STBI(P<0 .05) .The outcomes from the prognostic model were pretty much exactly the same as the ac-tual results(Kappa= 0 .640 ,P= 0 .026) .Conclusion Though using the prognostic model of STBI ,the present study found out prognosis was closely related to GCS ,age ,rescue time and reaction of pupil to light in patients with STBI ;thus it could provide guidance for prognosis evaluation .
2.Identification of HLA-A2 restricted cytotoxic T lymphocyte epitopes from tumor antigen PIWIL2

AIM: To identify the human leucocyte antigen A2 (HLA-A2) restricted cytotoxic T lymphocyte (CTL) epitopes from tumor antigen PIWIL2.METHODS:RT-PCR and Western blot was used to determine the expres-sion of PIWIL2 in cancer cell lines MCF-7, SW480 and HT-29.HLA-A2 epitopes from PIWIL2 protein were predicted by the software of BIMAS, RankPep, NetMHC, NetCTL1.2 and IEDB.The peptides were synthesized by standard solid-phase methods.The binding affinity of the peptides to HLA-A2 molecules was evaluated by T2 cells binding assay.ELISPOT as-say was used to investigate the levels of IFN-γ.The cytotoxicity assay in vitro was also used to determine the ability of indu-cing T cell response by the peptides.RESULTS:The expression of PIWIL2 was observed in MCF-7, SW480 and HT-29. The candidate peptide P485, P493 and P965 showed moderate affinity toward HLA-A2 molecule.ELISPOT assay showed P485 and P965 induced CTLs of IFN-γrelease form CTLs.The CTLs induced by P485 and P965 lysed the MCF-7 cells. CONCLUSION:The peptides P485 and P965 are excellent HLA-A2 restricted cytotoxic T lymphocyte epitopes from the tumor antigen PIWIL2, which could serve as new candidates towards antitumor peptide vaccines.

Trauma is the leading cause of death for people under 40 years old in the world. At present, the rescue and treatment system of trauma patients in China is not yet well established, and the mortality of trauma patients is higher than those in the developed countries. Improving the treatment system is the key to reducing the trauma mortality. In order to innovate the service mode of trauma first aid, further promote the establishment of regional trauma first aid system, improve the ability of trauma treatment, reduce the mortality and disability rate of trauma patients in Jiangxi Province, recently Health Commission of Jiangxi Province and the First Affiliated Hospital of Nanchang University have reached a consensus on the establishment of Jiangxi trauma first aid center. In order to provide reference for the construction of trauma treatment system, the author analyzes the following aspects including functional positioning, basic requirements, organization management, and evaluation of core indicators.
