1.CT and MRI findings of levamisole induced encephalopathy

Objective To investigate the CT and MRI features of levamisole induced encephalopathy. Methods The CT and MRI features of 6 cases with clinically proven levamisole induced encephalopathy were retrospectively analyzed. Gd-DTPA enhancement examinations were performed in 5 cases. CT examinations were performed in 4 cases before MRI. Results MRI features: Lesions were mainly located in bilateral periventricular and hypophloeodal white matter, scattered as multifocal lesions, and were different in sizes. Most of the lesions presented as irregular plaques (masses), with round/oval or spotty nodules in second. The lesions had low signal intensity on T 1WI and high signal intensity on T 2WI, and commonly without enhancement. Edema and mass effect were slight. CT features: Two cases showed multiple irregular plaque-sheet low density lesions scattered in bilateral periventricular white matter. One showed symmetrical low density in bilateral putamina nuclei. Another one was negative. Conclusion CT and MRI are valuable for the diagnosis and differentiated diagnosis of levamisole induced encephalopathy. MRI has higher sensitivity and specificity than CT does, and can favorably evaluate the treatment and prognosis.
2.The study on influence of complicated diabetes on the prognosis on kidney transplantation

Objective To investigate the difference in influerce of complicated diabetes on the prognosis of kidney transplantation.Methods Analyze the data of 507 times kidney transplantation and clinical tracing.Results Totally 507 patients with kidney function failure had received 539 times kidney transplantation,52 of whom were connected with diabetes[30 male,(51.4?7.1)age;22 female,(51.6?4.9)age],and 16 cases were pre-transplant,36 cases post-transplant.The other 455 cases were non-diabetes[293 male,(38.7?12.3)age;126 female,(43.5?10.1)age].The prevalence of diversified complications in the group with diabetes was all remarkably higher than the other group.There were 9 cases death in the group with diabetes,and 35 deaths in the other group after kidney transplantation;actually the mortality of diabetes group(17.3%,9/52)was 2.45 times as high as the group without diabetes(7.7%,35/455),especially in pre-transplant diabetes group.Another result was that there was no significant difference in the survival period of the dead patients in group with and without diabetes.Conclusion The possibility for the patients who,suffer from kidney function failure with diabetes to get Kidney transplantation postoperative complications is bigger compared with those without diabetes,and the prognosis is poor.To control diabetes more effectively before and after kidney transplantation perhaps can improve the prognosis of them.
3.Expression and characterization of an EGF-IL-18 fusion protein for targeting (therapy) in insect cells

Objective:To express and purify a new fusion protein harboring human epidermal growth factor receptor(EGFR) binding domain and Interleukin-18(IL-18), as well as preliminary assay biological activity of recombinant proteins.Methods:Fusion protein was expressed in insect cells Spodoptera frugiperda cell (Sf9) by using Bac-to-Bac system, and an abbreviation purification procedure was used to purify fusion protein. IFN-?induction assay and EGF Receptor competitive test was used to determine fusion protein's biological activity.Results:SDS-PAGE and western blot assay showed that purified EGF-IL-18 fusion protein had high purity in 20 kD as expected and had the same antigenic specificity as human IL-18. IFN-?induction assay and EGF Receptor competitive test showed that fusion protein induced production of IFN-?in human PBMC and bound with tumor cells.Conclusion:EGF-IL-18 fusion protein has been successfully expressed and purified in insect cells and shows potential to apply in targeting therapy for tumor.
4.Identification of recombination baculovirus and determination of virus titer with fluorescence quantitative PCR assay

Objective:To develop a real-time PCR assays based on TaqMan chemistry for the identification of recombinant baculovirus and determination of virus physical titers in Bac-to-Bac system.Methods:The recombinant baculovirus containing human IL-18 gene was produced using Bac-to-Bac system.A 10-fold series diluted primary viral stocks were used for plaque assay and DNA extraction.Bacmid(baculovirus plasmid) was 10-fold series diluted and served as standards.Real-time PCR amplification of the IL-18 gene was performed in triplicate for each diluted recombinant virus.At the same time,plaque assays were performed using overlay agarose method.Results:The standard linear(101 to 108 copies) from quantitation was achieved with the standard curve.We also find that the "vg/ml" titer value is generally about 10 times than "pfu/ml" titer of the same recombinant virus stock.Conclusion:A TaqMan real-time PCR method is established to identify the recombinant baculovirus and determine the "vg/ml" titer of virus.The method is rapid and quantitative over a wide range of virus titers.

In this review, we discuss approaches to targeting critical elements of the DNA damage response for radiosensitization and chemosensitization. In addition, we also discuss strategies for targeting DNA damage response and DNA repair defects in cancer cells for synthetic lethality.

Background The evaluation of visual function after laser in situ keratomileusis(LASIK) mainly focuses on the study of contrast sensitivity funetion (CSF).However,CSF measurement is a subjective method and therefore has a limiting in application.A point spread function (PSF) is becoming a study topic because of its objectivity in assessing visual quality.Objective Present study was to evaluate the change of the modulation transfer function (MTF) of eyes and compare the optical and visual quality of human eyes after LASIK.Methods Thirty-six patients(72 eyes) were included in this study.The patients were divided into low myopia(-2.72±0.52 D),moderate myopia (-4.89+0.80 D) and high myopia (-8.00+0.98 D) groups according to the spherical equivalent (SE)diopter before the sugery.The uncorrected visual acuity (UCVA),best corrected visual acuity (BCVA) were examined before and after operation,and PSF data was obtained with Topoeon PSF-1000 analyzer under the 3.0 mm and 6.0 mm pupil size conditions.The follow-up visits of the patients were scheduled at the 7th days,1 st and 3rd months.Written informed consent was obtained from each patient before this study. ResuIts The MTF values among the three refractive groups were significantly different between 3 mm and 6 mm pupil preoperatively (P<0.05).Under the condition of 3 mm pupil size,the MTF values in the spatial frequencies of 2.98 cpd and 14.88 cpd were significantly declined in different time points after operation in comparison with preoperative ones (P<0.05).However,the MTF changes had no statistical significance in the spatial frequencies from 18.85-37.70 cpd among various time points(P>0.05).Under the condition of the 6 mm pupil size,MTF values in low and moderate myopia groups were reduced after operation in comparison with before operation at the spatial frequencies from 2.98-7.44 cpd(P<0.05),however,there were no obvious difference was found in the spatial frequencies of 9.42-37.70 cpd(P>0.05).The MTF values of high myopia group was decreased significantly at all spatial frequencies(P<0.05).The MTF values improved gradually as the prolong of time after operation but was still lower at the 3rd postoperative month than that of preoperation. Conclusion The postoperative visual quality is associated with refractive power and pupil diameter.PSF is a feasible method in assessing the early visual quality after LASIK.

OBJECTIVE:
The aim of this study is to evaluate the development of auditory performance and speech intelligibility within the first year after hearing aid fitting in children with moderate or severe hearing loss, investigate the effects of hearing level on auditory performance and speech intelligibility and provide a clinical database for their hearing and speech habilitation.
METHOD:
Twenty-nine children participated in this study, ranging in age at hearing aid fitting from 3 to 8 years old with a mean of 5. 6 years old. 19 were boys and 10 were girls. According to their hearing level, they were divided into two groups. 14 children were in group of moderate hearing loss (41-60 dB HL). 15 children were in group of severe hearing loss (61-80 dB HL). The categories of auditory performance (CAP) and speech intelligibility rating (SIR) were used to evaluate their auditory performance and speech intelligibility. The evaluation was performed before hearing aid fitting and 1, 3, 6, 9, 12 months after fitting.
RESULT:
There was significant difference in mean score of CAP between group of moderate hearing loss and severe hearing loss before hearing aid fitting (P < 0.05). However, no significant differences were observed between these two groups at 1, 3, 6, 9, 12 months after fitting (P > 0.05). There was also significant difference in mean score of SIR between group of moderate hearing loss and severe hearing loss before hearing aid fitting (P < 0.05). How ever, no significant differences were also observed between these two groups at 1, 3, 6, 9, 12 months after fitting (P > 0.05). The mean scores of CAP for group of moderate hearing loss at 6, 9, 12 months after fitting were significantly superior in comparison with the score before hearing aid fitting (P < 0.05). The mean scores of SIR for group of moderate hearing loss at 6, 9, 12 months after fitting were significantly superior in comparison with the score before hearing aid fitting (P < 0.05); the mean score at 12 months after fitting was also significantly superior in comparison with the score at 1 month after fitting (P < 0.05). The mean scores of CAP for group of severe hearing loss at 3, 6, 9, 12 months after fitting were significantly superior in comparison with the score before hearing aid fitting (P < 0.05); the mean scores at 9,12 months after fitting were significantly superior in comparison with the score at 1 month after fitting (P < 0.05). The mean scores of SIR for group of severe hearing loss at 3, 6, 9, 12 months after fitting were significantly superior in comparison with the score before hearing aid fitting (P < 0.05); the mean scores at 6, 9, 12 months after fitting were also significantly superior in comparison with the score at 1 month after fitting (P < 0.05).
CONCLUSION
Auditory performance and speech intelligibility in children with moderate or severe hearing loss improved significantly within the first year after hearing aid fitting. The development followed different trajectory.

9.Effect of Thumb Pressing at Yangming Meridian on Function Rehabilitation After Total Knee Arthroplasty

Objective To observe the effect of thumb pressing at Yangming Meridian on the function rehabilitation after total knee arthroplasty (TKA) for knee osteoarthritis (KOA).Method Sixty KOA patients after TKA were randomized into a treatment group (thumb pressing at Yangming Meridian plus conventional rehabilitation) and a control group (single conventional rehabilitation),30 cases each.The treatment group was intervened by thumb pressing and kneading manipulation at the acupoints of Foot Yangming Meridian and Foot Taiyin Meridian plus rehabilitation training of passive flexion and extension by CPM machine,while the control group was intervened by rehabilitation training of passive flexion and extension by CPM machine.The American Hospital for Special Surgery (HSS) for knee joint,the American Knee Society score (KSS) and the Western Ontario and the McMaster Universities Osteoarthritis Index (WOMAC) of KOA were used to evaluate the function.Result After the treatment,the HSS,KSS and WOMAC scores were changed significantly in both groups (P<0.05).The improvements of HSS,KSS and WOMAC scores in the treatment group were superior to those in the control group,and the differences were statistically significant (P<0.05).Conclusion The efficacy of thumb pressing at Yangming Meridian plus conventional rehabilitation for post-TKA KOA is superior to single conventional rehabilitation.
10.Diagnosis and treatment of delayed intracranial hematoma after surgery induced by coagulation factor ⅩⅢ deficiency

Objective To investigate the relationship between coagulation factor ⅩⅢ deficiency and delayed intracranial hematoma after craniotomy. Methods The clinical data of 2 cases with delayed intracranial hematoma induced by coagulation factor ⅩⅢ after craniotomy were analyzed and compared with the data of 58 cases with delayed intracranial hematoma induced by other causes during the same period, so as to find out the clinical characteristics of the disease induced by coagulation factor ⅩⅢ. Results The delayed intracranial hematoma induced by the deficiency in coagulation factor ⅩⅢ was commonly located in the operative regions after surgery, and the process was insidiousand fast. Conclusion When the delayed intracranial hematoma is confirmed after craniotomy, the deficiency in coagulation factor ⅩⅢ should be taken into consideration. The prognosis will be satisfying if the diagnosis and treatment are in time.
