1.Mechanism of rat testis toxicity induced by organic extract from Jialing river water
Journal of Third Military Medical University 1984;0(01):-
Objective To study the toxicity of organic extract from river water on the male reproductive system in rats. Methods Organic pollutants were extracted from river water with XAD-2 resin and the extract from 2,16 and 80 L river water was administered daily into 30 male SD rats by gavage for consecutive 28 days.Corn oil served as the solvent control in another ten rats. After the last intake of water extract, all rats were killed, and the sections of testis were prepared according to routine procedure. The TUNEL assay for in situ detection of apoptosis and quantitation of apoptotic germ were performed on testicular sections.Immunohistochemistry and Western blotting were used to examine the changes of scf, fas, fasL and c-kit expressions in rat testis. Results The percentage of apoptotic spermatid cells significantly increased in rats receiving organic extract from 80 L water daily,as compared with the control rats. The spermatocytes in rats receiving organic extract from 80 L water daily overexpressed fas, fasL and c-kit.Conclusion The organic pollutants from river water result in spermatid apoptosis and spermatogenesis arrest.

Chinese Pharmacological Bulletin 1987;0(01):-
Aim To explore the effect of propofol preconditioning on myocardial ischemia/reperfusion injury and the role of phspho-ERK1/2 during the propofol preconditioning in renal hypertension rat(RHR).Methods The isolated RHR hearts perfused on langendorff apparatus were randomly divided into 8 groups(n=8 each). After 20 min of perfusion for equilibration, the CTRL group was subsequently perfused by 148 min.The ISCH group was submitted to 35 min of ischemia and 60 min of reperfusion(I/R) to induce ischemia/reperfusion injury. The DMSO group was given by once 18 min and twice 10 min K-H solution containing 20 ?mol?L~(-1) DMSO and 5 min K-H solution reperfusion prior to I/R procedure. The three propofol preconditioning groups were preconditioned by giving 2 cycles of 10 min K-H solution containing 30 ?mol?L~(-1),100 ?mol?L~(-1) or 300 ?mol?L~(-1) propofol and 5 min K-H solution reperfusion prior to the I/R procedure. The PD group and the PD+P100 group were given K-H solution containing 20 ?mol?L~(-1) PD98059,an ERK1/2 kinase specific inhibitor, for 18 min and 5 min K-H solution washout before I/R and 100 ?mol?L~(-1) PPC respectively.Cardiac functional indices were recorded.Activity of SOD and content of MDA were measured. The level of phspho-ERK1/2 protein expression was measured using Western Blotting. Results Compared with those in the CTRL group, the recovery of cardiac functional indices in ISCH group became worse(P

Chinese Ophthalmic Research 2009;27(12):1068-1072
Objective This study was to observe the effects of connective tissue growth factor(CTGF) on migration and transdifferentiation of bovine lens epithelial cells(BLECs). MethodsThe culture and identification of BLECs adopted the method of Hu(reference 1).The 2-3 passages of BLECs were collected and used in this experiment at the concentration of 1×10~6 cells/hole.The free-serum DMEM containing 0.1 ng/L,0.5 ng/L,1.0 ng/L of CTGF was added into medium for 24 hours in different experimental group respectively, and only equal volume of free-serum DMEM was added in control group.Expression of α-smooth muscle actin(α-SMA) mRNA and protein in the BLECs were examined by semiquantitative RT-PCR and Western blot, respectively. The transwell inserts were used to evaluate the migration ability of BLECs. ResultsThe expression of α-SMA mRNA in cultrued BLECs was gradually increased in different concentrations of CTGF groups.Compared with control group, the expression of α-SMA mRNA in experimental group was significantly enhanced (F=66.56,P<0.01).The expression of α-SMA protein followed the same pattern(F=65.43,P<0.01).The migration ability of BLECs was obviously elevated after CTGF stimulation under the light microscope. The migration rate of BLECs was considerably increased in experimental group compared with blank control group (t=51.7,P<0.01).ConclusionCTGF promotes the migration and transdifferentiation of BLECs at a dose-dependent manner in vitro.CTGF plays an important role in the formation of posterior capsule opacification.

Objective To compare the efficacy and influence on the immune function of oxaliplatin combined with tegafur gimeracil oteracil potassium capsule and mFOLFOX6 method in the treatment of gastric cancer with liver metastasis. Methods One hundred and thirty-four gastric cancer with liver metastasis patients who underwent treatment from Jan.2014 to Jan.2016 in Yulin First Hospital were selected as the participants and randomly divided observation group (67 cases) and control group (67 cases). The control group was treated by mFOLFOX6 method, while the observation group was treated by oxaliplatin combined with tegafur gimeracil oteracil potassium capsule. After treatment, the curative effects were evaluated. The life quality in each group as performed by KPS method before the treatment and in every period of treatment. Before and after the treatment, the levels of CD3 +,CD4 +,CD8 + and CD19 + were detected in two groups and the toxicity was compared. The SPSS 22.0 software was used to analyze relevant data, the measurement data was described by (x ± s) and using t-test,the enumeration data was described by percentage, using the Chi-square test. Results The total effective rate was 52.2% in the observation group, higher than the 38.8% in the control group (P < 0.05).After the third and fourth course of treatment, the karnofsky perfomance status score were (81.3 ± 9.7) and (83.2 ± 9.9),were significantly higher in the observation group than which in the control group (P < 0.05).After the treatment, the levels of CD3 +,CD4+,CD8+ andCD19+ were (54.21 ±6.38)%,(24.11 ±2.41) %,(21.33±3.16) % and (7.18 ±1.13) % in the observation group, and higher than the control group (P < 0.05).During the treatment, there were no serious toxicity occurs in the patients and the rates of toxicity were significantly lower in the observation group than which in the control group (P < 0.05).Conclusion Oxaliplatin combined with tegafur gimeracil oteracil potassium capsule has a better effect and safety for therapy in the treatment of gastric cancer with liver metastasis than mFOLFOX6.

Murine interleukin-4 gene(mIL-4) was transfected into mice glioblastoma cell line G422 by recombinant adenovirus vector. We detected mRNA transcription of target gene in gene modified tumor cells(G422-mIL4). High level of mouse IL-4 could be detected in the culture supernatant of G422-mIL4. When inoculated subcuatneously, the tumor growth of G422-mIL4 was significantly inhibited as compared to wild type G422 and LacZ gene modified G422( G422-LacZ) . The period of survival of mice inoculated with G422-mlL4 was significantly prolongated(p
