1.Mechanism of rat testis toxicity induced by organic extract from Jialing river water

Objective To study the toxicity of organic extract from river water on the male reproductive system in rats.Methods Organic pollutants were extracted from river water with XAD-2 resin and the extract from 2,16 and 80 L river water was administered daily into 30 male SD rats by gavage for consecutive 28 days.Corn oil served as the solvent control in another ten rats.After the last intake of water extract,all rats were killed,and the sections of testis were prepared according to routine procedure.The TUNEL assay for in situ detection of apoptosis and quantitation of apoptotic germ were performed on testicular sections.Immunohistochemistry and Western blotting were used to examine the changes of scf,fas,fasL and c-kit expressions in rat testis.Results The percentage of apoptotic spermatid cells significantly increased in rats receiving organic extract from 80 L water daily,as compared with the control rats.The spermatocytes in rats receiving organic extract from 80 L water daily overexpressed fas,fasL and c-kit.Conclusion The organic pollutants from river water result in spermatid apoptosis and spermatogenesis arrest.
2.Phspho-ERK1/2 involved in propofol preconditioning on ischemia/ reperfusion injury in isolated renal hypertension rat hearts

Aim To explore the effect of propofol preconditioning on myocardial ischemia/reperfusion injury and the role of phspho-ERK1/2 during the propofol preconditioning in renal hypertension rat(RHR).Methods The isolated RHR hearts perfused on langendorff apparatus were randomly divided into 8 groups(n=8 each).After 20 min of perfusion for equilibration,the CTRL group was subsequently perfused by 148 min.The ISCH group was submitted to 35 min of ischemia and 60 min of reperfusion(I/R) to induce ischemia/reperfusion injury.The DMSO group was given by once 18 min and twice 10 min K-H solution containing 20 ?mol?L~(-1) DMSO and 5 min K-H solution reperfusion prior to I/R procedure.The three propofol preconditioning groups were preconditioned by giving 2 cycles of 10 min K-H solution containing 30 ?mol?L~(-1),100 ?mol?L~(-1) or 300 ?mol?L~(-1) propofol and 5 min K-H solution reperfusion prior to the I/R procedure.The PD group and the PD+P100 group were given K-H solution containing 20 ?mol?L~(-1) PD98059,an ERK1/2 kinase specific inhibitor,for 18 min and 5 min K-H solution washout before I/R and 100 ?mol?L~(-1) PPC respectively.Cardiac functional indices were recorded.Activity of SOD and content of MDA were measured.The level of phspho-ERK1/2 protein expression was measured using Western Blotting.Results Compared with those in the CTRL group,the recovery of cardiac functional indices in ISCH group became worse(P
3.Comparative study of totally laparoscopic and open main bilateral femoral artery bypass for the treatment of advanced atherosclerosis

Objective To comparatively complete clinical effectiveness of laparoscopic and open-bifemoral bypass in the treatment of advanced atherosclerosis.Methods During January 2008 to January 2013,according to the treatment method,a total of 60 patients with advanced atherosclerosis patients were divided into group A and group B,and there were 30 cases in each group,and group A was treated with open master bifemoral bypass surgery,and group B was treated with laparoscopic totally master bifemoral bypass surgery,to compare the general information of the two groups,surgery related indicators,clinical efficacy and survival rate,and to analyze the effect of surgical treatment on the prognosis of patients by COX model.Results The operation time of patients in group B (365.3 ±41.3) min and aortic clamping time (59.5 ± 18.3) min were significantly higher than that of group A,(294.3 ±35.5) min and(26.5 ± 19.3) min (P <0.05),bleeding amount (400.0 ± 145.3) ml and length of hospital stay (10.5 ± 2.1) d of group B patients was significantly lower than that of group A (1 002.3 ± 324.3) ml and (16.4 ± 4.3) d (P < 0.05);postoperative mortality,thrombosis rate and the incidence of serious complications of patients in group B were significantly lower than group A (P < 0.05);survival rate at different time of group B was significantly higher than that of group A (P < 0.05);COX model analysis show that the surgical methods affect the postoperative mortality,thrombosis and the occurrence of severe complications.Conclusions Completely laparoscopic master bifemoral bypass can significantly decreased advanced main iliac artery occlusion disease mortality,and has small surgical trauma,less postoperative complications,rapid recovery and other advantages,especially for elderly critically ill patients,and it was worthy of clinical application.
4.Systematic review of accuracy of plasma matrix metalloproteinase-9 in predicting hemorrhagic transformation in ischemic stroke

Objective To study the accuracy of plasma matrix metalloproteinase-9 (MMP-9) predicting hemorrhagic transformation (HT) in ischemic stroke. Methods The studies on plasma MMP-9 predicting HT in ischemic stroke were included, in which diagnostic criteria, methods, and subjects were clearly described, regardless of language of publication and type of studies. MEDLINE (Apr, 1966 to 2006), EMBase (Apt, 1966 to 2006), CNKI (Feb, 1977 to 2006), and wanfangshuju (1989 to 2005) were searched and the references lists of eligible studies were manually searched. Two reviewers independently evaluated the quality of studies and extracted data. The data were analyzed using the Revman 4. 2 or SPSS 11.0. Results Five trials including 497 patients fulfilled the inclusion criteria, 109 of the 497 patients developed HT. The heterogeneity among studies was great (xspecialty 89. 9% (95% CI: 86. 4%-93.5%), sensitivity 86. 5% (95% CI: 77. 3%-96. 8%). Conclusions The values of plasma MMP-9 may be an independent predictor of hemorrhagic transformation. The number of patients in the included studies is not large enough, and the results need to be confirmed in further studies.

Murine interleukin-4 gene(mIL-4) was transfected into mice glioblastoma cell line G422 by recombinant adenovirus vector. Pseudomyxoma peritonei is a rare kind of clinical disease,mainly characterized by mucus exocrine cells grown in the peritoneum or omentum sticky which caused lots of jelly-like ascites.There is no obvious clinical specificity performance for pseudomyxoma peritonei,which is always misdiagnosed.By enhancing the awareness of pseudomyxoma peritonei,improving the relevant checks and reasonably extracting specimens for pathological examination can improve diagnosing rate of pseudomyxoma peritonei.Using complete cytoreductive surgery or major debulking surgery combined with intraperitoneal hyperthermic perfusion chemotherapy help to improve the survival of patients with pseudomyxoma peritonei.

To investigate the expression of ? adrenoceptors(? AR) and its possible function in rat detrusor. The relaxation response produced by ? AR subtypes(? 1 , ? 2 , ? 3 AR) agonists or antagonists were examined by means of the in vitro detrusor strip study, and the expression of ? AR subtypes mRNA was investigated in rat detrusor muscle by RT PCR. The selective ? 1 agonist T 0509, ? 2 agonist salbutamol, ? 3 agonist BRL37344A depressed the contractions of the detrusor muscle induced by KCl. The relaxant response to isoproterenol was antagonized by the ? 1 ,? 2 AR antagonists(100?mol/L) and the ? 3 AR antagonists SR59230(1?mol/L).RT PCR, showing that all three subtypes of ? AR subtypes mRNA were expressed in rat detrusor smooth muscles, in which ? 3 AR mRNA occupied 70%. All the ? 1 ,? 2 ,? 3 adrenoceptors participate in mediating relaxation of rat detrusor, among them ? 3 AR is the main one.

To investigate the role of ?and ? 3 adrenoceptors(? 3 AR) in rat detrusor instability secondary to bladder outflow obstruction(BOO). Animal models of detrusor instability were replicated in female rats, the relaxation response produced by ISO and ? 3 AR agonists BRL37344A were examined by means of in vitro detrusor strip study. ? and ? 3 adrenoceptors were detected in bladder specimens with radioligand binding assay. The concentrations of ISO and BRL37344A to produce relaxation were different in normal, stable and instable groups.
