1.Detection of circulating tumor cells and real-time personalized cancer therapy

Serious troubles were caused by the highly heterogeneous cancer cells in clinical cancer treatment.In recent years,molecular characterization of tumors and corresponding individualized cancer management has become hotspots for cancer research.Circulating tumor cells (CTCs) are shed from primary solid tumors or metastases into the peripheral blood.CTCs could serve as liquid biopsy for patients with cancer,with non-invasive,real-time and repeatable access.Serial monitoring of the amount and molecular characteristics of CTCs in the blood samples has significant clinical value for prognostic prediction,targeted drugs choice and real-time evaluation of clinical effectiveness in individualized cancer treatment.This review summarizes current methods for the CTC isolation and detection,and discusses the perspectives of CTC analyses in real-time personalized cancer therapy.Some future research directions in this field are proposed.
2.Dexamethasone inhibits cervical carcinoma cell xenografted tumors in nude mice

Objective To investigate the effects of over physiological dose of dexamethasone on cervical carcinoma cell xenografted tumors in nude mouse.Methods HeLa cells were inoculated subcutaneously in BALB/c female mice aged 5-6 weeks.Over physiological dose of dexamethasone(62.5 mg?kg-1?d-1) was injected into the peritoneum from the 16th day after inoculation for 10 d.Diameters of tumor were measured every two or three days.The volumes and the inhibition rates of tumor were calculated.Apoptotic bodies were observed by transmission electron microscopy and counted by TUNEL.VEGF and TNF-? were detected by immunohistochemistry.Results In vivo,over physiological dose of dexamethasone can repress the growth of tumor and induce cell apoptosis.Compared with the control group,the tumor sizes were significantly smaller(P0.05).Conclusion Over physiological dose of dexamethasone can repress cervical tumor growth in vivo.This action may be related to apoptosis and VEGF,but have no relationship with TNF-?.
3.Improvement of surgical technique for adult-to-adult living donor liver transplantation

Objective To investigate the modifications of surgical technique in adult-to-adult living donor liver transplantation (A-A LDLT) using right lobe liver grafts.Methods From Jan.2002 to Aug.2005,16 patients underwent living donor liver transplantation using right lobe grafts.During operation,a modification,designed to improve the reconstruction of right hepatic vein,the reconstruction of the tributaries of the middle hepatic vein by interpositing a vein graft,and the anastomosis of the hepatic arteries and bile ducts,was done.Results In these series,there were no donor severe complication and death.The graft and recipient weight ratio(GRWR) was between(0.72%) and(1.24%),among these,9 cases

Coronary heart disease (CHD) is a common gerontal disease, and percutaneouse coronary interangioplasty (PCI) has already become a main treatment in CHD. However, restenosis to some extend has limited clinical application of PCI. The advent of drug eluting stents brings optimistic prospect in preventing restenosis.

AIM:To investigate the effect of advanced glycation end products on inflammation in cultured cardiomyocytes.METHODS: Primary cardiomyocytes were isolated from Sprague-Dawley neonatal (1 to 2 days old) rats ventricles. The insulin resistant cardiomyocyte model was established. Neonatal rat ventricular myocytes were exposed to AGEs for 24 hours. TNF-? mRNA and PPAR-? mRNA expressions were determined by RT-PCR. Activation of NF-?B in the cells was examined by using immunocytochemistry. 7.Preliminary results of combined high intensity focused ultrasound and radiotherapy on rectal carcinoma in 121 cases

Objective To assess therapeutic effects and safety of high intensity focused ultrasound ( HIFU ) and radiotherapy for the treatment of rectal carcinoma. Methods In combination with low dose of radiotherapy, we treated 121 cases of rectal carcinoma with FEP BY01 type HIFU therapy from Jul.1998 to Dec.2000 and 43 cases were followed up for more than one year. ResultsComplete response (CR) was achieved in 27 cases(22%), partial response (PR) in 80 cases (66%),and no change(NC)was found in 14 cases (12%) with a total response rate of 88 4%.There were no complications. One year survival rate was 81%(35/43) and 2 year survival rate was 77%(23/30). ConclusionHIFU in combination with radiotherapy is a new method to treat rectal carcinoma with significant therapeutic effect and high safety.
8.Effect of Dexamethasone on tumor volume and apoptosis of transplanted HeLa cells in mice

Objective The aim of this study was to investigate the distribution and clinical significance of CD8+ T cells in bladder cancer tissues in situ.Methods Immunohistochemistry were used to examine the distribution of CD8+ T cells in bladder cancer tissues,which were obtained from January 2003 to December 2009 from 302 patients.Among all the patients,262 were male while 40 were female;mean age is 60 years;tumor size ≤ 3 cm was in 235 and tumor size > 3 cm was in 67;Unifocal tumor was in 214 and multifocal tumors were in 88.Amount of tumor stage Ta-T1 was 212 and T2-T4 was 90.Sixteen patients have lymph node metastasis.Histological low grade was diagnosed in 175 and histological high grade was diagnosed in 127.According to the differences between anatomic structure and cellular composition,bladder tumor tissues can be classified to two localization patterns:(1) intratumoral regions,defined as tumor cell nests;(2) stromal regions,defined as stromal areas that lack direct contact with tumor cells.Therefore,we divided 302 bladder cancer patients into two groups based on the median frequency of intratumoral CD8+ T cells (median,3/× 400 high resolution) and stromal CD8+ T cells (median,37/× 400 high resolution),respectively.x2 analysis was used to evaluated the correlation between CD8+ T cell density and clinicalpathological variables.Kaplan-Meier analysis and Cox proportional hazards regression models were applied to estimate overall survival (OS).Results CD8+ T cells were predominantly located in the intratumoral regions (mean,14 ± 2/× 400 high resolution) rather than in associated stromal regions (mean,50 ± 3/× 400 high resolution,P < 0.05).The density of intratumoral CD8+ T cells was inversely associated with age (P =0.026),tumor size (P < 0.05) and tumor stage (P < 0.05),and could represent a favorable prognostic predictor of OS (HR =0.427,P =0.003).However,the density of stromal CD8+ T cells was positively associated with age (P =0.004) and histological grade (P < 0.01),and could represent an adverse prognostic predictor of OS (HR =2.206,P =0.009).Conclusions Our findings suggest that intratumoral/ stromal CD8+ T cells could potentially serve as favorable/ adverse prognostic markers for bladder cancer patients,respectively.
