1.The Induction of hsp70 mRNA under Heat Stress

The induction of hsp70 mRNA in human osteosarcoma cells (HOS-8603) and intacl rats under heat stress was studied using hsp70 cDNA labeled with a-32P dATP. The results showed that the induction of hsp70 mRNA was evident in liver, lung, spleen and the most evident was found in brain when the core body temperature of rats was brought to 42℃ for 15 min. The induction of hsp70 mRNA in HOS-8603 was also significant when cells were cultured at 42℃ for 30 min. These results indicated that hsp70 mRNA could be induced both in vitro and in vivo under heat stress condition and the induction of hsp70 mRNA in intact rats was of tissue-specific fashion.
2.Investigation on the Quality of Life about Rural Elderly Population and Its Influencing Factors

Objective:To understand the quality of life about rural elderly population and its influencing factors.Methods:Using WHOQOL-BREF scale and self-designed questionnaire,a survey about quality of life was conducted among 1074 rural elderly in Shandong Province selected by stratified,random method.T test,variaace analysis and multiple linear regression were used to analyze the influencing factors.Results:The score of environment field was the highest (13.83 ± 1.05),while social relations field was the lowest (8.63± 0.45).Single factor analysis showed that age,education level,economic situation,pension source,chronic disease,pension mode,physical exanination had significant effect on quality of life scores in each dimension (P < 0.05).Marital status only had significant effect on psychological and social relations scores (P < 0.05).Gender and the number of children were not statistically significant on the quality of life score in each dimension (P > 0.05).Multiple regression analysis showed that the influencing factors of quality of life score in each dimension were chronic disease and economic situation.Physical examination only had significant effect on the physiology and psychological score.Pension mode had significant effect on psychological and social relations score.Age influenced physiology health score.Marital status influenced psychology health score.Conclusion:The government should pay attention to the health status of elderly and the psychological status of elderly without spouse;increase the support efforts to the poor rural elderly;strengthen the prevention and treatment of chronic diseases;improve the social support network and social security system.
3.Distribution of cardiocerebral vascular disease death and its life expectancy eliminating causes of death in Shandong Province in 2012

Objective To explore the impact of cardio cerebral vascular diseases (CVD) on life expectancy among residents in Shandong Province,develop effective preventive measures for the high-risk population and provide guidance for the health care resource allocation.Methods Based on the death monitoring data of the disease in Shandong Province in 2012,we calculated the crude mortality and standardized the mortality rate of the disease.We designed life tables with all causes of death and with eliminating causes of CVD death and analysed life expectancy with and without them examined.Results In 2012,crude mortality rate of CVD was 320.16/100 000 (SMR 252.85/100 000) in Shandong Province.The death rate of CVD was of 46.30% of all deaths reported by monitoring system.The mortality rate of CVD in male and female were 332.99/100 000 and 307.02/100 000,respectively.The mortality rate of CVD in rural and urban areas were 341.84/100 000 and 267.28/100 000,respectively.The mortality of CVD increased with age and growed rapidly after the age of 60.Life expectancy of urban and rural areas was 79.20 years and 78.14 years,respectively.After eliminating causes of CVD death,life expectancy for eliminating causes of CVD death in urban and rural areas increased 8.51 years and 10.02 years,respectively.Life expectancy for eliminating causes of CVD death in male and female increased 8.02 years and 11.10 years,respectively.Conclusions Life expectancy of urban and rural areas,the male and female for eliminating causes of CVD death than that of all causes of death were greatly extended.Reducing the death caused by CVD to increase health life expectancy have important public health significance.
4.Analyzing contradictions of video-case teaching in social medicine classroom

Based on practice of video-cases teaching,this paper put forwards four sorts of contradictions,which are the contradiction between time limitation of classroom and expressing limi-tation of video cases,the contradiction between relative independence of video cases (part)and con-sistency of teaching objective (entirety),the contradiction between apparentness of video cases (phe-nomenon)and depth of teaching content (essence)and the contradiction between variability of video case class and control of class by teachers. This paper suggested teaching principle of part conforming to entirety. Meanwhile,it indicated that classroom video case teaching should have strict design, closely coordinated class discussion and teachers should be good at guiding and summarizing. Through these efforts,the above four contradictions can be resolved and designed video case teaching obtained good effect.
5.Establishment of TaqMan probe real-time PCR for detection of Cryptococcus neoformans

Objective To establish TaqMan probe real‐time Fluorescent Quantitative PCR in detecting Cryptococcus neoformans genomic DNA ,and to provide important method for detection of cryptococcal meningitis .Methods According to the Cryptococcus neoformans ITS‐rDNA sequences obtained from NCBI ,specific primers and probe were designed based on the conserved sequences , a specific 114 bp fragment was amplified by primers and probe ,then recombinant plasmid was constructed .Eight different concen‐trations from 1 .42 × 10 copy/μL to 1 .42 × 10 copy/μL were used as templates by 2 μL .In the optimum reaction condition ,the sen‐sitivity ,specificity and repeatability were evaluated and standard curve was established .15 clinical cryptococcal meningitis strains i‐solated from clinical diagnosis patients were detected .Results The real‐time PCR showed high sensitivity and specificity and was a‐ble to detect 2 .84 × 102 copies of plasmid DNA .The detection sensitivity was 2 .84 × 102 copies plasmid DNA by real‐time PCR ,no amplification curve was detected with human genomic DNA ,other fungus ,bacterias and viruses .The CV of inter‐assay were 2 .86% ,1 .48% and 1 .36% respectively with excellent reproducibility .Fifteen clinical isolated strains could be detected accurately . Conclusion A method of detection of Cryptococcus neoformans DNA by real‐time PCR is established successfully with high sensi‐tivity and specificity ,and the results are stable ,could diagnose cryptococcal meningitis rapidly and early .
6.Staphylococcal protein A immunoadsorption plus nonmyeloablative chemotherapy with CD34+ autologous peripheral blood stem cell transplantation in the treatment of three cases of refractory systemic lupus erythematosns

Objective To observe the clinical efficacy of staphylococcal protein A immunoadsorption plus nonmyeloablative chemotherapy with CD34+ autologous peripheral blood stem cell transplantation in the treatment of refractory systemic lupus erythematosus (SLE). Methods Three patients with active SLE were enrolled into this study. All patients were diagnosed with lupus nephritis by renal biopsy and poorly responded to routine therapy. Before transplantation, patients were given 6 sessions of immunoadsorption apheresis using columns of staphylococcal protein A-silica with an interval of 3 days; each session processed 3 L plasma and a total of 18 L plasma was processed over the 6 treatments. Three days following the immunoadsorption apheresis, the mobilization of stem cells was realized by intravenous cyclophosphamide at a dose of 2 g per square meter of body surface area and subcutaneous recombinant human granulocyte colony-stimulating factor (G-CSF) at a dose of 5 g per kilogram of body weight per day for 5 days. Then, peripheral blood raonoclonal cells were obtained by CS-3000 Cell Separator, and passed through the Clini Macs CD34+ cell selection device, with the final concentration of CD34+ cells being 2.6×106, 2.1×106 and 2.4×106 per kilogram of body weight respectively, and that of CD3+ cells being 3×105, 2.1×105, and 2.0×105 per kilogram of body weight, respectively, in these three patients. The conditioning regimen consisted of oral fludarabine of 50 mg/d for 5 days plus intravenous pig anti-human thymocyte immunoglobulin (ATG) at a daily dose of 90 mg/kg for 5 days. After 72-hour treatment with ATG, the frozen stem cells were infused back to the patients. Clinical manifestations and lupus-correlated immune parameters were compared in patients at baseline and after transplantation. Results Following immunoadsorption apheresis, an obvious decrease was observed in the level of serum anti-dsDNA, antinuclear antibody and IgG antibodies, while an increase in the level of serum complement 3. All patients achieved the reconstruction of hemopoiesis 2-3 days after the transplantation. Also, an apparent clinical remission was achieved with the SLEDAI score being less than 3. Six months after the transplantation, serum anti-dsDNA and antinuclear antibodies as well as urine protein were undetectable, the level of complement 3 reached the normal range, and renal function was restored. Conclusions Staphylococcal protein A immunoadsorption plus nonmyeloablative CD34+ autologous peripheral blood stem cell transplantation are effective and safe for refractory SLE, but the long-term effect remains to be connfirmed by further studies.
7.Modulation on the P-glycoprotein in the jejunum by combined use of Glycyrrhiza inflata and Kansui.

To investigate the modulation on the P-glycoprotein in the jejunum by combined use of Glycyrrhiza inflata and Kansui with ussing chamber and rt-pcr, Rhodamine 123 (R123), a P-gp substrate and fluorescein sodium (CF), a model drug of non-P-gp substrate transported by a passive diffusion were taken as investigational drugs. Because these two drugs can be easily assayed and widely used in various research fields. The permeability of R123 or CF via Wistar rat jejunum membranes was evaluated by in vitro ussing chamber after oral administration of four different decoctions of Glycyrrhiza inflata and Kansui for 1 week. And the concentration of R123 or CF was determined by the fluorospectrophotometry in the receiving solution. Meanwhile the expression of mdr1a in P-glycoprotein was detected by real-time fluorescent quantitative PCR. After oral administration of combined decoction of the single drug, the absorptive directed permeability of R123 increased significantly (P < 0.01). On the other hand, Kansui and combine decoction of the two drugs also decrease the permeability of secretory directed transport (P < 0.05). No action of Glycyrrhiza inflata was found on the secretory transport of R123 [Papp = (2.56 +/- 0.38) x 10(-5), cm x s(-1)] across the jejunum tissues, while Papp of control group was found [Papp = (2.35 +/- 0.27) x 10(-5), cm x s(-1)]. After oral administration of Kansui decoction for 1 week and 2 weeks, the levels of mdr1a expression in Wistar rats were lower than that of the control group, but there were no significant difference in the results. Meanwhile, Glycyrrhiza inflata had no effect on transport of CF across the jejunum tissues, though the other three groups could decrease the permeability of CF, as compared with control group. Kansui may slightly inhibit P-glycoprotein function in the intestinal membrane. For another, some compositions in Kansui inhibit P-glycoprotein function, and some others strengthen the tight junction between cells in the intestinal membrane to decrease permeability of CF. As the inhibitory action to P-glycoprotein was enhanced by combination of Glycyrrhiza inflata and Kansui, based on the results, it may be one of the mechanisms of creating toxicity once co-administration of Glycyrrhiza inflata and Kansui.
8.CLINICAL ANALYSIS AND TREATMENT OF ACUTE LUNG INJURY AFTER LIVER TRANSPLANTATION

Objective To analyze causative factors and treatment of acute lung injury (ALI) after liver transplantation. Methods the correlative parameters, including electrocardiogram, blood oxygen saturation, blood pressure, hemodynamics, blood sugar, analysis of arterial blood gases, and other clinical examinations were monitored dynamically. The recipients were treated with oxygen inhalation, antibiotics, immunosuppressant, drugs for liver protection, nutritional support, diuretics, and vasodilators. Results Three of a total of 21 patients died within 48h post operation. Fourteen patients suffered from ALI (14/18, 77 8%), who recoverd after receiving appropriate treatment. Conclusion The occurrence of ALI is related to the primary disease, operative trauma, instability of the hemodynamics, and the quantity of blood loss. Systemic treatment is necessary for the patients with ALI besides the oxygen therapy.
9.Effects of Sorafenib on the cytotoxic sensitivity of nasopharyngeal carcinoma cells with high expression of ATP-binding cassette superfamily G member 2(ABCG_2)

Objective To investigate the mechanisms and effects of Sorafenib on cytotoxic sensitivity of allo-reactive natural killer(Allo-NK) cells against human multi-drug resistant nasopharyngeal carcinoma CNE2/DDP cells which expressing highly ATP-binding cassette superfamily G member 2(ABCG2)(abbr.as ABCG2HighCNE2/DDP cells).Methods ABCG2HighCNE2/DDP and Allo-NK cells were isolated by magnetic bead technique.The target cells were divided into 3 groups: a) treated group(ABCG2HighCNE2/DDP cells incubated with 10 ng/ml sorafenib for 4h);b) untreated group(conventionally cultured ABCG2HighCNE2/DDP cells);and c) control group(conventionally cultured K562 cells).Expression rates of ABCG2 in treated and untreated groups,and of five NKG2D-ligands(MICA,MICB,ULBP1,ULBP2,ULBP3) were evaluated by flow cytometry.The cytotoxic effects of NK cells against different groups of target cells were detected with LDH releasing assay.Results Expression rate of ABCG2 in isolated CNE2/DDP cells was 91.40%?2.32%.The purity of sorted CD3-CD16+CD56+ Allo-NK cells was 90% and higher.The expression rates of NKG2D-ligands(MICA,MICB,ULBP1,ULBP2 and ULBP3) in untreated group were 2.92%?0.33%,4.27%?0.33%,5.80%?0.62%,11.10%?3.15% and 7.75%?1.14%,respectively,which were remarkablely higher than that in treated group(10.38%?1.23%,10.68%?1.26%,11.62%?1.22%,43.24%?4.42% and 11.91%?0.88%,respectively,P
10.Regulation of blood sugar and electrolytes in nonage of liver transplantation

Objective To investigate the levels of blood sugar and electrolyte in nonage of 125 patients with liver transplantation.Methods According to the parameters of blood sugar and electrolytes,the usage of insulin and electrolyte was adjusted whenever necessary.The levels of blood sugar and electrolytes were maintained respectively in 6~10 mmol/L and normal limits.The oxygen inhalation,antibiotics,immunosuppressant,liver protectant,diuretics and vasodilators were also used in the preoperative period.Results Of 125 cases of liver transplantation recipients,there were 50(40%) cases with blood sugar heightened remarkably.Massive potassium chloride and sodium chloride was used to keep electrolytes in normal limits.Conclusion The levels of blood sugar and electrolytes are not stable in nonage of liver transplantation and it is very important to maintain them in normal limits for the success of liver transplantation.
