1.Surgical treatment and prognosis analysis of solid pseudo papillary tumor of the pancreas:a report of 45 cases

Objective To discuss the surgical treatment and prognosis of solid pseudo papillary tumor of the pancreas. Methods The clinical data of 45 patients with solid pseudo papillary tumor of the pancreas were analyzed retrospectively. Surgical treatment was performed in all cases,including 11 cases of local excision of the pancreas,20 cases of resection of the pancreatic body and tail,5 cases of resection of the pancreatic body,tail plus spleen,and 9 cases of pancreaticoduodenectomy. Results A total of 43 ca-ses were followed up for 6 ~ 160 months,with an average of(41. 5 ± 1. 5)months. There were 8 cases of dyspepsia,2 cases of pancreatic fistula,2 cases of pleural effusion,2 cases of incision liquefaction and in-fection,and 1 case of early hemorrhage of the digestive tract. All patients were cured by conservative treat-ment. No metastasis,recurrence or death occurred after the operation. Conclusion The pseudo papillary tumor of the pancreas is a potential low grade malignant tumor which mainly appears in females. Surgery is the only radical treatment and its prognosis is generally good.
2.Simultaneous Determination of 5 Effective Components in Luohua Zizhu Dry Extract by UPLC-MS/MS

OBJECTIVE:To establish a method for simultaneous determination of 5 effective components in Luohua zizhu dry extract. METHODS:UPLC-MS/MS was conducted. The separation was performed on an Zorbax Eclipse Plus C18 column with mo-bile phase of acetonitrile-water(gradient elution)at the flow rate of 0.3 mL/min. The column temperature was set at 40℃and sam-ple size was 2 μL. The analytes were detected in the multiple reaction monitoring(MRM)mode. Nitrogen was used as drying gas and atomized gas. The temperature and flow rate of drying gas were 325 ℃ and 6 L/min. The pressure of atomized gas was 45 psi. The temperature and flow rate of sheath gas were 350℃and 12 L/min. The voltage of capillary were 4000 V(+)and 3500 V(-). The voltage of nozzle was 500 V. RESULTS:The linear ranges of luteoloside,acteoside,quercetin,luteolin and rutin were 0.5048-252.4 ng/mL(r=0.9999),0.7124-356.2 ng/mL(r=0.9990),0.5094-254.7 ng/mL(r=0.9962),0.3030-151.5 ng/mL(r=0.9998) and 0.6022-301.1 ng/mL(r=0.9996),respectively. RSDs of precision,stability and reproducibility tests were all less than 3.0%. The limit of quantitation were 0.42,0.87,0.33,0.12,0.76 ng/mL. The recoveries were 97.99%-101.20%(RSD=1.3%,n=6), 96.50%-101.20%(RSD=1.7%,n=6), 94.81%-99.34%(RSD=1.7%,n=6), 97.54%-100.51%(RSD=1.2%,n=6), 93.37%-98.70%(RSD=1.9%,n=6),respectively. CONCLUSIONS:The method is simple,precise,stable and reproducible, and can be used for simultaneous determination of 5 effective components in Luohua zizhu dry extract.
3.Analysis in etiology and risk factors of severity in 500 patients with acute pancreatitis in Xinjiang area

Objective To explore the etiology and the risk factors of severity in patients with acute pancreatitis (AP) in Xinjiang area.Methods A total of 500 patients with AP were selected,and the age,gender,etiology,clinical pancreatitis types were analyzed.Patients were divided into different groups according to alcoholic,biliary,hyperlipidemia,diet,other causes,multiple factors,idiopathic pathogens,and the patients were also divided into different groups according to age such as 18 to 24,25 to 39,40 to 64 and over 65.The distribution of etiology in patients with different gender,age and severity were analyzed.Relationship between procalcitonin,C reactive protein and severity of acute pancreatitis were analyzed.Results There were 129 cases with biliary acute pancreatitis (25.8 %),which were significantly higher than those of patients with other pathogens (P < 0.05).The proportion of female patients with biliary disease was significantly higher than that of male patients (P < 0.05),the proportion of female patients with alcoholic etiology was significantly lower than that of male patients (P < 0.05),and the proportion of hyperlipidemia was high in the etiology of both male and female patients.The proportion of AP was the highest in the etiology in each age group (P < 0.05).The proportion of biliary was the highest in the etiology of MAP (P < 0.05),followed by idiopathic and other factors.Proportion of hyperlipidemia was the highest in the etiology of SAP (P < 0.05).The levels of PCT and CRP in patients with SAP were significantly higher than those in patients with MAP (P < 0.05).The levels of PCT and CRP were positively correlated with Ranson score,Balthazar CT score and APACHE score.Conclusion The occurrence of AP is related with multiple factors,in which biliary tract disease is the main risk factor of AP.

5.Molecular mechanism of regulating miR-223-3p/RHOB expression by circRNA SAMD8 to inhibit progression of pancreatic ductal adenocarcinoma

Objective To investigate the potential mechanism of circRNA SAMD8(circ-SAMD8)in development of pancreatic ductal adenocarcinoma(PDAC).Methods The expression profile of circRNAs in PDAC tissues was analyzed based on Microarray data(GSE79634).Quantita-tive real-time polymerase chain reaction(qRT-PCR)was used to verify the expression of circ-SAMD8(hsa_circ_0006148)in PDAC tissues and cells(CFPAC-1 and PANC-1).The target microRNA(miRNA)of circ-SAMD8 and its downstream mRNA were predicted by bioinformatics analysis,and identified by double luciferase reporter gene assay.The proliferation ability of PDAC cells was detec-ted by MTT assay and colony formation assay.The expression levels of anti-apoptotic protein Bcl-2 and pro-apoptotic protein Bax were detected by Western blot.The percentage of apoptotic cells was detected by flow cytometry.Results The expression levels of circ-SAMD8 in PDAC tissues and cells were significantly lower than those in paracancer tissues and normal cells(P<0.05).Overexpression of circ-SAMD8 could effectively promote apoptosis of PDAC cells and inhibited proliferation of PDAC cells.Dual luciferase reporter gene experiments showed that miR-223-3p was a potential interacting mol-ecule of circ-SAMD8,and the downstream mRNA target of miR-223-3p was RHOB.The miR-223-3p was abnormally overexpressed in PDAC tissues and cells,accompanied by low RHOB expression.In PDAC cells transfected with miR-223-3p mimics or sh-circ-SAMD8,RHOB expression was significantly decreased,proliferation ability was enhanced,and apoptosis rate was decreased.Conclusion Circ-SAMD8 regulates the expression of RHOB downstream by sponging miR-223-3p,and effectively inhibits the occurrence and development of PDAC.
