1.THE ROLE OF PROTEIN KINASE B IN THE APOPTOSIS OF GASTRIC CANCER CELLS INDUCED BY 12-O-TETRADECANOYLPHORBOL-1,3-ACETATE
Acta Anatomica Sinica 1954;0(02):-
Objective To study the role of protein kinase B(PKB) in gastric cancer BGC-823 cells treated by TPA. Methods Flow cytomety(FAC) and fluorescence microscope were used to detect the effect of TPA on gastric cancer BGC-823 cells, after cells were treated by BrdU and stained by DAPI,respectively. The effect of TPA on the expression level of PKB protein and its phosphorylation of Ser 473 and Thr 308 were investigated by Western blotting. When nuclear and cytoplasmic protein fractions were prepared through lysis of cell and centrifugation, the effect of TPA on the expression level of PKB protein and its phosphorylation of Ser 473 in cytoplasm and nuclei were detected by Western blotting.Meanwhile, the effect of TPA on the distribution of PKB was observed under laser-scanning confocal microscope with immunofluorescence technique. Results TPA induced the apoptosis of gastric cancer BGC-823 cells. It also induced the inhibition of PKB protein expression in a TPA concentration-dependent and time-dependent manner in gastric cancer BGC-823 cells, regardless of dephosphorylation of PP2A.TPA inhibited the phosphorylation of PKB at Ser 473,but it did not affect its phosphorylation at Thr308.TPA only attenuated the expression level of PKB and its phosphorylation of Ser 473 in nuclei, whereas it did not change the distribution of PKB in BGC-823 cells. Conclusion These data suggest that the inhibition of PKB plays an important role in mediating the effect of TPA in gastric cell line.Apoptosis induced by TPA is partly due to the inhibition of PKB and its phosphorylation of Ser 473 in nuclei.
2.Stem cells in optic nerve protection and restoration following injury
Chinese Journal of Tissue Engineering Research 2010;14(23):4367-4370
BACKGROUND: As a specially differentiated central nerve, optic nerve is once injured, retinal neurons are hard to restore and regenerate. As a result, visual function is lost permanently. Currently, an effective clinical treatment is not available. Due to the multi-directional differentiated potential of stem cells, their role in optic nerve protection and restoration following injury is becoming a hot research spot.OBJECTIVE: To review the classification and transplantation of stem cells in optic nerve protection study, as well as roles of stem cells transplantation in optic nerve protection and repair following injury.METHODS: The database of ISI Web of Knowledge platform was searched on computer from February 1994 to April 2009, with the key words "stem cells, optic nerve" in English. In the same way, the database of CNKI from February 1994 to April 2009 was also screened, with the key words "stem cells, optic nerve" in Chinese.RESULTS AND CONCLUSION: A total of 98 literatures related to stem cells in optic nerve protection and restoration after injury are collected, in which 23 papers are Chinese and 75 papers are English. The studies published earlier, repetitively and similarly are removed, and 38 papers which comply with the standards are involved into this review. Stem cells own the potential of self-renew physiologically, as well as potential of repairing the injuries. At present, most of the researches on stem cells in optic nerve protection and restoration after injury are represented in morphosis. How to implant stem cells safely and effectively and how to recover stable visual function still need our efforts. Stem cells derived from skin, peripheral blood, fat and other tissues are convenient to draw materials and are commendable subjects, we should study them deeply to make preferable use.

METHODS The experience of hospital infection control in disaster tent medical posts under special conditions was summarized. RESULTS Under the difficult conditions due to strict management system of infection control and strengthening the posts management, strict disinfection to reduce the spread of the infection was prevented. There were 15877 injured victims treated in disaster tents medical posts and 80 of them were seriously injured, but no infectious disease outbreak incidents were happened, and the safety of every victim and medical staff was ensured. CONCLUSIONS Strengthening management and strictly conducting disinfection and isolation system especially in primitive conditions could prevent the onset and spread of the nosocomial infection.
5.Epidemiological Characteristics of Anaphylactic Shock Caused by Qingkailing Injection
Traditional Chinese Drug Research & Clinical Pharmacology 1993;0(01):-
Objective To investigate the causations and rules of anaphylactic shock caused by Qingkailing Injection.Method The analysis was conducted on 66 cases of anaphylactic shock caused by Qingkailing Iinjection collected from medical journals in 1978~2005 by epidemiology and bibliometrics methods. Results There were a large number(40.91 %)of young sufferers from age 18 to 44 in the cases reported. The male cases was 29(44.62 %)and the female cases was 36(55.38 %)in the 65 cases with gender recordation. In primary disease, the upper respiratory tract infection rate was up to the highest 75.76 %.The dosage of Qingkailing Injection was in the range of 20~40 mL,the occurrence of anaphylactic shock was commonly within 30 mins after injection, and 68 %was within 10 min.The anaphylactic shock patients were cured after emergency treatment except 5 were died. Conclusion Qingkailing Injection can induce dangerous even fatal anaphylactic shock, its mechanism may be related to the constitution of the patient, ingredient of the drug self and diagnosis warp.Treatment based on syndrome differentiation combined with traditional Chinese medicine theory, dosage control, unnecessary compatibility decrease, prevention consciousness should be pay more attention in the clinical application.
6.Medical Records Database and Epidemiological Characteristics of Adverse Reaction Caused by Compound Salviae Miltiorrhizae Injection
Traditional Chinese Drug Research & Clinical Pharmacology 1993;0(02):-
Objective To investigate the epidemiological characteristics of adverse reaction(ADR) caused by compound Salviae miltiorrhizae injection. Methods By searching domestic medical periodicals issued in the years of 1978~2006,115 cases of ADR detailed records of compound salviae miltiorrhizae injection were collected and database of ADR cases was established by ACCESS.On the basis of database, epidemiological characteristics of the cases were analyzed by statistic methods. Results The incidence of ADR was the highest(70.43 %)in the middle-aged and old sufferers, and was 50.43 %(58) in the male and 49.57 %(57) in the female. In the primary diseases, coronary artery disease shared the highest of incidence(32.00 %).The incidence of generalized lesion(37.40 %) and that of lesion of skin and its appendages(25.95 %)dominated in ADRs,and ADRs usually occurred within 30 min after first injection(64.37 %).Conclusion The establishment of ADR database is beneficial to the storage and analysis of ADR information.Compound Salviae miltiorrhizae injection can induce multi-systemic lesion, and its mechanism is complicated. Therefore, clinician should pay more attention to the prevention of ADR of compound Salviae miltiorrhizae injection.
7.THE IMPLICATION OF PLC-?2 IN GASTRIC CANCER MGC80-3 CELLS TREATED BY TPA
Acta Anatomica Sinica 2002;0(05):-
Objective To study the implication of phosphoinositide-specific phospholipase C?2(PLC-?2) in gastric cancer MGC80-3 cells treated by TPA.Methods The effect of 12-O-tetradecanoylphorbol-1,3-acetate(TPA) on gastric cancer MGC80-3 cells was detected and analyzed by DAPI staining under fluorescence microscope. The effect of TPA on the expression level of PLC-?2 protein was detected by Western blotting, when nuclear and cytoplasmic protein fractions were prepared through lysis of cell and centrifugation.Localization and translocation of PLC-?2 were observed under laser scanning confocal microscope with immune-fluorescence technique. After MGC80-3 cells were pretreated by U73122(PLC-?2 inhibitor),the effect of TPA on PLC-?2 was detected by Western blotting and laser scanning confocal microscope with immunefluorescence technique, and the effect of U73122 on MGC80-3 cells followed by TPA was observed and analyzed by DAPI staining under fluorescence microscope. Results TPA induced the apoptosis of gastric cancer MGC80-3 cells.Meanwhile,TPA increased the expression level of PLC-?2 protein and induced its translocation from cytoplasm to nucleus.U73122 inhibited the effect of TPA on the expression level of PLC-?2 protein, but it did not affect on the cell apoptosis induced by TPA.However, the translocation of PLC-?2 protein induced by TPA was not inhibited by its inhibitor(U73122).Conclusion Although TPA could increase the expression level of PLC-?2 protein,it is not directly related to the cell apoptosis induced by TPA.However, its translocation might be related with the apoptosis of gastric cancer.
