Objective:To explore the application value of multislice CT perfusion imaging in normal adult cerebral hemodynamics. Methods:Routine CT and multislice CT perfusion imaging were performed by selecting the basal ganglia slice and its adjacent slice in 24 normal adults,These scan dynamic images were peocessed with the CT perfusion software package.Cerebral blood flow(CBF),cerebral blood volume(CBV) and mean transiting time(MTT) were measured within the specific regions of interest(ROI) of the brain,and quantitative analysis was performed. Results:A gradient of perfusion between gray matter and white matter showed on multislice CT perfusion imaging in normal adults.CBF,CBV,MTT were(52.8?10.0)ml/(100g?min),(2.7?0.6)ml/100g,(3.7?0.9)s respedtively in the gray matter and(22.0?3.3)ml/(100g?min),(1.3?0.4)ml/100g,(5?1.2)s respectively in the white matter. Conclusion:multislice CT perfusion imaging provided a new imaging method for measuring the cerebral hemodynamics.

Objective To study the expression of LIVIN and PTEN in human gastric cancer and evaluate their clinical significance. Methods The expressions of LIVIN and PTEN were detected by immunohistochemical method in the tissues of 65 primary gastric cancer and 15 normal gastric mucosae. Results The positive expression rate of LIVIN in gastric cancer tissues was significantly higher than that in normal tissues [73.85%(48165) vs 26.67%(4/15)], and the positive expression rate of PTEN was obviously lower than that in normal tissues [30.77%(20/65) vs 66.67%(10/15)](P < 0.05). The positive expressions of LIVIN and PTEN were closely correlated with tumor depth of gastric cancer, histological differentiation grade, lymph node metastasis and TNM stage. The expressions of LIVIN and PTEN had a significant positive correlation (r = -0.362,P =0.003). Conclusion LIVIN and PTEN may play important roles in carcinogenesis of gastric cancer, which may be considered as reliable markers in clinical diagnosis and the prognosis of gastric cancer.

Objective To establish a cisplatin (DDP)-resistant HepG-2 cell line,and to explore the role of Wnt/β-catenin signaling pathway on multidrug resistance in human hepatocellular carcinoma.Methods The HepG-2 cells were exposed in a gradually increasing dose of DDP to establish a cisplatin ( DDP)-resistant HepG-2 cell line.MTT assay was used to detect the cytotoxic activity of DDP against HepG-2 and HepG-2/DDP cells.The mRNA expression of β-catenin was determined by Real-time PCR assay.The small interfering RNA was used to specifically knockdown β-catenin expression in HepG-2/DDP cells.The protein expression was detected by western blot analysis.Results The DDP-resistant cell line HepG-2/DDP was established by gradient DDP induction successfully.The IC50 values of DDP against HepG-2 and HepG-2/DDP cells were (2.29 ± 0.14) μmol/L and ( 20.51 ± 0.84 ) μmol/L,respectively ( t=95.68,P＜0.01 ),HepG-2/DDP cells was 8.96 times than HepG-2 cells on the resistance of cisplatin.The result of real time PCR showed that 2-△Ct value of β-catenin in HepG-2 cells and HepG-2/DDP cells were (0.323±0.065) and (0.674 ±0.097) (P＜0.01 ).And the protein expression of cisplatin in HepG-2/DDP cells was also significantly higher than that in the HepG-2 cells.The expresssion of β-catenin was significantly and specifically depleted by siRNA duplexes(P＜0.01 ).The IC50 values of cisplatin against HepG-2/DDP cells were (21.02 ± 1.64) μmol/L in cisplatin control group,(6.23 ± 0.68 ) μmol/L in SiRNA targeting interference group and ( 20.44 ± 1.26 ) μmol/L in SiRNA negative interference group,and there was significant difference between control group and SiRNA targeting interference group ( P＜0.01 ).Conclusion The Wnt/β-catenin signaling pathway was activated on the cisplatin(DDP)-resistant HepG-2 cell line and down regulation of β-catenin increased the chemosensitivity of HepG-2/DDP cells against cisplatin.It provided a theoretical basis for finding the new targets of multidrug resistance in liver cancer.

Objective:To set up a eukaryotic system for high expressing human CD137 and to investigate the role of CD137 and CD137L on signals transduction of cells.Methods:The pCDNA3 plasmid containing full length of human CD137 cDNA sequence(CMV ILA SEN,CIS) and pSV2 dhfr plasmid were cotransfected into dhfr CHO cells by lipoid mediating method. The positive clone was selected with G418. Expression of CD137 on dhfr CHO cells were induced by MTX and detected by RT PCR, immunocytochemistry and flow cytometry.It's activity study was done by method of incorporating 3H TdR.Results:CD137 expressed on the surface of dhfr CHO, expression rate was 96.07%, it's activity study indicated that CD137 increase the proliferation of PBMC stimulated by anti CD3 monoclonal antibody.Conclusion:dhfr CHO cells that highly express CD137 were established. CD137 can increase the proliferation of PBMC stimulated by anti CD3 monoclonal antibody.

Objective To explore the relationship between Sirt1 single nucleotide polymorphisms ( rs2236319 ) and chronic obstructive pulmonary disease ( COPD ) susceptibility. Methods The subjects included 149 patients with stable COPD in the First People′s Hospital of Jining outpatient or ward from March 2014 to March 2015, and 160 cases of the same period health check. Sirt1?rs 2236319 genotype distribution frequency was detected by real?time PCR,and the levels of inflammatory factors were compared,including IL?6 and CRP. Results The AA,AG,GG genotype frequency of COPD group were 59. 1%( 88/149) ,35. 6%( 53/149),5. 4%(8/149) respectively,of control group were 45. 0%(72/160),48. 8%(78/160),6. 3%(10/160) respectively,the allele A,G frequency of COPD group were 76. 8%(229/298),23. 2%(69/298) respectively,of control group were 69. 4%( 222/320 ) , 30. 6%( 98/320 ) respectively, there were statistically significant differences between the two groups(χ2=6. 23,6. 06,P<0. 05) . Objective To evaluate the safety of mesohepatectomy for centrally located large hepatic tumors.Methods The clinical data of 37 patients who underwent hepatectomy for centrally located large liver tumors in our hospital from October 2010 to August 2015 were retrospectively analyzed.During the operation,the left and right hemilivers were mobilized.Slings for the improved liver hanging maneuver and for selective hepatic vascular occlusion were placed.These slings were used when necessary in order to minimize occlusion and ischemia time to the residual liver,and to maximize the volume of functional liver remnant.Mesohepatectomy was carried out using a microwave hemostatic separator.The feasibility,its effects on preventing hemorrhage,the degree of liver damage and the postoperative complications were evaluated.Results The mean diameter of the neoplasms was (12.6 ± 7.2) cm.The vascular inflow occlusion time of the left and right hemilivers were (12.2 ±3.5) min and (18.5 ±7.1) min,respectively.The blood loss was (487 ± 352) ml.The amount of red blood cell transfusion was (2.7 ± 1.9) U.The operation time was (215 ± 72) min.TBIL,ALT and AST reached their peak levels on the 1 st day after operation and they were higher than before surgery (P ＜ 0.01).The levels were then significantly decreased on the 3rd day after operation.However,the TBIL (P ＜ 0.05),ALT (P ＜ 0.01) and AST (P ＜ 0.05) were higher than the preoperative level,and only returned to normal or were close to the preoperative value on the 7th day after operation (P ＞ 0.05).The initial increase in ALT was quick and then it slowed down,while the initial increase in AST was slow but it decreased quickly.Conclusions Mesohepatectomy for centrally located large hepatic tumors could safely be carried out using the liver hanging maneuver combined with selective hepatic vascular occlusion and a microwave hemostatic separator.This method has the advantages of causing less bleeding,liver damage and rapid recovery.

[Absrt act] Obj ective To summarize the clinical experience with isolated complete resection of he-patic caudate lobe in 14 patients. Mte hods The clinical data of 14patients with isolated complete resection of hepatic caudate lobe carried out in our hospital from December 2007 to March 2015 were retrospectively analyzed .During the operation , selective hepatic vascular occlusion slings , and supra-and infra-hepatic in-ferior vena cava slings were placed after full mobilization of the liver .Isolated complete resection of the he-patic caudate lobe was performed through the left and /or the right , the anterior liver-splitting or the retro-grade approaches .Results The mean operation time was (227 ±64) min.The mean amount of blood loss was (530 ±325) ml.The mean vascular occlusion time was (19.5 ±18.6) min.The mean diameter of ne-oplasm was (15 4.±9 .2) cm. All the operations were successfully carried out .There were no massive hem-orrhage, bile leakage or perioperative death .Hydrothorax occurred in 3 patients and ascites in 2.The mean stay in hospital was (21 ±9) days.All the patients were cured and discharged home .Conclusion Isolated complete resection of hepatic caudate lobe was feasible in clinical practice .
