Objective To explore the role of connective tissue growth factor(CTGF)in the mechanism of liver fibrosis.Methods Liver fibrosis models of rats were made by subcutaneously injecting with CCl4.On the other hand,32 human liver cirrhosis and 12 normal liver tissues were collected too.Immunohistochemical staining was used to detect the expression of CTGF in these tissues.Results The expression of CTGF in rats after injection with CCl4 was found mainly in hepatic stellate cells(HSC) and hepatocytes are all significantly higher than those in normal rats.Moreover,it presents with progressive tendency for the expression of CTGF in rats respectively at 1st,4th and 8th week after injection of CCl4.The same tendency was found in human normal and fibrotic liver tissues(P
2.Clinical value of drainage in the treatment of acute obstructive suppurative cholangitis without X-ray guided

Objective To investigate the therapeutic value of duodenoscopy in the treatment of acute obstructive suppurative cholangitis without X-ray guided. Methods Clinical data of acute obstructive suppurative cholangitis were collected continuously in our hospital which can be used to analyze the safety and efficacy of duodenoscopy in treatment without X-ray guided. Results 146 patients were successfully treated with ERCP in bile duct cannulation, the successful rate was 93.6 % (146/156), all the patients' bilirubin decreased, the successful rate of endoscopic treatment was 93.6% (146/156), while minor pancreatitis, hyperamylasemia, cholangitis, duodenal perforation and other complications occurred in 18 patients, the incidence rate was 11.5% (18/156). 13 patients infected with obstructive suppurative cholangtis were successfully treated with duodenoscopy without X-ray guided, and the successful rate in bile duct cannulation was 86.7%(13/15), bilirubin decreased in 12 cases, the successful rate in endoscopic treatment was 80.0%(12/15), 3 patients suffered from complications, the incidence rate was 20.0% (3/15). Conclusions The bile duct cannulation technique without X-ray guided has progressed, it can be applied to cure several patients with biliary obstruction, especially in AOSC patients, and pregnant patients, and patients who are not suitable for the X-ray radiation.

4.Grain filling dynamics and germination characteristics of Bupleurum chinense seeds.

Bupleurum chinense used in the study were cultivated in the experimental fields of Gansu agricultural University for three years. The seeds of B. chinense were collected every 3 days 10 d after the blossom. The result showed that the 1 000-grain fresh weight reached the maximum 43 d after the blossom and then decreased rapidly, at the mature period the fresh weight of seeds were falling to the same level of the dry weight. The dynamic change of the grain dry matter accumulation showed as an S-shape curve, the rapid increase stage was 25-34 d following the flower, and the grain filling was ended 46 d after blossom. Grain filling rate was under the law "fast-slow-fast-slow". And there were two peaks of grain filling rate appeared, after reached the second peak 28 d after the flower the filling rate decreased rapidly and stayed steadily 43 d after flowering. The dehydration rate was also measured at its maximum 43 d following flower. The indexes of seeds all reached the top 52 days following the blossom, when the germination rate reached the peak (34.33%) and water content of seeds was near 10%. The rate of germination and the 1 000-graid weight of seed showed significant positive correlation, while the water content of seeds was found significant negatively correlation with germination percentage. So the best time for harvest should be 52 d after flowering (9 month), the seeds collected at that time showed both high quality and germination rate.

6.Suggestion of cell injury during islet cell transplantation

BACKGROUND: The incompatible reaction may occur after islet transplantation, which affects the survival and functions of cells. OBJECTIVE: To explore the islet cells injury and its causes during islet transplantation. METHODS: The pancreases of voluntary, brain death, donors were isolated by collagenase, and the islet cells injury was measured with different cold ischemia times. The islet cells were cultured with blood as follow: HLA matching group: recipient whole blood + islet cells, recipient whole blood + islet cells + heparin; HLA mismatching group: recipient whole blood + islet cells, recipient whole blood + islet cells + heparin; Control group: recipient whole blood + RPMI1640. The potential injury to islet cells was observed. RESULTS AND CONCLUSION: The pancreases were smoothly obtained. The activity of islets may be more than 80% within 5 hours of ischemia preservation time, which was less than 19% if the cold ischemia preservation time was over 8 hours. When human islets were exposed to human blood, it will induce a rapid consumption of blood cells, no matter HLA matching or HLA mismatching. After adding heparin into the blood, these events were avoided. At 24 hours of in vitro culture, the number of survival islet cells in the HLA matching group was greater than that of the HLA mismatching group (P < 0.05). The results described that cold ischemia time affects islet cells activity, reduce the cold ischemia preservation time within 5 hours and HLA typing are conductive to enhance the quantity of living islets.
7.Prevention of wrong absorption of pancreatic pseudocyst using guided transmural drainage under endoscopic ultrasonography

Objective To discuss how to preven wrong absorption of pancreatic pseudocyst using guided transmural drainage under endoscopic ultrasonography.Methods Retrospectively analyzed the clinical data of patients with pancreatic pseudocysts underwent operations of Endoscopic ultrasonography guided transmural drainage.Results 16 cases of pancreatic pseudocysts were finished with guided transmural drainage under endoscopic ultrasonography using needling to place bypass grafting with successful rate 100.0% of guided transmural drainage by needling. After needling, 2 cased happened regurgitation which led to wrongly absorbed, rate of occurrence is 12.5%. Generally, pancreatic pseudocysts of 16 cases disappeared completely with cure rate 100.0%. From above, stents were pulled out by endoscope in 3 cases while stents were removed voluntarily in another 13 cases.Conclusion Dorsal elevated position, detailed operation and esophageal annular tubes can effectively prevent wrong absorption of guided transmural drainage under endoscopic ultrasonography of pancreatic pseudocysts.
8.Shear bond strength of anterior teeth fragment reattachment using two different adhesive materials.

OBJECTIVETo compare the shear bond strength of the fractured anterior teeth reattached by two different adhesive materials.
METHODSForty crown fractured anterior modes were divided into two groups randomly, with 20 in each group. Group A were reattached by Clearfil SE Bond and Clearfil AP-X, while group B were reattached by Clearfil S3 Bond and Clearfil AP-X. Then the specimens were submitted to an axial compression test in a universal testing machine until tooth fractured. The strength was recorded.
RESULTSThe mean shear bond strength of group A and group B was (324.32 +/- 65.91) N and (263.08 +/- 55.88) N, separately. The mean shear bond strength of group A was statistically higher than group B(t = 3.17, P = 0.000).
CONCLUSIONThe shear bond strength of two-step adhesive Clearfil SE Bond is higher than one-step adhesive Clearfil S3 Bond for the reattachment of fractured anterior teeth.

9.Effect of liangxue shengji recipe on incidence of post-percutaneous coronary intervention restenosis and adverse cardiovascular events.

OBJECTIVETo observe the intervention effect of Liangxue Shengji Recipe (LSR) on incidence of post-percutaneous coronary intervention (post-PCI) restenosis and adverse cardiovascular events.
METHODSWith a randomized, single-blinded methods adopted, 100 patients with coronary artery disease (CHD) and underwent stent implantation were randomized into two groups, the control group and the treated group, conventional Western treatment was administered to them all, but with LSR to patients in the treated group additionally. They were followed up for at least six months. The incidences of post-PCI restenosis and adverse events, including cardiogenic death, acute myocardial infarction, recurrent angina pectoris, severe heart failure, further intervention and coronary artery bypass grafting, were observed to estimate the effect of LSR.
RESULTSNo statistically significant difference between the two groups was shown in terms of incidences of intra-stent restenosis, recurrent angina pectoris, estimator of restenosis and its cumulative risk, as well as in reducing the incidence of single adverse event, but did show statistically significant difference between groups in reducing the incidence of united cardiovascular event (P=0.032) and its cumulative risk (P=0.036).
CONCLUSIONAdministration of LSR in post-PCI stage could significantly reduce the probability and cumulative risk of united cardiovascular events, and the beneficial effect presents at about six months post-PCI.

10.Touchdown PCR and overlap extension PCR for generating CD133(+) cancer stem cell-selective adenovirus vector.

OBJECTIVETo construct a replication-incompetent adenovirus vector targeting cancer stem cells by modified touchdown PCR and overlap extension PCR and investigate its infection efficiency in CD133(+) SW480 cells in vitro.
METHODSThe two portions of the fiber gene encoding the Ad5 fiber knob domain with the HI loop deleted were amplified using two pairs of designed primers and then linked by overlap extension PCR. The product obtained was identified by sequencing and inserted into prokaryotic expression vector pEGFP-N1. The product, pEGFP-N1 KNOBδHI, contained a unique EcoRV restriction site in the deleted portion of the sequence encoding the HI loop. The gene sequences of the adenovirus fiber were amplified using both common PCR and overlap extension PCR, then identified by sequencing and inserted into pNEB193, resulting in pNEB-F5. CD133(+) SW480 cells were infected with the generated adenovirus vectors Ad5-GFP and Ad5FHI-GFP to investigate the infection efficiency using fluorescent microscope.
RESULTSThe target fragments of expected sizes were amplified by touchdown PCR and overlap extension PCR, but not by common PCR. Ad5FHI-GFP showed a higher infection efficiency than Ad5-GFP in CD133(+) SW480 cells.
CONCLUSIONCompared with common PCR, touchdown PCR and overlap extension PCR can significantly improve the specificity and efficiency of the PCR products for constructing CD133(+) cancer stem cell-selective adenovirus type 5 vector, which provides carriers for tumor-targeted gene therapy.
