4.Controlled clinical study on compound Decumbent Corydalis Rhizome and diclofenac in treatment of knee osteoarthritis.

To evaluate the efficacy and safety of compound Decumbent Corydalis Rhizome (DCR) in treating patients with knee osteoarthritis (OA). Totally 79 patients with knee osteoarthritis were selected from out-patient and inpatient departments of West China Hospital and randomly divided into the test group and the control group. The test group (n = 41) was given Compound DCR with the dosage of 1.8 g · d(-1), while the control group (n = 38) was administered with diclofenac sodium with the dosage of 75 mg · d(-1). After 12 weeks of treatment, the total efficacy rates based on patients/physicians evaluation for experimental and control groups were 68.29%, 63.41% and 71.05%, 63.16%, respectively, without significant difference between the two groups. Both of the two groups showed significant improvements in the main efficacy indexes (pain on walking 20 m) and minor indexes (tenderness on palpation, Western Ontario and McMaster Universities OA index (WOMAC) and Short-Form Health Survey (SF-36 ), but without significant difference in efficacy between them. The incidence of related adverse events was 24.39% in the test group and 47.37% in the control group, respectively, with significant differences between the two groups (P < 0.05). In the controlled study, compound DCR is as efficient as diclofenac sodium but more tolerable, with a good clinical application prospect.

5.Production of a corneal opacity mouse by N-ethyl-N-nitrosourea mutagenesis and chromosome mapping of the mutant gene

Background N-ethyl-N-nitrosourea (ENU)-induced mouse mutagenesis is a powerful approach for the study of gene function and the generation of human disease models.Objective This study was to create the corneal morphologic change and map the mutant gene of a kind of corneal opacity in ENU mutagenesis in mouse.Methods ENU was intraperitoneally injected in forty C57BL/ 6J (B6) male mice aged 8-10 weeks old.The male mice were mated with the same strain female mice.Their progenies were screened for visible eye mutation,and the mutant mice were mated with the same strain mice to confirm the heredity of mutation phenotypes.Hematoxylin & eosin staining was used to examine the histopathological change of cornea in one mouse with ENU-induced corneal opacity.To map the mutant gene,[(B6×D2)F1 ×B6] N2 mutant mice were bred,and the genome of the N2 mice was scanned by microsatellite markers distributed equally on the mouse chromosome.The microsatellite linked to the mutant gene was determined by the log odds score.This experimental procedure was approved by Ethic Committee about Experimental Animal Care and Use of Yangzhou University.Results The founder mouse,which was the progeny of an ENU-treated B6 male mouse and an untreated B6 female mouse,had a corneal opacity phenotype.After mating the mutant with B6 mice,19 of 59 descendants appeared corneal opacity phenotype.Thickening of corneal stroma,neoangiogenesis,infiltration of inflammatory cells and proliferation of fibroblasts were exhibited in cloudy cornea in ENU-induced mutated mice under the optical microscope.After linkage analysis between microsatellite markers and the mutant gene,the mutant gene was linked to D2Mi307,which was located at 63.42 cM.Three cases of 26 N2 mice underwent recombination with the LOD 3.79.The mutant gene associated with the cornea phenotype was located on chromosome 2.Conclusions This study map the mutant gene associated with the cornea phenotype on chromosome 2.The strain might be used as a mouse model for heritable human corneal opacity.
6.The expressions of B7-H1 and PD-1 signal pathway in patients with chronic hepatitis B

Objective To study the expressions of B7-H1 and PD-1 which are costimulatory molecules in the pathogenesis of chronic hepatitis B (CHB). Methods The heparin anticoagulant blood samples from 11 CHB patients and 16 healthy controls were collected. The expressions of B7-H1 and PD-1 on myloid dendritic cells (mDC) and T lymphocytes in the peripheral blood were analyzed by flow cytometry. The data comparison was done by t test. Results The expressions of B7-H1 on CD3~+ ,CD4~+ , CD8~+ T lymphocytes and mDC in CHB patients were (40. 69± 14. 49)%, (42. 84 ±11. 19)%,(33. 48±14.07)% and (16. 60±4. 04) % , respectively, and those were (14. 66±10. 11) % , (4. 62±3.84)%, (1.89±2.31)% and (0. 49±0. 37)% , respectively in healthy controls. The expressions of B7-H1 on T-lymphocytes and mDC in CHB patients were significantly higher than those in healthy controls, and the differences were significant (t=-2.884,t=-10.894, t=-7.378, t=-13. 182,respectively; all P<0. 05). The expressions of PD-1 on CD3~+ , CD4~+ and CD8~+ T lymphocytes in CHB patients were (12. 45±6. 36)%, (11. 42 ± 6. 20)% and (13. 03± 6. 71) %, respectively, and those were (7. 80±3. 53)%, (7. 12±2. 60)% and (7. 88±3. 74)% , respectively in healthy controls.The expressions of PD-1 on T lymphocytes in CHB patients were significantly higher than those of the healthy controls, the differences were significant (t = -2.323, t =- 2. 355, t =- 2. 439,respectively) all P<0. 05). Conclusion The expressions of B7-H1 and PD-1 on T lymphocytes and mDC in CHB patients are higher than the healthy people.
9.Low-dose tadalafil combined with Shuganyiyang capsules for mild-to-moderate erectile dysfunction.

OBJECTIVETo observe the clinical effect of low-dose once-daily tadalafil combined with Shuganyiyang Capsules in the treatment of mild-to-moderate erectile dysfunction (ED).
METHODSNinety patients with mild-to-moderate ED were equally randomized to groups A, B and C to receive Shuganyiyang Capsules, tadalafil, and tadalafil + Shuganyiyang Capsules, respectively. The scores of the patients on IIEF-5 and SF-PAIRS (15-Item Short Form of Psychological Interpersonal Relationship Scales) were recorded before and at 1 and 3 months after treatment.
RESULTSThe IIEF-5 scores of groups A, B and C were 10.13 +/- 1.55, 11.00 + 1.60 and 10.73 +/- 1.91 before treatment, and 13.77 +/- 2.11, 17.77 +/- 2.13 and 17.17 +/- 3.84 at 1 month after treatment, significantly higher in B and C than in A (P <0. 001) , but with no remarkable difference between B and C (P =0. 411). At 3 months after treatment, the IIEF-5 scores were 15.77 +/- 2.05, 18.07 +/- 2.24 and 19.37 +/- 3.76 in the three groups, dramatically higher in B and C than in A (P <0.001) as well as in C than in B (P<0.05). The scores on sexual self-confidence, sexual spontaneity and time concerns in SF-PAIRS were 3.90 +/-0.80, 8.67 +/- 1.94 and 14.43 +/- 1.92 before medication, 5.83 +/- 1.02, 9.90 +/- 1.75 and 11.17 +/- 1.68 at 1 month and 6.73 +/- 0.98, 11.07 +/- 2.08 and 10.67 +/-1.60 at 3 months after medication in group A; 4.17 +/- 0.87, 9.37 +/-1.43 and 14.47 +/-1.57 before medication, 6.47 +/-0.78, 10.83 +/- 2.18 and 10.20 +/-1.56 at 1 month and 6.83 +/-0.91, 11.30 +/- 1.88 and 9.47 +/- 1.57 at 3 months in group B; and 4.23 +/-0. 94, 9.50 +/- 1.89 and 14.67 +/- 2.91 before medication, 8.03 +/- 1.67, 13.43 +/-1.10 and 9.70 +/-1.21 at 1 month and 8.93 +/- 1.78, 14.70 +/- 1.26 and 8. 87 +/- 0. 97 at 3 months in group C. Compared with the baseline, the SF-PAIRS scores of the three groups were all significantly improved after treatment (P <0. 05) , and markedly higher in C than in the other two groups (P <0.05).
CONCLUSIONLow-dose once-daily tadalafil combined with Shuganyiyang Capsules is obviously effective in the treatment of mild-to-moderate ED, which not only improves the patients'erectile function, sexual self-confidence and sexual spontaneity, but also reduces their time concerns.

10.Effects of Gutuo Qingfu Decoction via gastro-enteric perfusion on 16SrRNA in blood of severe multitraumatic patients.

OBJECTIVETo observe the effects of Gutuo Qingfu Decoction (GQD) via gastro-enteric perfusion on blood level of bacterial 16S rRNA gene in severe multi-traumatic (SMT) patients at early stage.
METHODSSixty SMT patients were assigned to two groups, the 33 in the treated group and the 27 in the control group. They were treated with the same conventional treatment, but different in the gastro-enteric infusion with GOD for the former and saline for the latter. Blood 16SrRNA gene, body temperature, leukocyte count, C-reactive protein (CRP), and blood bacterial culture positive rate on the 3, 6, 9 post-trauma days were detected, and incidences of infective complication and mortality were observed.
RESULTSBody temperature on day 9 in the treated group was significantly lower than in the control group (37.6 +/- 0.12 degrees C vs 38.1 +/- 0.15 degrees C, P < 0.05); so did the CRP level on day 6 (52.4 +/- 6.3 mg/L vs 104.3 +/- 20.1 mg/L, P < 0.05) and day 9 (42.9 + 7.5 mg/L vs 92.5 +/- 17.1 mg/L, P < 0.05), as well as the positive rates of blood 16SrRNA gene on day 6 and 9 (33.3% vs 59.3% and 30.3% vs 77.8%, P < 0.05 and P < 0.01, respectively). However, the positive rates of blood culture were insignificantly different between the two groups ( P > 0.05). Besides, incidence of infective complication in the treated group was significantly lower than in the control group (30.3% vs 59.3%, P < 0.05).
CONCLUSIONEarly stage gastrointestinal administration of GQD is likely to have benefits for the improvement of intestinal mucosa barrier and reduction of enteric bacterial translocation in SMT patients, and it may also reduce the incidence of infective complication in these patients.
