3. Comparison of the osteogenic effect of three different bone graft materials
Chinese Journal of Tissue Engineering Research 2020;25(10):1507-1511
BACKGROUND: Dentin particles, tricalcium phosphate/collagen protein composites and Bio-oss particles can repair jaw defects, but the excellent osteogenic effect is not clear. OBJECTIVE: To compare osteogenic effects of three different bone graft materials on mandibular defects in beagle dogs. METHODS: Eight 1-year-old beagles were selected. A boxed bone defect area of 10 mm × 8 mm × 2 mm was prepared at the bilateral mandibular external oblique line and randomly divided into four groups with four bone defect models in each group. Undemineralized dentin particles were implanted in group A; tricalcium phosphate/ collagen protein composite materials were implanted in group B; Bio-oss particles were implanted in group C; and group D was used as blank control. Three months after transplantation, the samples were taken for histological observation. The experimental animals were approved by the Ethics Committee of the Experimental Animal Center of Dalian Medical University. RESULTS AND CONCLUSION: (1) In group A, dentin particles were slightly absorbed, surrounded by new bone tissue; new bone trabeculae and capillaries could be seen, and a large number of fibrous connective tissue surrounded dentin particles in the central area of bone graft. In group B, a small number of new bone trabeculae and osteoblasts could be seen; a large number of powdered β-tricalcium phosphate particles and a small amount of inflammatory cells could be seen in the fibrous connective tissue; and some β-tricalcium phosphate particles were surrounded by new bone tissue. Bone marrow cavity could be seen in the new bone. In group C, some Bio-oss particles were surrounded by new bone tissue; some Bio-oss particles were wrapped by surrounding fibrous connective tissue, and fibers, particles and new bone were intertwined. There was no new bone formation in group D, and many capillaries could be seen in a large number of fibrous connective tissue. (2) The rate of new bone formation in groups A, B and C was higher than that in group D (P < 0.05); the rate in groups A and C was higher than that in group B (P < 0.05). (3) The results show that all the three kinds of bone graft materials can promote the formation of new bone. The short-term osteogenic effects of undecalcified xenogeneic dentin particles and Bio-oss particles are better than tricalcium phosphate/collagen protein composites, but the long-term effects need to be further observed.

OBJECTIVE To observe the efficacy of specific immunotherapy (SIT) with Alutard and NHD in children with allergic rhinitis due to different level of skin prick test (SPT) with dermatophagoides farinae (DF) and dermatophagoides pteronyssinus (DP). METHODS A total of 178 children with persistent allergic rhinitis were included in this study. Their age ranged from 6 to 12 years. They were divided into 3 groups according to level of SPT. Group 1: The level of skin index (SI) of DF is greater than that of DP, Group 2: The level of SI of DF is equal to that of DP and Group 3: The level of SI of DF is less than that of DP. The children in each group were randomly divided into 2 subgroups: Alutard group and NHD group. The children were given SIT with Alutard or NHD for one year. Symptom and medication scores were recorded and analyzed. RESULTS After receiving therapy for 3 months and 6 months, symptom and medication scores of the Group 1 and 2 in the NHD group were lower than those in the Alutard group (P<0.05); Symptoms and medication scores of the Group 3 in the Alutard group were lower than those in the NHD group (P<0.05). After receiving therapy for 9 and 12 months, the symptom and medication scores of the NHD and Alutard group in all the three groups showed no statistical difference (P>0.05). CONCLUSION The efficacy of SIT with Alutard and NHD is different in children with allergic rhinitis with different levels of SPT due to DF and DP after 3 and 6 months, but is similar after 9 and 12 months. SIT with Alutard and NHD is effective in treating children with allergic rhinitis.

OBJECTIVE:
To explore the severity index of Meniere's disease during acute phase by a statistical method of principal component analysis (PCA).
METHOD:
Ninety-five patients with Meniere's disease in the acute phase who saw the doctor with 24 hours after onset were included in this study and their clinical data were analyzed retrospectively. The subjective symptoms of hearing loss (X1) and ear stuffy plug (X4) were evaluated by VAS scoring system, and the impact of vertigo (X2) and tinnitus (X3) on the patients were evaluated by DHI and THI score. The principal component analysis (PCA) method was used to analyze the quantified data and construct a synthetic evaluate function of subjective symptoms.
RESULT:
(1) The quantitative results of subjective symptoms in patients with Meniere's disease were as follow: VAS score (X1) of hearing loss was 0-91 points, with an average of 46.23 ± 18.80, DHI score (X2) of vertigo was 8-98 points, with an average of 49.66 ± 15.67, THI score (X3) of tinnitus was 10-100 points, with an average of 47.53 ± 17.44, and ear VAS score (X4) of stuffy plug feeling was 0-82 points, with an average of 21.55 ± 27.54. (2) The eigenvalue of principal components Z1, Z2 and Z3 were 1.876, 0.984 and 0.703 respectively, and the variance contribution were 46.898%, 24.592% and 17.574% respectively. (3) The constructed synthetic evaluate function of the disease was as follow: The Meniere's disease severity index ƒ = 0.213ZX1 + 0.398ZX2 + 0.370ZX3 + 0.455ZX4.
CONCLUSION
The method of PCA for the subjective evaluation of symptoms in Meniere's disease can be constructed as a model of comprehensive evaluation system, which may provide relatively comprehensive information of clinical original variables included in the four main symptoms, reflecting the severity of the disease.

Objective To study the effect of age on the hemodynamics of common carotid artery. Methods 47 healthy volunteers werestudied. Wall shear stress (WSS) values localized at common carotid artery 2 cm below the bifurcation were calculated with cine phase-contrastMR imaging combined three-dimensional paraboloid (3DP) model. On the spatial distribution, common carotid artery wall was dividedinto 24 equal parts. Calculate the mean WSS in a cardiac cycle on each part, and choose the minimal WSS value. Then the effects of age onthe average WSS and minimal WSS were observed. Results Total 94 common carotid arteries of 47 health volunteers were examined, inwhich 9 carotid arteries in 9 volunteers were excluded because of uninterpretable high-resolution MRI findings or volunteer's intolerance forthe detection. In the remaining 85 common carotid arteries, the average WSS and the minimal WSS during a cardiac cycle on the spatial distributionwere (7.45±2.12) dyne/cm2 and (5.98±1.93) dyne/cm2 respectively. Both average WSS and minimal WSS were negative correlatedwith age (P<0.05). Conclusion With the age increased, mean WSS and minimal WSS on the spatial distribution of common carotid arterydecreased.

BACKGROUND: As a candidate gene of congenital heart disease, ACTC1 gene is related to congenital atrial septal defect inhumans.OBJECTIVE: To perform a mutation screen of ACTC1 gene in 110 nuclear families of congenital heart disease.METHODS: A case-control study was conducted based on 110 nuclear families of congenital heart disease and 300 normalhuman beings with no reported cardiac malformation. Six fragments in the coding region of ACTC1 gene was amplified by PCR invitro using five primers pairs. PCR products were screened for gene mutations.RESULTS AND CONCLUSION: A novel G-to-A variant was found at the third nucleotide of the intron downstream from exon 5.This mutation existed in a 5-year-old female with an isolated ventricular septal defect and her 30-year-old father, who had noreported cardiac anomalies. This mutation was not detected in 300 normal controls. These findings indicate that the mutation maybe related with congenital ventricular septal defects in humans.
