1.Investigation on effect of dual-source CT scanning patterns on radiation dose and image quality of head and neck

Objective To study the impact on the radiation dose,organ doses of eye lens and thyroid and image quality with different scanning modes in head and neck CT scan.Methods The simulation phantom of head and neck was scanned by using fixed scanning condition (120 kV and 200 mAs),as well as the combinations of automatic tube current modulation (CARE Dose 4D),automatic tube voltage modulation technique (CARE kV) and partial angle scanning mode (X-CARE) respectively.Six kinds of scanning modes were adopted,including 120 kV +200 mAs,120 kV + 200 mAs + X-CARE,CARE Dose 4D + 120 kV,CARE Dose 4D + 120 kV + X-CARE,CARE Dose 4D + CARE kV,CARE Dose 4D + CARE kV + X-CARE.Two thermolumineseece dosimeters (TLDs) were exposed at the skin surface positions of eye lens and thyroid,and the values measured with two TLDs were averaged.The CT dose index volume (CTDIvol) and dose length product (DLP) for every scan were recorded,and the contrast to noise ratio (CNR) in eye lens section and thyroid section were measured.Results The crgan doses of lens and thyroid were 19.8 and 26 mGy at 120 kV and 200 mAs,as well as 13.3 and 22.2 mGv at X-CARE mode.Compared with the manual selection of 120 kV,the combination of CARE kV and CARE Dose 4D made the values of CTDIvol drop from 13.1 to 10.1 mGy,the doses of eye lens and thyroid from 16.6 and 20.8 mGy to 23.7 and 19.9 mGy respectively,while the image quality reducedsignificantly.Compared with CARE Dose 4D + 120 kV,the organ doses of eye lens and thyroid were reduced from 20.8 and 23.7 mGy to 9.6 and 15.1 mGy for with additional X-CARE,while CTDIvol dropped from 13.1 to 9.3 mGy.When the combination of CARE Dose 4D + CARE kV + X-CARE was used,CTDIvol and organ doses were reduced to a minimum,when the CNRs of head and neck were also minimized.Conclusions The scanning mode CARE Dose 4D + 120 kV + X-CARE for head and CARE Dose 4D + CARE kV for neck can effectively reduce the radiation dose while keeping good image quality.When requirements for image quality are not high,CARE Dose 4D + CARE kV + X-CARE mode can be selected to reduce the radiation dose significantly.
2.A meta analysis of p53 gene codon72 polymorphism and onset risk of prostate cancer among Asian population

Objective To systematically evaluate the relationship between p53 gene codon72 polymorphism and onset risk of prostate cancer (PCa) among Asian population by meta-analysis.Methods The databases of PubMed,Medline,Ovid,Wanfang and CNKI were retrieved for screening the case control trials on the relationship between p53 gene codon72 polymorphism and onset risk of PCa among Asian population.The obtained data were statistically analyzed by using the Stata 12.0 software,moreover the data reliability and publication bias of statistical literature were evaluated.Results The meta analysis showed that the p53 gene codon72 polymorphism had no obvious correlation with PCa onset risk in Asian population.The subgroup analysis results on the control source showed the coden72 polymorphism in P vs.A,PP vs.AA,PA+PP vs.AA models based on the hospital source subgroup could significantly decrease the Pca susceptibility among Asian population[P vs.A:OR =0.680,95 % CI(0.546,0.847),P=0.001;PP vs.AA:OR=0.409,95%CI(0.260,0.645),P=0.000;PA+PP vs.AA:OR=0.513,95%CI(0.350,0.749),P=0.001],whereas the codon 72 polymorphism in PA vs.AA and PA+PP vs.AA genotypes in the control source subgroup based on the common population increased the PCa onset risk among Asian population [PA vs.AA:OR=1.664,95 %CI(1.272,2.177),P=0.000;PA+ PP vs.AA:OR =1.314,95 % CI(1.020,1.693),P =0.003 6].The subgroup analysis was conducted according to whether conforming to the HWE equilibrium,the results showed p53 gene codon 72 polymorphosm was a protective factor for decreasing PCasusceptibility among Asian population in the subgroup unconforming to the HWE equilibrium [PP vs.AA:OR=0.251,95%CI(0.135,0.467),P=0.000;PA+PPvs.AA:OR=0.564,95%CI=(0.330,0.964),P=0.036].Conclusion p53 gene codon72 polymorphism has no relation with PCa susceptibility among Asian population.
3.Inhibitory effect of siRNA-mediated MSK1-silencing on the proliferation of CNE2 cells and its mechanisms

Objective Abnormal activation of mitogen-and stress-activated kinase (MSK1) plays an important role in the development of various cancers.This study was to explore the effect of small interfering RNA (siRNA)-mediated MSK1-silencing on the proliferation of human nasopharyngeal carcinoma (NPC) cells and its underlying mechanism.Methods The siRNA vector targeting MSK1 was constructed and transfected into CNE2 cells, and the NPC cell line stably expressing MSK1 was established.Then the cells were divided into a blank control (without transfection of the plasmid), a negative control (with stable transfection of the negative control plasmid), and an experimental group (with stable transfection of the positive recombinant plasmid).The expressions of MSK1 mRNA and protein were detected by real-time quantitative PCR and Western blot, respectively, the proliferation of the cells determined by CCK-8 and colony formation assays, the cell cycles analyzed by flow cytometry, the level of histone H3 phosphorylation at Ser10 examined by Western blot, and The transcriptional activity and expression of the c-jun protein measured by dual-luciferase reporter gene assay and Western blot.Results Compared with the blank control, the inhibition rates of cell proliferation at 48, 72 and 96 hours were significantly reduced in the experimental group (P<0.05), and so were the colony formation ability of the cells (P<0.01) and the expression and transcriptional activity of the c-jun protein (P<0.05).In comparison with the negative control, the experimental group showed significant decreases in the rate of cell growth after 24 hours, the inhibition rates of cell proliferation at 48, 72 and 96 hours (P<0.05), the number of formed colonies ([221.00±20.08] vs [99.67±15.57] / 300 cells, P<0.01), the proportion of S-phase cells (P<0.01), and the expression of the c-jun protein in the CNE2 cells ([100.00±0.00] vs [48.77±10.71] %, P<0.05), but a remarkable increase in the percentage of G0/G1-phase cells (P<0.01).Furthermore, histone H3 phosphorylation at Ser10 was markedly reduced (P<0.01) but no significant change was observed in the expression of the total c-jun protein in the experimental group.Conclusion Knockdown of MSK1 using siRNA can significantly inhibit the growth and proliferation of CNE2 cells, which may be closely related to the decreased phosphorylation of histone H3 and subsequently down-regulated transcriptional activity of c-jun.
4.The compounds from n-butanol fraction of Alpinia oxyphylla.

Nine compounds were isolated from the n-butanol fraction of 95% ethanol extract of the fruit of Alpinia oxyphylla Miq. with a combination of various chromatographic approaches, including MDS resin, silica gel, reverse phase C18 and preparative HPLC. On the basis of spectroscopic data analysis, they were elucidated as (1R, 4R, 10R)-1β, 4α-dihydroxy-11, 12, 13-trinor-5, 6-eudesmen-7-one (1), 1β, 4β-dihydroxy-11, 12, 13-trinor-8, 9-eudesmen-7-one (2), oxyphyllenone A (3), oxyphyllenone B (4), rhamnocitrin (5), staphylionoside D (6), benzyl-1-O-β-D-glucopyranoside (7), 2-O-β-D-glucopyranosyl-(1S)-phenylethylene glycol (8), and (S)-1-phenylethyl-β-D-glucopyranoside (9). Among them, compound 1 is a new sesquiterpene, named as oxyphyllenone C; compounds 8 and 9 are new natural products; compounds 2 and 6 were isolated from the genus Alpinia for the first time, and compound 7 was isolated from A. oxyphylla for the first time.
5.Investigation on species and community ecology of Acaroid mites breeding in stored traditional Chinese animal medicinal materials

Objective To investigate the species of Acaroid mites breeding in the stored traditional Chinese animal medici?nal materials and the relationship between its community and habitats. Methods A total of 30 samples of traditional Chinese animal medicinal herbs were collected from Wuhu City,Anhui Province,China. The mites were isolated by the directly micro?scopic and floatation microscopic examinations,and then identified and counted under a light microscope. Results Acaroid mites was represented in 28 of the 30 samples,and the breeding rate accounted for as high as 93.3%(28/30). Totally,13 spe?cies of Acaroid mites were identified,which belonged to 4 families and 9 genera. The densities of Acaroid mites were top in 6 Chinese herbal medicines,such as corium erinacei,aspongopus,hirudo,pheretima aspergillum,Apostichopus and huechys. The diversity parameters of these six traditional Chinese animal medicinal herbs were calculated. The highest richness indexes were in aspongopus and hirudo,the highest diversity index was in hirudo,and the highest evenness index was in Apostichopus. Conclusions There are Acaroid mites breeding in parts of the traditional Chinese animal medicinal herbs stored in Wuhu. In the storage and processing of Chinese herbal medicines,we should pay attention to the prevention and control of mites.
6.Heat shock improves Sca-1+ cell transplantation for treatment of myocardial infarction in mice

BACKGROUND:Recent studies have found that stem cels can directly differentiate into mature myocardial cels or promote their regeneration, providing a new therapeutic strategy for the treatment of myocardial infarction. However, the low cel transplantation rate reduces the myocardial differentiation ability and myocardial repair.
OBJECTIVE: To study the role of heat shock treatment in Sca-1+ cel transplantation for treatment of myocardial infarction in mice.
METHODS:Sca-1+ cels were isolated from the bone marrow of mice using magnetic bead sorting method, and were subjected to heat shock treatment. Animal models of myocardial infarction were made in mice, and then randomized into two groups: heat shock group and non-heat shock group, which were given 1 mL heat shock-treated Sca-1+ cels and 1 mL non-heat shock-treated Sca-1+ celsvia the tail vein, respectively. After transplantation, cel survival, heart function, myocardial cel apoptosis and myocardial fibrosis were detected. Meanwhile, the expressions of heat shock factor (HSP), HSP70 and miR-34a in the left ventricle were measured.
RESULTS AND CONCLUSION:(1) The expression of sry gene in the heat shock group was significantly higher than that in the non-heat shock group. (2) The left ventricular ejection fraction and fractional shortening in the heat shock group were significantly higher than those in the non-heat shock group. The left ventricular end-diastolic diameter and systolic diameter in the heat shock group were significantly lower than those in the non-heat shock group. (3) The cardiac fibrosis and myocardial cel apoptosis in the heat shock group were significantly lower than those in the non-heat shock group. (4) The HSF and HSP70 expression in the left ventricle was significantly higher in the heat shock group than the non-heat shock group, and the miR-34a expression in the left ventricle was significantly lower in the heat shock group than the non-heat shock group. These findings indicate that heat shock-treated Sca-1+ cel transplantation can reduce myocardial apoptosis and infarct size, and improve heart function of mice with myocardial infarction.
7.Prevalence of work-related musculoskeletal disorders in neck and shoulder among workers in pharmaceutical and chemical enterprises

Objective:
To investigate the prevalence and influencing factors of work-related musculoskeletal disorders (WMSDs) in neck and shoulder among workers in pharmaceutical and chemical enterprises, so as to provide the basis for prevention of WMSDs.
Methods:
Workers in 8 pharmaceutical and chemical enterprises in Jinhua City, Zhejiang Province from July to October 2023 were sampled using a stratified random sampling method, and demography, work conditions and WMSDs prevalence were collected using questionnaire surveys. Factors affecting WMSDs in neck and shoulder were analyzed using a multivariable logistic regression model.
Results:
A total of 1 343 workers were surveyed, including 496 males (36.93%) and 847 females (63.07%). The mean age of workers was 38 (15) years, and the length of service in current posts was 6 (8) years. There were 356 workers with WMSDs (26.51%), with 206 workers with neck WMSDs (15.34%) and 167 workers with shoulder WMSDs (12.43%). Multivariable logistic regression analysis showed that gender (female, OR=1.692, 95%CI: 1.203-2.381), sufficient rest time (OR=0.660, 95%CI: 0.471-0.926) and maintaining same neck posture for a long time (OR=1.690, 95%CI: 1.148-2.510) were factors affecting neck WMSDs; gender (female, OR=1.572, 95%CI: 1.060-2.311), sufficient rest time (OR=0.619, 95%CI: 0.422-0.906), repetitive daily work (OR=1.678, 95%CI: 1.064-2.647), uncomfortable work posture (OR=3.791, 95%CI: 1.330-10.803), multiple repetitive movements per minute (OR: 1.988-2.413, 95%CI: 1.060-4.161) and maintaining same neck posture for a long time (OR=1.926, 95%CI: 1.218-3.043) were factors affecting shoulder WMSDs.
Conclusion
The prevalence of WMSDs in neck and shoulder are high among workers in pharmaceutical and chemical enterprises in Jinhua City, and mainly affected by gender, rest time and adverse ergonomics.
8.Clinical significance expression of MMP-7 in patients with primary non-small cell lung cancer

Objective To study the relationship between expression of matrix metalloproteinases-7 (MMP-7) and clinicopathological characteristics in patients with primary non-smaU cell lung cancer(NSCLC). Methods MMP-7 in 20 normal people and 60 advanced NSCLC patiens were detected with reverse-transcription-polymerase-chain-reaction. Gelatum image analysator analyzed the result. Results The amount of MMP-7 was less in normal people (30.000) than in NSCLC patients(41.231) significantly(P<0.05); the level of MMP-7 was no correlated with gender, age, pathology pattern, tumor size, was inverse correlation with differentiation, and was positive correlation with clinical stages(P <0.05). Conclusion The level of MMP-7 is closely correlated with tissue differentiation and clinical stages of NSCLC, which may serve as a parameter for determining tumor invasion and metastatic.
9.18F-FDDNP positron emission tomography in differentiating Alzheimer disease and vascular dementia

BACKGROUND:At present, some neurological imaging methods, including MRI, fMRI, 2-(1-(6-[(2-[18F]fluoroethyl)(methyl)amino]-2-naphthyi) ethylidene) malononitrile (18F-FDDNP) positron emission tomography (PET), are helpful but not specific for the diagnosis of Alzheimer disease (AD). 18F-FDG is a special marker of beta-amyloid (Aβ), thus AD can be diagnosed by 18F-FDDNP PET at early period.OBJECTIVE: To evaluate the role of 18F-FDDNP PET in the diagnosis of AD, and establish reliable clinical biological indexes for the diagnosis of AD patients.DESIGN: A controlled analysis.SETTINGS : Department of Geriatric Neurology and Department of Nuclear Medicine, the General Hospital of Chinese PLA. PARTICIPANTS: Patients visiting the General Hospital of Chinese PLA from May 2004 to March 2005 were selected. Informed consents were obtained from all the participants. ① AD group (n =7): (74.88±12.03) years old; Accorded with the criteria related to diagnosis of AD in NINCDS/ADRDA (National Institute of Neurological and Communicative Disorders and Stroke/Alzheimer Disease and Related Disease Association) and revision of Diagnostic and statistical Manual (DSM-Ⅳ, 4th ed.); brain CT or magnetic resonance imaging (MRI) examination displayed that inter-uncus distance of temporal lobes was ≥ 30 mm. ② Vascular dementia group (n =6): (73.83±4.75) years old; Accorded with the diagnostic criteria of NINDS-AIREN (National Institute of Neurological Diseases and Stroke, USA) and DSM-Ⅳ for vascular dementia; Inter-uncus distance of temporal lobes < 30 mm. ③ Control group (n =6): (71.17±3.71) years old; Without rarefaction of white matter; Intelligence examination was normal.METHODS: PET was performed in all the subjects. PET scanner type was SEIMENS ECAT EXACT HR. The tracer selected was 18F-FDDNP which had radiochemical purity higher than 95% and error of radioactivity measurement lower than 10%. The images were collected at 5, 25 and 45 minutes after injection of 18F-FDDNP. Horizontal and coronary tomograms of brain were obtained after reconstruction.MAIN OUTCOME MEASURES: Characteristics of 18F-FDDNP brain PET images.RESULTS: ① In the control group, signs of obvious atrophy of brain were not seen. At about 45 minutes, the radioactivity in cortex and subcortical nucleus groups was essentially cleared and the structures of brain could not be differentiated clearly. ② In the vascular dementia group, brain atrophy and enlargement of ventricular system to various degrees could be seen. The clearance of radioactivity at three time points was similar to that in the control images. ③ In the AD group, the brain was obviously atrophied and the ventricular system was enlarged. The clearance of radioactivity at the three time points was significantly different from the images of other two groups. The radioactivity in cortex and hippocampus was cleared slower. At 45 minutes, the gray matter could still be clearly differentiated from the white matter, but the radioactivity in corpus striatum and thalamus was not higher than that in cortex and much radioactivity retention could be seen in cortex and hippocampus.CONCLUSION: 18F-FDDNP PET brain images can differentiate AD and vascular dementia, and it is an effective imaging index for the diagnosis of AD.
10.Evaluation of value of indirect method in establishing reference intervals of CA72-4

Objective To establish the reference interval for CA 72-4 in indirect method.Methods All results for CA72-4 that were stored in our laboratory information system of Zhongshan hospital between Jan.2010 and Dec.2012 were included in this study.Outliers were identified and omitted using Stem-and-Leaf&Box Plots in SPSS statistical software.The treated data was divided into several groups according to gender and age.Nonparametric rank sum test was used to observe the difference between male and female participants and Spearman correlation analysis was used to examine the correlation between CA 72-4 and age.Nonparametric reference intervals for CA 72-4 were estimated statistically in two gender sub-groups.Results After 139 cases excluded, there were 1 548 cases of male (Median 1.7, 0.4 to 18.9) and 773 cases of female ( Median 1.8, 0.2 to 18.9 ).There was a significant difference in serum CA 72-4 between male and female participants.No significant difference was found in serum CA 72-4 among age sub-groups.Indirect reference values for CA 72-4 of male and female were respectively 0 to 8.9 U/ml and 0 to 11.6 U/ml.Conclusion Indirect method to establish biological reference interval is a relatively simple and less expensive method under the high rapid development of the hospital information network .It can be used in the periodical review and establishing the reference intervals where the direct method can not be used.
