1.Expression of Cyclooxygenase-1 in Rat Spinal Cord of a Neuropathic Modal: Sciatic Cryoneurolysis

ObjectiveTo observe behavioral changes and the dynamic expression of cyclooxygenase-1(COX-1) in the spinal cord of rats after unilateral sciatic cryoneurolysis(SCN).Methods72 male SD rats were randomly divided into 3 groups: normal control group(N, n=6), sham group(S, n=33) and cryoneurolysis group(C, n=33). The mechanical withdrawal threshold and autotomy assession were assessed before and 1, 2, 4 weeks after operation and COX-1 immunohistochemistry was performed on L4-6 spinal cord in 6 rats.ResultsFollowing SCN, rats exhibited significant bilateral tactile hypersensitivity until 4 weeks after operation. Furthermore, we observed autotomy peaked in severity and incidence (in 40% of rats, 1 weeks and 43.8%, 2 weeks ) 1 to 2 weeks after SCN. The COX-1 expression in dorsal horn of the spinal cord in group C was significantly higher than group S 2 and 4 weeks after operation (P<0.05).ConclusionCOX-1 plays an important role in spinal cord pain processing and central sensitization after SCN.
2.Experimental study of ginsenosides in reversing the down-regulation of glucocorticoids on glucocorticoid receptor

Objective To observe the effect of ginsenosides (GS) on the action of glucocorticoid (GC) in down-regulating glucocorticoid receptor (GR), and to explore the mechanism. Methods The rat model with hepatic artery ligation (HAL) was established and treated with GS and dexamethasone (DEX). Serum corticosterone level, maximal GK binding capacity (GR Bmax), K<,d> and GR mRNA in liver cells were detected 12 h after HAL. Results The levels of corticosterone in GS group and model group were higher than those in GS + DEX group, DEX group and normal group (all P < 0.01). The levels of corticosterone in GS + DEX group and DEX group were higher than that of normal group (both P <0.01). Compared to model group, GR Bmax in GS group was higher (P < 0.05), GR Bmax in GS + DEX group was lower (P < 0.05), but higher than that of DEX group (P <0.01). The expression of GR mRNA in GS group was significantly higher than model group (P < 0.01), but GR mRNA expression in DEX group was lower than that in model group, normal group and GS + DEX group (P <0.01). Conclusion GS partially reverses the down-regulation of GC on GR Bmax and GR mRNA, but it has no effect on serum corticosterone level in the rat model with HAL.
3.Quality Standard Study on Danqi Tablets

[Objective] To set up standard method of quality to Danqi Tablets.[Method] Take HPLC to make nature authentication,take reverse HPLC to test the content of salvianolc acid B in the preparation.[Result] The authentication is strong in attribute and has good repetition,the measurement has good linear relation,the recycle rate is 101.7%,RSD 1.77%.[Conclusion] The quality standard can effectively control the quality of Danqi Tablets.
4.Optimization of Ultrasonic-Assisted Extraction of Epimedin C and Icariin from Epimedii Wushanensis Herba by Using Response Surface Methodology

Objective To optimize ultrasonic-assisted extraction Epimedin C and Icariin from Epimedii Wushanensis Herba by response surface methodology. Methods On the basis of single factor tests, Box-Behnken experimental design and response surface methodology were adopted to optimize extraction conditions with the concentration of ethanol, ultrasonic time and solid-liquid ratio as factors. HPLC was used to determine the content of Epimedin C and Icariin from Epimedii Wushanensis Herba. Results Optimal ultrasound-assisted extraction technology was as following: the concentration of ethanol was 73%; the ultrasonic time was 22 min; the ratio of liquid to material was 1:32. The contents of Epimedin C and Icariin from Epimedii Wushanensis Herba were 15.90% and 0.75%, respectively. Conclusion This extraction technology can improve the extraction efficiency of Epimedin C and Icariin from Epimedii Wushanensis Herba, which is in accordance with predicted value.
5.Screening anti-human IgG for colloidal gold conjugate with dot immunogold filtration assay

Objective:To screen the appropriate anti-human IgG (secondary antibody) for conjugating with colloidal gold by dot immunogold filtration assay,and put forward the method of using multiple evaluation indicators for secondary antibody comparing and analyzing. Methods:Three different secondary antibodies A,B and C from three different companies were screened. Firstly the titration was used to determine the optimal reaction conditions. Then three secondary antibodies were conjugated to the colloidal gold,tested with a dot immunogold filtration assay for hydatidosis specific antibody detection. The optimal conjugated secondary antibody were compared with the standard indirect enzyme-linked immunosorbent assay. Results:The optimal pH of three secondary antibodies were 8. 5,the binding capacity were 38. 4,24 and 19. 2μg /ml colloidal gold. According to the comprehensive evaluation,the diagnostic effects of sec-ondary antibody B was better than others. Its diagnostic effects in dot immunogold filtration assay was compared with enzyme-linked im-munosorbent assay. The Kappa was 0. 895(P<0. 05) and showed the two methods were in good agreement. Conclusion:The appropriate secondary antibody for conjugating with colloidal gold could be screened by dot immunogold filtration assay and the evaluation indicators which put forward by this study,the screening method would have an important reference value for all kinds of colloidal gold test kit to screen the suitable secondary antibody.
6.Correlation among childhood trauma, obsessive-compulsive symptoms, and implicit and explicit memory in patients with obsessive-compulsive disorder

Objective:To explore the correlation among childhood trauma, obsessive-compulsive symptoms, and implicit and explicit memory in patients with obsessive-compulsive disorder(OCD).Methods:Fifty-two OCD patients were enrolled, and the childhood trauma was investigated by using the childhood trauma questionnaire short-form(CTQ-SF). The degree of obsessive-compulsive symptoms was assessed by using the Yale-Brown obsessive symptoms scale (Y-BOCS). In addition, the abilities of implicit memory and explicit memory were tested by vocabulary perception speed tasks and vocabulary recognition tasks.According to the scores of CTQ-SF, the patients were divided into abuse group( n=26) and neglect group( n=26). SPSS 22.0 software was used for t-test and Pearson correlation analysis. Results:Results of obsessive-compulsive symptoms, implicit memory, and explicit memory showed no differences between the abuse group and the neglect group( t=-1.959-1.839, P>0.05). The scores of obsessions symptoms(12.52±4.61) were positively correlated with the total scores of CTQ-SF (40.10±10.20)( r=0.331, P<0.05). On the subscale, the scores of obsessions were positively correlated with the scores of physical abuse(7.89±3.02), sexual abuse(6.47±2.28)( r=0.373, P<0.01, r=0.356, P<0.05). There was a negative correlation between the scores of physical abuse and the accuracy of explicit memory(68.75±13.33)( r=-0.281, P<0.05). The scores of physical neglect(8.98±2.67) was positively correlated with implicit memory response time(4 285.94±2 067.42)( r=0.314, P<0.05). Conclusion:Obsessions in patients with OCD are related to traumatic childhood experiences, especially physical abuse and sexual abuse.Physical trauma may influence the level of implicit and explicit memory in patients with OCD.
7.Curative Effects of Laparoscopic Radical Prostatectomy for Prostate Cancer and Its Effect on Serum Hormones Levels

Objective:To analyze the curative effects of laparoscopic radical prostatectomy for the prostate cancer and the its effect on the level serum hormones levels.Methods:86 patients with prostate cancer were selected and divided into the control group and the observation group with 43 cases in each group according to the draw method.The control group was treated by open radical prostatectomy,while the observation group was treated by laparoscopic radical prostatectomy.The operation indicators,serum follicle stimulating hormone (FSH),luteinizing hormone (LH),dihydrotestosterone (DHT),free testosterone (FT),total testosterone (T),prostate specific antigen (T-PSA,F-PSA) levels,CD3+,CD4+,CD8+,CD4+/CD8+ before and after the surgery as well as the occurrence of postoperative complications were compared between two groups.Results:The operation time of observation group was significantly longer than that of the control group,and the blood loss,bowel function recovery time,hospital stay,pain scores of observation group was significantly lower or shorter than those of the control group (P<0.05).No significant difference was found in the serum FSH,LH,DHT,FT,T,T-PSA,F-PSA levels before and after the surgery between two group(P>0.05).The CD3+,CD4+,CD4+/CD8+ of observation group was significantly higher than those of the control group (P<0.05),the CD8+ of observation group was lower than that of the control group(P< 0.05).The incidence rate of postoperative complication in the observation group was significantly lower than that of the control group (P<0.05).Conclusion:Laparoscopic radical prostatectomy could play a similar effect with the open surgery on the tumor control,it could improve the level of serum testosterone,immune function with high safety.
8.Influence of cervical curvature change on the nerve root and vertebral artery of patients with cervical spondylosis and clinical observation of manipulation treatment

OBJECTIVE: To observe the influence of the change of cervical curvature on the nerve root and the vertebral artery of the patients with cervical spondylosis and the curative effect of therapeutic manipulations. METHODS: Sixty cases of cervical spondylosis with the change of cervical curvature, 60 cases of cervical spondylosis without the change of cervical curvature and 60 normal people were chosen. They were clinically observed and the cervical spondylosis was treated by manipulations and the data was processed according to the principle of statistics. RESULTS: The change of cervical curvature affected the nerve root and the vertebral artery and therapeutic manipulations produced a satisfactory curative effect. CONCLUSION: Cervical spondylosis with change of cervical curvature has special characteristics in the etiology, course and state of the disease. The change of cervical curvature produces an effect on the nerve root and the vertebral artery. Selected therapeutic manipulations produce a better curative effect.

BACKGROUND:Bone marrow mesenchymal stem cel s are considered to have good proliferation and differentiation potentials. Sox9 is a transcription factor that is essential for chondrogenesis and has been termed as a“master regulator”of the chondrocyte phenotype. OBJECTIVE:To study the therapeutic effects of Sox9-transfected bone marrow mesenchymal stem cel s on knee osteoarthritis. METHODS:The bone marrow mesenchymal stem cel s were transfected with Lenti-Sox9-EGFP in vitro. The model of murine knee osteoarthritis was established by cutting off the anterior cruciate ligament. Thirty model mice were randomly divided into three groups, as normal saline group, bone mesenchymal stem cel group and Sox9-transfected bone mesenchymal stem cel group. 0.1 mL of normal saline, 0.1 mL of normal saline containing non-transfected bone marrow mesenchymal cel s (non-transfected group), or 0.1 mL of normal saline containing Sox9-transfected bone marrow mesenchymal cel s (Sox9-transfected group) was injected into the knee joint cavity of mice in the corresponding group, respectively. After 4, 8, 12 weeks, the repair of articular cartilage lesions was evaluated by toluidine blue and immunohistochemical staining. RESULTS AND CONCLUSION:The lesions of articular cartilage were more serious in the normal saline group, compared with the other two groups, and the difference became more obvious over time. Damaged articular cartilage was improved in the non-transfected group, but the improvement was less than that in the Sox9-transfected group. Immunohistochemistry staining revealed that in the Sox9-transfected group, the positive type II col agen expression was stronger than that in the other two groups, but this positive expression was decreased over time in al the three groups. These results suggest that Sox9-transfected bone marrow mesenchymal stem cel s promote the repair of damaged cartilage in mice with knee osteoarthritis.
