1.Effects of yohimbine on norepinephrine release by locus coeruleus in stressed rats

Objective To study the cellular and molecular modulation mechanisms of norepinephrine (NE) release in hypothalamus induced by electrical stimulation in locus coeruleus (LC) in the rat model of depression-like behavior.Methods The depression-like behavior model was established by combining separation and chronic unpredictable stress stimulations.After the model establishment,behavior tests were used to verify the success of modeling.NE release in the hypothalamus induced by electrical stimulation in LC was studied in real time and NE signal was recorded with carbon fiber electrode.The peak value,the time to peak and half-life period of NE signal in both group rats were measured and analysed.Results The bodyweight,score of open-field test,percentage of sucrose preference of model group rats ((263.9± 16.4) g,(19.4±7.9),(44.3± 10.8) %) were significantly lower than those((314.3±24.3) g,(53.3± 19.0),(60.6± 13.3) %) of control group(P<0.01).The peak value of NE signal in the hypothalamus induced by electrical stimulation in locus coeruleus in depression-like behavior model rats((176.9±50.4) pA)was less than that((361.6±88.6) pA)in control group(P<0.01),and the time to peak of NE signal was also shortened in depression-like behavior model group rats (P<0.05).Intraperitoneal injection of yohimbine (3 mg/kg) potentiated electrical stimulation induced NE release in depression-like behavior model rats but not in control group.Conclusion The chronic unpredictable stresses attenuate the secretion of NE in the hypothalamus induced by electrical stimulation in LC in rats.Yohimbine,a presynaptic α2 receptor antagonist,increased NE release in hypothalamus in depression-like behavior model rats.These findings suggest that the LC projects functionally to the hypothalamus and the projection may contribute to the pathogenesis of depression.
2.Protective effects of polydatin on focal cerebral ischemia in rats

BACKGROUND: Free radicals are produced during ischemia, which can strengthen activity of lipid peroxidation; induce lesion of cell and cellular barrier, result in necrosis or apoptosis of neurons; and aggravate edema of ischemic cerebral tissue.OBJECTIVE: To observe the effects of polydatin (PD) on free radicals, lipid peroxidation, water contents and pathological morphology of brain tissue in rats with focal cerebral ischemia so as to explore its protective mechanisms.DESIGN: A randomized controlled trial.SETTING: Intensive Care Unit, Second Affiliated Hospital, Chongqing University of Medical Sciences; Pediatric Research Institute, Children's Hospital Chongqing University of Medical Sciences.MATERIALS: The experiment was carried out at the Pediatrics Medicine Institute of Chongqing Medical University from October 2001 to July 2002.Totally 48 healthy adult male Wistar rats were divided into 3 groups randomly,with 16 in each group. Group Ⅰ was sham-operated group: rats were anaesthetized, the right common carotid arteries were separated instead of being occluded. Group Ⅱ was ischemia group: to establish the right middle cerebral artery occlusion model of rats. Group Ⅲ was PD pretreatment group: polydatin (6 g/L, 12 mg/kg) were intravenously administrated 30 minutes before the onset of ischemia. Saline substituted for PD, besides, were intravenously administrated with the same way and dosage on Group Ⅰ, Group Ⅱ and Group Ⅲ.The rats were decapitated and the brains were immediately removed after cerebral ischemia 2 hours. In each group, 8 rats were chosen to be determined water contents of brain tissue, the other 8 rats were chosen to be determined levels of lipid peroxidation and free radicals in brain tissue.METHODS: According to the formula which was: wet weight-dry weight/wet weight×100%, water content of cerebral tissue was assayed. Superior liquid was taken to assay MDA with spectrophotometer thiobarbituric acid method (TBA) method, SOD activity assayed by xanthiue oxidase enzyme method, the activities of GSH-Px, CAT and NOS determined by colorimetry,the amount of protein determined by the method of Lowry. All the procedures were carried out strictly according to the instruction.malonaldehyde (MDA) and activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) and nitric oxide synthase chemia, contents of SOD, GSH-Px and CAT in cerebral tissue of PD group were obviously higher than those of ichemia model group [(226.43±8.69),(193.37±11.14) NU/mg; (244.38±12.34), (211.71±16.50) μkat/g; (59.85±9.67),water in cerebral tissue of PD group were obviously lower than those of ichemia model group [(6.38±0.54), (8.63±0.78) μmol/g; (78.72±0.43)%,tivity in ischemic tissue but the results were similar to that in ischemia model group [(12.00±1.00), (12.84±1.17) μkat/g, P > 0.05] in brain tissue.ed that PD alleviated the ischemia edema of cerebral ischemia.CONCLUSION: PD can alleviate the reaction of lipid overoxidation, improve the activities of antioxidant-enxymes, reduce ischemia brain edema,protect the function of cell member, bring down the damage to ischemia neurons. It shows that PD has significant cerebral protective role on focal ischemia brain damage.

OBJECTIVE:To prepare the compound chlorhexidine acetate ear drops for treating anaerobic and aerobic infections of antrum auris METHODS:The compound chlorhexidine acetate ear drops was prepared with mixed solvent of glycerin,alcohol and distilled water The contents of two main ingredients were determined by dual-wavelength isobestic point spectrophotometry and the stability of preparation was examined RESULTS:The average recovery of metronidazole was 99 34%(RSD=0 57%,n=6) and that of chlorhexidine acetate was 101 17%(RSD=0 88%,n=6) CONCLUSION:The new preparation is rational in formula,simple in quality control and good in stability and has good prospects in development
4.The clinical analysis of diabetic peripheral neuropathy

Objective To investigate the clinical characteristics of diabetes complicated with peripheral neu- ropathy and increase the cognition of diabetic peripheral neuropathy in clinical work.Methods The clinical data of 160 diabetic patients were analyzed with retrospective analysis.Results 118 diabetic patients had different levels neuroelectrophysiologic abnormality,and the incidence rate was 73.8 %.The cure rate of diabetic peripheral neuropa- thy was 11.0%.Conclusion The incidence of peripheral neuropathy is high in diabetic patients,neuroelec- trophysiologic examination is a sensitive index in diagnosis of diabetic peripheral neuropathy,and the therapeutic ef- fect of diabetic peripheral neuropathy is poor.
5.Effect of physical stimulus on periosteal chondroplasia

Objective To study the impact of physical stimulus on periosteal chondroplasia with an in- tention of reconstructing cartilage tissue that has a biomechanical function and can be created either entirely in vivo or in vitro and subsequently implanted into the patient.Methods A series of biomechanical test methods were designed in order to investigate the mechanical properties of engineered cartilage under a variety of mechanical stress stimulations.The periosteal explants were harvested from proximal medial tibias of New Zealand rabbits and secured onto poly-?-caprolactone(PCL)scaffolds.The composites(periosteum/PCL)were suspended in the spinner flask bioreactor and the spinner flask was located on the stirring machine.The stem cells were stimulated by fluid shear stress under magnetic bar stirring.Results In gross observation,the thickness and length of neo-cartilage with shear force stimulation were significantly greater than those without shear force.The direction of elongation was parallel with fluid flow direction,regardless of composite orientation.Secondary,two different zones based on various cell morphology and distribution were clearly observed.The thickness of the superficial zone was greater in samples exposed to high shear force compared with low shear force.The superficial zone protein and lubricant were found in the engineered cartilage under shear stress stimulation.Conclusion The results reveal that shear stress can not only influence proliferation and differentiation of stem cells,but also change chondrocyte morphology and increase extracellular matrix production.
6.Midterm follow-up results on Asian femoral intramedullary nail for the treatment of segmental and comminuted femoral fractures.

OBJECTIVETo investigate midterm follow-up results on Asian femoral intramedullary nail in treating segmental and comminuted femoral fractures.
METHODSBetween June 2011 and October 2012,16 patients with segmental and comminuted femoral fractures were treated with minimally invasive reset and Asian femoral intramedullary nail under extension table. Among them, there were 10 males and 6 females aged from 21 to 49 years old with an average of 34.5 years old; the time from injury to operation ranged from 3 to 24 d with an average of 9.1 d. There were 6 cases were type C1,2 cases were type C2 and 8 cases were type C3 according to AO classification. X-ray of femoral segment at 3,6 and 12 months after operation were applied for evaluating fracture healing. Harris score of hip joint and HSS score of knee joint were used to evaluate postoperative function.
RESULTSAll patients were followed up from 24 to 36 months with an average of 28.4 months. Operative time was from 88 to 112 min with an average of 90.7 min; blood loss ranged from 150 to 200 ml with an average of 188.75 ml; the time of fracture healing was from 5 to 9 months with an average of 5.4 months. All incision were healed at stage I. No loosening, breakage of internal fixation and displacement of fracture were occurred. There were no significant differences in Harris score of hip joint at 3, 6 and 12 months after operation (F = 0.07, P = 0.893 > 0.05), 10 cases obtained excellent results, 5 good and 1 moderate. There was no obvious meaning in HSS score of knee joint (F = 0.08,P = 0.876 > 0.05), 9 cases obtained excellent results, 6 good and 1 poor.
CONCLUSIONAsian femoral intramedullary nail could treat segmental and comminuted femoral fractures by using variety of less invasive ways,which has advantages of less trauma, quick recovery of function and satisfied midterm following-up results. But long term following-up effects remains to be seen.

7.A systematic review of animal models for Staphylococcus aureus osteomyelitis

Staphylococcus aureus ( S. aureus ) osteomyelitis, a significant complication for patients un-dergoing fracture fixation, is a great challenge for orthopaedic surgeons due to its extreme difficulty in mangae-ment. Animal models play an important role in exploring the pathogenesis of osteomyelitis and determining the efficacy of prophylactics and therapeutic treatment. To help understand current animal models of S. aureus os-teomyelitis, we conduct a systematic search to identify animal experiments that have investigated the management of S. aureus osteomyelitis. Experimental studies are categorized by animal species and are further classified by the setting of infection. Study methods are summarized and the advantages and disadvantages of each species and model are discussed.
8.Correlation between upper airway MRI parameters and the severity of patients with obstructive sleep apnea hypopnea syndrome

[ABSTRACT]OBJECTIVETo investigate the relationship between upper airway MRI parameters and the severity of patients with obstructive sleep apnea hypopnea syndrome (OSAHS). Then the values of upper airway MRI parameters in waking state were assessed.METHODS The MRI upper airway parameters of 37 OSAHS patients and 18 nonsnoring normal subjects were measured and compared. Then data analysis were carried out to find out significant correlations between the MRI upper airway parameters and the severity of patients (the apnea hypopnea index, AHI; the lowest oxygen saturation, LSaO2; the oxygen desaturation index, ODI) with OSAHS.RESULTSThe correlation between cross-sectional areas and left-right diameter in the narrowest retropalatal region, the thickness of lateral pharyngeal wall, the length of the tongue, the length and thickness of the soft palate and the cross-sectional areas of the soft palate on midsagittal planes and severity of OSAHS were found (P<0.05).CONCLUSIONThe MRI upper airway parameters can be used in evaluating the severity of OSAHS in some ways, locating the obstruction sites and choosing the best treatment planning.
9.Orthodontic tooth movement at different stages of adolescent female menstrual cycle

BACKGROUND:Estrogen has an effect on orthodontic tooth movement. Currently, only animal experiments show that the lower estrogen levels, the greater the amount of orthodontic tooth movement;the higher estrogen levels are, the smal er the amount of orthodontic tooth movement is.
OBJECTIVE:To investigate effects of orthodontic force applied on orthodontic tooth movement at different stages of menstrual cycle among young female patients.
METHODS:Twelve young female patients were included in this study, aged 14-18 years old. They already have regular menstrual cycle, and need to extract the first premolar in the maxil a. By using self-control method, these female patients with their maxil ary canine at both sides were randomly divided into two groups:orthodontic force at ovulatory period and orthodontic force at menstrual period. Micro-implant anchorage was implanted to the distal y moving canine. Orthodontic force was given to the group of ovulatory period 2 weeks after the force was given at menstrual period. Dentition models were taken at day 0 and 28 after force, to prepare a superhard plaster model using silastic impression materials. Between these two groups, the distances of the canine distal movement were measured and statistical analysis was performed with GraphPad Prism5 software.
RESULTS AND CONCLUSIONS:The distances of the canine distal movement in the group of orthodontic force at menstrual period were greater than that in the other group (P<0.05). The orthodontic teeth with the application of orthodontic force at menstrual period move faster than that with the application of orthodontic force at ovulatory period, thus effectively shortening orthodontic treatment.
10.Comparison of Inhibition Effect of Compound Danshen Tablets on Platelet Activation and That of Danshen Tablets in Patients with Coronary Atherosclerosis

OBJECTIVE:To compare the inhibition effect of Compound danshen tablets on platelet activation and that of Danshen tablets in patients with coronary atherosclerosis.METHODS:90 patients with coronary atherosclerosis were randomized into compound danshen tablet group,danshen tablet group and control group with treatment course of 2 months.30 cases were included into normal group.Radioimmunoassay and realtime RT-PCR were applied to detect mRNA expression of PTGIS,cAMP,TXA2 and PGI2 in plasma respectively and compared with control group.RESULTS:As compared with normal group,the mRNA expression of cAMP,PGI2,PTGIS were decreased markedly while expression of TXA2 was increased remarkably.In compound danshen tablets group and danshen tablets group the mRNA expression of cAMP,PGI2 and PTGIS were increased while the expression of TXA2 was decreased.The increase and decrease of mRNA expression in compound danshen tablets group was higher than that in danshen tablet group.CONCLUSION:Both of Compound danshen tablets and Danshen tablets could inhibit platelet activation and prevent acute coronary syndrome.The effect of Compound danshen tablets on inhibiting platelet activation is better than that of Danshen tablets.
