1.The relationship between constant work rate exercises and dynamic hyperinflation in patients with stable COPD

Objective To study the relationship between the constant work rate exercises and dynamic hyperinflation in patients with stable COPD. Methods Routine lung ventilation function and 80%Wmax constant power bicycle ergometer exercises were tested in 30 stable COPD patients. We analyzed the correlation coefficient of Time of tolerant exercise , degree of dyspnea , VO2 max%, VO2 @kgmax% and IC%. Results All COPD patients existed varying degrees of exercise capacity accompaning with dynamic hyperinflation in the bicycle ergometer exercises. By Spearman correlation analysis , the VO2 @kg% of constant power exercises in patients with COPD is correlated with the DH% (r = -0.480, P = 0.007) and ICq% (r = 0.667, P = 0.01). Conclusion Dynamic hyperinflation is probably the prominent factors for the capability of constant work rate exercises in patients with stable COPD. Inhibition of dynamic hyperinflation may help improve exercise capability in patients with constant power exercise.

Objective To investigate the significance of antifilaggrin autoantibodies (AFA) in rheumatoid arthritis (RA).Methods Filaggrin was extracted from the skin of the patients who had breast cancer undergoing mammectomy.AFA were detected at the sera titer 1∶10 with Western blotting,and its clinical significance was investigated with the corresponding clinical materials.Results Antifilaggrin autoantibodies (AFA) were detected in 39 9%(71/178) of RA patients,and 6 cases of control group,with a specificity of 0 969.AFA were present in 48 4%(15/31) of the early and 29 2% of seronegative RA.AFA positive patients exhibited higher frequency of Sjgren′s syndrome;The average and onset ages of AFA positive patients were older than those of the negative ones,and there was correlation between AFA and age.The frequency and average titer of RF patients were higher in AFA positive than in negative ones.AFA was significantly correlated to RF,AKA and APF.Conclusion AFA are valuable diagnostic parameter for RA,especially in the early and seronegative RA.AFA is significantly correlated to AKA and APF,but they are not totally overlapped,None of the three antigens used bear all the epitopes recognised by antifilaggrin autoantibodies.Immunoblot detection of AFA,a simple and reliable test,may be complement to conventional IIF detection of AKA and APF.

Objective To study the differentiation of embryonic stem cells(ESCs) induced by transforming growth factor-?_1(TGF-?_1) and co_cultured with visceral endoderm like END_2 cells,and explore cardiomyocytes induction effects of the combined techniques Methods Day 2-3 embryoid bodies(EBs) were derived from ESCs,and then TGF-?_1 was added or/and co_cultured with END_2 cells or END_2 cells conditioned medium.Spontaneous differentiation was as a control.The expression of cardiac specific ?-sarcmeric actin(?-actin) and cardiac troponin_T(TnT) was detected by immunofluoresence staining.The ultrastructural analysis for ESCs_derived cardiomyocytes was scanned by transmission electron micrograph. 5.Effect of cataract surgical incision on original astigmatism and tear film stability of corrected cornea

AIM:To investigate the effect of the size of cataract surgical incision on original astigmatism and tear film stability of corrected cornea.METHODS: Totally 92 cataract patients (92 eyes) who were admitted to our hospital from July 2014 to July 2016 were randomly divided into the control group and the observation group,46 cases (46 eyes) in each group.Both groups were treated by clear corneal tunnel incision phacoemulsification combined with intraocular lens implantation.The incision of the control group was 3.0mm while of the observation group was 1.8mm.The uncorrected visual acuity,corneal astigmatism,Schirmer I test (SⅠt) and break-up time (BUT) were detected before surgery and at 1d,1wk,1mo and 3mo after surgery.The surgery induced astigmatism (SIA) was recorded at 1d,1wk,1 and 3mo after surgery.RESULTS: There were significant differences in the uncorrected visual acuity between the two groups at 1 and 3mo before surgery (P<0.05),but there were no significant differences at different time points before and after surgery (P>0.05).At 1wk,1 and 3mo after surgery,SIA of two groups decreased continuously,and the SIA of the observation group was significantly lower than that of the control group at 1d,1wk and 1mo after surgery (P<0.05).At 1wk after surgery,SⅠt and BUT in the two groups were less or shorter than those before surgery (P<0.05),but there were no significant differences at 1mo,3mo after surgery,compared with those before surgery (P>0.05).SⅠt and BUT in the observation group were less or shorter than those in the control group at 1wk after surgery (P<0.05) but there were no significant differences at 1 and 3mo after surgery (P>0.05).CONCLUSION: Compared with 3.0mm standard incision,1.8mm clear corneal incision can reduce SIA and shorten the time for corneal stability recovery.
6.Expressions and clinical significances of IL-10 and IL-18 levels in patients with cervical intraepithelial neoplasias and cervical carcinoma

Objective To explore the expressions and.significancse of interleukin (IL)-10 and IL-18 in serum and tissues of patients with cervical intraepithelial neoplasias (CIN) and cervical cancer,and to analyze their relationships with clinical pathological characteristics and prognosis.Methods One hundred and eight patients' tissues and clinical blood samples (68 cases of CIN and 40 cases of squamous cell carcinoma) were enrolled in this study,including 25 with low-grade cervical intraepithelial neoplasias (CIN Ⅰ),43 with high-grade cervical intraepithelial neoplasias (CIN Ⅱ-Ⅲ),and 40 with invasive cervical squamous cell carcinoma (SCC);meanwhile,20 cases normal cervical tissues were enrolled.The expressions of IL-10 and IL-18 in cervical tissues and serum of the studied patients were measured by enzyme-linked immunosorbent assay and polymerase chain reaction.Meanwhile,second generation hybrid capture was used to detect human papilloma virus(HPV) DNA in cervical smears obtained from the studied patients with the different lesions.The relationships among IL-10 and IL-18 with HPV DNA and clinical prognosis were analyzed.Results The expressions of IL-10 and IL-18 in normal control serum were (212.75 ± 62.09),(187.84 ± 81.03)pg/ml respectively.The expressions of IL-10 in CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC serums were (324.71 ±75.87),(397.43 ±68.56),(482.77 ± 104.05) pg/ml,with statistically significant difference (F =17.657,P =0.001).The expressions of IL-18 in CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC serums were (305.53 ± 64.08),(392.74 ± 87.38),(499.86 ± 92.04) pg/ml,with statistically significant difference (F =13.309,P =0.003).The relative expressions of IL-10 in normal control,CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC tissues were 0.99 ±0.01,3.24 ±0.68,7.32 ±0.99,13.24 ± 1.03,with statistically significant difference (F =21.694,P =0.000).The relative expressions of IL-18 in normal control,CIN Ⅰ,CIN Ⅱ-Ⅲ and SCC tissues were 0.98 ± 0.01,2.02 ± 0.84,7.01 ± 1.59,14.38 ± 2.10,with statistically significant difference (F =19.912,P =0.001).The expressions of IL-10 and IL-18 were positively associated with HPV infection (r =0.696,P =0.001;r =0.852,P =0.001).The median survival time of patients with high expressions of IL-10 was 9.74 months,which obviously shorter than those patients with low expressions of IL-10 (24.47 months),with statistically significant difference (x2 =21.363,P =0.001).The median survival time of patients with high expressions of IL-18 was 12.32 months,which obviously shorter than those patients with low expressions of IL-18 (22.88 months),with statistically significant difference (x2 =7.457,P =0.006).Conclusion High expressions of IL-10 and IL-18 are observed in patients with CIN and SCC,which can be used as biomarkers for predicting early cervical lesions.

【Objective】In order to establish the foundation for transgenic mouse model,the human thalassemic gene(β654) was cloned and sequenced.【Methods】The human β654 gene was amplified by PCR,and cloned into the plasmid BGT51 in which the human β gene was cut out aforehand.The recombinant plasmid was certified by enzyme-digestion,reverse dot hybridization and sequencing.【Results】A recombinant plasmid was obtained,which contained the human β654 gene in the correct recombinant direction.Sequencing showed that the cloned insert was correct.【Conclusions】The recombinant plasmid constructed is useful for establishing a transgenic mouse.

【Objective】 To investigate the incidence of β-th alassemia (β-thal) heterozygote carrying α-thalassemia (α-thal ) 1 gene in Guangdong area. 【Methods】 β-thal genes were screened by reve rse dot blotting (RDB). In β-thal DNA samples α-thal 1 genes were am plified using gap-PCR method. 【Results】43 α-thal-1 cases were identifi ed among the 500 β-thal traits. The rate is 8.6%. 【Conclusion】 In Guangd ong area the incidence of β-thal heterozygote carrying α-thal-1 gen e is 8.6%, which should be paid much attention in the genetic counselling and pr enatal diagnosis.
