2.Determination of Vancomycin and Norvancomycin in human serum by HPLC

Objective To establish HPLC method for fast, effective, simultaneous determination of Vancomycin and Norvancomycin in serum. Methods The separation was achieved on the Hypersil ODS2(250 mm ×4.6 mm,5μm)with methanol-acetonitrile-a mixture of potassium phosphate (9.5:2.5:88) as the mobile phase.The flow rate was 1.0 mL/min.The UV detecting wavelength was 280 nm.The temperature of the column was 35 ℃. Vancomycin and Norvancomycin were used as the internal standard for each other .Results Vancomycin and Norvancomycin blood concentration were in well linearity in the range of 1.25-100μg/mL,R2 =0.9996.The average method recovery rate of three concentrations were 96.88%,99.50%,105.01%respectively, the average method recovery rate of three concentrations were 94.40%,103.90,103.81%respectively, the intra-day and inter-day RSD were less than 5%.Conclusion The HPLC method is proved to be simple,sensitive, accurate and precise in which little blood sample is taken, thus it is suitable for the therapeutic drug monitoring(TDM) and pharmacokinetic study of Vancomycin and Norvancomycin.
3.Expression level and significance of TGF-β1 and CTGF in cerebrospinal fluid of patients with subarachnoid hemorrhage

Objective To explore the expression and significance of TGF-β1 and CTGF in the occurrence and development of patients with subarachnoid hemorrhage. Methods Cerebrospinal fluid TGF-β1 and CTGF concentrations in all subjects were serially detected by ELISA. Results Mean total TGF-β1 and CTGF in patients were elevated more than that in controls at the first day(P<0.05).Thereafter the levels of TGF-β1 and CTGF fell by the third day, then rose to the second peak at the tenth day and remained significantly increase until the sixteenth day(P<0.05).The levels of TGF-β1 was positive correlated CTGF. Conclusion The expressions of TGF-β1 and CTGF in patients with subarachnoid hemorrhage showed two peaks character. After SAH, the concentration of TGF-β1 and CTGF concentration is related to chronic communicating hydrocephalus.
5.Research advances on second primary malignancies of oral cavity following radiotherapy for nasopharyngeal carcinoma

Radiotherapy is the primary treatment modality for nasopharyngeal carcinoma(NPC) which can effectively control the disease.Oral cavity,anatomically near nasopharyngeal region is the main area for the occurrence of complication of radiotherapy.Second primary malignancy (SPM) in oral cavity is an important factor interferencing NPC patients survival rate.The etiology of oral SPM is unclear and,the prognosis is poor.The research of it is still in exploration.
6.The influence of puerarin on the expression of TNF-α in rats with focal cerebral ischemia and reperfusion

Objective To study the nerve protective mechanism of puerarin on ischemic brain injured and observe the expression change of TNF-α cytokines in ischemic brain area. Methods 90 healthy male SD(Spraque-Dawley)rats were used to built the animal model of brain medium sized artery focal cerebral ischemia reperfusion with the patching methods, and the rats were randomly divided into three groups: Sham operation group(n=30),operation group(n=30),operation with puerarin interfered group(n=30).There were five different observing time in every group(6h,12h,24h,48h,72h after operation).The operation with puerarin group rats were treated with puerarin solution, the sham operation and operation group rats were injected the same volume Sodium Chloride. To observe the expression of TNF-α cytokines in different time, immunohistochemistry staining in ischemic region was used. Results Compared with the sham operation group. the expression of TNF-α cytokine in cerebral isehemic reperfusion brain tissue in the operation and the operation with puerarin interfered groups was higher(P<0.01).Compared with the operation group, the expression of TNF-αcytokine was significantly decreased in different time after cerebral ischemic-reperfusion in the operation with puerarin interfered group(P<0.01 or P<0.05).Conclusions The puerarin can restrain the expression of the TNF-α cytokine.
7.Effects of Transgenic Biocontrol Bacterium 308R(pCPP430) on Tomato Rhizosphere Bacterial Community

The study aimed at investigation the effect of the transgenic biocontrol bacteria 308R(pCPP430) on the tomato rhizosphere bacterial communities. Strain 308R (Pantoea agglomerans), an adnascent bacterium isolated from tomato, was a biocontrol bacterium. The recombinant cosmid pCPP430 contained the hrp gene cluster of Erwinia amylovora was transformed into 308R Strain. Transgenic biocontrol bacterium 308R(pCPP430) produced Harpin protein which induce hypersensitive response and disease resistance in plants and, surprisingly, increase plant growth. Two complementary methods, sole-carbon-source utilization tests (SCSU) and ERIC-PCR, were used to compare the characterization of bacterial communities from three groups of tomato roots dipped in sterilized water, 308R suspended liquid and 308R(pCPP430) suspended liquid. The cluster analysis and principal components analysis of the number of colonies from SCSU indicated consistent differences in the rhizosphere of each group. Cluster analysis of SCSU data showed that there were consistent differences in the rhizosphere of each group. The rhizosphere bacteria communities of root dipped with 308R(pCPP 430) and 308R have a relative good replication, but the differences of corresponding replicates in dipped with distilled water was increased by degrees. Cluster analysis of ERIC-PCR fingerprints revealed that most of results (eight in ten) have overlaps between roots dipped in water and those dipped in 308R liquid. The methods used in this study may prove a useful approach for the comparison of bacterial communities.
9.The study of risk factors bringing on death and the conversion of renal function in elder acute renal failure

Objective To investigate risk factors bringing on death and the conversion factors of renal function in the elder acute re-nal failure (ABF) and establish the prevention and treatment measure in clinic to reduce mortality. Methods The clinical information of 143 elder ARF cases which were treated by two hospitals for many years was studied. The risk factors possible bringing on death in the elder ARF were studied by using binary logistic regression pattern. The clinical factors possible affecting the conversion of renal function in the eld-er ARF were analyzed. Results Binary logistic regression pattern study showed that heart failure, respiratory failure, gastrointestinal bleed-ing, hepatic failure, coma, organ failure number and lower blood pressure among 25 study factors were found to be the risk factors bringing on death, and heart failure, respiratory failure, organ failure number and whether chronic renal insufficiency were the most important factors of affecting recover of renal function in the elder ARF. Conclusion The effective prevention and treatment measure should be adopted in the elder ARF with these risk factors so as to reduce mortality and increase the recover rate of renal function.
10.The curative effect of Azithromycin Sequential Therapy in the treatment of mycoplasma pneumonia in chil-dren

Objective To observe the clinical efficacy and safety of Azithromycin Sequential Therapy in the treatment of mycoplasma pneumonia for children.Methods 112 children with mycoplasma pneumonia were randomly divided into observation group and control group,each 56 cases.The observation group was treated with Azithromycin Sequential Therapy in the treatment,while the control group was treated with erythromycin lactobionate anti infection treatment.The clinical efficacy was compared between two groups after treatment,the clinical symptoms disappeared time and drug adverse reaction occurred the situation.Results The total effective rate of the observation group was 96.43%,which was significantly higher than 80.36% of the control group,the difference was statistically significant (χ2 =7.04,P <0.05).The fever,cough,pulmonary rales disappearance time of the observation group were (3.74 ± 1.58)d,(3.24 ±0.47)d,(5.58 ±2.23)d,which were significantly shorter than (4.68 ±1.71 )d,(4.64 ± 0.97)d,(7.82 ±2.70)d of the control group(t =4.27,4.68,4.59,all P <0.05).The gastrointestinal reactions, local pain,rash,the elevation of ALT of the observation group were significantly lower than the control group,the difference was statistically significant (χ2 =4.26,7.04,7.03,5.23,all P <0.05 ).Conclusion Azithromycin Sequential Therapy for the treatment of mycoplasma pneumonia in children has the advantages of good clinical effica-cy,fast effect,high safety.
