2.Clinical study of t-PA and u-PA expression in patients with gastric or intestinal cancer

Purpose:To study the changes of t-PA ?u-PA expression and fibrinolysis molecular mark ers in patients with gastric or intestinal cancer in order to elucidate their cl inical significance. Methods:The plasma levels of t-PA ?u-PA ?u-PAR and PAP were measured by ELISA . Gen e transcription of t-PA?u-PA mRNA were detected by Real-time RT-PCR. Results:The plasma levels of u-PA?u-PAR and PAP were elevated in gastric or intestina l cancer patients ,while u-PA was markedly elevated in patients with local infi ltration?lymph node involvement or distal metastasis. u-PA mRNA was higher and t-PA was lower in gastric or intestinal cancer compared to normal tissue. Conclusions:Hyperfibrinolysis was an important factor related with m etastasis potential of gastric or intestinal cancer. t-PA may be a character of well differentiated tissue.
3.Role and change of IL-1?,IL-6 and TNF-? during rabbit lumbar cartilage endplate degeneration

[Objective]To study the role of nitric oxide on the degeneration of the cartilage endplate(CEP).[Method]Twenty-four New Zealand rabbits were divided into experiment and control groups.In the experiment group,the model of lumbar CEP degeneration was established by resection of all lumbar supraspinous and interspinous ligaments,excision of parts of zygapophysial joints and detachment of the posterior paravertebral muscles from lumbar vertebraes.Mechanical instability of the lumbar spine could induced the process of CEP degeneration.The X-ray examination of lumbar spine was examined by at 12,24 and 36 weeks with or without operation,respectively.Changes of the ultrastructure of CEP were also observed by scanning electron microscope(SEM) during this process,and the content of IL-1?,IL-6,TNF-? in the CEP were also detected during the periods.[Result]X-ray graph showed that the CEP calcified gradually with time increasing,and those of the experiment groups calcified more obviously than those of the control groups.The SEM showed that collagen fibers became thin,irregular and fissured and the content of proteoglycan decreased severely with the elapse of time.The changes in experiment groups were more obvious than those in control groups.The content of IL-1?,IL-6,TNF-? had a significant difference between the experiment groups and the control groups(P0.05).[Conclusion]It implies that inflammatary cytokin may play an important role in the development and progression of the degeneration of cartilage endplate.

Objective To observe the effect of inhibition of polo like kinasel (Plk-1) gene expression on 5-Fu and CDDP induced apoptosis of gastric cancer cell line-MKN45 in vitro. Methods The Plk-1 expression was inhibited by chemically synthesized siRNA, the Plk-1 mRNA and protein level were measured by real-time PCR and Western blotting, MKN45 cells proliferation was measured by MTT method, the apoptosis rate was detected by flow-cytometry. Results After treatment by siRNA targeting Plk-1, Plk-1 mRNA level decreased by 51.91% .89.45%.91.03% at 24 h.48 h and 72 h, and Plk-1 protein level decreased by 38.43% and 60.66% at 48 h and 72 h, compared with control group. 8.Effect of moutan cortex extract on Th17 cells of patients with systemic lupus erythematosus

Objective To study the effect of moutan cortex extract on Th17 cells of systemic lupus erythematosus (SLE) patients, so as to provide reference for clinical practice.Methods A total of 168 SLE patients admitted to our hospital from March 2012 to August 2013 were selected as subjects, andrandomized into the observation group and control group.84 patients in observation group received moutan cortex extract treatment and 84 patients in control group received conventional medical treatment.The Th17 and Th1 cell percentage of two groups before and after treatment were compared, and the incidence of adverse reactions were observed.Results All the indicators before treatment showed no statistically significant differences between two groups.After treatment, the Th17 cell percentage of observation group decreased to (0.27 ±0.15)%, and the Th1 cell percentage increased to (2.53 ± 1.72)%, showing more significant changes than control group (P<0.05).The reduction of IL-6 level, SLEDAI score and ESR after treatment were more significant in observation group than in control group(P<0.05).The incidence of adverse reactions was lower in observation group than in control group ( P <0.05 ) .Conclusion Moutan cortex extract can effectively improve the condition of SLE through reducing the Th17 cell percentage and increasing Th1 cell percentage with less adverse reactions.
9.Clinical analysis of Laparoscopic combined fiber choledochoscope common bile duct cholelithotomy in 25 cases

Objective To evaluate the Laparoscopic combined fiber choledochoscope common bile duct cho-lelithotomy for my own clinical experience in rencent five years.Methods Clinic data of 25 cases with choledocholi-thiasis undergoing laparoscopic common bile duct cholelithotomy from June 2010 to March 2014 were retrospectively analyzed.Results The operation was performed successfully in 24 cases,a case conversions to laparotomy,21 cases with T-tube drainage,3 cases with primary ductal submucosa closure.The operative time was 105 to 305min,mean (184.0 ±12.2 ) min, postoperative hospitalization time was 6 to 10 day, mean ( 7.7 ±0.2 ) day, a case residual stones,no bile leakage.Conclusion The beginning of Laparoscopic combined fiber choledochoscope common bile duct cholelithotomy,one-stage suture of common bile duct should be selected carefully, timely transfer laparotomy, should not aim for laparoscopic surgery success rate.
10.Experimental Study of Multiple-Mouse MRI in Evaluating the Therapeutic Effect of Endostatin for Colorectal Carcinoma in Mice

Objective To explore the value of multiple-mouse MRI in evaluating the therapeutic effect of endostatin for transplantation tumor models of colorectal carcinoma in mice.Methods 24 subcutaneously transplantation tumor’s models of colorectal carcinoma (CT-26) in mice were established, 1 week later, 16 tumor-bearing mice were sieved out and divided randomly into two groups: endostatin (ES) group and normal saline (NS) group, treated with intraperitoneal injection of endostatin 6 mg/kg?d 0.2 ml and equal volume of saline respectively for 14 days. Subsequently, MMMRI was performed, and then the mice were killed immediately and the tumors were cut into sections which were stained with hematoxylin and eosin (H&E). Results Subcutaneous fat layer in NS group presented thinner or disappeared on T1WI,while subcutaneous fat layer in ES group presented thicker. The tumors presented inhomogeneous high signal and intratumoral stippled necrosis on T2WI. The tumor’s volumes measured by MRI and pathology were(2723.26?1136.91) mm3 and (3505.76?1350.12) mm3 respectively,there was no difference between these two measures. And there was correlation between MRI results and pathological results. There was no difference of absolute signal intensity between ES group and NS group on T1WI and T2WI. The signal intensity ratio of ES group (3.19?0.28) was higher than which of NS group (2.60?0.47) on T2WI, and there was no difference on T1WI. Conclusion The therapeutic effect for endostatin on transplantation tumor models of colorectal carcinoma in mice can be displayed distinctly in MMMR image, and the inhibition rate of results can be displayed exactly and noninvasively.
