1.The clinical probation of new type oral MRI contrast agent D98A

Objective To evaluate the efficacy of D98A,a newly developed superparamagnetic oral MRI contrast agent,in the diagnosis of abdomen.Methods Plain and contrast MRI after taking 500~1000 ml D98Ain 80 patients were performed by using different field strength MR scanners,and the sequences included T 1WI,T 2WI,and fat suppressed sequences.The contrast effect,distribution,influence on demonstration of contours of abdominal organs,and side effects were studied concerning this suspension.Results It showed that this oral contrast agent had good security character,wide consideration area,low cost,good contrast effect,and high value of clinical application.The gastrointestinal tracts appeared as hypointensity on various MR sequences,and thus,made them easy to be identified.The margins of abdominal organs were easier to be located after taking the contrast,and the distinction had statistical denotation.The demonstration rate of gastrointestinal tracts was raised from 10% to 89% after using D98A.There were no side-effects in all patients. Conclusion D98A is safe and effective in facilitating the localization of the abdominal organs.It can improve the accuracy in delineating the gastrointestinal tracts and in diagnosing the lesions of abdominal organs.

3. The regulatory effects of quercetin combined with DDP on Bcl-2 and Bax expressions in transplanted lung adeno-carcinoma in mice and their significance

Objective: To observe the inhibitory effects of quercetin combined with DDP on the growth of transplanted lung adenocarcinoma LA795 cells in T739 mice and detect the expression levels of Bel-2 and Bax and apoptotic index to explore the action mechanism for the combination therapy. Methods: Thirty-two T739 mice inoculated with LA795 lung adenocarcinoma cells were randomly divided into four groups with 8 mice in each group: control group (A), DDP treatment group (B), quercetin treatment group (C), DDP plus quercetin treatment group (D). The tumor growth was observed on day 16 after drug administration. All mice were sacrificed on day 24 after inoculation. The weight and volume of transplanted tumors were recorded. The expression levels of Bcl-2 and Bax were quantified using immunohistochemical method and image analyzing system. The apoptotic index of transplanted tumors was measured by TdT-mediated dUTP-biotin nick end labeling(TUNEL) method. Results: The growth of transplanted tumors in groups B, C, and D were significantly inhibited. The weight of tumor body in the three groups were markedly lower than that of group A. The tumor inhibitory rates were 38.57%, 26.03%, and 51.58% in groups B, C, and D, respectively. The inhibitory effect of quercetin combined with DDP on tumor growth was enhanced compared with quercetin or DDP alone treatment. The expression of Bcl-2 was significantly higher but Bax was significantly lower in group A compared with group B, C, and D. The difference between the drug therapy groups and control group was significant (P< 0.05 or P<0.01). The apoptotic index was significantly increased in the three drug therapy groups compared with control group and the difference was significant (P<0.01). Conclusion: The combination therapy for quercetin plus DDP significantly suppresses the growth of lung adenocarcinoma cells in mice. The action mechanism may be due to regulation of Bcl-2 and Bax expressions, thereby, inhibiting cell proliferation and inducing apoptosis.
4.Changes of visual quality after Nd:YAG laser posterior capsular dissection to the patient with after-cataract

AIM:To observe and analyze visual quality changes of the patients with posterior capsule opacification ( PCO ) Nd:YAG laser posterior capsular dissection, including the change of the best corrected vision acuity ( BCVA ) , total high- order aberration ( tHOA ) , and the modulation transfer function ( MTF) .
METHODS:In this prospective observational study, 100 cases of patients ( 100 eyes ) with posterior cataract underwent Nd:YAG laser posterior capsular dissection ( posterior capsular diameter dissected was 5mm or higher). The mean age was 65. 52±7. 01 years old. The change of the BCVA was collected. The tHOA and MTF under the 3mm and 5mm pupil diameter were assessed by iTrace respectively before and after Nd:YAG laser posterior capsular dissection.
RESULTS:All the surgery went well without obvious intraoperative and postoperative complications happened. The preoperative BCVA was 0. 451 ± 0. 023 while the postoperative BCVA was 0. 763±0. 025. The difference of BCVA before and after Nd: YAG laser surgery was statistically significant (P<0. 01). At 3mm pupil diameter, the tHOA preoperative was 0. 551 ± 0. 031 while the postoperative tHOA was 0. 214± 0. 011, the differences were significance (P<0. 05). At 3mm pupil diameter while the spatial frequencies ( 5cpd, 10cpd, 15cpd, 20cpd, 25cpd, 30cpd ) respectively, the MTF tHOA value postoperative (0. 644±0. 023, 0. 49±0. 011, 0. 311±0. 015, 0.202±0. 018, 0. 056±0. 027, 0. 041±0. 011) were significantly higher than that preoperative (0. 401±0. 021, 0. 261±0. 026, 0. 179±0. 012, 0. 108±0. 014, 0. 031±0. 016, 0. 022±0. 021), and the difference has statistical significance (P<0. 05). At 5mm pupil diameter, the tHOA preoperative was 0. 752±0.028 while the postoperative tHOA was 0. 361±0. 014, the differences were significance (P<0. 01). At 5mm pupil diameter while the spatial frequencies ( 5cpd, 10cpd, 15cpd, 20cpd, 25cpd, 30cpd) respectively the MTF tHOA value postoperative (0. 426±0. 027, 0. 209±0. 018, 0. 172±0. 013, 0. 116±0. 015, 0. 049±0. 010, 0. 034±0. 014 ) were significantly higher than that preoperative (0. 234±0. 021, 0. 102±0. 019, 0. 088±0. 016, 0. 058±0. 022, 0. 021±0. 014, 0.016 ± 0. 011 ), and the difference had statistical significance (P<0. 05).
CONCLUSION: Patients with posterior capsule opacification ( PCO ) Nd:YAG laser posterior capsular dissection can help improve BCVA, reduce tHOA, increase MTF tHOA values, and significantly improve visual quality of patients.
5.Construction of lentiviral vector of microRNA targeting IGF1R gene and its inhibition effect on liver cancer cell growth

Objective To study the effects of lentiviral vector of microRNA targeting IGF1R gene on growth of liver cancer cells.Methods The complementary DNA containing both sense and antisense Oligo DNA of the targeting sequence was designed,synthesized and cloned into the pPRIME vector,named pPRIME-IGFIR-miR30-shRNA.The viruses were propagated on 293T cells.Viruses were purified by CsCI gradient according to standard techniques,and functional PFU titers were determined by plaque assay on 293 cells.The effect of pPRIME-IGFIR-miR30-shRNA on IGFIR expression of Hep3B、SMMC7721 cells was detected by RT-PCR and Western blot.The antitumor potential of PRIME-IGFIR-miR30-shRNA to Hep3B、SMMC7721 cells was evaluated by CCK-8 assay and Tunel.Results PRIME-IGFIR-miR30-shRNA was constructed successfully.Functional PFU titers of pPRIME-IGFIR-miR30-shRNA were 4.58?109PFU/ml.PRIME-IGFIR-miR30-shRNA inhibited IGFIR expression and the proliferation of Hep3B、SMMC7721 cells.Conclusions PRIME-IGFIR-miR30-shRNA expressing IGFIR-siRNA can inhibit IGFIR expression and may be used for further investigation of gene therapy of liver cancer.
7.The improvement on surgical treatment of primary hyperthyroidism

Objective To improve the quality of thyroid surgery though the special surgical instruments and operating skill.Method Sub-total thyroidectomy for 504 pafients with primary hyperthyroidism were done through MPBS illuminating instruments and dissection,scrape absorption and electric coagulation skills of PMOD.Results Five hundred and four eases of sub-total thyroidectomy were successfully performed.The operation time was(50±20)minutes.The blood loss volume Was(60±30)ml.The hospital duration was(54±1)days.The incisions were short and the sears wore even.There was 1 case recurrence of hyperthyroidism and 2 cases of hypothyroidism after(5±3)years follow-up in 482 patients.There were no postoperative complications of hypoparathyroidism and injury of nerve.Conclusion The MPBS and PMOD instruments can be safely applied in thyroid surgery with the advantage of less blood loss,shorter operation time,smaller incision and less side-injuries.
8.Evaluation of pedicle screws applied in the affected vertebra for the treatment of thoracolumbar burst fracture

[Objective]To investigate curative effect for thoracolumbar burst fractures by the method of vertebral pedicle screw fixation applied in the with transpedicular bone graft,and consider its indications.[Method]Thirty cases of thoracolumbar burst fractures were treated by vertebral pedicle screw fixation combining transpedicular bone graft.Among them 22 were male and 8 were famale,age ranged 18-64.All of cases were single segment fractures.The average height of anterior border was 35% before the operation,the average of Cobb's angle was 26?,the percentage of midsagital diameter was 55%.According to Frankel's neurological function classification,preoperative neurological function was Grade A in 4 cases,B in 8,C in 5,D in 6,E in 7.[Result]All cases were followed up for 6 to 30 months.All patients' pain relieved distinctly,the average height anterior border was increased to 97%,the average of Cobb's angle was 3.5?,the percentage of midsagital diameter was 92% postoperatively,showing significant difference(P
9.Inducing bone mesenchymal stem cells of rabbits into chondrocytes using the technology of micromass culture in vitro

[Objective] To study the methods of inducing bone mesenchymal stem cells(BMSCs)of rabbits into chondrocytes in vitro and the interaction of transforming growth factor ?1(TGF-?1),insulin-like growth factor-Ⅰ(IGF-Ⅰ)and basic fibroblast growth factor(bFGF).[Methods]BMSCs of rabbits were primarily cultured and subcultured in vitro,and then divided into four groups according to the difference of factors:group A receiving TGF-?1 and bFGF;group B receiving TGF-?1 and IGF-Ⅰ;group C receiving TGF-?1;group D receiving no cell growth factor.After three weeks all the four groups were detected by methyl thiazolyl tetrazolium(MTT)assay,measurement of glycosaminoglycan(GAG)and immunohistochemistry.[Results]Immunohistochemical detection of collagen Ⅱ was positive in groups A,B and C.The results of the MTT assay and the GAG content in groups A and B were obviously higher than those in groups C and D.[Conclusion]Rabbit BMSCs can be induced into chondrocytes under certain conditions.TGF-?1,IGF-Ⅰ and bFGF have synergy effect in the differentiation from BMSCs into chondrocytes.
10. A new monoterpenoid glycoside from aerial parts of Siegesbeckia pubescens

Objective: To study the chemical constituents of the aerial parts of Siegesbeckia pubescens. Methods: A systematic separation of chemical constituents was conducted. Detailed chemical investigation of S. pubescens led to the isolation of six compounds by comprehensive chromatographic Methods: (Extraction, Silic gel C.C, Sephadex LH-20, ODS C. C). The structures of them were fully determined based on spectroscopic analysis including IR, HR-ESI-MS, ESI-MS, 1H-NMR, 13C-NMR, DEPT, HSQC, and HMBC spectrum. Results: A total of six compounds were isolated from the fraction of n-butanol extract of S. pubescens including one new monoterpenoid glycoside named (2Z,5E)-7-hydroxy-3,7-dimethyl-2,5-octadiene-1-O-[α-L-rhamnpyranosyl (1→6)]-β-D-glucopyranoside (1), four known diterpenoid glucosides named pubeside D (2), ent-15,16,19-trihydroxypimar- 8(14)-en-2-one-19-O-β-glucopyranoside (3), ent-15,16-dihydroxypimar-8(14)-en-2-one-19-oic-β-glucopyranoside (4), and neodarutoside (5), and eleutherazine B (6). Conclusion: Compound 1 is a new compound, named as siegeside F, and compound 6 is isolated from this species for the first time.
