1.The analysis of bacteria strains and sensitivity of bacteris to antibiotics in acute obstructive cholangitis with suppuration

Objective To analyze the changes of bacteria stains in acute obstructive cholangitis with suppuration (AOSC) and sensitivity of different bactera strains to antibiotics in recent decade.Methods The data of bacterial susceptibility of AOSC patients and sensitivity of different bacteria strains to antibiotics in our hospital collected from 1999 to 2001 (group A,n =54) and from 2009 to 2011 (group B,n =62) were analyzed.Resules In group A,there were 29 male and 24 female with age range of 35 ~ 82 and mean age of 57.5 years,and in group B,there were 23 male and 39 female with age range of 39 ~ 87 and mean age of 68.2 years.There were no differences in bacteria strains infected between two groups.However,there was a trend of increase in the proportion of the Pseudomonas aeruginosa and Staphylococcus aureus infection,and a trend of decrease in the proportion of Escherichia coli infection.The degrees of sensitivity of Escherichia coli to ciprofloxacin,ceftazidime,cefaclor and ceftriaxone were statistically different from those observed ten years ago ; and the degrees of sensitivity of Pseudomonas aeruginosa to ceftazidime and cefaclor were statistically different from those detected ten years ago as well.In recent years,the sensitivity of bacteria to antibiotics was on a downwards trend.Conclusions The pathogens of acute obstructive and suppurative cholangitis did not obviously change in recernt decade,but the sensitivity of bacteria to antibiotics was lowered.
2. Effect and Mechanism of Ferulic Acid Against Apoptosis of HUVECs Cells Induced by High-Glucose

OBJECTIVE: To study the protective effects of ferulic acid on apoptosis of human umbilical vein endothelial cells (HUVECs) induced by high-glucose and its mechanisms. METHODS: HUVECs cells were cultured, survival rate of HUVECs cells was determined by MTS colorimetric method. Cell intracellular ROS were detected with fluorescence probe DCFH-DA.Expressions of VPO1, Bax and Bcl-2 were detected by Real-time PCR and Western Blot. RESULTS: Compared with normal control group, the survival cell number in the high glucose-treated group were obviously decreased, the expressions of VPO1 and Bax were obviously up-regulation and the expressions of Bcl-2 were obviously down-regulation. Compared with high-glucose group, the level of intracellular ROS and the expression lever of VPO1 and Bax were obviously down-regulation, HUVECs proliferation rate and the expression of Bcl-2 significantly increased after incubated with ferulic acid. CONCLUSION: Ferulic acid can protect HUVECs against the apoptosis induced by high-glucose in vitro, and its mechanism could be related with the reduced VPO1 expression and the inhibition of the oxidative stress.

6.Changes of cAMP and cGMP contents in the central nervous system in morphine dependent and withdrawal rats

Objective To investigate the contents of cAMP and cGMP in the central nervous system in morphine dependent and withdrawal rats.Methods A physical morphine dependent model in rats was established by subcutaneous injection of morphine in gradually increasing doses.The cAMP and cGMP contents in the brain regions were determined by radioimmunoassay.Results Compared with control group,morphine dependence could significantly decrease the cGMP content or profoundly increase the cAMP content and the cAMP/cGMP ratio in rat striatum,diencephalons,midbrain,pons and hippocampus,but these changes described above were not detected in cerebellum.Compared with morphine dependent group,naloxone induced withdrawal could significantly decrease the cGMP contents or increase the cAMP contents and the cAMP/cGMP ratio in striatum and hippocampus,but these changes described above were not observed in the other regions.Conclusion The changes of the cAMP and cGMP contents may be one of the important molecular mechanisms leading to morphine dependence and abstinence.
7.Therapeutic Effect of Compound Glycyrrhizin on Acute Biliary Tract Infection Combined with Abnormal Liver Function

OBJECTIVE:To observe the therapeutic effect of compound glycyrrhizin on acute biliary tract infection com?bined with abnormal liver function.METHODS:The patients were randomly divided into trial group(141cases)and control group(120cases).The control group received ATP,CoA and inosine,and the trial group received compound glycyrrhizin60~120ml/d in addition to the treatment for control group.RESULTS:In trial group,113cases were remarkedly effective and28cases effective with a total effective rate of100%,and in control group,79cases were remarkedly effective and29cases effective with a total effective rate of90%.CONCLUSION:Compound glycyrrhizin has the anti-inflammatory effect and can protect the cell membrane of liver cells,shorten the hospital stay duration and decrease the occurrence of postoperative complications.
8.Expression of estrogen receptor and progesterone receptor in different tumor tissues of invasive ductal carcinoma of breast

Objective To investigate the expressions of estrogen receptor (ER) and progesterone receptor (PR) in different tumor tissues of invasive ductal carcinoma(IDC) of the breast. Methods A total of 32 cases of IDC samples were collected. ER and PR expressions in 4 different sites of every sample were measured by immunohistochemical(IHC) assays. Results Overall, a high concordance in both estrogen receptor and progesterone receptor expressions are observed between different tumor tissue sites. The best Kappa values were 0.789 and 0.810 for ER and PR expression respectively. The worst Kappa values were 0. 563 and 0.672,respectively. Conclusion ER and PR expression is not significantly different in different tumor tissues of the same IDC sample. Repeated testing in different tumor tissue sites has little contributions in clinical settings.
9.Treatment of alveolar cleft by autogenous cancellous bone graft obtained with mini incision

Objective: To sammarize the methodology of the treatment of alveolar cleft by autogenous cancellous bone graft obtained with mini incision(ABGM).Methods: 22 cases of alveolar cleft received ABGM by one operator. The cleft area was restored with autogenous cancellous bone from iliac crest by cylinder osteotomes for extraction of bone transplants(COEBT) with mini incision. All the patients were follow up for at least six months postoperatively. The result of the bone grafting was evaluated on the radiographs. Results: ①The overall survival rate of ABGM was 95.8%,and clinical successful rate was 91.7%;②The successful rate of ABGM in group of patients above 18 year old, or with complete cleft palate or bilateral cleft lip and palate was lower than that in other groups.③The wound for extracting autogenous cancellous bone from iliac crest by COEBT with mini incision was smaller than that by osteotomy. Conclusion: ABGM with COEBT is an effective way in the treatment of alveolar cleft. The successful rate of ABGM is related with the age and cleft style of the patients.
10.Rapamycin induces apoptosis of lung adenocarcinoma cell A549 under hypoxia and its molecular mechanism

Objective:To investigate the molecular mechanism underlying apoptosis of lung adenocarcinoma cell line A549 induced by rapamycin, a type of mammalian target of rapamycin (mTOR) signal pathway inhibitor, combined with hypoxia treatment. Methods:Rapamycin was applied to treat A549 cell line under hypoxia for 16 h. Then the discrepancy of cell apoptosis between treated and untreated control was compared by Hochest 33258 staining. After being treated with rapamycin combined with hypoxia, the mRNA and protein expressions of survivin, a kind of anti-apoptotic molecule, were tested by real-time PCR and Western blotting. Western blotting was employed to test the difference of hypoxia-inducible factor (HIF)-1α expression level before and after rapamycin was added. In additon, chromatin immunoprecipitation(ChIP) assay was further applied to explore the mechanism underlying the effects of rapamycin under hypoxia in inducing apoptosis of A549 cells. Results:Rapamycin induced apoptosis of A549 cell line under hypxia in vitro. In A549 cell line, the expressions of survivin at mRNA (P<0.01) and protein levels, were both decreased by rapamycin combined with hypoxia treatment. Besides, ChIP assay revealed that rapamycin inhibited the expression of survivin at the transcriptional level through decreasing the hypoxia-induced up-regulation of HIF-1α and its binding with the promoter of survivin gene. Conclusion:Rapamycin exerted anti-tumor effects by inhibiting the expression of HIF-1α and decreasing the binding of HIF-1α with the promoter of survivin gene, thereby down-regulating the increase in the expression of apoptosis inhibiting gene survivin.
