1.Development of fluorescent quantitation PCR method for detection of urinary CK20

Objective:To develop a novel real-time fluorescence PCR method using Taqman probe for convenient,fast,sensitive and specific detection of urinary CK20 and use in early diagnosis and follow-up of transitional cell carcinoma of the bladder(TCCB).Methods:CK20 gene from cultured T24 cells was amplified by conventional RT-PCR,the standard quantitative plasmid was constructed by T-Aclone method.Taqman probe and primers was designed according to the sequence of CK20 cloned gene,the real-time PCR method for determination of CK20 mRNA was established and evaluated.Results:The developed real-time PCR method showed high sensitivity(10 2copies /?l) and good specificity;the linear range was 102 ~109 copies /?l,the coefficient variation(CV) was 0.89 % in intra-assay and 2.45% in day to day.Conclusion:A novel real-time fluorescence quantitative PCR method for detection of CK20 was established.It provided a basis for developing new diagnostic kits using the Taqman probe for clinical application.
2.Long no-coding RNAs and prostate cancer

Long non-coding RNAs ( lncRNAs) are a class of RNA molecules which are unable to codify proteins. They express abnormally in tumor cells including prostate cancer cells and play a significant role in development of tumor. Some lncRNAs have been proven to affect physiological processes of prostate cells and act as tumor suppressor or activator and could be regulated through several methods. Thus, lncRNAs have the potential to be therapeutic targets of prostate cancer. This review introduces expressed abnormally ln-cRNAs in prostate cancer and their functions and mechanisms in prostate cancer.

BACKGROUND:With the development of sports medicine and research of patel ofemoral disorder, patel a alta has been paid increasing attention as the exact pathogenesis of patel ofemoral disorder. There are many radiographic measurement indexes of patel a alta. A large number of clinicians and radiography physicians are trying to diagnose and treat this disease as early as possible.
OBJECTIVE:To review the radiographic measurement and research progress of patel a alta, find out the advantages and disadvantages of different indexes, and look into the future development prospects.
METHODS:A computer-based online search of Pubmed and CNKI databases between 1989 and 2013 was performed for articles addressing radiographic diagnose and measurement patel a alta. The key words were“patel a alta, patel a height, measurement, diagnosis”in English and“patel a alta, patel a height, patel a position, diagnosis, measurement”in Chinese.
RESULTS AND CONCLUSION:Now there are many measurement indexes of patel a alta and they are divided into two groups:indirect index and direct index. The indirect index measures the relative location of patel a and tibia, such as Insal-Salvat index, Blackburne-Peel index, and Caton-Deschamps index. The direct index measures the relative location of patel a and femur, such as patel otrochlear index, and patel ophyseal index. The latest study indicates that the biomechanics of patel ofemoral joint is the key to understand the pathogenesis of patel ofemoral disorder. As the MRI technique develops, direct index for the location of articular cartilage of patel ofemoral joint, such as patel otrochlear index, seems to be the best measurement to patel a alta. However we stil have many problems in the study of patel otrochlear index.
5.Efficacy of microlaryngoscopy radiofrequency in treatment of patients with tongue base cysts

Objective To investigate the clinical efficacy of microlaryngoscopy radiofrequency in treatment of patients with tongue base cysts. Methods 42 patients with tongue base cysts from January 2013 to December 2014 were randomly divided into treatment group and control group, the treatment group underwent microlaryngoscopy radiofrequency treatment, the control group underwent direct laryngoscopy under the tongue cystectomy, then the operation time, blood loss and postoperative recurrence rate were observed and recorded, and self-evaluation method using visual analog (VAS) to evaluate the effect of surgery. Results The operative time of treatment group was significantly shorter, and the blood loss was significantly less than the control group, the difference between the two groups was statistically significant (P < 0.01); for 1-year follow-up, six-month recurrence rate in control group was 9.52%, the treatment group had no recurrence cases, the difference was not statistically significant (P > 0.05); 12 months, the recurrence rate was 28.57% in the control group, the treatment group, the recurrence rate was 4.76%, the difference between the two groups was statistically significant (P < 0.05); Preoperative VAS score of the two groups have no significant difference, while after surgery, the VAS scores' difference was statistically significant, even treatment group compared with the control group VAS scores after surgery was statistically significant (P < 0.01). Conclusion Microlaryngoscopy radiofrequency in treatment of tongue base cysts have advantages of significant efficacy, safe and reliable, It's worthy of clinical promoting.
6.Anti-angiogenic Activity of Ray Cartilage Glycosaminoglycans:An Empirical Study

OBJECTIVE:To investigate the anti-angiogenic activity of ray cartilage glycosaminoglycans(RCG).METHODS:Primary culture of human umbilical vein endothelial cells(HUVEC)was performed,and MTT was adopted to evaluate the effects of RCG on the proliferation of HUVEC;Corneal neovascularization(CNV)was induced by sutures on Wistar rats,and CNV area was calculated to evaluate the effects of different concentration of RCG on CNV in Wistar rats.The model of mice with Lewis lung carcinoma was made to observe the effects of different concentration of RCG on tumor growth,with inhibition ratio of primary tumor measured and microvessel density(MVD)quantitated by immunohistochemical staining.RESULTS:RCG obviously inhibited the proliferation of HUVEC in vitro,with IC50 value at 62.93 mg?mL-1.As compared with normal saline group,RCG groups showed smaller areas of new blood vessels(P
7.Hemodynamic effects of rocuronium in patients with rheumatic heart disease during induction of anesthesia

Objective To evaluate the hemodynamic effects of rocuronium in patients with rheumatic heart disease during induction of anesthesia.Methods Twenty patients of either sex (ASA classes Ⅲ -Ⅳ ;NYHA classes Ⅱ -Ⅲ) scheduled for valve replacement were included in this double blinded study. Their ages ranged from 34 to 62 yr and weights from 48 to 70 kg. The patients were premedicated with intramuscular morphine 0.2 mg?kg-1 and scopolamine 0.3mg. Radial artery was cannukted for direct BP monitoring and Swan-Ganz catheter was placed via right internal jugular vein for hemodynamic monitoring before induction of anesthesia.TOF,BIS,ECG and SpO2 were also monitored throughout anesthesia. Anesthesia was induced with midazolam 0.05-0.1 mg?g-1 and fentanyl 10-15?g?kg-1. The patients were randomized to receive either rocuronium 0.6 mg?kg-1 (group Ⅰ,n=10) or vecuronium 0.1 mg?kg-1(group Ⅱ,n=10) to facilitate tracheal intubation when BIS value dropped to 60%.The patients were mechanically ventilated (VT 8-10 ml?kg-1 ,RR 10-12 bpm).Systolic arterial pressure (SAP),MAP,HR,cardiac output (CO), PCWP, CVP, mixed venous blood O2 saturation (SvO2), BIS and TOF were recorded and stroke volume (SV), stroke index (SI), LVSWI and rate-pressure product (RPP) were calculated before anesthesia (To), 1 min after administration of muscle relaxant (T1), when TOF reached 0 (T2) and 1,2,3,4,5,7,10,15,20,25,30 min after tracheal intubation (T3-13).Results The demographic data including age, sex and body weight were comparable between the two groups. 8.Can procalcitonin differentiate sepsis from noninfectious systemic inflammatory response syndrome?

Sepsis is featured with acute onset, rapid progress and high mortality.Because its clinical manifestations is very similar to noninfectious systemic inflammatory response syndrome, it is difficult to diffferentiate the two diseases, which results in delayed use or abuse of anti-infective drugs.Procalcitonin is widely recognized as a sepsis biomarker, but it has its limits.Procalcitonin must be interpreted cautiously, in conjunction with the clinical picture.Dynamic monitoring of procalcitonin can give clinicians more help.
9.Rutin inhibits hydrogen peroxide-induced apoptosis through PI3K/AKT signaling pathway in human lens epithelial cells

Objective To explore whether PI3K/ AKT signaling pathway participates in the inhibiting effect of Ru-tin on H2 O2-induced apoptosis in human lens epithelial cells( HLEC). Methods HLEC were divided into four groups: control group,H2 O2 group,rutin group,LY294002 group. Cell survival rates were determined by a 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay; cell apoptosis rates were monitored by flow cytometry with Annexin V-FITC and propidiun iodide(PI) staining. Western blot was used to measure the expres-sion levels of AKT and p-AKT. Results H2 O2 induced HLEC apoptosis. Compared with H2 O2 group,rutin group not only increased the expression lever of p-AKT,but also reduced cell apoptosis rate(P < 0. 01). In LY294002 group, LY294002, an inhibitor of PI3K/ AKT signaling pathway,could significantly block the change of these inde-xes produced by rutin group(P < 0. 01), but no significant change compared with H2 O2 group. Conclusion Rutin inhibits H2 O2-induced cell apoptosis and may be associated with PI3K/ AKT signaling pathway.
10.Determination of EPA and DHA in Ray Oil by HPLC

OBJECTIVE:To establish a method for the determination of EPA and DHA in ray oil.METHODS:The determination was carried out on Diamonsil C18 column.The mobile phase consisted of acetonitrile-water(95 :5) with flow rate of 1.0 mL?min-1 and detection wavelength set at 220 nm.The column temperature was set at 25 ℃.RESULTS:The linear ranges were 1~4 ?g for EPA(r=0.999 1) and 2~8 ?g for DHA(r=0.999 4).The average recoveries were 99.23% for EPA(RSD=0.49%) and 99.30% for DHA(RSD=0.27%).CONCLUSION:The method is simple,rapid,accurate and reproducible for the detection of EPA and DHA in ray oil.
