1.Detection on early gene expression of tissues around hematom in rats with cerebral hemorrhage with the technique of DNA microarray

BACKGROUND: Changes of physiological structure, changes of phenotype and first basic excision are all the changes of gene expression. The technique of DNA microarray is a new method to filtrate target genes fleetly and largely by using the theory of base-partnershin, which can holistically and magnificently study the expression and function of organics genes. OBJECTIVE: To study early differential expression genes of rats with cerebral hemorrhage with DNA microarray and establish academic foundation for exploring mechanism of cerebral hemorrhage.DESIGN: Randomized controlled research.SETTING: Department of Neurology, Affiliated Hospital of North Sichuan Medical College. MATERIALS: The experiment was conducted at the Affiliated Hospital of North Sichuan Medical College from October 2002 to December 2003. Twenty Wistar rats, of either gender, with body mass of 220-260 g, without special pathogen, provided by Experimental Animal Center of Chongqing University of Medical Sciences, were selected and randomly divided into control group and cerebral hemorrhage group, with 10 in each group. METHODS: Animal models with cute cerebral hemorrhage of rats were established with type Ⅶ collagenase tridimensional localization method,and 4 hours later tissues around hematom and normal cerebral tissue at the same part were detected with gene chip. Fluorescent signal was scanned with scanning apparatus and analyzed with computer. Result of genic expressive pattern was researched with reverse transcriptase-polymerase chain reaction (RT-PCR). MAIN OUTCOME MEASURES: Result of gene chip in cerebral tissue of rats and result of RT-PCR.RESULTS: Four hours after acute cerebral hemorrhage, 129 differential expression genes were screened out, in which there were 114 up-regnlation genes and 15 down-regulation genes. Those genes were mostly related to the following aspects: stress, immunological response, apoptosis, energy metabolism and signal transmitting. Genes related with inflammatory impairment were mostly obvious. The result of RT-PCR suggested that the level of genic expression was as the same as the result of Cdna chip, which indicated that genic expressive pattern based on gene chip had great reliability.CONCLUSION: Early cerebral hemorrhage has many differential expression genes, which can play an important role in hemorrhagic brain damage.

Objective To observe the protective effect of aprotinin on the cerebral ischemia-reperfusion injury. Methods Rat model of cerebral ischemia-reperfusion injury was created by the middle cerebral artery embolization. The changes of IL-1β,IL-6 and IL-8 content in the brain tissue were measured by ELISA. The neurological scores were made on Zea Longa 5-point scale. Results In model group, the IL-1β,IL-6 and IL-8 protein content began to increase slightly at 3 hours compared with sham group, reached the peal[at 12 hours. In the aprotinin treated group, aprotinin reduced the protein content. It showed no significant difference between model group and aprotinin treated group at 3 hours, however and there was greatly difference at 6,12 and 24 hours between these two groups. Conclusion The present study provides in vivo evidence that aprotinin protects brain against cerebral ischemia-reperfusion injury. The mechanism is related to the decrease of cerebral levels of IL-1β,IL-6 and IL-8.
5.Effect of aprotinin on nuclear factor-?B expression in brain regions around the hematoma of intracerebral hamorrage in rats

Objective To investigate the effect of aprotinin on nuclear factor-?B p65 expression in brain regions around the hematoma of intracerebral hamorrage (ICH)in rats. Methods ICH models were induced utilizing the local injection of collagenase into the basal ganglia with sterile technique and divided into ICH group, aprotinin group(ATG) and Pyrrolidine dithiocarbamate(PDTC) group. The expression levels of NF-?B p65 protein and mRNA were detected by immunohisochemistry or RT-PCR at 6 h,24 h,72 h,7 d after injection. Serum anti-M2 mitochondrial antibody (AMA-M2) was screened with immune colloidal gold technique in 6 008 individuals aged over 60 years who underwent health check-up in Xuhui District of Shanghai,the positive samples were retested with ELISA method.The results showed that 1.31% (79/6 008) individuals were AMA-M2 positive; the positive rate was 0.82% (18/2 186) in males and 1.60% (61/3 822) in females.Eleven subjects with positive AMA-M2 underwent further investigation,8 cases were diagnosed as primary biliary cirrhosis (PBC) and the remaining 3 were highly suspected as PBC.All 11 individuals were abnormal in biochemical tests of liver function and/or clinical symptoms.

OBJECTIVE: To study protection effect of trasylol on cerebral injury resulting from cerebral ischemia-reperfusion in the interference of immunological response.METHODS: 105 SD rats were randomly assigned into sham operation group,model group and trasylol group (n=35).Focal ischemia-reperfusion model were established by occluding middle cerebral artery.24 h before ischemia and at the 0 h,24 h,4 d,6 d,8 d,10 d,12 d and 14 d of reperfusion,trasylol group were given trasylol via tail vein and other two groups were given normal saline.The infiltration of CD4+ and CD8+ T lymphocytes was observed using immunohistochemistry at different stages of cerebral ischemia-reperfusion.RESULTS: As compared with sham operation group,CD4+and CD8+ T lymphocytes count in model group were increased significantly after cerebral ischemia-reperfusion injury and obtained maximum value at 10th day.CD4+ and CD8+ T lymphocytes count in trasylol group were lower than in model group after cerebral ischemia-reperfusion injury and obtained maximum value at 8th day (P

Objective To explore the effects of one time loading of different doses of simvastatin before percutaneous coronary intervention (PCI)on post-PCI inflammation,oxidation stress and the endothelium function in (ACS) patients. Methods Totally 124 cases with ACS were randomly divided into two groups:high dose simvastatin group(40 mg,62 cases),low dose simvastatin group (20 mg,62 cases). Each group was given the same basic treatment. Blood samples were obtained from all the patients before and 12 h after PCI,and endothelin-1 (ET-1),nitric oxide (NO),interleukin-10(IL-10),high sensitive - C reactive protein(hs-CRP),superoxide dismutase (SOD) and malondialdehyde(MDA) were detected. Results The baseline information,distribution of sex,age,and implanted frames had no significant differences between the two groups (P>0.05).Before PCI,the levels of ET-1,NO,IL-10,hs-CRP,SOD and MDA had no significant differences (P>0.05) between the two groups.After PCI,ET-1,IL-10 and hs-CRP levels in simvastatin 40 mg group were significant lower while NO level was higher than in simvastatin 20 mg group[(4.4 ± 1.1)ng/L vs.(4.8±1.2)ng/L,t=2.03,P=0.044; (15.0±6.3) ng/L vs.(18.7±9.0)ng/L,t=2.68,P=0.008;(26.9±10.0)ng/L vs.(31.5± 11.7)ng/L,t=2.52,P =0.022;(51.9± 10.9)μmol/L vs.(47.1±11.8)μmol/L,t=2.37,P=0.020].There were no significant differences in MDA and SOD levels between the two groups.For safety,all the patients had no abnormality in liver and kidney function after treatment. Conclusions Compared with 20 mg simvastatin loading before PCI,the 40mg simvastatin loading may decrease the inflammatory cytokines and improve the endothelium function more effectively.
10.Analysis of lead level in 1963 cases of children's blood in Beilun district of Ningbo city

Objective In order to understand the children's blood lead load and presence status of blood lead levels,provide the first-hand information on further preventive intervention in high lead hyperlipidemia.Methods This study routinely collected 5 to 9-year-old children's blood,a total of 1963 cases.Peripheral blood lead levels was detected by atomic absorption spectrometry.Results Blood lead ≥ 100μg/L is 45 cases in 1963 cases of children, the rate is 2.3%, the highest is 439μg/L,only one.Other 44 persons' blood lead level is 100 ~ 200μg/L,average 149.2μg/L.Conclusion The children's blood lead levels in Beilun district of Ningbo city is in normal state in general,not high blood lead load.
