1.Influence of the sperm DNA integrity on ART result

Objective To explore the relationship between the sperm DNA integrity and ART result.Methods Employ improved SCD test for sperm DNA integrity assay.Meanwhile,statistical methods was used for data analysis.Results After compaed the infertility rate( IVF:77.4% vs 84.8%,P =0.366;ICSI:90.1% vs 92.1%,P =0.899),cleavage rate ( IVF:97.1% vs 96.4%,P =0.958 ; ICSI:96.0% vs 99.0%,P =0.845 ),high-quality embryo rate ( IVF:42.8% vs 38.6%,P =0.485 ; ICSI:25.8 % vs 46.2%,P =0.002 ) and pregnancy outcome ( IUI:13.8% vs 16.6%,P =1.000;IVF:35.3% vs 73,7,P =0.150;ICSI:49.5% vs 42.8,P =0.832) between the populations with normal and abnormal sperm DNA integrity during ART therapy,there is no statistical difference was found except high-quality embryo rate with ICSI therapy.Conclusion Sperm DNA integrity value can't be used to predict the ART outcomes independently.
2.Relationship between homocysteine,C-reactive protein and type 2 diabetes peripheral vascular disease

Objective To observe the relationship between homocysteine (Hcy),C-reactive protein (CRP) and type 2 diabetes peripheral vascular disease (PVD).Methods Fifty-eight patients with type 2 diabetes according to the condition of PVD were divided into simple type 2 diabetes mellitus group (DM group,30 cases) and PVD group (28 cases).Twenty-five healthy people were chosen at the same period as control group.The fasting blood glucose (FBG),glycosylated hemoglobin (HbA1c),total cholesterol(TC),triglyceride (TG),high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C),Hcy and CRP levels were detected among 3 groups,and the body mass index (BMI),ankle brachial index(ABI) was calculated.Results The BMI,TG,TC and HDL-C levels among 3 groups had no statistical significance (P >0.05).The FBG,HbA1c and LDL-C in DM group and PVD group were significantly higher than those in control group [(7.54 ± 1.25),(8.12 ± 1.57) mmol/L vs.(4.68 ±0.54)mmol/L; (6.92 ± 0.89)%,(7.54 ± 0.97)% vs.(4.80 ± 0.54)% ; (3.79 ± 0.73),(4.41 ± 0.93) mmol/L vs.(2.68 ± 0.54) mmol/L],and the difference was statistically significant (P < 0.05).But the difference between PVD group and DM group had no statistical significance (P > 0.05).Hcy and CRP in DM group and PVD group were higher than those in control group [(10.26 ± 3.62),(19.83 ± 5.54) μ mol/L vs.(8.92 ± 2.41)μ mol/L; (4.12 ±0.57),(5.47 ±0.85) mg/L vs.(2.73 ±0.54) mg/L],ABI was lower than that in control group(0.92 ± 0.18,0.81 ± 0.16 vs.1.02 ± 0.23),and the difference was statistically significant (P < 0.05);Hcy,CRP in PVD group were obviously higher than those in DM group,ABI was lower than that in DM group,and the differences were statistically significant (P< 0.05).Spearman analysis showed that Hcy was negatively correlated with ABI (r =-0.635,P < 0.05),CRP was negatively correlated with ABI (r =-0.529,P < 0.05),and Hcy was positively correlated with CRP (r =0.528,P < 0.05).The partial correlation analysis results showed that Hcy and ABI still showed a negative correlation (r =-0.315,P < 0.05),CRP and ABI still showed a negative correlation (r =-0.243,P < 0.05).Conclusion Hcy and CRP participating in the occurrence and development of type 2 diabetes PVD,can be used as a predictor of type 2 diabetes PVD.
3.X-ray evaluation of alpha-calcium sulfate hemiydrate/biphasic bioceramics bone for repairing bone defects of rabbit radius

BACKGROUND:Alpha-calcium sulfate hemiydrate/biphasic bioceramics (α-CSH/BCP) as a bone repair material has been widely used in the clinic, but there are stil some deficiencies. OBJECTIVE:To observe the effect of α-CSH/BCP on the repair of bone defects of the radius in rabbits. METHODS:The unilateral radius of 18 adult New Zealand white rabbits was selected to establish bone defect models, and then these rabbit models were randomized into three groups that were treated with α-CSH,α-CSH/BCP, BCP materials, respectively. At 4, 8, 12 weeks after implantation, bone samples were taken for gross and X-ray observations. RESULTS AND CONCLUSION: (1) Gross observation: At 12 weeks of implantation, the bone defect was completely repaired in the α-CSH/BCP group, and the defected radius recovered basicaly; in the α-CSH group, bone defects were visible; and in the BCP group, there was stil some material for filing and coverage. (2) X-ray observation: In the α-CSH group, the degradation of implanted materials was faster than new bone formation, and bone defects were visible; in the α-CSH/BCP group, bone defects were healed, cortical bone remodeling was good, and the bone marrow cavity was re-canalized; in the BCP group, the implanted materials were stil visible, and bone defects were not repaired. These findings indicate that the α-CSH/BCP bone repair material can significantly promote the repair of bone defects of rabbit radius, with a better repair effect than α-CSH or BCP alone.
4.Biocompatibility of coralline hydroxyapatite/bone morphogenic protein composite for bone tissue engineering

BACKGROUND: Besides good biocompatibility, ideal scaffold for bone tissue engineering should possess the characters of promoting cell proliferation and differentiation inducing the regeneration of bone tissue. However, the osteogenetic ability is limited when the materials of scaffold is used alone.OBJECTIVE: To observe the biocompatibility of coralline hydroxyapafite (CHA)/bone morphogenic protein (BMP) composite and osteoblasts.DESIGN: A comparative observation.SETTING: Department of Orthopaedics, Guangzhou General Hospital of Guangzhou Military Area Command of Chinese PLA.MATERIALS: Composite of CHA/BMP (CCHA) and CHA were cut into blocks of 4 mm×4 mm×3 mm and 10 mm×10 mm ×3 mm respectively, immersed in complete medium for 4 days after autoclaving.HETHODS: The experiments were carried out in the Central Laboratory of the Guangzhou General Hospital of Military Area Command of Chinese PLA from March to August in 2005. ① The bone marrow of New Zealand rabbits were extracted to culture osteoblasts. ② The osteoblasts were co-cultured with CCHA (CCHA group) and CHA (CHA group) respectively, and the blank control group was untouched. ③ The proliferative indexof osteoblasts and activity of alkaline phosphatase were measured at 2, 4, 6 and 8 days after co-cultureMAIN OUTCOME MEASURES: ① Results of osteoblasts observed under phase contrast microscope;② Results of CCHA material observed under scanning electron microscope; ③ Activity of alkaline phosphatase; ④ Proliferative index of osteoblasts.RESULTS: ① Osteobtasts observed under phase contrast microscope: The cell appearance was fusiform, oblate and multi-angles with processes of various number and size, and the cells connected with each other by the processes. ② CCHA observed under scanning electron microscope: The CCHA surfacewas granule-like with micropores. The osteoblasts attached to the surface of the materials and grew in monolayer at first, then overlapped as the cells further differentiation and proliferation High-density granules appeared around the cells,most of them tended to a certain direction, and finally formed bone nod as the crystals of calcium salt increased. The cells were embedded in matrixes. ③ Activities of alkaline phosphatase: The activities of alkaline phosphatase [absorbance (A) value] at 2, 4 6 and 8 days after co-culture in the CCHA group were significantly higher than those in the CHA group and blank control group (4 days: 101.6±7.7, 93.7±6.8, 94.4±5.0; 8 days: 120.1±8.2, 108.3±8.8, 110.4±7.5;P < 0.01 or 0.05=, and there was no obvious difference between the CHA group and blank control group. ④ Proliferative index: The proliferative indexes were increased as the culture time prolonged in the groups, but there was no obvious difference CONCLUSION: The inside of CHA is coral-like with many micropores, cells can well attach to its surface, and has good biocompatibility. Besides, the combined bone morphogenesis protein can obviously enhance the activity of alkaline phosphatase, which may play a certain role in bone induction.
5.Disposal of the small saphenous vein trunks during sural neurocutaneous island flap transplantation for repairing soft tissue defects of the foot and ankle

BACKGROUND: Scholars disagree with each other about the small saphenous vein effects on skin flap and how to dispose vascular proximal pedicles.OBJECTIVE: To analyze effects of different methods of the small saphenous vein disposal on flap survival using sural neurocutaneous island flap retrograde metastasis for repairing defects of soft tissue of instep, heelstick and ankle.DESIGN, TIME AND SETTING: The case control observation experiment was performed at the General Hospital of Guangzhou Military Area Command of Chinese PLA from March 1998 to April 2007.PARTICIPANTS: A total of 56 patients with defects of soft tissue of instep, heelstick and ankle were divided into 2 groups,small sapbenous vein deligation in proximal pedicle flap group (group A) (n=38), and proximal small saphenous vein and great saphenous vein or tributaries at recipient site anastomosis flap group (group B) (n=18).METHODS: During sural neurocutaneous island flap retrograde metastasis for repair,(3.5×4.0)cm-(4.0×4.5) cm flap was obtained in 35 cases, and (4.0×4.5) cm -(10.0×12.0) cm in 21 cases.MAIN OUTCOME MEASURES: Outcome of flap survival at different incision area and implanted methods.RESULTS: No vein articulo occurred in 21 cases, which flap areas were (4.0×4.5) cm -(10.0×12.0) cm. Five of 35 cases which flap areas were (3.5×4.0) cm -(4.0×4.5) cm developed vein articulo.The necrotic rate of flaps in group B was significantly lower compared to the group A (P=0.017 67).CONCLUSION: When the area of skin flap is smaller than (4.0×4.5) cm, the proximal end of the small saphenous vein should be anastomosed with the great saphenous vein or tributaries connecting with the great saphenous vein at recipient site.The small saphenous vein is not a superficial vein, which only cross the skin flap, but it has trophic action on the skin flap.
6.Histological study of sciatic nerve regeneration repaired in different size by artery sleeve briding

ObjectiveBy the study of histological feature of the regenerated nerve at different time after operation,to explore the mechanism of the peripheral nerve regenerationwhen the distal nerve stump is repaired by a smaller proximal end with artery sleeve briding.MethodsFour Wistar rats were served as the donors of arteris for bridging.Sixteen Wistar rats were randomized into 2 groups (n =8 per group).In control group,the right sciatic nerves were cut off and received epineurial suture.In experimental group,the right sciatic nerves received artery sleeve bridging in different size.HE staining was taken to observe the histological changes of the regenerated nerve at 7,14,21 and 28 days after operation.ResultsThe new regenerated axons need about 21 days to pass the bridging gap.Twenty-eight days after operation,the number of the regenerated axons in distal segment was greater than that of the proximal in the experimental group.The regenerated axons were matured and more regular.Conclusion The functional reserve of repaired nerve is exists when the proximal end is smaller than the distal in size by means of amplification effect.And the method of artery sleeve bridging provides a better situation for functional reserve.
7.Effect of astragalus polysaccharides on the cellular immune function in rats with sciatic nerve Wallerian degeneration

Objective:To explore the effect of astragalus polysaccharides on the cellular immune functions in rats with sciatic nerve Wallerian degeneration.Methods:10 female Wister rats were established as the sciatic nerve injury model,which were randomly divided into astragalus polysaccharides group and control group.Then 20 mg/kg of astragalus polysaccharides were peritoneal injected every postoperative day in experimental group for 7 days and the same volume of saline for the control group.The content of IL-1? in serum and in supernatants of spleenocytes and macrophages was measured by Sandwich ELISA.Results:The proliferation ability of splenic T cells and macrophages in astragalus polysaccharides group was higher than that of control group(P0.05).Conclusion:Astragalus polysaccharides induces the cellular immuno-regulation in sciatic nerve Wallerian degeneration rats and by this way to promote nerve regeneration.
8.CD4~+CD25~+Foxp3~+ and CD4~+CD25~-Foxp3~+T cells in peripheral blood from new-onset SLE patients

Objective:To investigate the expression of Foxp3 and CD25 on CD4+ T cells from patients with new-onset systemic lupus erythematosus(SLE)and assess their clinical significance.Methods:Twenty-one new-onset and untreated SLE patients including ten with active(SLEDAI≥10)and eleven with inactive disease(SLEDAI≤5)were enrolled in this study.Eleven age-and sex-matched healthy volunteers were also included as controls.Peripheral blood samples were collected and mononuclear cells isolated.The expression of CD25,Foxp3 and CD127 by CD4+ T cells was analyzed by flow cytometry(FACS).Proliferation assays were performed with the isolated CD4+CD25+ and/or CD4+CD25-T cells.Results:There was no significant difference in number of CD4+CD25+Foxp3+T cells in subjects with either active(1.91%-6.75%)or inactive(2.74%-7.01%)SLE compared to normal controls(2.11%-9.90%)(P=0.524 and P=0.794 respectively),Moreover,suppressive capacity of CD4+CD25+T cells in new-onset lupus patients was not impaired as measured by the capacity to inhibit proliferation of CD4+CD25-effector T cells(P=0.174,P=0.689).While CD4+CD25-Foxp3+T cells in both active(3.71%-10.94%)and inactive(2.97%-7.69%)patients with new-onset SLE were significantly more frequent than in normal controls(1.01%-3.62%)(P
9.ANALYSIS OF PYROLYSIS GAS CHROMATOGRAPHY OF SALMONELLA TYPHI

The chromatograms of pyrolysis gas chromatography (PGC) of fifteen strains of Salmonella typhi were reported to see if there is any possible relation of the graphs to serological typing and drug-resistance spectra of the bact. In the fifteen strains there were fourteen clinical strains and one reference strain (Hg01). The results indicated that the clinical strains might be divided into group A and B (7 strains each) by PGC. The graphs drawn from group A organisms were entirely the same in shape while those of group B showed only basic similarity. According to the diversity of graphs of group B organisms, the group B organisms, may be subdivided into group B1 (4 strains), B2 (2 strains) and B3 (1 strain). The graphs within each subgroup were completely the same and those of subgroup B1 exhibited close similarity to that of the reference strain. Albeit no regular connection of chromatograms with serological types and drug-resistance could be seen, the graphs yet provided useful reference for founding a bank of chromatograms, compaed with which salmonella typhi graph could then be identified.
