1.Clinical study on auricular plaster therapy in the auxiliary treatment of insomnia caused by liver fire disturbance in a sub-healthy population

Objective:To investigate the clinical efficacy of psychotherapy combined with auricular plaster therapy in the auxiliary treatment of insomnia caused by liver fire disturbance in a sub-healthy population.Methods:Eighty sub-healthy patients with insomnia caused by liver fire disturbance who received treatment in Wenzhou Hospital of Traditional Chinese Medicine from June 1, 2017 to December 31, 2018 were included in this study. These patients were randomly assigned to receive cognitive behavioral therapy (control group, n = 40) or cognitive behavioral therapy combined with auricular plaster therapy (observation group, n = 40) for 4 weeks. Polysomnography was used to evaluate the sleep condition before and after treatment. The Pittsburgh Sleep Quality Index was used to evaluate the severity of insomnia. The World Health Organization Quality of Life Scale Brief Version (WHOQOL-BREF) was used to evaluate the quality of life of patients. The therapeutic effect was compared between the two groups. Results:After treatment, total sleep time [(421.25 ± 11.58) min vs. (456.25 ± 11.23) min], sleep efficiency [(70.32 ± 4.44)% vs. (76.58 ± 5.89)%], non-rapid eye movement stage 2 (N2) sleep [(43.25 ± 4.44)% vs.(45.62 ± 4.36)%], N3 sleep [(14.25 ± 4.52)% vs. (18.52 ± 5.55)%], rapid eye movement (R) sleep [(21.98 ± 5.21)% vs. (24.66 ± 4.87)%] in the control group were significantly shorter or lower than those in the observation group ( t = 13.723, 5.367, 9.035, 2.408, 3.772, 2.376, all P < 0.05). After treatment, sleep quality [(1.85 ± 0.54) points vs. (1.10 ± 0.42) points], sleep-onset latency [(1.52 ± 0.51) min vs. (1.08 ± 0.35) min], sleep duration [(1.30 ± 0.41) min vs. (0.75 ± 0.41) min], sleep efficiency [(1.53 ± 0.44) points vs. (1.02 ± 0.33) points], sleep disturbance [(1.20 ± 0.41) points vs. (0.78 ± 0.31) points], hypnotics [ (0.11 ± 0.02) points vs. (0.04 ± 0.01) points], daytime dysfunction [(1.65 ± 0.47) points vs. (1.03 ± 0.28) points] and total scores [(9.11 ± 1.33) points vs. (5.80 ± 1.01) points] in the control group were significantly higher than those in the observation group ( t = 6.933, 4.499, 5.999, 5.864, 5.167, 19.789, 7.167, 12 535, P < 0.05). After treatment, the scores in physical health [(13.85 ± 2.22) points vs. (17.86 ± 2.01) points], psychological domain [(15.52 ± 1.89) points vs. (16.35 ± 1.33) points], social relationships [(16.01 ± 1.58) points vs. (19.52 ± 1.85) points] and environment domain [(17.52 ± 1.90) points vs. (20.89 ± 2.22) points] in the control group were significantly lower than those in the observation group ( t = 8.468, 4.499, 9.124 and 7.294, all P < 0.05). After treatment, total effective rate in the observation group was significantly higher than that in the control group [95.0% (38/40) vs. 72.5% (29/40), χ 2 = 7.440, P < 0.05]. Conclusion:Psychotherapy combined with auricular plaster therapy can significantly increase the sleep duration and sleep efficiency and improve the quality of life of sub-healthy patients with insomnia caused by liver fire disturbance, which is worthy of clinical promotion.

The hemodynamic effects of deliberated hypotension induced by PGE1 or SNP on 14 dogs anesthetized by ketamine were studied separately.MAP was decreased by 30%~40% after infusing PGE,or SNP Hemodynamic variables were measured before and 15 min 30 min after hypotension and 15 min after the discontinuation of infusion.The results indicated that SVRI, PVRI, MPAP, LVSWI and RVSWI all decreased significantly during PGE1-or SNP-induced hypotension. CVP decreased and HR increased significantly during SNP-induced hypotension, but with no significant change during PGE1-induced hypotension and with SVI. CI. PCWP remained unchanged in these two groups throughout the experiments.
3.Ghrelin participates in bone metabolism:the newest research progress

BACKGROUND:Ghrelin is a newly discovered brain-gut peptide from the stomach of human and rats. As an endogenous ligand for growth hormone secretagogue receptor (GHSR), ghrelin can notably stimulate the release of growth hormone. Although GHSR is expressed in many peripheral tissues, little is known about the influence of ghrelin on bone metabolism and GHSR expression in bone tissue. OBJECTIVE:To review the research progress of ghrelin in bone metabolism.METHODS:The first author retrieved CNKI, WanFang, PubMed, and Springerlink databases with the keywords ofGhrelin, bone metabolismin Chinese and English, respectively. The studies regarding ghrelin and its involvement in bone metabolism were included, and repetitive ones were excluded. A total of 53 eligible literatures were selected through skimming abstracts. RESULTS AND CONCLUSION:Ghrelin is a peptide composed of 28 amino acids discovered in gastric endocrine cells and hypothalamic arcuate nucleus in mice and human, which makes a great effect on digestive, nervous, immune and endocrine systems, and also plays a role in hormone secretion, glucose metabolism, immunity, cell proliferation, and inflammation. Serum ghrelin makes a certain influence on bone growth and development, and promotes the differentiation and proliferation of osteoblasts, and inhibits its apoptosis. Additionally, ghrelin suppresses the early osteogenic differentiation of C3H10T1/2 cells by upregulating the expression of Runx2 protein, and attenuates adipogenic differentiation by downregulating PPARγ2 expression, thus inducing osteogenic differentiation. However, few studies have addressed the expression of GHSR in bone tissue.
4.Inhibition of ovarian cancer growth by small interfering RNA targeting X-linked inhibitor of apoptosis gene

Objective To construct an RNA interference vector to down-regulate X-linked inhibitor of apoptosis(XIAP)gene and study the RNA interference effect on the cell cycle and growth of ovarian cancer.Methods Oligonucleotides of 64 base pairs for hairpin RNA targeting XIAP were designed, chemically synthesized,annealed,and cloned into the pSUPER vector.After identification by restriction digestion,the correct vectors were transiently transfected into SKOV3 cells,a human ovarian cancer cell line.The XIAP mRNA was detected by RT-PCR.The proteins were detected by western blot and indirect immunofluorescence staining.Flow cytometry(FCM)analysis and methyl thiazolyl tetrazolium(MTT)assay method were applied to measure cell cycle,cell growth and sensitiveness to cisplatin.Results SKOV3 cells had a high level expression of XIAP.The vector of RNA interference,which can interfere with XIAP gene was successfully constructed.After transient transfection,the expression of XIAP protein was significantly decreased in SKOV3 cells and the value of relative density was 3584?124,2138?65,1973?80 and 110 ?12,respectively(P=0.0334).At the same time,the expression of XIAP mRNA was decreased accordingly and the value of relative density was 6674?274,4532?107,2322?57 and 1864?78, respectively(P=0.0127).The FCM results showed that,the vector could increase the number of cells in G_1 phase compared with parent cells and compared with the cells transfected with pSUPER(P
5.Study on molecular etiology of respiratory tract virus infection in patients with community-acquired pneumonia in Lianyungang area

Objective:To explore the distribution characteristics of respiratory pathogens in patients with community-acquired pneumonia in Lianyungang.Methods:A total of 612 patients admitted to the second people′s Hospital of Lianyungang City because of community-acquired pneumonia (CAP) in 2019 were selected as subjects. Sputum or pharyngeal swabs were collected to extract nucleic acids, and 13-fold nucleic acids of respiratory pathogens were detected by PCR capillary electrophoresis fragment analysis. SPSS statistical software and GraphPad5.0 statistical mapping software were used for statistical analysis.Results:The physical examination rate of respiratory pathogens in the adult group was 82.0% in winter, 48.4% in spring, 28.0% in autumn, 20.0% in summer, χ 2=38.473, P=0.000. The positive rate of nucleic acid detection was significantly different in different seasons, among which the physical examination rate of respiratory pathogens in winter was the highest. The physical examination rate of respiratory pathogens in the juvenile group was 86.0% in spring, 76.2% in winter, 71.3% in summer and 66.7% in autumn, χ 2=7.946, P=0.047 . The positive rate of nucleic acid detection was calculated according to gender grouping. The comparison of nucleic acid positive rate between adult group and juvenile group in different seasons: 86.0% vs 48.4% in spring, χ 2=19.436, P=0.000; 71.3% vs 20.0% in summer, χ 2=22.180, P=0.000; 66.7% vs 28.0% in autumn, χ 2=13.485, P=0.000; 76.2% vs 82.0% in winter, χ 2=0.758, P=0.384. Except in winter, the detection rate of nucleic acid of pathogens in the juvenile group was significantly higher than that in the adult group. Conclusions:The nucleic acid detection rate and etiological distribution characteristics of respiratory pathogens are different in patients with community-acquired pneumonia in different seasons and different age groups. 13 kinds of multiple detection methods of respiratory pathogens can provide favorable laboratory data support for the diagnosis and treatment of clinical CAP patients.
6.The changes of peripheral blood lymphocyte subsets in patients with acute hydrogen chloride gas poisoning.

OBJECTIVETo explore the changes of peripheral blood lymphocyte subsets in patients with acute hydrogen chloride gas poisoning.
METHODSWhen the patients were admitted or on the secondary day, the percentages of total T-cell lymphocyte subsets (CD(3)(+)CD(19)(-)), CD(4)(+)T cells (CD(3)(+)CD(4)(+)), CD(8)(+)T cells (CD(3)(+)CD(8)(+)), B cells (CD(3)(-)CD(19)(+)) and NK cells (CD(3)(-)CD(16)(+)CD(56)(+)), and the ration of CD(4)(+)/CD(8)(+) in 37 cases with acute hydrogen chloride gas poisoning and 49 healthy controls were detected with flow cytometer.
RESULTSThe total T-cell percentage and total CD(4)(+)T cell percentage in 37 cases were significantly lower than those in 47 controls (P < 0.05). The percentages of NK cells and B lymphocytes in 37 cases significantly increased, as compared with controls (P < 0.05). The ration of CD(4)(+)/CD(8)(+) in 37 cases significantly decreased, as compared with controls (P < 0.05).
CONCLUSIONThe lymphocyte subsets in the patients with acute hydrogen chloride gas poisoning changed, which could influence the immune function of patients.

7.Study on apoptosis of human ovarian cancer cell induced by siRNA on Rab25

Objective To study the effect of siRNA of Rab25 on apoptosis induction in ovarian carci- noma cell.Methods According to Rab25 mRNA sequence in the Genebank,the plasmids expressing siRNA against Rab25 were constructed and stably transected into A2780 cells,MTY method were applied to measure cell growth and proliferation.Apoptosis rate and cell cycle phase distribution of A2780 cells were measured by flow cytometry(FCM).Apoptosis was confirmed by agarose gel electrophoresis(AGE)of DNA.Results Cells transected with the plasmids expressing siRNA targeting Rab25 gene effectively decreased cell growth, proliferation;blocked A2780 cells in the G_1 phase of cell cycle and induced cell apoptosis.A typical DNA ladder pattern appeared on AGE.Conclusion Rab25 gene siRNA can inhibit growth and induce apoptosis in ovarian carcinoma cell line,which will facilitate further studies of Rab25 function and its application in the treatment of ovarian cancer.
8.Magnetic resonance imaging of lymph node metastasis in nasopharyngeal carcinoma

Objective To explore the distribution rule of metastatic lymph node in nasopharyngeal carcinoma (NPC) by magnetic resonance imaging (MRI).Methods 315 histopathologically proved NPC patients were studied retrospectively.All patients had had their nasopharynx scanned by MRI with plain and contrast enhanced sequences.The distribution of lymph node was divided into six cervical levels plus retro- pharyngeal nodes(RN) according to RTOG guidelines proposed in 2003.Results 254 out of 315 patients (80.6%) had lymph node involvement,with 81 in the right neck alone,72 left neck alone,and 101 both necks;73 in RN alone,21 neck node alone,and 160 both necks and RN node.Skip metastasis was found in only 4 patients (1.6%).There was significant difference in BN metastasis between the primary tumor be- ing located merely on the superior/posterior wall and lateral wall (78% vs 49%,P<0.01).The incidence of lymph node metastasis in T1,T2,T3 and T4 patients was 73.5%,91.2%,71.9%,73.5% (P>0.05), respectively,without significant difference between early or advanced T stage in node distribution (P>0.05).Conclusions The incidence of lymph node metastasis is high in nasopharyngeal carcinoma,with retropharyngeal node being the most commonly involved,but the incidence of skip metastasis is very low. There is no significant difference between T stage and the incidence of lymph node metastasis.So is the dis- tribution of metastatic node.
9.Morphological evaluation of lumbar dorsal root ganglion on three-dimensional magnetic resonance imaging

Objective To investigate the morphological features of normal lumbar dorsal root ganglia using a three-dimensional(3D)coronal MR imaging.Methods One hundred and fifteen volunteers were included.Ages ranged from 15 to 75 years,with a mean of 40 years.Coronal 3D fast field echo(FFE) with water selective excitation(Proset)MR examination of 1150 dorsal root gangha were underwent at nerve root levels from L1 to L5.The source coronal images were further reconstructed into a series of rotational alignment coronal images with an interval angel of 12 degree using maximum intensity projection(MIP) technique.All DRGs of bilateral spinal nerve from L1 to L5 were morphologically analyzed on the original and MIP images including qualitative evaluation of the location,signal intensity,architecture and quantitative dimensional measurement.Results There were 225,225,219,210 and 160 foraminal ganglia from L1 to L5 level,respectively.The incidence of intraspinal ganglia from L3 to L5 gradually increased with a maximum at L5 level of 29.1%(X~2=188.371,P
10.Degradation kinetics of larotaxel and identification of its degradation products in alkaline condition

Larotaxel, a new taxane compound prepared by partial synthesis from 10-deacetyl baccatin Ⅲ, is active against tumors. In this research, a selective LC–MS method was developed and validated for the study of degradation kinetics of larotaxel, which was carried out in aqueous solutions with different pH (1.5, 3.0, 5.0, 6.5, 7.4, 9.0, 10 and 11.0) and temperature (0, 25, 37 and 45 °C). The linear range was 0.5–25μg/mL, the intra-and inter-day precisions were less than 7.0%, and accuracy ranged from 97.4–104.5% for each analyte. The observed rate obtained by measuring the remaining intact larotaxel was shown to follow first-order kinetics. The activation energies for degradation were 126.7 and 87.01 kJ/mol at pH 1.5 and 11, respectively. Although larotaxel was stable in pH 5, 6.5 and 7.4 buffers at 37 °C for 24 h during our study, increasing or decreasing the pH of the solutions would decrease its stabilities. Moreover, three main degradation products in alkaline condition were separated by HPLC and identified by Q–TOF–MS. The three degradation products were confirmed as 10-deacetyl larotaxel, 7, 8-cyclopropyl baccatin Ⅲ and 10-deacetyl-7, 8-cyclopropyl baccatin Ⅲ.
