1.Construction and identification of recombinant adenovirus vector containing human ?-catenin

Objective:To construct the recombinant adenovirus vector containing human?-catenin gene by homogenous recombination in E.coli for later research of the mechanism of Wnt/Wingless signal pathway in tumorigenesis.Methods:The target gene was amplified from pcmv-?NS-FC plasmid by PCR,then cutted by two endonucleases and directional cloned to the pAd track-cmv vector.Afterward,the correct recombinant was linearized by PmeI,following co-transformation with the backbone vector pAd Easy-1 in E.coli BJ5183.The homologous recombinant was linearized by PacI,and then transfected into HEK293 cell line to pack the adenovirus.Results:Through PCR,endonuclease cutting and gene sequencing,the target gene was verified to be correctly cloned in adenovirus vector.The high expression of green fluorescence protein in 293 cell line was found under fluorescent microscope.Conclusion:The recombinant adenovirus vector containing human ?-catenin gene was successfully constructed and the adenovirus was packaged in HEK293 cell line.
2.An Association Analysis of Different Types of Dyslipidemia and Heart Rate Variability in Patients with Type 2 Diabetes

Objective To study the effect of different types of dyslipidemia on heart rate variability in patients with type 2 diabetes.Methods Cohort study was carried out at Renmin Hospital of Wuhan University during January 2014 to May 2016.Total 725 patients with type 2 diabetes were recruited,including 231 patients without dyslipidemia and 494 patients with dyslipidemia.Clinical parameters were collected from electronic medical records system.Using variance analysis,chi square test,Pearson correlation analysis and partial correlation analysis,we explored an association of different types of dyslipidemia and heart rate variability in patients with type 2 diabetes.Results TG,LDL-C,HDL-C,TG + HDL-C group significantly decreased heart rate variability compared group without dyslipidemia (P < 0.05).But the partial correlation analysis showed that only atherogenic index of plasma and Triglyceride (TG) were negatively correlated with heart rate variability,HDL cholesterol was positively correlated with heart rate variability,and the difference was statistically significant (P < 0.05).Conclusion Triglyceride (TG),atherogenic index of plasma(AIP),HDL cholesterol were related to the reduction heart rate variability in patents with type 2 diabetes.

Many alimentary tumor patients are characterized by malnutrition resulting from various elements and disorders of metabolism.So it is necessary to combine together with parenteral nutrition(PN) and enteral nutrition(EN).It can improve the nutritional status and reduce morbidity and mortality in patients with alimentary tumors in perioperative period.
4.Relationships between serum resistin, adiponectin levels and bone mineral density in Chinese men: Data analysis of 232 volunteers

OBJECTIVE:To investigate the correlation of serum resistin and adiponectJn Ievels with bone mineral density(BMD)in 232 healthy male volunteers in Changsha area.METHODS:A totaI of 232 healthy Chinese men of Han population were selected randomly from Changsha area.All subjects gave their informed consent to participate in this study.Subjects were excluded from the study if they had conditions or received treatment that affecting bone metabolism.Serum adiponectin and resistin levels were determined by ELISA.BMD was measured by dual energy X-ray absorptiometry in total body,lumbar spine,and total hip.Body fat mass and lean body mass were measured by the whole body scanning.The correlation of serum adiponectin and resistin levels with fat mass and BMD at various sites were analyzed;the predictors of BMD at various sites was assayed using multiple linear stepwise regression analysis.RESULTS:Adiponectin was negatively correlated with fat mass(r=-0.216.P<0.05),while resistin had no correlation with fat mass.The correlation disappeared after adjustment for age and fat mass(r=-0.006,P>0.05).The significant negative correlations between adiponectin and BMD were found,and remained significant after adjustment for age and fat mass.There was no correlation between resistin and BMD at various sites.In the multiple linear stepwise regression analysis.adiponectin was an independent predictor of BMD in men.CONCLUSION:Resistin js not correlated with fat mass and BMD at various sites.Adiponectin is correlated with fat mass,and has a negative correlation with BMD at various sites,it is an independent predictor of BMD in Chinese men.

Advancing medical technology and numerous clinical experiences contribute to great improvement in treatment methods and effects on patients who have suffered bile duct injuries. At the same time, There are many new questions should be confronted with. The current conditions of treatments of iatrogenic bile duct injuries are reviewed in this article.

7.SGLT2 versus DPP4 Inhibitors for Type 2 Diabetes: A Meta-analysis

Objective To compare the efficacy of empagliflozin and DDP4 inhibitors in the treatment of type 2 diabetes mellitus.Methods With Computer,we retrieval PubMed,Cochrane Library,EMbase,CNKI,CBM and Wanfang database to find all of the randomized controlled trials about the effectiveness and safety of empagliflozin and DDP4 inhibitors in the treatment of type 2 diabetes mellitus.The retrieval time was from the establishment of database to March,2016.References screen were performed manually.Meta-analysis was carried out using RevMan 5.2 software.Results A total of five studies included 1935 cases.The Meta analysis results showed that:HbA1c level (MD =-0.07,95% CI:-0.13--0.01,P <0.05),fasting blood glucose (MD =-16.73,95% CI:-21.02--12.43,P < 0.01),weight loss level (MD =-2.43 95% CI:-2.68--2.19,P < 0.01) in the two groups showed statistically significant differences.Empagliflozin group was better than DDP4 inhibitors group in reducing HbA1c level,fasting blood glucose,weight loss level.Safety:hypoglycemia between the two groups were similar (OR =0.86,95% CI:0.46-1.6,P > 0.05).Empagliflozin group's genital tract infection rate was statistically significant higher than that of DDP4 inhibitors.(OR =2.85,95% CI:1.71-4.76,P < 0.01).Conclusion Empagliflozin can effectively reduce the fasting blood glucose,HbAIc,and body weight,but increase the incidence of genital infection compare with DDP4 inhibitors.
8.Effect of two surgical treatments for primary angle-closure glaucoma combined with cataract

AIM: To compare the effect of trabeculectomy and combined surgery of glaucoma and cataract treating primary angle-closure glaucoma ( PACG) with cataract. METHODS: It was retrospectively reviewed that 80 patients (90 eyes) with PACG and cataract were treated in our hospital from January 2012 to October 2013. They were divided into observation group ( combined surgery of glaucoma and cataract group, 45 eyes ) and control group ( trabeculectomy group, 45 eyes) . Postoperatively, all the patients were followed up for 6mo. Intraocular pressure ( IOP ) , visual acuity and complications were observed.
RESULTS: Compared to preoperative IOP, postoperative IOP was significantly reduced in both observation group and control group, while IOP was reduced more greatly in observation group (P=0. 003). Visual acuity was significantly improved in observation group, while it was not improved significantly in control group ( P = 0. 036 ). Compared to control group, complications such as shallow anterior chamber with hypotony, macular edema and anterior chamber inflammation were less observed in observation group, with significant statistical difference ( P=0. 002, P=0. 003, P=0. 001).
CONCLUSION: For patients with PACG and cataract, combined surgery of glaucoma and cataract is an effective surgery that can improve visual acuity and reduce IOP with fewer complications.
10.Clinical following study of modified anterior decompession approach to treat cervical spondyllosis

[Objective]To summarize and evaluate the curative effect of anterior decompression approach by using cervical retractor systems to treat cervical spondylosis.[Method]From April 2002 to October 2004,68 cases were performed anterior undermined far-reaching decompression and fusion with autograft and titanium plate internal fixation by using removing disc merely at the single-level or separately at the multilevels employing CCR self-retractor and Caspar cervical retractor systems via interspinal approach.The cases were followed up,and serial roantgcnographic evaluations being applied.Then the height of involved intcrspinal space was measured preoperatively and after 12 months postoperatively,and the spinal function was evaluated in accordance with the standard of Japanese Orthopeadic Association(JOA),and then all results were compared statistically.[Result]51 of all cases were followed up,of which 50 were better,1 was improved,no one worsened.After 12 months postoperatively,roentgenographic appearance showed that the allograft healing and interbody fusion of all patients were achieved,and the reserving height of involved interspinal space and JOA evaluation postoperatively were significantly superior to both preoperatively.No complications such as cervical spinal cord injury,internal fixation unfastening,and hematoma turned up.[Conclusion]Anterior decompression approach by using cervical retractor systems to treat cervical spondylosis could better reserve the height of involved interspinal space,and improve the spinal function significantly.
