2.Differential expression of nucleus-cytoplasm transport receptor protein importin13 in limbal neoplasms

Background The study on normal stem cell markers provides a new way of thinking of that pathogenesis of cancer research and looking for specific markers of cancer stem cells.Importin13 (IPO13) is a novel nucleus-cytoplasm transport receptor protein of importin β family,the study on the biological behavior of IPO13 in ocular tissue and limbal neoplasms is lacking.Objective This study was to investigate the differential expression of IPO13 and p63 in human benign and malignant conjunctiva-cornea neoplasms.Methods The specimens of normal donor limbal and conjunctival tissues (6),conjunciva-cornea papilloma (CCP) (6) and conjunctiva-cornea intraepithelial neoplasia(CCIN) (6)were collected from Xiangya Hospital of Center South University.The expressions of IPO13 and p63 in the corresponding tissue were qualitatively and quantitatively detected using immunochemistry.The A values of IPO13 and p63 positive response were calculated and compared among the 3 types specimens.Results The immunohistostaining on frozen sections showed that IPO13 was expressed in nuclei of basal cells of limbal and conjunctival epithelium and the cellular nuclei of basal cells and suprabasal cells of CCP and CCIN epithelium.The A values of IPO13 positive expression were 1687± 1014,3546± 1375 and 7635 ±2854 in the normal limbal specimen,CCP and CCIN,respectively,showing a significant difference among them (F =7.23,P<0.05),and the A value was higher in the CCP and CCIN than that in the normal limbal tissue (q =4.02,5.13,P<0.05),and that in the CCIN was significantly elevated in comparison with CCP(q =3.45,P<0.05).p63 was expressed in nuclei of basal cells and suprabasal of limbal,conjunctival and CCP epithelium,and was expressed in nuclei of the entire CCIN epithelium.The expressions of p63 in the normal conjunctiva-cornea tissue,CCP and CCIN were 2110± 1229,3966± 2129 and 6650± 2136 respectively,with significant difference among the three different specimens (F =6.17,P< 0.05),and the A value of p63 positive expression was significantly elevated in the CCP and CCIN compared with normal limbal specimen (q =4.33,5.01,P<0.05),and that in the CCIN was significantly elevated in comparison with CCP(q=3.83,P<0.05).Conclusions IPO13 is more specifical in marking the poorly differentiated cells in limbal epithelial proliferative lesions than p63.Compared with the normal limbal specimen,the expressions of IPO13 and p63 in CCP and CCIN specimens gradually upregulat,which suggests that IPO13 and p63 may play a positive regulatory role in conjunctiva-cornea proliferative lesions.The differential expression of IPO13 and p63 is predominant between benign and malignant limbal epithelial proliferative lesions,indicating that IPO13 and p63 may play an important role in regulating the abnormal proliferation and differentiation of limbal epithelial proliferative lesions.

4.Construction of nude mouse models bearing subcutaneous tumors:human osteosarcoma cell lines MG63, U2OS and 143B

BACKGROUND:A variety of cel lines participate in the construction of animal models of osteosarcoma. OBJECTIVE:To compare the abilities of subcutaneous tumorigenesis of human osteosarcoma cel lines MG63, U2OS and 143B in nude mice, and to establish the basis for experimental studies on osteosarcoma. METHODS:A total of 18 nude mice were randomly divided into MG63, U2OS and 143B groups. The back of nude mice was subcutaneously injected with MG63, U2OS and 143B cel lines in corresponding groups, and the process of tumorigenesis was observed. RESULTS AND CONCLUSION:MG63 and U2OS could not induce subcutaneous tumorigenesis in nude mice. Tumors were observed after injection with 143B cels for (6±1) days, with the tumor formation rate of 100% (6/6). The tumor grew very fast. The mean volume of tumors was (3 475±1 544) mm3 at 2 months. The survival time of nude mice burdened with tumors was (68±10) days. Hematoxylin-eosin staining revealed that in tumor tissue, cancer cels formed nest shape, nuclei were big and darkly stained, and split mostly. These results suggested that 143B cel line could wel induce subcutaneous tumorigenesis, which could be used for animal research of osteosarcoma.
5.The influence analysis of public health emergencies on mood fluctuations of depression and related factors in the elderly in Shanghai

Objective:To analysis the impact of public health emergencies on mood fluctuations of depression and the related factors in the elderly in Shanghai, in order to provide a new evidence for early psychological intervention.Methods:A cross-sectional survey was conducted among 983 elderly in the Department of Geriatrics of Xinhua Hospital Affiliated to Shanghai Jiaotong University and two communities in Changning and Hongkou Districts of Shanghai by using a self-compiled social life questionnaire and Geriatric Depression Scale(GDS-30)during corona virus disease(COVID)-19 outbreak from 1 February to 15 February 2020.The mood fluctuations of depression and the related influencing factors were analyzed by comparing the current GDS-30 scores with the scores within previous six months.Results:Of 983 questionnaires, 867 were valid.The incidence of mild fluctuations of depression was increased from 20.9%(181/867)to 27.2%(235/867)during the public health emergencies( P<0.05), with 3 cases of newly emerged severe depression.The GDS-30 scores were higher during the public health emergencies than before the emergencies(9.88±3.85 vs.7.67±3.54, P<0.05). The four risk factors inducing fluctuations of depression in the elderly were the number of coexisted chronic diseases ≥2( P=0.036), the lack of visits or communication from families( P=0.015), the family members exposing a risk to COVID-19( P<0.01), and the daily viewing of epidemic news more than 4 h( P=0.023). Linear regression analysis showed that cough, sore throat, dizziness, sleep disturbance and dyspnea were significantly related to the aggravation of depression(based on the increase of GDS-30 score)( P<0.05). Conclusions:Public health emergencies may exacerbate depression in the elderly.They are more likely to experience the aggravation of depression when they suffer from multiple chronic diseases, lack the child visits or communications, have the family members exposing a risk to COVID-19 and pay excessive attention to the epidemic.
6.Quantitative assessment of ventricular-arterial coupling in patients with uremia

Objective To investigate the clinical value of quality intima-media thicknes (QIMT),quality arterial stiffness(QAS) and XStrain in assessing the ventricular-arterial coupling (VAC) in patients with uremia.Methods Sixty-five patients with uremia and 30 normal subjects were enrolled in this study as the uremic group and control group respectively.Ultrasound examination for the cardiac and carotid artery was performed and some parameters were obtained,such as left ventricular ejection fraction(LVEF),E/e,Tei index,stiffness (β),compliance coefficinet (CC),pulse wave velocity (PWV),intima-media thicknes (IMT),strain,and so on.The sum of left ventricular systolic radial strain and carotid arterial diastolic radial strain was used as a new parameter (VACs) for assessing VAC.The correlation between VACs and VACv,a traditional method to evaluate VAC,was analyzed.The parameters obtained by ultrasonic techniques were compared between two groups.The intra-and inter-observer reliability of XStrain was assessed by intraclass correlation coefficient.The multiple linear regression and ROC curve were used to analyze the independent factor of cardiovascular dysfunction.Results ①The pulse pressure,E/e,Tei index,as well as β,PWV and IMT were larger in uremic group than control group significantly (P <0.05).②The function of VAC was decreased in uremic group,and the VACv and VACs were lower than control group significantly (P <0.05).③There was strong positive relation between VACs and VACv (r =0.908,P =0.000).The reproducibility of XStrain technique was well.④VACs,β,VACv and PWV could be considered as independent factor of cardiovascular dysfunction and performance of VACs was the largest (standardized coefficient was-0.582).A cutoff value of VACs for assessing cardiovascular dysfunction was less than 23.60,which had both higher sensitivity and specificity (96.4%,81.1%).Conclusions The QIMT,QAS and XStrain can be used to assess cardiovascular structure,function and VAC accurately and globally,which can be consider as an effective method for detecting cardiovascular complication and improving prognosis of uremic patients.
7.Practice on the case-based teaching innovation of Science of Meridians, Collaterals and Acupoints.

The case-based teaching method which is applied to the teaching of Science of Meridians, Collaterals and Acupoints is discussed in this paper, in which the typical cases such as the growth of the acupuncture-moxibustion eminent physicians, the application of acupoints by the eminent physicians and the experiences in the acupoint combination are integrated. The students are instructed to launch the clinical practical activity through establishing the clinical base of Science of Meridians, Collaterals and Acupoints, in association with the true cases. It is proved that the case-based teaching method can promote the training of the manipulation techniques of the students and the inheritance of the experiences of eminent physicians in higher education.

8.The application of X -1 2 -ARIMA model in analysis on incidence trend of typhoid

Objective To study the function of X -1 2 -ARIMA model in analysis on incidence trend of typhoid. Methods Secular trend,seasonal periodicity and random fluctuations of the monthly morbidity data in Zhejiang province from 2005 to 201 3 were analyzed by X -1 2 -ARIMA model.Results The seasonal fluctuation showed a narrowing trend year by year during 2005 to 201 3.After September,2007,the incidence of typhoid showed a downward trend.After 2008,the annual peak of incidence changed from August to July.The irregular factor may well represent the outbreak. Conclusion The X -1 2 -ARIMA model showed clear secular trend and seasonal periodicity,and the random fluctuation was of great value.
9.Protective immune mechanisms induced by cellular vaccine made of the erythrocytic Plasmodium yoelii.

OBJECTIVETo explore the characteristics of protective immunity against Plasmodium yoelii (P.y.) infection by asexual blood-stages cellular vaccine.
METHODSThe particulate vaccines were constructed by saponin or double-distilled-water lysed parasitic red blood cells and inoculated into BALB/c mice by intraperitoneal injection (i.p.). Each group was challenged by the lethal erythrocytic P.y. parasites, and then their parasitemia and survival rates were detected. Expressions of interleukin-4 (IL-4) and interferon-gamma (IFN-gamma) were detected by RT-PCR. ELISA showed the serum antibodies against the malaria challenge and their-subclasses. Special membrane protein was recognized by immunofluorescence assay.
RESULTSThe vaccination with saponified erythrocytic parasites protected the immunized mice against P.y. challenge, while double-distilled-water lysed vaccine did not (P < 0.01). This protection was characterized by the increase of both IFN-gamma/IgG2a and IL-4/IgG1. Meanwhile, MHC class I alpha chain molecule was recognized on the membrane of infected-erthythrocyte.
CONCLUSIONSaponified P.y. asexual blood-stage cellular vaccine has a significantly high protective immunity against this lethal P.y. malaria, and the immunity may be associated with the expression levels of IgG2a and IFN-gamma. MHC class I alpha chain on infected erythrocytes may play an important role in the successful immunization.

10.Quantitative analysis of pulegone in Herba Schizonepetae.

OBJECTIVETo establish the determination method of pulegone in Herba Schizonepetae.
METHODHPLC method was developed for the determination of pulegone in Herba Schizonepetae. The separation was performed on C18 column with MeOH-H2O (80:20) as a mobile phase. The detection wavelength was set at 252 nm.
RESULTThe quantitation of pulegone in Herba Schizonepetae varied from 0.2-0.7 mg x g(-1). The average recovery is 96.2%, RSD 0.81%.
CONCLUSIONThe method is simple, rapid and accurate.
