[Objective] To observe the therapeutic effect of heat-clearing and diuresis-promoting herbal medicine for hyperuricemia induced by pyrazinamide. [Methods] Eighty-six patients with pyrazinamide-induced hyperuricemia were randomized into 2 groups: group A ( n = 43) received antiphthisic drugs (including isoniazid, rifampin, pyrazinamide and ethambutol) , allopurinol tablets and Modified Simiao Powder (one dose per day); group B ( n = 43) was treated with antiphthisic drugs and allopurinol tablets. Fourteen days constituted one treatment course and the two groups were treated for 2 courses. After treatment, the therapeutic effect was assessed and the changes of blood uric acid level were observed. [Results] In group A, 28 (65.12%) were cured, 12 (27.91%) effective, 3 (6.97%) ineffective and the total effective rate was 93.02%; in group B, 16 (37.21%) were cured, 17 (39.53%) effective, 10 (23.26%) ineffective and the total effective rate was 76.74% . Objective To investigate the effect of microminiature water halloon combined with misoprostol per rectum for delivery of full-term pregnancy. Methods 305 full-term pregnant women with planned delivery were divided into three groups at random.The observation group (n=99) was given microminiature water balloon combined with misoprostol per rectum.The first control group (n=96) was only given misoprostol per rectum. The second control group (n=110) was given 0.5% oxytocin in a continuous infusion.Their results were taken a com-parative study,including the cervical ripening,dilivery process,delivery ways and the condition of the newbom. Results The observation group was better than other groups for cervical ripening(P<0.00).Tune from induction to the onset of regular contraction activity of the uterus in the observation group was significantly shorter than other groups (P<0.00).The rates of cesarean section and the total stage of lahor between the observation and the first control group were not statistically significant(P>0.05),but they were significantly different from the second con-trol group.There were no significant differences among the groups in neonatal asphyxia and amount of postpartum blood loss. Conclusions The efficiency of microminiature water balloon combined with misoprostol per rectum for delivery of full-tarm pregnancy was better than other groups.It has a short delivery process and shows no side effects on both mother and fetus,which is worthy of popularizing in obstetrics.

Aim To study the protective effects of sodium magnesium fructose diphosphate (FDPM) on the kidney in hemorrhagic shock rats. Methods The study utilized Wiggers hemorrhagic shock model. Sixty Wistar rats were divided into six groups randomly: normal saline control group,three doses of FDPM group (90.0,45.0,22.5 mg?kg -1 ),1,6-fructose diphosphate group(37.5 mg?kg -1 ) and magnesium sulfate group (3.4 mg?kg -1 ). Mean arterial pressure was monitored before shock, 10, 30 and 60 minuts after drug administration and 0, 30 and 60 minuts after shed blood reinfusion Blood urea nitrogen and creatinine, the content of malondiadehyde, and the activity of Na+-K+-ATPase, Ca 2+ -ATPase, Mg 2+ -ATPase, and superoxide dismutase in kidney were also measured. Results FDPM significantly improved mean arterial blood pressure of hemorrhagic shock rats, reduced blood urea nitrogen and creatinine, decreased the content of malondiadehyde and increased the activity of Na+,K+-ATPase,Ca 2+ -ATPase,Mg 2+ -ATPase and superoxide dismutase in kidney. Conclusion FDPM had protective effects on the ischemia-hypoxia injury of kidney by improving energy metabolism and attenuating tissue injury caused by free radicals in hemorrhagic shock.

Objective To analyze the protein expression changes of retina in diabetic mice using isobaric tags for relative and absolute quantitation (iTRAQ) approach and to study proteins of apoptosis.Methods 8 diabetic mice were chosen as the diabetic model group (DM group),8diabetic mice as the normal control group.The animals were housed in wire-bottomed cages and received normal pellet chow and tap water in a constant environment.After 10 weeks,all mice were killed,and their retina were dissected.After hematoxylin and eosin(H&E) staining,the sections of retina were examined using light microscopy.The changes of protein expression in retina were studied using iTRAQ approach.Expression of apoptosis associated proteins was analyzed using ingenuity pathway analysis (IPA).Results Compared with control group,mice retina in DM group developed looser structures,tissue edema and obvious telangiectasia under light microscopy.Using iTRAQ approach,a total of 348 differential proteins were identified.Among those proteins,16 proteins were related with apoptosis,including Ataxin-10,Protein NDRG1,mucin-4,Aquaporin-1 and annexin A4,etc.There were 8 apoptosis-related proteins in retina with up-regulation,and the other 8 proteins with down-regulation in the DM group.The relationship between these proteins were analyzed and charted by IPA.Conclusions Apoptosis may be involved in the development of diabetic retinopathy.The identification of the apoptosis-related proteins will be helpful for the further study.

Objective To study the effect of multi-disciplinary teamwork (MDT) nursing mode on perioperative care to breast cancer patients. Methods One hundred and twenty breast cancer patients undergoing surgical operations were evenly randomized into the observation and control groups by random digit number table. The control group was treated with routine nursing care and the observation group with MDT nursing mode. Result The rate of complications (like subcutaneous effusion, lymphatic edema and skin flap necrosis) in the observation group was significantly lower than that of the control group and the hospital stay was significantly shorter than that of the control group as well (all P<0.05). Conclusion The MDT nursing mode can reduce the rate of complications, shorten the hospital stay and relieve patient′s depression.

Objective To explore the differentiation of rat bone marrow mesenchymal stem cells into dopaminergic neuron.Methods The mesenchymal stem cells were isolated from bone marrow. They were induced to differentiate into dopaminergic neurons by freated with bFGF, VitC and EGF at the third generation. Dopamine-associated protein and genes in the treated cells were examined by immumofluorescence and RT-PCR. Dopamine in the supernatant and endoplasm from culture system was determined by ELISA kit. Results The results showed that tyrosine hydroxylase, dopamine transporter and nerve neucleoprotein and Nestin,Nurr-1 genes were found. And the dopamine existed in the supernatant and cytoplasm from inducing culture system. Conclusion The rat bone marrow mesenchymal stem cells have the capacity of differenting into dopaminergic nurons.

ObjectiveTo investigate the effects of grape seed proanthocyanidin extracts (GSPE) on advanced glycation end product receptor (RAGE),NF-Κb and connective tissue growth factor (CTGF) in the myoeardium of diabetic rats.MethodsTotal 30 streptozotocin (STZ) induced diabetic rats were randomly divided into 2 groups:diabetic group (DM1,n-15) and GSPE (250 mg/ kg,i.g) treated diabetic group (DM2,n= 15).Another two control groups:normal rats(C1,n= 10)and normal rats treated with GSPE (250 mg/kg,i.g) (C2 group,n= 10) were also observed.After 24 weeks,blood was collected to measure fasting plasma glucose (FBG) and RAGE.The protein expression of NF-Κb was determined in myocardial tissue by immunohistochemical staining and Western blot.The protein expressions of RAGE and CTGF were measured by Western blot.Results The levels of FBG and RAGE were significantly higher in diabetic rats than in control rats (P＜0.05).After GSPE treatment,RAGE level significantly reduced (P＜0.05),but FBG had no change in diabetic rats.The protein expressions of RAGE,NF-Κb and CTGF in the myocardial tissue of diabetic rats had marked increase compared with control rats (P＜ 0.05),however,their levels significantly reduced after GSPE treatment (P＜0.05).ConclusionsGSPE may protect diabetic rats against cardiomyopathy,possibly by decreasing the protein expressions of RAGE,NF-Κb and CTGF.

Objective:To study the effect of Bcl-2 short hairpin RNA (shRNA) in enhancing methotrexate (MTX)-induced apoptosis of Raji cells. Methods:Expression plasmid containing Bcl-2 shRNA was transfected into Raji cells by lipofectmine 2000 and then the transfected cells were treated with MTX. The expression levels of Bcl-2 mRNA and protein were evaluated by RT-PCR and immunofluorescence method at 48 h of transfection. MTT assay was used to analyze cell proliferation at 24, 48 and 72 h. Apoptosis was detected by Giemsa staining and flow cytomertric cell cycle analysis. Results:After transfection with Bcl-2 shRNA, the expression levels of Bcl-2 mRNA and protein in Raji cells were significantly decreased (P<0.05). Bcl-2 shRNA transfection plus MTX treatment induced marked apoptosis, decreased in cell proliferation activity, and increased in apoptotic rate. The difference was significant compared with MTX group, negative shRNA plus MTX group, Bcl-2 shRNA group, and empty plasmid plus MTX group (P<0.05). Conclusion:Bcl-2 shRNA could enhance MTX-induced apoptosis and inhibition of cell proliferation in Raji cells.

AIM: To examined the effects of hypoxic preconditioning ( HPC) on oxygen-glucose deprivation ( OGD)-induced PC12 cells, and to investigate its possible mechanisms of autophagy .METHODS: Cultured PC12 cells were randomly divided into control group , HPC group, 3-methyladenine (3-MA) group, HPC+OGD group, 3-MA+HPC+OGD group and OGD group .CCK-8 assay was used to detect the cell viability .The caspase-3 activity was also tested . TUNEL staining and flow cytometry were used to detect the cell apoptosis .The protein levels of apoptosis-related protein caspase-3 and autophagy-marked protein LC3-2 and beclin-1 were determined by Western blot .RESULTS:Compared with control group, the viability of PC12 cells was significantly reduced , and the activity of caspase-3 was significantly increased in OGD group.Compared with 3-MA+HPC+OGD group and OGD group , the viability of PC12 cells was significantly in-creased, and the activity of caspase-3 was significantly reduced in HPC +OGD group (P<0.05).The PC12 cell injury was apparent after OGD with a great increase in the apoptotic rate (P<0.05).Compared with OGD group, the apoptotic rate significantly decreased in HPC +OGD group ( P<0.05 ) .Compared with control group , the protein level of cleaved caspase-3 was significantly increased in OGD group ( P<0.05) .Compared with OGD group , the protein level of cleaved caspase-3 was significantly decreased , and the levels of LC3-2 and beclin-1 were significantly increased in HPC +OGD group (P<0.05).CONCLUSION:OGD decreases cell survival and induces apoptosis .Activation of cell autophagy may be the mechanism by which hypoxic preconditioning protects the PC 12 cells from OGD induced injury .

Objective To observe therapeutic effects of acupoint catgut embedding therapy on ulcerative colitis.Methods 55 cases of ulcerative colitis were randomly recruited into a treatment group and a control group. The treatment group was treated with catgut embedding, and the control group was treated with oral administration of salicylazosul fapyridine. Results The therapeutic effect of the treatment group was better than those of the control group (P=0.002＜0.05). Conclusion Acupoint catgut embedding therapy has a better therapeutic effect on ulcerative colitis.
